The Pharmacological Activity of Coffee Fermented Using Monascus purpureus Mycelium Solid-state Culture Depends on the Cultivation Area and Green Coffees Variety (원산지 및 품종에 따라 조제된 홍국균 균사체-고체발효 원두커피의 생리활성)

  • In previous work, we fermented coffee beans using solid-state culture with various fungal mycelia to enhance the physiological activity of the coffee. The coffee fermented with Monascus sp. showed a higher physiological activity than non-fermented coffee or other coffees fermented with mushroom mycelium. The aim of this study was to characterize the various fermented coffees with respect to their area of cultivation and their variety using Monascus purpureus (MP) mycelium solid-state culture. Thirty types of green coffee beans, which varied in terms of their cultivation area or variety, were purchased from different suppliers and fermented with MP under optimal conditions. Each MP-fermented coffee was medium roasted and extracted further using hot water (HW) under the same conditions. Of the HW extracts, those derived from MP-Mandheling coffees had the highest yield (13.6-15.5%), and MP-Robusta coffee showed a significantly higher polyphenolic content (3.03 mg gallic acid equivalent/100 mg) and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) free radical scavenging activity (27.11 mg ascorbic acid equivalent antioxidant capacity/100 mg). Furthermore, in comparison to other MP-fermented coffees at $1,000{\mu}g/mL$, MP-Robusta coffee showed not only the most effective inhibition of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) production in LPS-stimulated RAW 264.7 cells (67.1% of that in LPS-stimulated control cells), but also an effective inhibition of lipogenesis in 3T3-L1 adipose cells (22.2% of that in differentiated control cells). In conclusion, these results suggest that Vietnam Robusta coffee beans solid-state fermented with MP mycelium are amenable to industrial applications as a functional coffee beverage or material.

The Analysis of Management and the Method of Cultivation of Lentinus edodes I. for Full-Development of Mycelium in Bed Logs (표고재배(栽培)의 관리분석(管理分析)과 종균활착(種菌活着)을 위한 골목관리(管理)에 관(關)한 연구(硏究))

  • This study was carried out to offer the successive method of cultivation and increase the productivity of mushroom yield with good quality through the elevation of rate of spawn development for Lentinus edodes. Studied about the analysis of current management of actural cultivation, a base of these, researched and presented for the upward method of productivity through an experiment of the high rate of spawn development and cultivation, putting first cultural environment. The results obtained were as follows ; 1. As the result of the analysis of current management in actural cultivation, many cultivators had a tendency to neglect managements of cultivation. These were reason for the deficiency of labour, funds and the lack of knowledge of cultivation, etc. 2. Water contents in bed logs according to the date of inoculation was shown as the decreasing order of 28.63%(3/12), 25.20%(3/25) and 23.19%(4/10). For the purpose of the maintenance of the water contents, the full-development of mycelium in bed logs and the dispersion of labour, the date of inoculation should be started in the early March. 3. The difference of the rate of spawn development among species was not shown, 100%(Mori 465). 98.98%(Mori 3046) on the spawn in high temperature and 98.97%(Mori 290) on the spawn in low temperature. The relative rate of spawn development was 97.70%(Mori 465), 82.45%(Mori 3046) on the spawn in high temperature and 88.87%(Mori 290) on the spawn in low temperature, it showed the difference. The spawn should be selected carefully in the future, as the spawn of cultivater's preference showed the difference for the development of mycelium. 4. The rate of spawn development following the date of inoculation was 100.0%(3/12), 98.98%(3/25) and 96.79%(4/10) on the spawn in high temperature and 99.09%(3/12), 98.97%(3/25) and 97.89% (4/10) in low temperature, it showed little difference. And the relative rate of spawn development was 97.70%(3/12), 82.45%(3/25) and 81.42%(4/10) on the spawn in high temperature and 93.27%(3/12), 89.67%(3/25) and 88.87%(4/10) that in low temperature, As the result of the relative rate, the time of inoculation of spawn should begin in the early March. 5. The height of stock logs on temporary placing should be less than 60cm at most on the surface, because of the low rate of water contents.

Optimization of the Expression of the Ferritin Protein Gene in Pleurotus eryngii and Its Biological Activity (큰느타리버섯에서 석충 페리틴 단백질 유전자의 발현 최적화 및 생물학적 활성)

