Vasodilation of BCT is Associated with Inhibition of PKC$\alpha$ Translocation and LC20 Phosphorylation

  • We have previously reported that the vasodilatory effect of BanhabackchuIChunma-tang(半夏白朮天麻湯; BCT), a herbal formula, and its mechanism might be associated with at least in part NO pathway. In the present study, we studied the influence of BanhabackchulChenuma-tang (BCT) on the phosphorylation of LC20, in parallel, the distribution of α-protein kinase C(PKCα) by phenylephrine was monitored using laser scan confocal immunofluorescent microscopy in freshly isolated ferret portal vein smooth muscle living single cells. Phenylephrine stimulation induced LC20 phosphorylation and translocation of PKCα. However, BCT dephosphorylated LC20 phosphorylation and inhibited the translocation of PKCα. Our results demonstrate that the mechanism of relaxant effect of BanhabackchulChunma-tang inhibition is associated with inhibition of PKCα activation and LC20 phosphorylation.

The Effects of Leg Raising with Voluntory Lumbo-pelvic Contraction on Abdominal Muscle Activation (허리 골반의 수의적 조절을 병행한 다리 들기가 배근육 활성에 미치는 영향)

  • 연구목적: 본 연구는 허리 골반의 수의적 조절을 동반한 다리 들기 운동이 배 근육에 미치는 영향을 알아보기 위하여 엉덩 관절의 벌림 각도에 따라 다리 들기 시 배근육의 활성도를 확인하였다. 연구방법: 20대 정상성인을 대상으로 엉덩 관절 벌림각도($0^{\circ}$, $30^{\circ}$)에서 압력 센서 사용 그룹과 사용하지 않는 그룹으로 나우어 다리 들기를 실시하는 동안 복부 근육의 근활성도를 측정하였다. 연구결과: 엉덩 관절 벌림 $0^{\circ}$$30^{\circ}$ 모두에서 오른쪽 다리를 들어 올리는 동안 반대측 내복사근과 동측 외복사근에서 수의적 조절을 한 그룹과 하지 않은 그룹 사이에 통계적인 유의한 차이를 보였다(p<.05). 결론: 허리 골반의 수의적 조절을 통한 다리 들기 시 몸통 안정성의 증가를 통하여 안정적인 배 근육 강화 훈련을 수행 할 수 있을 것으로 생각되며 요통 환자에게도 안정적인 임상적용이 기대된다.

Effect of Samhwasan on Na-K-ATPase Activity in Microsomal Fraction of Rabbit Heart Ventricles (삼화산(三和散)이 심장(心臟) Na-K-ATPase 활성(活性)에 미치는 영향(影響))

  • This study was carried out to evaluate the effect of Samhwasan on the Na-K-ATPase activity of heart muscle. The Na-K-ATPase activity was prepared from rabbit heart ventricles. Samhwasan markedly inhibited the Na- K - ATPase activity in a dose-dependent manner with an estimated $I_{50}$ of 0.56%. Hill coefficient was 1.70, indicating that the enzyme has more than one binding site for the Samhwasan. Inhibition of enzyme activity by Samhwasan increased as pretreatment time was prolonged. Inhibition by the drug was not affected by a change in enzyme protein concentration. Kinetic studies of substrate activation of the enzyme indicated classical noncompetitive inhibition, showing significant reduction in Vmax without a change in Km value. Inhibitory effect by Samhwasan was not altered by changes in concentration of $Mg^{2+}$, $Na^+$ or $K^+$, dithiothreitol. a sulfhydryl reducing reagent, did not protect the inhibition of Na-K-ATPase activity by Samhwasan combination of Samhwasan and ouabain showed a cumulative inhibition fashion. These results suggest that Samhwasan inhibits Na-K-ATPase activity of heart ventricles with an unique binding site different from that of ATP, $Mg^{2+}$, $Na^+$ or $K^+$ and ouabain.

