Susceptibility to FAS induced apoptosis in mycoplasma contaminated HCE cells (Mycoplasma가 오염된 배양 각막상피 세포의 FAS 유도 세포고사의 민감성)

  • The aim of the present study was to determine mechanisms of corneal epithelial cell apoptosis in vitro following exposure to anti-FAS and anti-FAS ligand antibody and during infection with mycoplasma sp.. A cultured human corneal epithelial(HCE) cell line was treated with anti-FAS antibody or anti-FAS ligand antibody for 2 and 4 days. The original cell line was found to be contaminated by mycoplasma removal agent(MRA) was used to eliminate the bacterium from the cell line. MRA($0.5{\mu}{\ell}$ tissue culture medium) was added to the cell line and incubated for 1 week. The cell line underwent multiple passages in media not contaminating MRA and cells were grown to 50-80% confluency on coverslips and stained using the Hoechst stain provided in the kit to ensure mycoplasma removal. Apoptosis experiments were performed before and after mycoplasma removal. The apoptotic index of anti-FAS and anti-FAS ligand antibody on mycoplasma contaminated cell line was studied using Hoechst 33342 staining and Annexin V-FITC and Propidium Iodide Staining. In conclusion, anti-FAS antibody induces apoptosis in HCE cells in a time and concentration-dependent mechanism. Cell lines contaminated with mycoplasma have an incresed susceptibility to FAS induced apoptosis.

Generation and Characterization of a Monoclonal Antibody with Specificity for Mycoplasma arginini

  • Previously, we generated monoclonal antibodies (MAbs) that bound to the surface of human embryonic stem cells (hESCs) in an attempt to discover new hESC-specific surface markers. In this study, MAb 47-235 (IgG1, ${\kappa}$) was selected for further characterization. The MAb bound to the surface of undifferentiated hESCs but did not bind to mouse ESCs or mouse embryonic fibroblast cells in flow cytometric analysis. The antibody immunoprecipitated a 47 kDa protein from the lysates of cell surface-biotinylated hESCs. Identification of the protein by quadrupole time of flight tandem mass spectrometry revealed that 47-235 binds to Ag 243-5 protein of Mycoplasma arginini. BM-Cyclin treatment of the hESCs that reacted with 47-235 resulted in loss of mycoplasma DNA and the reactivity to 47-235. Nevertheless, the hESCs that were reactive to 47-235 maintained self-renewal and pluripotency and thus could be differentiated into three embryonic germ layers.

The Clinical Aspects of Pneumonic Patients with Positive Mycoplasma Antibody (Mycoplasma 항체 양성인 폐렴 환자의 임상상)

  • Background : Mycoplasma pneumonia (M. pneumonia) is a major cause of atypical pneumonia, and its incidence is predominantly at childhood and early adulthood. In contrast, the incidence of adult patients with M. pneumonia has been known to be low. Furthermore the clinical aspects of M. pneumonia are different from those of community acquired pneumonia. Thus, we evaluated the clinical aspects of M. pneumonia in the adult patients. Method : Mycoplasma antibody and cold agglutination tests were performed in patients with clinically suspected pneumonia who had abnormal infiltrations on chest P-A. The 12 patients with pneumonia, who fulfilled entry criteria of more than 1:64 of cold agglutination titer and 1:40 of mycoplasma antibody titer or four-fold increase of mycoplasma antibody titer during one week, were analyzed in terms of clinical aspects. Results : 1) Twelve patients, male 3 and female 9, were included in this study. The peak incidence was teenager. 2) M. pneumonia occured perennially, but predominantly between June to October in eight patients. 3) The main symptoms were fever, coughing, sputum. 4) The main patterns of chest P-A were bronchopneumoina in 8 cases, and involved lesion were nearly both lower lobe. Conclusion : The clinical aspects with Mycoplasma pneumonia in adult patients were different from those of community acquired pneumonia.

  • The Guillain-Barre Sydrome (GBS) is post-infectious autoimmune disease and it could be caused by auto-antibodies produced after infections. Mycoplasma pneumoniae is one of rare cause of GBS and known to be associated with antibody to galactocerebroside (GalC) which is a major neutral glycolipid constituent of myelin. We report a case of GBS with immunoglobulin M GalC antibody after M. pneumoniae infection.

