Type IV phosphodiesterase inhibitor(CJ-10882)의 개에 대한 2주간 경구반복투여 독성시험 (Toxicity Study of CJ-10882, a Type IV Phosphodiesterase Inhibitor: 2 Weeks Repeated Oral Administration in Beagle Dogs)

  • CJ-10882, (E)-[(3-Cyclopentyloxy-4-methoxyphenyl)methylene]hydrazine-carboxamide, is a newly developed type IV phosphodiesterase isozyme (PDE IV) inhibitor. To investigate the subacute toxic effects of CJ-10882, it was administered to both male and female dogs at 0, 25, 50, 100 or 200 mg/kg/day orally for up to 2 weeks. During the test period, clinical signs, mortality, body weight, food consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, gross finding, organ weight, and histopathology were evacuated. Several clinical signs were observed in treated dogs at above 25 mg/kg, including salivation and vomiting. A reduction in the body weight was observed in both sexes at above 50 mg/kg. There were no treatment-related effects on mortality, ophthalmoscopy, urinalysis, hematology, sect biochemistry, necropsy findings, and histopathology in any treatment group. The results of this study demonstrate that CJ-10882, a selective Inhibitor of the type IV class of PDE, may cause effects on gastrointestinal tract and salivary glands. Therefore, these organs should be closely examined in studies with other PDE IV inhibitors.

Single Oral Dose Toxicity Test of Water Extracts of Radix Araliae Cordatae in ICR Mice

  • The object of this study was to evaluate the acute toxicity of lyophilized water extract of Radix Araliae Cordatae (RA) in male and female mice. The extract was administered to female and male ICR mice as an oral dose of 2000 mg/kg (body wt.) according to the recommendation of KFDA Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy, organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, changes in the body weight and gross findings except for increases of hypertrophy of lymph nodes in male RA extracts-dosing group. In addition, no RA extracts-treatment related abnormal changes in the organ weight and histopathology of principle organs except for some sporadic accidental findings. The results obtained in this study suggest that the RA extracts does not cause any toxicological signs. The LD$_{50}$ and approximate LD of RA extracts in both female and male mice were considered as over 2000 mg/kg.

마우스를 이용한 십전대보탕(十全大補湯) 발효물의 단회투여 독성 연구 (Single Oral Dose Toxicity Test of Fermented Sipjeondaebo-tang Extracts in Mice)

  • Objectives : Sipjeondaebo-tang is a medicine traditionally prescribed as a restorative. The aim of this study was to investigate the single oral dose toxicity and safety of extract of fermented Sipjeondaebo-tang in ICR mice. Methods : In single oral dose toxicity study, non-fermented or fermented Sipjeondaebo-tang were administered by oral gavage to ICR mice (5 males, 5 females) at single doses of varying concentrations: 1250, 2500 and 5000 mg/kg. Changes of body weight, general behavior, adverse effects and mortality were determined throughout the experimental period. Hematological parameters, organ weights and necropsy findings were evaluated at the end of the experiment. Results : There were no mortality or signs of toxicity in single oral dose toxicity studies. There were also no significant differences in body weight, organ weight, or hematological parameters between the treatment and control groups. Conclusions : Fermented Sipjeondaebo-tang did not cause remarkable adverse effects in ICR mice. The oral lethal dose of fermented Sipjeondaebo-tang is more than 5000 mg/kg and no-observed-adverse-effect level (NOAEL) of the extract for both male and female mice is 5000 mg/kg.

A Study on Subchronic Inhalation Toxicity of 1-Chloropropane

  • This study was conducted to measure toxicity of 1-chloropropane (CAS No. : 540-54-5). According to the OECD Test Guideline 413 (Subchronic inhalation toxicity: 90-day study), SD rats were exposed to 0, 310, 1,250, and 5,000 ppm of 1-chloropropane for 6 h/day, 5 day/week for 13 weeks via whole-body inhalation. Mortality, clinical signs, body weights, food consumption, motor activity, ophthalmoscopy, hematology, serum chemistry, urinalysis, organ weights, gross and histopathological findings were compared between control and all tested groups. No mortality or remarkable clinical signs were examined during the study. No gross lesions or adverse effects on body weight, food consumption, motor activity, ophthalmoscopy, urinalysis, hematology, organ weights were observed in any of male or female rats in all tested groups. In serum biochemistry, glucose was significantly decreased in males of 1,250 and 5,000 ppm groups compared to control group in dose-dependent relationship. In histopathological examination, vacuolation of acinar cells was observed in pancreas of all male and female groups exposed to 1-chloropropane. In conclusion, no observable adverse effect level (NOAEL) was considered to be below 310 ppm/6 h/day, 5 day/week for rats.

Nematicidal Activity of Some Fluorescent Pseudomonads on Cyst Forming Nematode, Heterodera cajani and Growth of Sesamum indicum var. RT1

  • Among 24 isolates of fluorescent Pseudomonads, 5 isolates named as LPT1, LPT2, LPT3, LPT4 and LPT5 were screened in vitro for their nematicidal activity against cyst forming nematode, Heterodera cajani causing patchiness, poor and stunting growth besides discoloration in Sesamum indicum. Second stage juveniles of H. cajani hatched from egg masses were collected from roots of host plant and subjected to fresh and heat-treated culture filtrate of isolates for 24 h. Mortality of H. cajani was recorded on the basis of parameters used for test organism bioassay. Among these isolates, Pseudomonas aeruginosa LPT5 caused maximum mortality towards second stage juvenile of H. cajani in vitro. Five isolates were used as seed coating for the management of cyst forming nematode H. cajani on sesame in green house condition. The strains LPT5 was better than the other strains in reducing the population of H. cajani both in vitro and in vivo. The reduction in cyst and juveniles population was found to be 49 and 60%, respectively when seeds were coated with strain LPT5. Among other strains, LPT4 was also found to inhibit the cyst and juveniles population 12 and 36% respectively. Increases in early vegetative plant growth parameters recorded in both in vitro and in vivo further revealed the significance of indigenous bacteria in comparison to introduced strain.