Effect of dietary supplementation of grape skin and seeds on liver fibrosis induced by dimethylnitrosamine in rats

  • Grape is one of the most popular and widely cultivated fruits in the world. Although grape skin and seeds are waste product of the winery and grape juice industry, these wastes contain large amounts of phytochemicals such as flavonoids, phenolic acids, and anthocyanidins, which play an important role as chemopreventive and anticancer agents. We evaluated efficacies of grape skin and seeds on hepatic injury induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN significantly increased levels of serum alanine transaminase, aspartate transaminase, alkaline phosphatase, and bilirubin. Diet supplementation with grape skin or seeds (10% daily for 4 weeks) prevented these elevations. The grape skin and seeds also restored serum albumin and total protein levels, and reduced the hepatic level of hydroxyproline and malondialdehyde. Furthermore, grape skin and seeds reduced DMN-induced collagen accumulation, as estimated by histological analysis of liver tissue stained with Sirius red. Grape skin and seeds also reduced hepatic stellate cell activation, as assessed by ${\alpha}$-smooth muscle actin staining. In conclusion, grape skin and seeds exhibited in vivo hepatoprotective and antifibrogenic effects against DMN-induced liver injury, suggesting that grape skin and seeds may be useful in preventing the development of hepatic fibrosis.

EMG Activities of Vastus Muscles are Related to the Shoe-heel Heights in Female Patients with Patellofemoral Pain

  • The aim of this study was to investigate the effect of different shoe-heel heights on the surface electromyographic (EMG) activity of vastus medialis (VM) and vastus lateralis (VL) during treadmill walking in female patients with patellofemoral pain (PFP). Nineteen women with PFP participated in this research. EMG signals were recorded from the VM and VL of both sides and were compared during the treadmill walking. The subjects walked on a treadmill wearing shoes of three different heel heights: 1 cm, 3 cm and 7 cm. Each subject walked on a treadmill for five minutes at a speed of 2 km/hour with three minutes resting intervals between consecutive trials. The data were analyzed by one-way repeated-measures analysis of variance. The results of the present study indicate that EMG data of the VM and VL of female patients with PFP did improve with an increase in the height of the shoe heel, which were statistically significant. Additionally, the EMG activity of VM increased more dramatically than that of VL associated with the task of walking with high-heeled shoes on the treadmill. This study suggests that the type of high-heeled shoes is related to the VM and VL muscle activation patterns contributing to knee joint pathologies in female patients with PFP.

Effects of Shoulder Abduction in Opposite Directions on EMG Activity in the Abdominal Muscles during Single Leg Raising in the Supine Position on the Foam Roller in Healthy Subjects

  • Purpose: The purpose of this study was to examine the electromyographic (EMG) activity of the abdominal muscles and to compare the activity ratios of the bilateral rectus abdominis (RA) to oblique abdominal muscles during shoulder abduction in opposite directions with single leg raising (SLR) performed in the supine position on a foam roller. Methods: Fifteen healthy subjects were recruited to the study. Each subject lay on the foam roller and performed left single leg raising with right or left shoulder $90^{\circ}$ abduction (Abd); performed in a random order. Surface EMG recordings of selected abdominal muscles (i.e., the RA, external oblique abdominis [EO], internal oblique abdominis [IO], and transverse abdominis [TrA]) were normalized to maximum voluntary isometric contraction. EO/RA and IO and TrA/RA ratios were determined with surface EMG. Data were analyzed by Independent t-test. The statistical significance level was p<0.05. Results: The results were as follows: (1) the right RA, left EO, and right IO and TrA muscle activities increased significantly at the left SLR with left Abd compared to the left SLR with right Abd (p<0.05); and (2) the ratio of right EO/RA activity increased significantly at the left SLR with right Abd compared to left Abd (p<0.05). Conclusion: These findings suggest that left SLR with left Abd on a foam roller is an appropriate exercise for activation of specific oblique abdominal muscles.

The Study of Lumbar Erector Spinea and Rectus Abdominis Activations according to the Different Gait Velocities in Young Healthy Adults

  • Purpose: The purpose of this study was to investigate the lumbar erector spinea and rectus abdominis activations, according to the different gait velocities in young healthy adults. Methods: We recruited 6 young male and 10 young female (mean age=21.43 years; range 19~23) in this study. We used a wireless surface electromyogram (Telemyo 2400T G2, Noraxon, USA) and a treadmill unit for the experiment. EMG activity from the lumbar erector spinea, and rectus abdominis of the dominant side was record with surface electrodes. On different day, all subjects gaited on 2.7 km/h, 4.5 km/h, and 6.3 km/h of speed in random order. They gaited at the same velocity, three times, on the treadmill unit. To reduce fatigue, sufficient rests were given between the measurements. Results: As the gait speed increased, lumbar erector spinea and rectus abdominis activations were significantly increased (p<0.05). Conclusion: In the current study, we found lumbar erector spinea and rectus abdominis activations were changed, according to the gait velocity. We suggested that rehabilitation intervention should be focused on the exercise velocity for the patients with problem of the trunk control.