Analysis of antibody titer of transovarian transmitted diseases in Korean native breeder chickens (토종닭 종계의 난계대 전염병 항체 양성률 분석)

  • This study was carried out to identify the occurrence of transovarian transmitted diseases and antibody positive rates among Korean native breeder chickens. The infection rates with Salmonella pullorum and Salmonella gallinarum among 16-week-old, 36-week-old, and 56-week-old breeder chickens and the antibody positive rates to Egg Drop Syndrome '76, Mycoplasma gallisepticum and Mycoplasma synoviae among 16-week-old, 18-week-old, and 56-week-old breeder chickens were identified, and the antibody positive rates to seven major transovarian transmitted diseases among 1-day-old chicks were investigated. As a result, no infection with Salmonella pullorum and Salmonella gallinarum was found among the investigated subjects of all ages. Vaccinated breeder chickens showed the 100% antibody positive rate to Egg Drop Syndrome '76, and unvaccinated breeder chickens showed the 100% antibody negative rate to Mycoplasma gallisepticum and Mycoplasma synoviae, confirming that there was no infection with Mycoplasma gallisepticum and Mycoplasma synoviae. As a result of the antibody tests of the 1-day-old chicks for transovarian transmitted diseases, it was found that vaccinated chicks showed good antibody positive rates to avian encephalomyelitis, chicken infectious anemia, and avian reovirus, confirming that they had power of defense against the relevant infectious diseases, and that unvaccinated chicks showed the 100% antibody negative rates to avian leukosis, chicken reticuloendotheliosis, and Mycoplasma synoviae, confirming that there was no infection with the relevant diseases. Given that the results of this study showed that among the transovarian transmitted diseases of chickens, there was no history of infection with diseases against which vaccination was not administered and high antibody positive rates were found with diseases against which vaccination was administered, it is judged that chickens with good power of defense against diseases were bred, and it is deemed that constant monitoring and vaccination against transovarian transmitted diseases will be necessary for the control and prevention of the diseases.

Studies on Enzyme-Linked Immunosorbent Assay(ELISA) for Detection of antibody to Mycoplasma hyopneumoniae (돼지의 유행성폐렴 원인균(Mycoplasma hyopneumoniae)에 대한 항체가 분포도 조사)

  • Mycoplasmal pneumonia of swine(MPS) cause by Mycoplasma hyopneumoniae has been recognized as a serious impediment to swine production due to chronic respiratory disorder which result in the weight loss and decreased feed conversion. The disease causes a great economic losses in pig industry by characterizing with high morbidity, low mortality, growth retardation and low feed efficiency. The present study was conducted to investigate the titers of antibody against M hyopneumoniae from the regional and seasonal groups of the slaughtered pigs by enzyme-linked immunosorbent assay(ELISA). The result have shown that the average seropositive rate of M hyopneumoniae infection was 84.6% . The regional seropositive rate in Korea showed 87.4% in Kyonggj, 83.4n in Kangwon, 89.2% in Chungnam and 77.6% in Chungbuk area, respectively. Also the seasonal seropositive rate was appeared as 78.6% in spring,90.1% in summer, 76.9% in autumn and 83.8% in winter, respectively.

The Study of Mycoplasma pneumoniae Infection in Children with Respiratory Tract Infection (호흡기 감염증 소아에서 Mycoplasma pneumoniae 감염에 관한 연구)

  • Purpose: This study was performed for analysis of the results of polymerase chain reaction(PCR) and antibody test of Mycoplasma pneumoniae(M. pneumoniae) in children with symptoms of respiratory tract infection. In the cases of both positive antibody test and PCR for M. pneumoniae, the chest X-ray findings were assessed. Methods: The antibody test was done in 1,979 cases who have been admitted to Wonkwang university hospital department of pediatrics with symptoms of respiratory tract infection from January, 2000 to December, 2001. The positive antibody test was defined as titer of 1 : 80 and over 1 : 80. The PCR of M. pneumoniae were done in randomly selected 131 cases of respiratory tract infection. The chest X-ray findings were assessed in the cases of positive antibody test and PCR. Results: The numbers of cases of the positive antibody test for M. pneumoniae were 499 cases(25%). The PCR for M. pneumoniae were performed in 131 cases and the 45 cases(34%) were positive and 86 cases(66%) were negative. The 56 of 86 PCR negative cases were also negative antibody test, but 30 cases were positive antibody test. The 36 cases of 45 PCR positive cases were antibody positive, and 9 cases were antibody negative. The sputum Gram stain and culture for M. pneumoniae were negative in all the 499 cases of mycoplasma antibody positive respiratory infection. In these antibody positive 499 cases, the most common X-ray findings was interstitial pneumonic infiltration in 266 cases(53%), and pleural effusion were detected in 22 cases(4%), but nonspecific chest X-ray finding showed in 129 cases(26%). In PCR positive 45 cases, the most common chest X-ray finding was interstitial pneumonic infiltration in 32 cases(71%). Conclusion: The PCR for M. pneumoniae is more useful method for detection of mycoplasma infection in children with respiratory tract infection. The M. pneumoniae is a important etiologic agent for respiratory infection in children.

