Protein secretion in filamentous fungi has been shown to be restricted to actively growing hyphal tips. To determine whether an increase in the amount of growing surface area of a fungus can lead to an increase in the amount of protein secretion, we examined secretion in a temperature-sensitive Neurospora crassa mcb mutant that shows a loss of growth polarity when incubated at restrictive-temperature. Incubation of the mcb mutant at restrictive-temperature results in a three- to five-fold increase in the level of extracellular protein and a 20- fold increase in carboxymethyl cellulase activity relative to a wild-type strain. A mutation in the cr-l gene has been shown previously to suppress the apolar growth phenotype of the mcb mutant, and we find that the level of extracellular protein produced by a mcb; cr-l double mutant was reduced to that of the wild-type control. Immunolocalization of a secreted endoglucanase revealed that proteins are secreted mainly at hyphal tips in hyphae exhibiting polar growth and over the entire surface area of bulbous regions of hyphae that are produced following a shift of the mcb mutant to restrictive-temperature. These results support the hypothesis that secretion of extracellular protein by a filamentous fungus can be significantly increased by mutations that alter growth polarity.

Ansamitocin P-3 is a potent antitumor agent produced by A. pretiosum. A deletion mutant of A. pretiosum was constructed by deleting the asm2 gene, a putative transcriptional repressor. The deletion mutant showed a 9-fold enhanced ansamitocin P-3 productivity. The response surface method with central composite design was employed to further optimize the culture medium composition for ansamitocin P-3 production by the deletion mutant. The concentrations of four medium ingredients, dextrin, maltose, cotton seed flour, and yeast extract, which have been reported as major components for ansamitocin production, were optimized through a series of flask culture experiments. The optimum concentrations of the selected factors were found to be dextrin 6.0%; maltose 3.0%; cotton seed flour 0.53%; and yeast extract 0.45%. The maximum titer of ansamitocin P-3 was 78.3 mg/l with the optimized composition, about 15-folds higher than the unoptimized titer of 5.0 mg/l obtained with YMG medium.
Ile-224 in I269S mutant horse liver alcohol dehydrogenase isoenzyme S (HLADH-S) was mutated to serine by site-directed mutagenesis in order to study the role of the residue in c oenzyme binding to the enzyme. The specific activity of the I269S and I224S mutant enzyme to ethanol was increased 6-fold and all Michaelis constants($K_a,\;K_b,\;K_p,\;and\;K_q$,/TEX>) were larger than those for the wild-type and I269S enzyme. The substitution decreased the afffinity to coenzymes and increased the specific activity of the enzyme. The mutant enzyme showed the highest catalytic efficiency for octanol among the primary alcohols. But it didn`t have activities on retinoids and 5${\beta}$-cholanic acid-3-one. From these results, it was confirmed that the hydrophobic interaction of Ile-224 residue with coenzyme was related to coenzyme affinity in ADH reaction. The substitution also affected the substrate affinities to the enzyme.
Quorum sensing and the stringent response are well-known regulation systems for the expression of virulence genes in enterohemorrhagic Escherichia coli (EHEC). However, how these two systems interact is not well known. E. coli strains with mutations in two regulation systems, ${\Delta}luxS$ (ECM101) and ${\Delta}luxS{\Delta}relA{\Delta}spoT$ (ECM201), and the ${\Delta}luxS$ complement strain to ECM201 (ECM202) were created from EHEC O157:H7 EDL933 to investigate how the regulatory systems interact. The phenotypic changes of the mutant strains were characterized and compared with the wild type. The mutant strains exhibited no obvious growth defects, although acid resistance and cellular cytotoxicity were decreased significantly in all the mutant strains. Phenotypic characterization revealed that mutations in the stringent response system (ECM201 and ECM202) influenced the metabolic (defective utilization of arabinose and L-sorbose) and enzymatic activities (decreased trypsin activity, and increased ${\alpha}$-glucosidase activity). In contrast, the quorum sensing system mutant (ECM101) did not display these phenotypes. The motility of the quorum sensing system mutant (ECM101) was unchanged, but mutation in the stringent response system influenced the motility. Our results suggest that quorum sensing interacts with the stringent response regulation system.

