언어습득기 이전 청각장애인의 후두소견 및 음성학적 특성 (Laryngeal Findings and Phonetic Characteristics in Prelingually Deaf Patients)

  • Background and Objectives : There are few studies reported that specifically examine the laryngeal function in patients with profound hearing loss or deafness, This study was designed to examine videostroboscopic findings and phonetic characteristics in adult patients with prelingually deaf. Materials and Method: Sixteen patients (seven males, nine females) diagnosed as prelingually deaf aged from 19 to 54 years, and were compared with a 20 normal control group with no laryngeal pathology and normal hearing group, Videostroboscopic evaluations were rated by experienced judges on various parameters describing the structure and function of the laryngeal mechanism during comfortable pitch and loudness phonations. Acoustic analysis test were done, and a nasalance test performed to measure rabbit, baby, and mother passage. CSL were measured to determine the first and two formant frequencies of vowels /a/, /i/, /u/, Statistical analysis was done using Mann-Whitney U or Wilcoxon signed ranks test. Results: Videostroboscopic findings showed phase symmetry but significantly more occurrences decrement in the amplitude of vibration, mucosal wave, irregularity of the vibration and increased glottal gap size during the closed phase of phonation, In addition, group of prelingually deaf patients were observed to have significantly more occurrences of abnormal supraglottic activities during phonation. The percentage of shimmer in the group of prelingually deaf patients were higher than in the control group. Characteristics of vowels were lower of the second formant of the vowel /i/. Nasalance in prelingually deaf patients showed normal nasality for all passages, Conclusion: Prelingually deaf patients show stroboscopic abnormal findings without any mucosal lesion, suggesting that they have considerable functional voice disorder. We suggest that prelingually deaf adults should perform vocal training for normalized laryngeal function after cochlear implantation.

  • Background: Lung cancer is usually diagnosed at an advanced stage, resulting in a poor prognosis. The detection of these lesions at an earlier stage would be a clear benefit to patients. However, it is extremely difficult to detect carcinomatous lesions in the bronchial mucosal sites during a routine bronchoscopy. Methods: This study employed a novel optical technique, known as narrowband imaging (NBI), which allows noninvasive visualization of the microvascular structure of an organ's surface using reflected light. Results: Narrow band imaging was performed on 10 patients who were radiologically suspicious or had a high risk of lung cancer. The median age of the patients was 57.5 years (range, 44~81 years), and 80% of the patients were male. All lesions showed a microvascular proliferation pattern (dotted, tortuous and abruptly ending vessel) on the magnified NBI. Two lesions were confirmed histologically to be adenocarcinoma and the remaining lesions were squamous cell carcinomas. Two lesions were confirmed histologically to be a carcinoma in situ. Conclusion: NBI is a promising and potentially powerful tool for identifying carcinomas at an earlier stage or a central lesion during a routine bronchoscopy examination.

Characterization of Surface Layer Proteins in Lactobacillus crispatus Isolate ZJ001

  • Lactobacillus crispatus (L. crispatus) ZJ001 is highly adhesive to epithelial cells and expresses S-layer proteins. In this study, S-S-layer layer genes were sequenced and expressed in E. coli to characterize the function of proteins with this particular strain. L. crispatus ZJ001 harbored two S-layer genes slpA and slpB, and only slpA gene was expressed in the bacterium, as revealed by RT-PCR and immunoassays. The mature SlpA showed 47% amino acid sequence identity to SlpB. The SlpA and SlpB of L. crispatus ZJ001 were highly homologous at the C-terminal region to other Lactobacillus S-layer proteins, but were substantially variable at N-terminal and middle regions. Electron microscopic analysis indicated that His-slpA expressed in E. coli was able to form a sheet-like structure similar to the natural S-layer, but His-slpB formed as disc-like structures. In the cell binding experiments, HeLa cells were able to bind to both recombinant His-slpA and His-slpB proteins to the extent similar to the natural S-layer. The cell binding domains remain mostly in the N-terminal regions in SlpA and SlpB, as shown by high binding of truncated peptides SlpA2-228 and SlpB2-249. Our results indicated that SlpA was active and high binding to HeLa cells, and that the slpA gene could be targeted to display foreign proteins on the bacterial surface of ZJ001 as a potential mucosal vaccine vector.