  • We investigated the serological prevalence of avian metapneumovirus (AMPV), avian reovirus (ARV), Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) in 760 broiler breeder (38 flocks), in the Jeonbuk province in 2013. This study was conducted to evaluate the immune and infection status of the broiler breeder flocks against AMPV, ARV, MG, MS by an enzyme-linked immunosorbent assay (ELISA). Serological test for AMPV were positive 37 (97.3%) flocks and 712 (93.6%) broiler breeder and geometric mean antibody titers were $16,350{\pm}10,195$, ARV were high positive rate 100% (38/38) flocks and 97.8% (743/760). The seropositive flocks against MG were 71.1% (27/38) and the geometric mean antibody titers were $2,474{\pm}2,045$, whereas the rates of positive flocks against MS were 50.0% (19/38) and the geometric mean antibody titers were $1,469{\pm}1,230$.

Prevalence of antibody titers for poultry disease in laying hens in Incheon area, Korea (인천지역 산란계 가금질병 항체 보유율 조사)

  • This study was conducted from May to December 2016 to investigate the prevalence of antibody titers for Newcastle disease (ND), Infectious Bronchitis (IB), Egg drop syndrome (EDS), Avian pneumovirus (APV), Fowl Typhoid (FT), Mycoplasma gallisepticum (MG), Mycoplasma synoviae (MS) in laying hens in Incheon area. In ND, 418 (99.5%) of the 420 outbreaks were positive. IB showed 330 (78.5%) positive antibody out of 420 cases. EDS showed 419 (99.7%) positive seropositive rates among 420 cases. In the case of APV, 357 cases (85%) were positive in 420 cases. In the case of PT, the positive rate of antibody was 128 (30.4%) out of 420 cases. Among the 420 cases of MG, 222 cases (52.8%) were positive for antibodies. In the case of MS, 395 out of 420 cases (94.0%) were positive. This result suggests that the positive rate of antibody for PT in Incheon area was low. And it was also clear that MS without vaccine is now rampant in Incheon.

Comparison of Eosinophil Markers between Acute and Recovery Stages in Children with Mycoplasma pneumoniae Pneumonia (Mycoplasma pneumoniae 폐렴 환아에서 급성기 및 회복기의 호산구 지표의 비교)

  • Purpose : Several studies have shown that increases of eosinophil markers are common findings of asthma and Mycoplasma pneumoniae infection, and eosinophil markers reflect the clinical stage of asthma. The purpose of this study was to investigate the change of eosinophil markers according to the clinical stage of Mycoplasma pneumonia. Methods : The patient group consisted of 33 outpatient children with Mycoplasma pneumonia. Peripheral blood total eosinophil count(TEC) and serum eosinophilic cationic protein(ECP) level were measured at both acute and recovery stages and were compared between both stages. The patient group was subdivided into the wheezing(n=16) and the nonwheezing group(n=17), and the TECs and the ECPs of one group were compared with those of the other group. The correlation between Mycoplasma antibody titer and the eosinophil markers of acute stage were analyzed. Results : In the whole patient group, the TECs and the ECPs of the acute stage were significantly higher than those of the recovery stage(P=0.018, P=0.005), but there were no differences in the TEC and the ECP between the wheezing and the nonwheezing group. In the wheezing group, there were no significant differences in the TEC and the ECP between acute and recovery stages. There were no correlations between acute stage Mycoplasma antibody titer and the eosinophil markers. Conclusion : Eosinophil markers reflect the clinical stage of Mycoplasma pneumonia and eosinophilic inflammations may continue even after the acute stage in wheezing patients with Mycoplasma pneumonia.
