Cultivation Charateristics of Neolentinus lepideus Strains on Sawdust Media (잣버섯 수집균주의 톱밥재배 특성)

  • We investigated the cultivation characteristics of Neolentinus lepideus (21 strains derived from Korea and Japan) on Pinus densiflora sawdust medium as follows; period of mycelial growth, fruiting period, productivity of fruit body and color and morphology of fruit body. The periods for mycelial growth and fruiting on the sawdust medium of N. lepideus were 40~65 days and 6~22 days, respectively. It was produced approximately 48.3 g of fresh weight and 9 fruit-bodies per 600 g of sawdust medium. The productivity of fruit body was reduced by scratching off the surface of sawdust medium. We suggest that three strains (KFRI 1199, 1302, 1331) might be good candidates for sawdust cultivation.

Selection of parental strain on the sawdust cultivation and mycelial growth and cultural characteristics of Lentinula edodes hybrid strains (표고 톱밥재배용 모균주 선발과 교배균주의 균사배양 및 생육 특성)

  • In order to breed new strains of oak mushroom, Lentinula edodes, on the sawdust cultivation, we collected 10 from Korea, Taiwan and China respectively and examined somatic incompatibility, morphological features of fruiting body and productivity. Four strains(SANJO701HO, FMRI2534, FMRI2337 and FMRI2613) shown remarkable results, were confirmed with parental strains. 80 monokaryotic strains derived from the selected 4 parental strains, were selected and 117 hybrid strains were made by mono-mono mating. Aslo, Physiological characteristics were investigated. Average of the mycelial growth of hybrid strains of mating combination SANJO701HO-FMRI2337 was approximately 10% lower than other mating combinations. Overall, This study was founded to develop new varieties of L. edodes. The productivity of new 117 strains on sawdust cultivation needs continued research.

Cultivation of Oyster Mushrooms Using the Garlic Peel as an Agricultural by-product (마늘껍질을 이용한 느타리버섯의 인공재배)

  • High prices of raw materials used as media for the mushroom cultivation increased the cost of commercial production of oyster mushroom (Pleurotus ostreatus). In this study, garlic peels (Allium sativum f. pekinese) as an agricultural by-product were investigated to replace the saw dust for the bottle cultivation of oyster mushroom. Mycelial growth of oyster mushroom were examined by the extracts made from the sawdust, rice bran and garlic peels. The mycelial growth was very poor in the agar media containing the extracts of sawdust or garlic peels, but was good when those of the rice bran were added. In the polypropylene bottle experiment, the sawdust medium which ammended with minerals vitamin was essential for the production of the mushroom fruitbodies. The rice bran was considered to stimulate the mycelial growth, but not the development of basidiocarps. The garlic peel was not a factor to stimulate the production of mushroom fruitbodies, but a raw material ammended with the rice bran produced much amounts of mushroom. In this work, garlic peels ($10{\sim}70%$ v/v) added to the mixture of sawdust and rice bran (4 : 1, v/v) was considered to help the productions of mushroom fruitbodies. Based on the result, the replacement of expensive saw dust with inexpensive garlic peels was a good example to reduce production cost of the bottle cultivation of mushroom.

Breeding of a new cultivar of Lentinula edodes 'Charmjon' (표고버섯 신품종 '참존' 육성 및 특성)

  • We aimed to develop outstanding domestic varieties suitable for both columnar and cylindrical-shape substrates, aiming to replace foreign varieties. and bred a high-quality new strain named 'Charmjon', using genetic resources collected from Japan and China. The optimal cultivation temperature for Charmjon's mycelial growth was found to be 25℃, and its mycelial growth at 15℃ and 25℃ was superior to the control variety. In terms of mycelial growth characteristics based on the substrate, Charmjon exhibited excellent mycelial strength on PDA medium compared to the control variety. Through columnar and cylindrical-shape substrates cultivation, we assessed the quantity and morphological characteristics of the fruiting bodies. The results confirmed that Charmjon can be produced stably using both cultivation methods, and it showed higher yields and individual weights than the control variety. In addition, the color of the pileus was notably darker, and the shape of the pileus varied depending on the cultivation method. The test of genetic diversity revealed that Charmjon has distinct genetic characteristics compared to the control varieties.

Screening of-Lyophyllum Decastes-Highly Productive Cultivable Strains

  • In order to explore mycelial growth and fruiting body formation of Lyophyllum decates on different media, ten cultivation media by using cottonseed hull, sawdust, corn cob etc. as main components were designed for seven strains. The results showed that the mycelial colour of all strains are mainly snow-white, and the formula of media using corn cob as main materials was better than that using cottonseed hull and sawdust for mycelial growth, but no fruiting body was formed. The cottonseed hull medium with a small amount of sawdust, plant leaves, humus or fermented material and wheat was beneficial for fruiting formation. The incubation period for fruiting formation of strain 3001 was 108 days and the highest yield was-214.80 g/bag. Fructification of the strains tasted occurs successively in order of 3001, 1035, 1004 and 1013. It was concluded that different medium composition had significant effect on the mycelial growth and fruiting body formation.