  • This study describes the cell type, ultrastructure and histochemical characteristics as a preliminary study for the research on integument of the smooth lumpsucker, Aptocyclus ventricosus in accordance with the physiological and environmental changes using light and electron microscopes. The SEM revealed the presence of well-developed finger printing structure in the skin surface. The skin surface of the smooth lumpsucker showed an irregular folds in cross section of light microscope. Integument is composed of outer epidermal and inner dermal layer. The epidermal layer is a stratified layer composed of epithelial cells, mucous cells, vacuolar cells, and granular cells. Epithelial cells are classified into superficial, intermediated, and basal cell. The superficial cells were the squamous with well-developed microridges on the free surface, and the microridges were covered with glycocalyx. The mucous cells of unicellular gland were mainly distributed in the apical layer of epidermis and contained mucosal materials of neutral glycoprotein. The vacuolar cells of unicellular gland were mainly distributed in the mid and basal layer of epidermis. The proportion of mucous cells and vacuolar cells were $7.0({\pm}1.07)%$ and $40.6({\pm}3.31)%$ of epidermal area, respectively. The granular cells contained membrane bounded secretory granules with high electron density and developed cell organelles in the cytoplasm. The dermal layer was loose connective tissue layer and composed of mainly collagen fibers. It also contained blood vessels and chromatophores of melanophores and reflecting platelets.

Preventive Effects of Spirogyra neglecta and a Polysaccharide Extract against Dextran Sodium Sulfate Induced Colitis in Mice

  • Ulcerative colitis (UC) results from colonic epithelial barrier defects and impaired mucosal immune responses. In this study, we aimed to investigate the modifying effects of a Spirogyra neglecta extract (SNE), a polysaccharide extract (PE) and a chloroform fraction (CF) on dextran sodium sulfate (DSS)-induced colitis in mice and to determine the mechanisms. To induce colitis, ICR mice received 3% DSS in their drinking water for 7 days. Seven days preceding the DSS treatment, oral administration of SNE, PE and CF at doses of 50, 25 and 0.25 mg/kg body weight (low dose), 200, 100 and 1 mg/kg body weight (high dose) and vehicle was started and continued for 14 days. Histologic findings showed that DSS-induced damage of colonic epithelial structure and inflammation was attenuated in mice pre-treated with SNE, PE and CF. Furthermore, SNE and PE significantly protected colonic epithelial cells from DSS-induced cell cycle arrest, while SNE, PE and CF significantly diminished apoptosis. Proteome analysis demonstrated that SNE and PE might ameliorate DSS-induced colitis by inducing antioxidant enzymes, restoring impaired mitochondria function, and regulating inflammatory cytokines, proliferation and apoptosis. These results suggest that SNE and PE could prevent DSS-induced colitis in ICR mice by protection against and/or aiding recovery from damage to the colonic epithelium, reducing ROS and maintaining normal mitochondrial function and apoptosis.

Development of a Novel Subunit Vaccine Targeting Fusobacterium nucleatum FomA Porin Based on In Silico Analysis

