Multi-locus Phylogeny Analysis of Korean Isolates of Phytophthora Species Based on Sequence of Ribosomal and Mitochondrial DNA (핵 및 미토콘드리아 DNA 염기서열을 이용한 국내 Phytophthora 속의 Multi-locus phylogeny 분석)

  • To investigate genetic relationships either interspecies or intraspecies of 14 Korean Phytophthora species, sequence analyses of nuclear DNA (ypt gene and rDNA-IGS region) and mitochondrial DNA (Cox gene, $\beta$-tubuline gene, and EF1A gene) were performed. All of 14 Korean Phytophthora species clearly clustered into foreign isolates of each species. These Korean isolates in Phytophthora species also showed no correlation between molecular classification and morphological classification like as in case of foreigners. P. palmivora KACC 40167 reported previously from genetic groups of Phytophthora species in Korea was not consistent with the classification system, and therefore was required re-examination for the genetic group analysis. Korean isolates of P. drechsleri KACC 40195 showed very close relationship with P. cryptogea KACC 40161 above 94% bootstrap value in P. cryptogea-P. drechsleri complex group. Identification of these isolates is still unclear, because P. cryptogea and P. drechsleri were not differentiated in this study. On the other hand, it was required to unify species for these two species, since P. parasitica and P. nicotianae were clustered into a group on the level of 99 to 100% sequence homology. Comparing to the sequences of foreigners, Korean isolates were newly divided to ten groups in the phylogenic system. These results could be prepared useful informations to understand genetic diversity of Phytophthora species in Korea.

SELECTIVITY OF DRIET NET FOR SPANISH MACKEREL SCOMBEROMORUS NIPHONIUS (삼치 유자망 어구의 선택성에 관하여)

  • During the period from 1966 to 1968, total catches of Spanish mackerel averaged 6,000 to 7,000M/T per annual in Korea, and approximately 70 per cent of this amount was captured by drift nets. In an effort to improve the efficiency of drift nets, some experiments were conducted in 1969 to investigate the selectivity of material and mesh sizes. Seven different mesh sizes (80,85,95,100,105,110 and 115 mm) of both multi- and mono-filament netting were used, and the following results were obtained : 1, The body weight of Spanish mackerel taken with the seven different mesh sizes ranges from 0.5kg to 2.9kg, and the mode of body weight consists of three groups, 1kg ($21%$), 1.3kg($15\%$) and 1.5kg($19\%$). 2. For multi-filament net, 80 to 105mm mesh sizes were suitable to catch those three groups, and a little smalter than these for mono-filament net. 3. For Spanish mackerel only, the mono-filament material proved to have 1.5 times better selectivity than multi-filament : however, the latter proved superior for miscellaneous fish species due to its different size and shape. 4. Multi-filament net showed better selectivity for smaller species than mono-filament. (and mono-filament in general indicated opposite phenomenon.)

Age-related Changes in Plasma Leptin from Early Growing to Late Finishing Stages of Castrated Holstein Steers: Utilizing Multi-species Leptin RIA

  • This experiment was performed to understand the changes in plasma leptin in association with plasma IGF-1, body weight and ADG from early growing to late finishing stages of Holstein steers. Blood collection was performed by arterial vein puncture at selected monthly ages of 1 (54 kg), 2.6 (103 kg), 7.2 (205 kg), 13.5 (314 kg), 16.9 (414 kg), 22.2 (550 kg), 24.9 (626 kg) and 27.4 months (695 kg). The blood was analyzed for leptin using the multi-species leptin RIA with recombinant bovine leptin (rbleptin) as standard, plasma IGF-1 was also measured using RIA. Against the standard rbleptin, the multi-species Leptin RIA system's sensitivity, cross reactivity, slope and recovery of 41.0 ng/ml rbleptin in plasma were 4.9 ng/ml, 11.22%, -1.396 and 97.8%, respectively. Plasma leptin measured were more than 5.0 ng/ml, which enable multi-species RIA system to investigate plasma leptin in normal growing steers. Body weight resulted to a highly significant second-degree polynomial relationship with plasma leptin (q=0.54, p<0.0001) and plasma IGF-1 (q=0.44, p<0.0001) from 1 to 27.4 monthly ages. However, the second-degree polynomial curve of plasma leptin and IGF-1 differs showing a concave and convex curvilinear relationship, respectively. ADG was not significantly associated to plasma leptin (r=0.06, p>0.05) and plasma IGF=1 (r=0.06, p>0.05) from 1 to 27.4 monthly ages. Low coefficient, but significant associated increase of plasma leptin and IGF-1 (r=0.12, p<0.008) from 1 to 27.4 months was observed. The uncoordinated increases of plasma IGF-1 at growing and plasma leptin at fattening period, may indicate (1) indirect involvement of endogenous IGF-1 on leptin secretion, and (2) IGF-1 level may signify lean and bone accretion while plasma leptin may mirror body fatness across the monthly ages of Holstein steers.

Confirmation of Two Undescribed Fungal Species from Dokdo of Korea Based on Current Classification System Using Multi Loci

  • Using dilution plating method, 47 fungal isolates were obtained from a soil sample collected from Dokdo in the East Sea of Korea in 2013. In this study, two fungal isolates, EML-MFS30-1 and EML-DDSF4, were confirmed as undescribed species, Metarhizium guizhouense and Mortierella oligospora in Korea based on current classification system using multi loci including rDNA internal transcribed spacer, large subunit, small subunit, and ${\beta}$-tubulin (BTUB) genes. Herein, detailed morphological descriptions on characters of the undescribed fungal species as well as their molecular phylogenetic status are provided with comparisons to related species.