  • To optimize the expression and secretion of ferritin protein associated with ion storage in the mushroom, Pleurotus eryngii, a recombinant secretion vector, harboring the ferritin gene, was constructed using a pPEVPR1b vector under the control of the CaMV 35S promoter and signal sequence of pathogen related protein (PR1b). The ferritin gene was isolated from the T-Fer vector following digestion with EcoRI and HindIII. The gene was then introduced into the pPEVPR1b secretion vector, and it was then named pPEVPR1b-Fer. The recombinant vector was transferred into P. eryngii via Agrobacterium tumefaciens-mediated transformation. The transformants were selected on MCM medium supplemented with kanamycin and its expression was confirmed by SDS-PAGE and western blotting. Expression of ferritin protein was optimized by modifying the culture conditions such as incubation time and temperature in batch and 20 L airlift type fermenter. The optimal conditions for ferritin production were achieved at 25℃ and after incubating for 8 days on MCM medium. The amount of ferritin protein was 2.4 mg/g mycelia, as measured by a quantitative protein assay. However, the signal sequence of PR1b (32 amino acids) seems to be correctly processed by peptidase and ferritin protein may be targeted in the apoplast region of mycelia, and it might not be secreted in the culture medium. The iron binding activity was confirmed by Perls' staining in a 7.5% non-denaturing gel, indicating that the multimeric ferritin (composed of 24 subunits) was formed in P. eryngii mycelia. Mycelium powder containing ferritin was tested as a feed additive in broilers. The addition of ferritin powder stimulated the growth of young broilers and improved their feed efficiency and production index.

  • Background: The biological activities of ginseng saponins (ginsenosides) are associated with type, number, and position of sugar moieties linked to aglycone skeletons. Deglycosylated minor ginsenosides are known to be more biologically active than major ginsenosides. Accordingly, the deglycosylation of major ginsenosides can provide the multibioactive effects of ginsenosides. The purpose of this study was to transform ginsenoside Rb2, one of the protopanaxadiol-type major ginsenosides, into minor ginsenosides using ${\beta}$-glycosidase (BG-1) purified from Armillaria mellea mycelium. Methods: Ginsenoside Rb2 was hydrolyzed by using BG-1; the hydrolytic properties of Rb2 by BG-1 were also characterized. In addition, the influence of reaction conditions such as reaction time, pH, and temperature, and transformation pathways of Rb2, Rd, F2, compound O (C-O), and C-Y by treatment with BG-1 were investigated. Results: BG-1 first hydrolyzes 3-O-outer ${\beta}$-$\text\tiny{D}$-glucoside of Rb2, then 3-O-${\beta}$-$\text\tiny{D}$-glucoside of C-O into C-Y. C-Y was gradually converted into C-K with a prolonged reaction time, but the pathway of Rb2 ${\rightarrow}$ Rd ${\rightarrow}$ F2 ${\rightarrow}$ C-K was not observed. The optimum reaction conditions for C-Y and C-K formation from Rb2 by BG-1 were pH 4.0-4.5, temperature $45-60^{\circ}C$, and reaction time 72-96 h. Conclusion: ${\beta}$-Glycosidase purified from A. mellea mycelium can be efficiently used to transform Rb2 into C-Y and C-K. To our best knowledge, this is the first result of transformation from Rb2 into C-Y and C-K by basidiomycete mushroom enzyme.

Mycelial Growth and Fruiting Body Formation of Hericium erinaceum in Sawdust and Agricultural By-product Substrates (톱밥 및 농업부산물 이용 배지상에서 노루궁뎅이버섯(Hericium erinaceum)의 균사생장 및 자실체형성)

  • This study was carried out to investigate the suitability of various agricultural by-products as basal substrates for the mycelial growth and fruiting body formation of Hericium erinaceum. For this aim, oak sawdust, cotton waste, sugarcane bagasse, Job's tears, rice hull, Chinese cabbage, and coconut waste were used as sole or mixed substrate(s). Corn waste and rice bran were used as nutrient supplements. The growth and density of mycelium, yield of fruiting body, and biological efficiency were compared among tested substrates colonized by Hericium erinaceum. The best measurement of mycelial growth and density, yield of fruiting body, and biological efficiency in a laboratory test was found in a spawn substrate composed with oak sawdust 80% and rice bran 20%. The suitability of this spawn substrate composition for Hericium fruiting body production was testified through practical tests in plastic bottles (850 ml) in a mushroom farm which had bottle cultivation facility. However, test in a mushroom farm which had plastic bag cultivation facility, best production of Hericium fruiting body (520 g per one bag) was observed in a spawn substrate composed of cotton waste 40%, saw dust 40%, corn waste 10%, and rice bran 10%.

Effect of solid grain media on the mycelial growth of Pleurotus ostreatus and its biofunctional activity (곡물배지가 느타리버섯의 균사배양과 생리활성에 미치는 영향)

  • Effect of solid grain media on the mycelial growth of Pleurotus ostreatus and its biofunctional activities were investigated using seven kinds of grains. Foxtail millet and barley were good for growth of hypha of P. ostreatus. However, growth of the mycelium was very slow in the solid grain media containing wheat, corn and brown rice. Mycelial growth of P. ostreatus according to water content of solid grain media was good at 25% to 30%. Mycelial growth of P. ostreatus according to heating-time and temperature of solid grain media was good for 30 min at $121^{\circ}C$. Anticancer activities against lung cancer cell line of the myceial extracts from P. ostreatus grown on several grain media were strong in the corn, defatted soybean, brown rice, barly and black bean in order. Fibrinolytic activities of the myelial extracts were strong in order defatted soybean, wheat, foxtail millet, barley, brown rice and black bean. The mycelial extracts were showed good antibacterial activities against Pseudomonas aeroginosa and Staphylococcus aureus.

