Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.
The occurrences of the major diseases and the densities of air-born microbes were surveyed in the cultivation facilities for oyster mushroom (Pleurotus ostreatus), king oyster mushroom (Pleurotus eryngii), and enoki mushroom (Flammulina velutipes) in different areas of Korea. Green mold disease was most often developed in oyster mushroom bed cultivation with the disease incidence rate of approximate 10% while the disease incidences from bottle and plastic envelop cultivation were less than 1~2%. In the bed cultivation, the major air-born microbes in the growth room were Aspergillus, Penicillium, Trichoderma, and Curvularia with the total fungal population density of 567~1,297 CFU/$m^3$ . However, only Trichoderma and Penicillium were detected in the growth rooms and innoculation rooms of bottle and plastic envelop cultivation with the densities of 350~700 CFU/$m^3$ and 160~260 CFU/$m^3$, respectively. The bacterial diseases become evident in the growth rooms of bottle and plastic envelop cultivation with the approximate incidence rate of 10%. The identified bacterial species were Brevibacillus levelkil, Rhizobium radiobacter, Brevundimonas vesicularis, Pseudomonas mosselii, Microbacterium testaceum. Sphingomonas panmi, Sphingomonas yabuuchiae, Paracocus dinitrificans, Curtobacterium flaccumfaciens pv. flaccumfaciens and some unidentified bacteria with the densities of 40~6,359 CFU/$m^3$ in the growth rooms and 9 CFU/$m^3$ in the inoculation room. This study indicated that the green mold disease by fungal strains was the major mushroom disease in the bed cultivation and suggested that the contamination of bacteria and fungi together in the growth media could result in severe production loss. The plastic envelope and bottle cultivation were evidenced to be less susceptible to such contaminations.
본 연구에서 생력화가 가능하면서 친환경적인 재배법을 개발하고자 병, 상자를 이용하여 표고의 재배적 특성 및 자실체 생산성을 분석하였다. 재배용기별 배양기간은 병에서 배양된 처리구가 30일, 상자에서의 배양이 35일로 나타났고, 갈변기간은 병 재배가 25일, 상자 재배가 40일이었으며, 병 배양 후 상자재배가 50일로 가장 길었다. 전체 재배기간은 병이 75일, 상자가 90일, 병 배양 후 상자 재배방식이 105일로 가장 길었다. 재배방식별 버섯 발이수는 상자 17.8개, 병 배양후 상자재배 14.7개, 병 1개 순으로, 자실체 생산성에도 영향을 끼쳐 상자재배는 945 g, 병 배양 후 상자재배 638 g, 병재배가 70 g으로 나타났다. 건조 배지량별 신선버섯 수량(BE, %)은 상자재배가 32.7%, 병 재배가 24.3%, 병 배양 후 상자재배가 22.1% 순으로 상자재배의 자실체 수량성이 가장 우수하였다. 따라서 상자재배는 갈변기간의 단축과 버섯 수량성에서 병 재배, 병 배양후 상자재배 방식보다 유리한 장점이 있어 친환경 생력화 재배법 개발 가능성을 보여주었다.
Perforated vinyl mulching technique was performed on oyster mushroom beds for controlling mushroom diseases. Mycelium under vinyl sheets were safely protected from outside undesirable microorganisms. One of two mushroom farms showed 75% of disease incidence, the other 40% and National Institute of Agricultural Science and Technology(NIAST) 13% in the conventional growing method, whereas 12%, 14%, and 5% in the vinyl mulching cultivation method. Waterlogging caused mushroom bed worse, and Trichoderma spp. were infested on the conventional mushroom bed. Disease incidence investigated in other case was 25% to 30% in the conventional growing method, whereas 5 to 9% in the vinyl mulching cultivation method. Yields in conventional method were 6.5 to 7.2 $kg/m^2$ and those in vinyl mulching method were 7.6 to 8.1 $kg/m^2$. So it was suggested that vinyl mulching technique was good for prevention from disease and elevation of productivity.