The dlm (disease lesion mimic) mutant of soybean (Glycine max L. Merr) shows the similar lesion of a soybean disease caused by a fungus, Corynespora cassilcola. The lesion was examined at cellular and molecular level. Trypan blue staining result indicated that cell death was detectable in the entire region of leaves excluding veins when the lesions had already been developed. We found that the mesophyll cells of palisade layer in the dim mutant appeared to be wider apart from each other. The chloroplasts of the dim mutant cells contained bigger starch granules than those in normal plants. We also found that the lesion development of dlm plant was light-dependent and the starch degradation during the dark period of diurnal cycle was impaired in the mutant. Three soybean pathogenesis-related genes, PR-1a, PR-4, and PR-10, were examined for their expression patterns during the development of disease lesion mimic. The expression of all three genes was up-regulated to some extent upon the appearance of the disease lesion mimic. Although the exact function of DLM protein remains elusive, our data would provide some insight into mechanism underling the cell death associated with the dim mutation.
Food intake is regulated by both central and peripheral mechanisms. In the central nervous, the hypothalamus acts for autonomic and endocrine homeostasis. The paraventricular nucleus(PVN) of hypothalamus is an imprtant site of interaction in central feeding pathways. Neuroepetide Y(NPY)is one of the most powerful neurochemical stimulants of food intake known. Also brain nitric oxide(NO), known as neurotransmitter, is involved in the mechanisms that regulate food intake. In this experiment, 24h fasting mice and anorexia mutant mice have been to examine the expression of NPY, which is the major neuropeptide increasing food intake. Double staining with NPY and nicotinamide-adenine-dinucleotide-phosphate diaphorase(NADPH-d), followed by immunohistochemical method and image analysis, have been used to observe coexisting neurons and the level of expression of each neurons. The results were as follows. 1) NPY-immunoreactivitys reduced immune response of the hypothalamus, particularly paraventricular nucleus(PVN), in anorexia mutant mice. Decreased level of NPY is assumed to be a major pathological factor in anorexia mutant mice. On the other hand, PVN in hypothalamus of fasting mice showed increased immunoreactivity which is in agreement of other researchers. 2) NPY and NADPH-d double staining revealed coexisting neurons in the cerebral cortex. Fasting mice had a tendency to have increased level of coexisting neurons compared to the control group. Compared to the control group, fasting mice express is not increase level of NPY-immunoreactivity, while anorexia mutant mice tended to have a decreased level.

To isolate a mutant which overproduces threonine and tryptophan, mutants of Saccharomyces cerevisiae were screened after UV and EMS mutagenesis. Hydroxynorvaline, a Thr analogue was used for selection of a Thr-overproducing mutant after UV mutagenesis. Among 31 mutants, TC 5-1 was selected as the strain candidate, based on amino acid analysis. TC 5-1 was then treated by EMS mutagenesis for Trp overproduction. Eight mutants were selected using fluorotryptophan for Thr and Trp overproducing strains. Amino acid analysis results showed that TC 6-1 was the best strain since it had the highest amount of Thr and Trp among mutants.

After random mutagenesis, the mutant Lactobacillus plantarum GNS300 showed improved exopolysaccharide production as determined by the quantification of total sugar. The mutant L. plantarum GNS300 produced 2.82 g/l of exopolysaccharide which showed 79.62% improved exopolysaccharide production compared with the parental strain. When exopolysaccharide of L. plantarum GNS300 was analyzed, the exopolysaccharide is composed of galactose (93.35%) and glucose (6.65%). Through the optimization of fermentation conditions using a bioreactor, 2.93 g/l of exopolysaccharide was produced from 20 g/l of glucose at 35℃, 500 rpm, and 0.1 vvm for 12 h. The mutant L. plantarum GNS300 exhibited 69.18% higher antioxidant activity than that from the parental strain, which might be caused by higher exopolysaccharide production. The concentrated supernatant of the mutant L. plantarum GNS300 inhibited the growth of gram-positive bacteria (Bacillus cereus and Staphylococcus aureus) and gram-negative bacteria (Escherichia coli, Vibrio parahaemolyticus, and Salmonella typhimurium).
S. pneumoniae recP mutant was compared with rec-8 mutant to identify whether they are the same gene or not by determining sensitivity to DNA damaging agents. recP and rec-8 mutant have almost same sensitivity to UV, ethylmethane sulfonate, and methylmethane sulfonate, suggesting that recP has the same function as the rec-8 gene in DNA repair.
Kinetics of anaerobic ethanol fermentation by Clostridium thermohydrosulfuricum were investigated for the one-step production of ethanol from starch. A mutant strain with a high ethanol yield was induced from C. thermohydrosulfuricum. The mutant, designated as ME4, produced anaerobically 6.1 g/l of ethanol, 3.1 g/l of lactate and 0.1 g/l of acetate from 20 g/l of starch at $68^{\circ}C.