  • Selecting an appropriate antigen with optimal immunogenicity and physicochemical properties is a pivotal factor to develop a protein based subunit vaccine. Despite rapid progress in modern molecular cloning and recombinant protein technology, there remains a huge challenge for purifying and using protein antigens rich in hydrophobic domains, such as membrane associated proteins. To overcome current limitations using hydrophobic proteins as vaccine antigens, we adopted in silico analyses which included bioinformatic prediction and sequence-based protein 3D structure modeling, to develop a novel periodontitis subunit vaccine against the outer membrane protein FomA of Fusobacterium nucleatum. To generate an optimal antigen candidate, we predicted hydrophilicity and B cell epitope parameter by querying to web-based databases, and designed a truncated FomA (tFomA) candidate with better solubility and preserved B cell epitopes. The truncated recombinant protein was engineered to expose epitopes on the surface through simulating amino acid sequence-based 3D folding in aqueous environment. The recombinant tFomA was further expressed and purified, and its immunological properties were evaluated. In the mice intranasal vaccination study, tFomA significantly induced antigen-specific IgG and sIgA responses in both systemic and oral-mucosal compartments, respectively. Our results testify that intelligent in silico designing of antigens provide amenable vaccine epitopes from hard-to-manufacture hydrophobic domain rich microbial antigens.

  • 관강내 십이지장 게실은 드문 선천성 이상으로, 십이지장 점막으로 구성된 낭성 병변이며, 위장 출혈, 십이지장 폐색 또는 췌장염과 함께 나타날 수 있다. 이 논문에서는 25세 성인 여성에서 십이지장-십이지장 장중첩증과 재발성 췌장염의 합병증을 일으킨 드문 관강내 십이지장 게실의 사례를 보고하는 바이다. CT, MRI, 상부위장관조영술등의 영상 검사에서 십이지장의 중복낭을 의심하였으나, 수술로 절제된 조직에서 병리학적으로 고유근육층이 없는 관강내 십이지장 게실로 확진되었다.

  • Turtle bladder 상피세포(上皮細胞)의 수송기작(輸送機作)을 in vitro에서 효과적으로 연구하기 위하여 Lucite chamber 한가운데 상피조직을 두고 전압고정법(電壓固定法)을 적용하여 상피 세포층의 막전위(膜電位)를 측정한 후 급속 동결(凍結)하고 투과 및 주사형 전자현미경(電子顯微鏡)으로 탄산 탈수효소를 함유하는 세포의 표면막 특성을 분석(分析)하였다. 방광(膀胱)의 점막층(粘膜層)은 두 타입의 탄산탈수효소를 함유한 세포가 특징적인데 정단부(丁端部)와 기저부(基底部) 세포막에서는 각기 다른 수송의 특성을 나타내고 있다. 즉 ${\alpha}$${\beta}$형 탄산탈수효소가 풍부한 세포는 정단세포막(丁端細胞膜)의 proton 펌프를 이용하여 $H^+$ 분필(分泌)에 관여하거나 bicarbonate의 재흡수(再吸收) 기능을 가지는 것으로 믿어진다. 본 연구에서 탄산탈수효소를 함유한 ${\alpha}$형의 세포의 proton 분필수송(分泌輸送)과 세포막 투과성 변화와의 상관관계를 관찰하였는 바, 이들 세포에서 $H^+$을 분비하는 과정에서 정단부의 표면세포막(表面細胞膜) P-face에는 특이한 구조로서 세포막내(細胞膜內) 입자(粒子)들이 다량으로 분포하였다. 이와같은 세포막내(細胞膜內) 입자(粒子)들은 proton 펌프를 함유하는 것으로 생각되며 ${\beta}$형의 세포에서는 기저세포막(基底細胞膜)에서 관찰되고 있다. 이와같은 결과는 방광상피(膀胱上皮) 세포내 탄산탈수효소는 $H^+$$HCO_{3}^{-}$의 생성에 관여하지만, 특히 ${\alpha}$형 세포에서 정단세포막의 proton 펌프를 이용한 $H^{+}$ 분필수송(分泌輸送)과 기저세포막을 통한 bicarbonate의 재흡수(再吸收) 기능을 설명해 주는 중요한 사실로서 사료된다.