  • CFD analysis has been conducted to find the two stage impeller configuration which is the most suitable for a stirred tank with an internal helical cooling coil and a cooling jacket, which is frequently used in chemical industries for highly exothermic reactions ranged from low to medium viscosity. Two typical types of impellers are considered; pitched paddle impellers and Rushton turbine impellers. Interestingly, pitched paddle impellers show a good mixing performance for multi-species, whereas Rushton turbine impellers achieve a good mixing performance for multi-phases. Besides the type of an impeller, the location of an impeller is another important factor to be considered in order to accomplish an effective mixing. The best set of types and locations of two impellers is recommended based on the coefficient of variation(CoV) value and the heat removal capability obtained from CFD results. The former is a measure to quantify the degree of mixing.

Characteristics of Wildbirds Habitat of Artificial Green Corridor in Gangseo-gu, Seoul (서울시 강서구 조성녹지축의 야생조류 서식처 특성 연구)

  • This study was to examine the characteristics of wirdbirds habitat for improvement plan in green corridor. The target site, Gangseo-gu artificial green corridor was set up with the structure in which small scale of core green space with Goongsan and Yeomchang neighborhood parks in urbanized city was connected with the artificial green space with Gongamnaru, Hwanggeumnae neighborhood parks with 28~42.5 m in width. Wild birds six~eleven species; Dendrocopos spp, Paradoxornis webbiana, Parus major, Phasianus colchicus, etc. were observed in core green, but wild birds of two~five species: Columba livuia, Passer montanus, Pica pica, Hypsipetes amaurotis, etc. were observed in artificial green space. Thus wild birds of artificial and generalist species only moved in artificial green space. The artificial green space where vegetation structure was consisted of single-layer with poorness chose target species laying stress on generalist species and edge species of Parus major, P. palustris, Paradoxornis webbiana etc. for short-term and interior species of Dendrocopos major, Picus canus, etc. for long-term. The result suggested enhancement methods for target species's habitat in green corridor: to secure at least a corridor 30 meters in artificial corridor, to secure ecological pond, to offer the various shelterer and environment of prey-resources through the multi-layer structure.

  • This paper presents a multi-scale hybrid simulation for the design of a catalytic multi-tubular reactor with high performance. The multi-tubular reactor consists of shell and a large number of tubes in which various catalytic chemical reactions occur. To consider fluid dynamics in the shell-side and kinetics in the tube-side at the same time, commercial CFD package and process simulation tool are coupled. This hybrid approach allowed us to predict many kinds of meaningful results such as tube center temperature profile, heat transfer coefficients on the tube wall, temperature rise of cooling medium, pressure drop through shell and tube side, concentration profile of each chemical species along the tube, and so on., and to achieve the optimal reactor design.

The influence of chemical water quality on fish trophic guilds, pollution tolerance, and multi-metric ecological health in the main streams of Mangyeong River (만경강 본류의 어류 트로픽 길드, 오염 내성도 및 다변수 생태건강도에 대한 화학적 수질영향)

  • The objectives of this study were to analyze the influence of chemical water quality on fish guilds, pollution tolerance and the multi-metric ecological health, based on the Fish Assessment Index (FAI) in the main stream of Mangyeong River between 2009-2016. The quality of water with specific conductivity, TP, and $NH_4-N$ got worse dramatically in the down region. During the study, a total of 50 species were collected and the most dominant species was Zacco platypus. Also known as tolerant species, accounted 22.9% of the total abundances, thus indicating a trophic degradation. The downstream region (S5) had the highest number of fish external abnormalities, indicating a degradation of ecological health, based on the fish assemblages. Pearson correlation analysis indicated that relative abundance of tolerant fish species and omnivore fish species had a significant positive correlation(r>0.30, p<0.05) with values of BOD, conductivity and $NH_4-N$. Whereas, the relative abundance of the sensitive species and insectivore species had a significant negative relations (r<-0.30, p<0.001) with the parameters. The mean obtained from the multi-metric fish model, based on the FAI of all sites was 47 (n=40). This indicated a "fair condition" in the ecological health, and the downstream regions (S3-S5) were judged as "bad condition", indicating an influence of the chemical degradation on the ecological health.

Zooplankton Biomass and Size Estimation Using a Multi-frequency Acoustic System (고주파 다주파 음향시스템을 이용한 동물성 플랑크톤의 크기별 생물량 추정)

  • High- and multi-frequency acoustic systems can measure a zooplankton patch successively and estimate the spatial distribution and abundance of zooplankton according to size using a multi-frequency inversion (MFI) method. This study measured zooplankton distribution to a depth of 150m using a multi-frequency acoustic system (TAPS-6), installed on a CTD system with a fluorometer and analyzed it using the MFI method. Simultaneously, zooplankton samples were collected by north pacific standard (NORPAC) net to confirm the species composition. The results showed that the combined method is valuable for estimating the zooplankton profile in detail and investigating the relationship between the zooplankton and phytoplankton profiles.