  • Rice hull was used as a additive in order to find the effect for incresing of mushroom growth and yield in Chungnam Provincial techinical institution. 1 Treatment of 80% rice hull in small Neutaribeosut mycelial grow duration is shorter about 11 days and yield increased about 7% than conventional culture. 2. In case of Chongpung Neutaribeosut bottle culture, mycelial growth duration is shorter about two to three days in additive of 30 to 80% rice hull compared to conventional but yield similar to conventional. 3. Treatment of 30% rice hull in Agrocybe aegerita bottle culture, mycelial growth and yield increased 6days and 6% than convrntional, respectively 4 Treatment additived of 30% to 40% rice hull in Ganoderma lucidum bottle culture, similar to 454ays demand in mycelial grow duration and 38g yield/bottle in conventioal culture methods.

Morphological Changes of Hansenula anomala B-7 by Cadmium Ion (카드뮴이온에 의한 Hansenula anomala B-7의 형태 변이)

  • Yeast-form cells of cadmium ion-tolerant Hansenula anomala B-7 were changed to mycelial cells in medium containing more than $400\mu$g/ml of cadmium. Moreover, the mycelial cells were exchanged into clumped cells in a medium containing more than $1,000\mu$g/ml of cadmium. Optimal conditions of mycelial cell formation were achieved in the presence of .$1,000\mu$g/ml of cadmium with shaking cultivation for 7 days. Glucan and mannan contents of the yeast cell wall frown with $1,000\mu$g/ml of cadmium decreased by 10% compared with those grown without cadmium. However, protein and lipid contents increased about 20% respectively. By cadmium, no significant findings in specific amino acid contents were discovered.

Effect of Supplement nutrition on the Mycelial Growth of Lentinus edodes

  • Mycelial growth of L. edodes by supplement nutrition of softwood was studied on a sawdust medium. The sawdust used was from the following softwood species : Larix leptolepis, Pinus densiflora and Pinus koraiensis. The added nutritions consisted of carbon nutritions(sucrose, active carbon, xylose, glucose, paper pellet), nitrogen nutritions(potassium nitrate, ammonium chloride, asparagine, glutamic acid) and vegetable oil(rice bran oil). The sawdust medium was a mixture of 76% sawdust, 20% rice bran, 3% carbon nutrition, 0.4% nitrogen nutrition and 0.6% calcium carbonate. Following addition of carbon and nitrogen nutritions on the sawdust medium proved most suitable : L. leptolepis (glucose, glutamic acid), P. densiflora (active carbon, asparagine) and P. koraiensis (xylose, glutamic acid). The highest mycelial growth was obtained from sawdust medium of optimum condition with 97% of L. leptolepis, 110% of P. densiflora and 98% of P. koraiensis. This study has provided useful preliminary information for the cultivation of L. edodes.

Selection of Effective Fungicides Against Xylogone sphaerospora, a Fungal Pathogen of Cultivated Mushroom, Ganoderma lucidum (영지 노랑병 방제에 효과적인 살균제의 선발)

  • A fungal disease of the cultivated mushroom, Ganoderma lucidum, caused by Xylogone sphaerospora was epidemic throughout all cultivation areas in Korea which caused a lot of yield losses in the mushroom production. For controlling the disease, the screening of effective fungicides against the pathogenic fungus were conducted. Thirty seven commercially available fungicides were tested for their inhibitory activities on potato dextrose agar media supplemented with these fungicides at various concentrations. Twenty one fungicides significantly inhibited mycelial growth of the pathogen, Xylogone sphaerospora, but 16 fungicides had no inhibitory effect. Among these 21 fungicides, 17 fungicides also inhibited mycelial growth of Ganoderma lucidum as well, but imazalil, procymidone, triforine, and vinclozolin had no inhibitory effects. However, vinclozolin showed no inhibitory effect on mycelial growth of the mushroom even at the concentration of 50 $\mu\textrm{g}$/ml vinclozolin solution for 2 hours, and then the pathogen was inoculated. After two month-cultivation of the mushroom, over 90% of logs treated with vinclozolin without pathogen inoculation produced fruiting bodies. However, fruiting bodies were not produced form the logs inoculated with the pathogen, but not treated with vinclozolin. Fifty seven percent of logs. which were pre-treated with vinclozolin and then inoculated with the pathogen produced fruiting bodies. Based on the results, vinclozolin is effective for the control of yellow disease of the Ganoderma lucidum caused by Xylogone sphaerospora.

영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 수용성 다당류의 분획 및 항암활성

  • Exo-polysaccharide (BWS) obtained from submerged cultivation of Ganoderma lucidum mycelium was fractionated. Antitumor activity of their fractions was investigated in comparison with the mycelial polysaccharide fractions. Eight kinds (BWS-DN, BWS-DA, BWS-DN-GI, BWS- DA-GI, MWS-DN, MWS-DA, MWS-DN-GI and MWS-DA-GI) of polysaccharide fractions were obtained by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel chromatography from BWS and MWS, which were isolated from culture fiuid and mycelial cell, respectively. The anticomplementary activities (ITCH$_{50}$%) of the exo-polysaccharide fractions showing 15% to 30% were lower than those of mycelial polysaccharide fractions showing 15% to 70%. The acidic fractions of BWS-DA and BWS-DA-GI fractionated from BWS, showed the highest activity of 30%. In the MTT assay, BWS-DN and MWS against mouse leukemia L1210 exhibited high inhibition ratio of 86 and 89%, respectively at the concentration of 600 $\mu$g/ml. High inhibition ratio of 50% (IC$_{50}$) was achieved for BWS, BWS-DA and MWS-DA fractions against human colon adenocarcinoma COLO-205 and for BWS-DA, BWS-DN and MWS-DN fractions against human leukemia HL-60 at the concentra- tion of 300 $\mu$g/ml among the six polysaccharide fractions, respectively.