Di-(2-ethylhexyl)phthalate (DEHP)가 흰쥐 저정낭의 미세구조에 미치는 영향 (Effects of Di-(2-ethylhexyl) phthalate (DEHP) on Ultrastructure of Rat Seminal Vesicle)

  • 플라스틱 제품의 가소제 성분으로 쓰여지며, 내분비 교란물질로 알려져 있는 di-(2-ethylhexyl)phthalate (DEHP)가 흰쥐 부속생식선의 발달에 미치는 영향을 알아보기 위하여, 미성숙한 상태의 웅성 흰쥐에 DEHP(1g/kg/day, 2g/kg/day, 3g/kg/day)를 15일간 구강으로 삽관 투여한 후, 저정낭 상피세포들을 중심으로 광학 및 전자 현미경적 관찰을 실시하였다. DEHP 투여에 의한 광학현미경적 변화로서는 분비선의 크기와 상피층의 두께가 감소하고, 강소 주변부위에서 교원섬유를 비롯한 결합조직 성분들이 증가하는 경향을 나타냈다. 저정낭 분비세포들의 미세구조를 보면, 원주형의 대조군 세포들내 전자밀도가 높은 분비과립들은 소포내 가장자리에 치우쳐 있었고, 세포질내에서 조면소포체와 골지복합체가 잘 발달하였으며, 다수의 미세융모들이 존재하고 있었다. DEHP 1g을 처리한 실험군에서 분비세포들의 미세구조상의 뚜렷한 변화는 관찰되지 않았으나, 미세융모와 분비소포의 분포가 다소 감소한 것으로 나타났다. 2g을 처리한 실험군에서는 분비세포의 세포질이 감소하여 세포의 형태가 원주형에서 입방형으로 변화되었으며, 핵막이 불규칙한 모습을 보였고, 조면소포체의 발달이 미흡하였으며, 분비소포의 수도 크게 감소하였다. 3g을 처리한 실험군에서는 세포소기관들의 미세구조가 뚜렷한 변화를 나타냈다. 핵내 이질염색질이 크게 증가하였고, 세포질내에서 분비소포를 거의 관찰할 수 없었으며, 2차 리소솜이나 공포들도 관찰할 수 있었다. 이상과 같이, DEHP는 저정낭내 분비세포들의 분화 및 기능을 농도 의존적으로 억제하는 것으로 나타났으며, 이러한 저해작용은 DEHP에 의한 테스토스테론의 농도 감소와 연관이 있는 것으로 판단된다.

양식산 돌돔, Oplegnathus fasciatus 소화관의 형태학적 구조 및 조직화학적 특징 (The Morphological Structure and Histochemical Features of the Alimentary Tract in Parrot fish, Oplegnathus fasciatus)

  • 돌돔, Oplegnathus fasciatus의 소화관은 식도, 위, 장 및 항문이 일련의 관을 이루고 있고,전체적으로 지방 덩어리가 싸고 있다. 어체 체장에 대한 식도에서 항문까지의 소화관 길이의 비(relative length of gut: RLG)는 일반적인 잡식성 어류와 비슷한 평균 1.78 (n=30)의 값을 가진다. 식도는 근육층이 잘 발달되어 있으며, 점막주름의 상피층은 입방상피 또는 원주상피로 구성되고 상피세포 사이에는 다수의 산성 점액분비세포들이 분포하며, 상피세포의 정단부에서 미세융모의 관찰은 어렵다. 위의 근육층은 외부로부터 종주근, 환상근층으로 구성된다. 위의 점막주름은 규칙적인 형태로서 상피층에는 pit, 상피 및 위선이 아주 잘 발달되어 있으며, 점막상피 세포질의 정단부에서는 영양분 흡수계통의 물질들이 관찰된다. 50∼60개의 유문수를 가지는 돌돔 유문수의 점막상피층에서는 원주형의 상피세포와 중성의 점액분비세포들이 관찰되며, 상피세포의 정단면에는 미세융모가 발달되어 있다. 장의 점막상피층에서는 원주상피세포와 중성과 산성의 점액분비세포들이 관찰되며, 종주근층의 두께가 전장, 중장, 후장으로 갈수록 두꺼워진다. 산성의 점액분비세포의 출현빈도는 중장부에서 가장 발달된 양상을 보이며 후장부로 갈수록 감소하는 경향을 보인다.