보중익기탕(補中益氣湯)의 마우스 경구 단회 투여 독성시험 (Single Oral Dose Toxicity Test of Bojungikkitang (Buzhongyiqi-tang) Aqueous Extract in ICR Mice)

Method : Aqueous extracts of BJIKT were administered to female and male ICR mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy ; organ weight and histopathology of 12 principle organs were examined. Results : we could not find any mortality, clinical signs, and changes in the body and organ weight. In addition, no BJIKT-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. Conclusion : The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of BJIKT aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines, and can be safety used in clinics.

육미지황탕가미방(六味地黃湯加味方)의 마우스 경구 단회 투여 독성 평가 (Single Oral Dose Toxicity Test of Yukmijihwangtanggamibang, a Polyherbal Formula in ICR Mice)

  • The object of this study was to evaluate the single dose toxicity of Yukmijihwangtanggamibang (YMJHTGMB), a polyherbal formula have been traditionally used as prevention or treatment agent for various lung diseases including chronic obstructive pulmonary disease (COPD), in male and female mice. Aqueous extracts of YMJHTGMB (Yield = 16.33%) wasadministered to female and male ICR mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changesin the body and organ weight except for soft feces restricted to YMJHTGMB 2,000 mg/kg treated two male mice (2/5; 40%) at 1 day after administration. In addition, no YMJHTGMB-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of YMJHTGMB aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines.

KJD was administered to female and male ICR mice as an oral dose of 2000, 1000 and 500 mg/kg (body wt.) according to the recommendation of KFDA Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy, organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changes in the body and organ weight except for increases of lymphoid organ weights in KJD-dosing groups. These increases of lymphoid organ weights considered that related to the immune modulate effect of KJD not toxicological signs. In addition, no KJD-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. The results obtained in this study suggest that the KJD does not cause any toxicological signs. The $LD_{50}$ and approximate LD of KJD extracts in both female and male mice were considered as over 2000 mg/kg.

경북서부지역에서의 개 파보바이러스 감염상황 조사 (Epizootiologic Surveys on Canine Parvovirus Infections of Dogs Kyungbuk West Area)

  • The disease syndrome characterized by the acute vomiting and diarrhea with high mortality had been greatly epidemic in Kyungbuk West Area since March 1990 and it was followed serologically for the classification of the agent. The agent present in feces of dogs associated with this syndrome had characteristic feature in agglutinating pig red blood cells that was specifically inhibited by anti-CPV reference dog serum. This also showed the serological identity with the reference CPV antigen in Hemagglutinating inhibition test. The result obtained were summarized as follows : 1. During 5 years(March. 1990∼September. 1994), 1,470 dogs were investigated on the actual condition of CPV infections. The Infection rate of CPV from dogs was 62.5% and mortality rate was 59.8%. 2. Among 24 fecal samples collected from the dogs with enteric disease, all showed the hemagglutinating activity to porcine erythrocyte ranging from 40 to 5,120 of HA titers. 3. Among 12 sera samples collected from the dogs with enteric disease, all showed the serological identity with the reference CPV antigen from 5 to 5,120 of HI titers. 4. Bacteriologic examination of fecal specimens resulted in the isolation of pathogeric bacteria such as Staphylococcus sp, Streptococcus sp, Escherichia coli and Bacillus. Cultures for salmonella sp and Clostridium remaind negative. 5. The prevalence and identification of internal parasites were determined by fecal examination using the floatation methods. From 20 fecal samples 12(60.0%) were isolated and their species were Toxacara canis, Toxascaris leonina, and coccidium.

Replacement of Soyflakes with Cottonseed Meal in Diets of Angora Rabbits

  • This experiment was conducted with 30 adult wool producing Angora rabbits of either sex, to evaluate the effect of replacing soyflakes (SF), on equal protein basis, with low gossypol containing cottonseed meal (CSM) either alone or in combination with lysine and methionine; on the biological performance, total wool yield, wool yield per shearing, wool characteristics and mortality. Three experimental mash diets were prepared by incorporating SF (6%) as standard / control protein source $(T_1)$ and CSM (9%) as test protein source ($T_2$ and $T_3$). In $T_3$, amino acids-lysine and methionine (0.1 % each) were added. Animals were given the experimental diets about $150g{\cdot}day^{-1}{\cdot}head^{-1}$, for a period of 225 d or three shearing, and ad libitum Kudzu-vines. No significant effect of $T_2$ or $T_3$, on the body weight gain, total wool yield, wool yield per shearing and wool characteristics, was observed compared to $T_1$. However, the digestibility of dry matter, crude fibers, ether extract, acid detergent fibers, neutral detergent fibers, cellulose and hemicellulose was significantly (p<0.05) depressed in CSM based diets. Mortality of about 20% was recorded in $T_2$ and $T_3$, but not related to the addition of CSM or gossypol toxicity. More studies are needed to standardize the safe level of CSM, duration of safe feeding of CSM, and level of amino acids supplementation in CSM based diets.
