Mycological Characteristics of Nine Unrecorded Yeasts from Flowers in the Orchard of Yesan-gun, Chungcheongnam-do and Hanbat Arboretum in Daejeon City, Korea

  • Six unrecorded yeasts, Cryptococcus festucosus 41-3, Cryptococcus heveanensis 56-4, Debaryomyces nepalensis 95-4, Issatchenkia occidentalis 142-1, Dioszegia zsoltii 39-1, and Kwoniella europala 47-2 were screened from 108 yeasts isolated from flowers and fruits in orchards of Yesan-gun, Chungcheongnam-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. They had various shapes, including ellipsoidal, globose, and oval, and also had budding mode in vegetable reproduction, except I. occidentalis 142-1 (fission mode). K. europaea 47-2 only formed pseudomycelium. D. zsoliti 39-1 did not grow in yeast extract-malt extract medium, potato dextrose medium, and vitamin-free medium. C. festucosus 41-3 grew well in 5% NaCl-containing yeast extract-peptone-dextrose medium and had a growth pH range of 7.0~10.0. Three unrecorded yeasts Ogataea polymorpha HB45-1, Rhodotonula hinnulla HB62-2, and Cryptococcus rajasthanensis HB80-4 were screened from 51 yeasts isolated from flowers in Hanbat arboretum in Daejeon city, Korea. They were globose in shape and did not form pseudomycelium. In addition, O. polymorpha HB45-1 and C. rajasthanensis HB80-4 had budding mode in vegetable reproduction. All of them grew well in vitamin-free medium and C. rajasthanesis HB80-4 also grew in 50% glucose and 5% NaCl-containing YPD medium.

  • Gray mold of blueberry caused by Botrytis sp. is reported for the first time in Korea. A detailed description of the fungus is given, along with its rDNA internal transcribed spacer sequence. The fungus was identified as Botrytis cinerea based on mycological characteristics and molecular data.

  • Stem rot was found for the first time on the Asiatic dayflower plant (Commelina communis L.) in Korea. A detailed description of this Korean specimen is given, along with its rDNA internal transcribed spacer sequence. The fungus was identified as Sclerotium rolfsii Saccardo based on mycological characteristics and molecular data.

  • Rhizopus oryzae is reported for the first time on Cucumis melo L. var. makuwa Makino. A detailed description of this Korean specimen is given, along with its rDNA internal transcribed spacer sequence. On the basis of mycological characteristics and molecular data, the fungus was identified as R. oryzae Went & Prinsen Geerligs.

  • Cultivation of a native wild vegetable, Ligularia fischeri, is hampered by Phytophthora sp. Infected plants showed wilt and eventual death due to rots on the root and basal portion. Eight isolates collected were all identified as P. drechsleri based on their mycological characteristics. The fungi showed relatively strong pathogenicity to L. Fischeri, mild to Aster scaber and Codonoposis lanceolata, and none to Circium setidens and Pimpinella brachycarpa. This is the first report of Phytophthora root rot on wild vegetables in Korea.

Silver Scurf of Potato Caused by Helminthosporium solani

  • Potate tubers with slver scurf lesions were collected from the cold storage at Pyungchang, Kangwon province in Korea. The causal agent of the silver scurf was identified as Helminthosporium solani by mycological characteristics of conidia and conidiophores. Pathogenicity of the fungus was confirmed by artificial inoculation on the potato tuber. This is the first report of potato silver scarf by Helminthosporium solani in Korea.

  • Soft rot in apple caused by Rhizopus oryzae was found for the first time in Korea. A detailed description of the specimen is given along with its internal transcribed spacer rDNA sequence. The fungus was identified as Rhizopus oryzae based on the mycological characteristics, molecular data, and pathogenicity testing.

  • A Pythium species was isolated from roots of lisianthus (Eustoma grandiflorum) showing wilt symptoms and reduced growth in a greenhouse at Ichon, Kyonggi-do in 1997. The Pythium species was identified as Pythium spinosum Sawada based on various mycological characteristics. The isolate was strongly pathogenic when inoculated to root of lisianthus plants in pots. The diseased plants showed typical symptoms of root and crown rot, resulting in reduced growth of roots and shoots, and consequently wilting of the above ground part of plants.

벼키다리 병원균(病原菌)인 Fusarium moniliforme의 균주특성(菌株特性)에 의(依)한 Strain 분류(分類) (Classification of Fusarium moniliforme Isolates into Four Strains Based on Mycological Characteristics)

  • 벼키다리병(病)에 이병(罹病)된 벼에서 531균주를 분리(分離)하여 5가지 기준(基準)을 두고 분류(分類)한 결과(結果) Strain I, II, III과 IV로 분류(分類)한 수 있었다. 그중에서 Strain IV가 가장 많이 우리나라 분포(分布)되었으며 병원성도 강(强)한 것으로 나타났다. 5가지기준(基準)은 기주(寄主)의 병징(病徵), 포자형성(胞子形成), 대형분생포자격막수(大型分生胞子隔膜數), 균총의 색깔과 약제(藥劑)의 반응(反應)이었다. Strain I은 병원성(病原性)이 약하게 나타났으며 pionnotes안에 $6{\sim}7$개(個)의 격막(隔膜)을 가진 대형분생포자(大型分生胞子)를 형성(形成)하며 균총의 색깔은 표면(表面)과 이면(裏面)에 전부(全部) reddish purple을 보인다. Strain II는 병원성(病原性)은 약(弱)하나 pionnotes안에 소형분생포자를 형성(形成)하며 균총의 색깔은 표면(表面)과 이면(裏面)에 전부(全部) reddish purple을 보인다. Strain III는 병원성(病原性)이 강(强)하여 심하게 도장(徒長) 시키며 sporodochia안에 4개의 격막(隔膜)을 가진 대형분생포자(大型分生胞子)를 형성(形成)하며 균총의 색깔은 표면(表面)에는 yellowish red 이면(裏面)에는 reddish purple를 보인다. Strain IV은 병원성(病原性)이 강(强)하여 심하게 도장(徒長)시키기도 하고 고사(枯死)시킨다. sporodochia안에 주로 3개(個)의 격막(隔膜)을 가진 대형분생포자(大型分生胞子)를 형성(形成)하며 균총의 색깔은 표면(表面)과 이면(裏面) 전부(全部) yellowish red를 보이는 것이 특징이다.

  • 1995년 12월 전라북도 지역 내에 유통되고 있는 시클라멘의 꽃잎 중앙부에 진회색~흑색 원형 반점을 형성하거나, 꽃잎 주변에 흑색 수침상을 나타내는 탄저병의 발생이 관찰되었다. 병반으로 붜 분리된 균은 PDA 배지상에서 회백색을 띄었으며, 8.3~15.0${\times}$2.5~6.3 ${\mu}m$ 크기의 양끝이 둥근 무색 단포의 원통형~타원형의 분새오자를 형성하였고, 갈색 곤봉형의 부착기와 이병조직에서 갈색 둥근형의 분생자층을 형성하였다. 이에 따라 시클라멘 탄저병징에서 분리한 병원균은 국내에서 미보고된 Colletotrichum gloeosporioides로 동정되었으며, 균사생장 및 분생포자 형성을 위한 배양적정 온도는 25~30$^{\circ}C$였다.