  • Phellinus linteus (Sang Hwang mushroom; P. linteusis) has been widely used for a traditional medicine in Japan, Korea and China. In traditional oriental applications, P. linteus and their extracts have been exclusively used to treat various diseases like insomnia, neurasthenia, gastric ulcers, arthritis, nephritis, stress, asthma, bronchitis, neuromuscular disorders, hypertension and also as an immune system stimulator in cancer therapy. In a number of articles, it is indicated how tremendous their therapeutic values are. According to these studies, P. linteus and their extracts show immunomodulatory activities like enhancing immune system function including anti-tumor activity and inhibiting the growth of existing tumors. P. linteus and their extracts have been being studied for use as immunotherapy agents and biological response modifiers (BRMs) for the possible treatment of cancer without side effects. Recently, the cultured mycelium product of P. linteus has been developed and approved as a medicine in Korea. This review shows not only the current status of methods of developmental technology like artificial culture and various biological applications of P. linteus but also recent market trends of functional foods made from P. linteus.

Development of an apple/pear pomace fermented with Lentinus edodes Mycelia (사과/배 부산물 및 표고버섯균사체를 이용한 발효물 개발)

  • The purpose of this study was to investigate the possibility of enhancing the functional compounds in apple and pear pomace (APP) by fermentation with mycelia from the mushroom Lentinula edodes. A 30% (w/v) APP with added rice bran and Biji was fermented with L. edodes at $24^{\circ}C$ and 80% humidity. The cellulase and pectinase activities in the fermented APP (FAPP) were higher than those in the non-fermented control. In addition, the physiological activities of the FAPP, including DPPH, ABTS radical scavenging, and SOD-like activity, as well as the total polyphenol and ${\beta}-glucan$ contents were higher than those in the control. FAPP treatment significantly reduced LPS-induced nitric oxide (NO) levels in Raw 264.7 cell. Therefore, FAPP treatment was considered to more effectively suppress cell injury caused by inflammatory cytokines through inhibition of LPS-induced NO production. These results suggest that the levels of functional components in APP can be increased by fermentation with this mushroom mycelium. However, further studies are needed before it can be used as a functional material.

Comparison of Mycelium Cultivation and Fruiting Body Characteristics of Lentinula edodes According to the Sawdust Media Nutrients and Inoculation and Cultivation Conditions (표고 톱밥배지 영양원, 접종 및 배양조건에 따른 균사배양 및 자실체 특성 비교)

  • The cultivation conditions of shiitake mushrooms (Lentinula edodes) influence the production and quality of fruiting bodies. We conducted this study to improve the productivity and quality of shiitake mushrooms by modifying the cultivation conditions. Two types of spawns (sawdust and liquid spawn) were used, and corn flour was used as a nutritional source for the sawdust medium. A blue light-emitting diode (LED; 300 lux) was also used instead of a white LED during the incubation period. Sanbaekhyang was used as the experimental variety. When using corn flour, the mycelial growth rate increased 1.1 to 2.7 times the growth rate of the control up to 21 days of incubation, and the weight loss rate of the media was also higher. Mushroom productivity increased 1.2 times when the liquid spawn was used compared to when the sawdust spawn was used, and the blue LED also increased fruiting body production by 1.1 times compared to the white LED. Mushroom productivity increased when the liquid spawn was used, and the blue LED also increased fruiting body production. Fruiting body weight and the size of the cap were greater when sawdust spawn was used. The fruiting body weight and the stipe diameter were greater when the blue LED was used. Taste analysis showed that the saltiness increased when corn flour was used, and the sourness increased when the blue LED was used.

The Mycelia Isolated from the Basidiocarps of Tricholoma matsutake in Korea (한국 산 송이에서 분리된 균사)

  • Tricholoma matsutake (TM=T. caligatum var. nauseoum) is, for an agricultural income earned by Forestry by-products, a very important mushroom in Korea. The mycelia isolated from the basidiocarps were compared with basidiocarps of TM by the random amplified polymorphisms of RAPD-DNA bands. The mycelia were confirmed to be originated from the basidiocarps of TM by cluster analyses of the DNA-bands made from RAPDs and Southern blotting with the band (0.75 kb) identified. The mycelia defined were observed to grow very slowly at the rate of 10 cm per month at $25^{\circ}C$ and also to be semi-transparent and submerged in on PDA. The method developed in this work was considered to be very useful for confirming the mycelia originated from the ectomycorrhizal mushrooms and also to be applied for the fungal mycelia isolated from the commercial useful mushrooms.