표고 재배사내 진균류의 발생특성을 분석하기 위해 골목에서 발생한 진균류를 분리·동정하고 재배사내 온·습도 환경을 분석하였다. 표고골목에서 발생한 진균을 분석한 결과 Cladosporium sp., C. cladosporioides, C. anthropophilum, Pleosporales sp., Trichoderma harzianum, Acremonium sp. 6종으로 확인되었다. 재배사내 환경을 조사한 결과는 5월 1일부터 10월 12일 사이에 재배사내 최고·저 온도가 14.0~30.1℃, 평균습도는 82.8%으로 나타났다. 이러한 결과는 표고 재배과정 중 진균류의 발생이 활발한 초여름부터 가을철 사이에 철저한 재배관리가 중요하며, 한번 발병한 진균류는 연중 재배사내 생존이 가능하기 때문에 주기적으로 모니터링하여 사전에 유해 진균류 발생에 유리한 생육환경을 예방해야 한다.
Cotton waste is usually used for cultivating agaric mushroom after outdoor fermentation for a few months. Electron beam was used to break down the polymer chaims of cotton waste for increasing low molecular weight soluble sugars, which may enhance the agaric mushroom cultivation. By increasing electron beam radiation, alpha cellulose content of the cotton waste was decreased while beta cellulose content and hot water solubles were increased. Electron beam radiation over 240 kGy on cotton waste caused significant increase of mushroom yield without lowering mushroom quality.
표고는 활엽수(濶葉樹)의 유익(有益)한 활용방안(活用方案)으로서, 산촌지역(山村地域)의 유일(唯一)한 소득원(所得源)으로서, 건강식품(健康食品)으로서 그리고 수출품목(輸出品目)으로 각광을 받고 있는 임산품(林産品)이다. 그러나 재배규모(栽培規模) 재배기술(栽培技術) 재배관리(栽培管理)등 제측면(諸側面)에서 많은 문제점(問提點)을 안고 있으며 生産水準(생산수준)은 저조(低調)한 실정이다. 따라서 표고생산(生産)의 증수(增收)와 아울러 收支問題(수지문제)를 배려한 합리적(合理的)인 표고재배(栽培) 관리(管理)의 방향(方向) 제시(提示)를 서둘러야 할 시점(時點)에 이르고 있다. 본(本) 연구(硏究)는 표고생산(生産) 과정(過程)의 이같은 당면과제(當面課題) 해결을 위한 방안(方案)으로 표고재배(栽培)의 투자효과(投資效果) 분석(分析)을 시도(試圖)했다. 그 결과(結果) 표고재배(栽培)의 투자수익율(投資收益率)(FRR)은 10.3%로서 농업부문(農業部門)의 투자사업(投資事業)에 비(比)해 비교적(比較的) 높게 나타났다. 또한 표고재배(栽培)의 규모(規模)가 커짐에 따라 투자수익율(投資收益率)은 현저하게 증가하는 경향(傾向)을 보였다. 투자효과(投資效果)의 중요(重要)한 변수(變數)가 되는 임목가(林木價)나 표고가격(價格)을 적정수준(適正水準)에서 안정화(安定化) 시키고 표고생산량(生産量)의 증산(增産) 기술(技術)이 확립(確立)된다면 투자효율(投資效率)은 더욱 증가될 수 있다. 그와 같은 재배(栽培) 여건(與件)의 변화(變化)는 표고생산(生産)에 임하고자 하는 재배자(栽培者)들의 투자의욕(投資意慾)을 고취 할 수 있는 것으로 판단(判斷)된다.
Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.
잣버섯에 적합한 액체종균용 주배지 재료는 대두분, 배양 기간은 12일, 통기량은 0.9 vvm이 적합하였다. 또한, 잣버섯 재배용 톱밥종균용 배지는 미송과 옥분을 부피비로 95 : 5로 혼합하는 것이 적합하였다. 액체종균 및 톱밥종균을 사용하였을 경우 잣버섯 생산성을 비교한 결과 액체종균으로 재배한 경우 재배일수 43일로 톱밥종균으로 재배할 때보다 재배기간 보다 2일 단축되었고, 유효경수 11.3개, 수량 111.9 g로 우수하였다.
느타리버섯의 병재배를 위한 톱밥 대체재료를 선발하기 위해 팽화왕겨, 코코피트, 콘코브, 코코넛나무톱밥을 비트펄프와 면실박에 혼합하여 실험한 결과 화학적 특성은 4가지 배지재료 모두 느타리버섯 재배에 적합하였고, 균사배양 및 재배기간도 지연되지 않았으나, 코코피트 혼합배지에서 수량이 133 g/850ml으로 미송톱밥과 대등하였고 생물학적 효율이 77%로 가장 높아 코코피트를 톱밥 대체 재료로 선발하였다.