Effect of McKenzie Exercise on Temporomandibular Joint Disorder and Body Balance (맥켄지운동이 턱관절 장애 및 신체균형에 미치는 효과)

  • Purpose : This study was aimed at investgating the effect of McKenzie exercise on temporomandibular joint disorder (TMJD) and body balance. Methods : Thirty subjects with TMJD were selected. They were randomly assigned to one of two groups (15 in each group) : namely the conservative treatment group and McKenzie exercise group. The conservative treatment group were treated physical therapy for 6 weeks (3 times a week). The McKenzie exercise group performed McKenzie exercise for 6 weeks (3 times a week). We measured mouth opening, pain, muscle activation and body balance before and after exercise by using the maximum mouth opening MMO), visual analog scale (VAS), electromyography and Biodex Balance System, respectively. Results : We found statistically significant differences in pain and mouth opening in the conservative treatment group and McKenzie exercise group before and after (p<.05). Conclusion : We confirmed the effect of McKenzie exercise on TMJD. Thus we thought these results could be used as basic data and reference for TMJD. But we need more study effect of McKenzie exercise on TMJD and body balance.

Signaling Mechanisms on the Vascular Relaxation of HMC05 (HMC05의 혈관이완 활성과 신호전달 작용기전)

  • This study investigated the signaling mechanisms contributed to the vasodilatory effects of HMC05, a herbal prescription. HMC05 acted in an endothelium-independent manner. To elucidate the fundamental mechanisms of its vascular actions, we focused on the signaling molecules involved in actin-myosin filament regulation including 20 kDa myosin light chains (LC20), Rho-associated kinase (ROCK), PKC, JNK and extracellular signal-regulated protein kinase (ERK) in the endothelium-denuded thoracic aorta or isolated smooth muscle cells (SMCs). It lowered the phosphorylation level of LC20 and showed that ROCK, ERK, JNK and $PKC{\alpha}$ pathways played important roles in the effects, as confirmed by the observations with a specific inhibition or activation, and with the activity and the subcellular localization of these molecules. In particular, HMC05 dramatically inhibited the activity of ERK and the downstream signaling of ROCK. It also changed the subcellular localization of the phophorylated $PKC{\alpha}$ as well as the amount of phosphorylation. Taken together, these data indicate that the vascular relaxation effects of HMC05 are attributed to the regulation of these signaling mechanisms.

Vasorelaxant and hypotensive effects of trazodone in Guinea pig (기니픽에서 trazodone의 혈관 이완 및 혈압 하강 효과)

  • We studied the effects of trazodone on arterial blood pressure in anesthesized guinea pigs, and on vascular responses in isolated thoracic aorta. Trazodone produced a concentration-dependent relaxation in phenylephrine-precontracted endothelium intact (+E) rings, but not in a KCl-precontracted aortic rings. These relaxant effects of trazodone on +E rings were significantly greater than those on denuded (-E) rings. The trazodone-induced relaxation was suppressed by glibenclamide and tetrabutylammonium, but not by N(G)-nitro-L-arginine (L-NNA), N(omega)-nitro-L-arginine methyl ester (L-NAME), methylene blue (MB), nifedipine, indomethacin, 2-nitro-4-carboxyphenyl-n,n-diphenylcarbamate (NCDC) and clotrimazole. In vivo, infusion of trazodone elicited a significant decrease in arterial blood pressure. Trazodone-induced blood pressure lowering was markedly inhibited by intravenous pretreatment of prazosin but not by pretreatment of saponin, L-NNA, L-NAME, MB, nifedipine, glibenclamide, clotrimazole and NCDC. In addition, trazodone produced an increase in twitch force of isolated papillary muscle and left ventricular pressure of perfused heart. These findings suggest that the endothelium-independent vasorelaxant effect of trazodone may be explained by activation of $Ca^{2+}$-activated and ATP-sensitive $K^+$ channels, and the hypotensive effect of trazodone is not associated with cardiac contraction.
