Background: Proliferative verrucous leukoplakia (PVL) is an oral potentially malignant disorder, characterized by multifocal expression, progressive clinical evolution, and a high rate of malignant transformation. Evidence-based information regarding optimal PVL management is lacking, due to the paucity of data. The present report describes a case of PVL associated with HPV-16 infection and epithelial dysplasia treated by diode laser surgery, and the outcome of disease clinical remission over a 2-year follow-up period. Case report: A 61-year-old Caucasian male with oral verrucous hyperkeratosis presented for diagnosis. The lesions were localized on the maxillary gingiva and palatal alveolar ridge. Multiple biopsy specimens have been taken by mapping the keratotic lesion area. Microscopic examination was compatible with a diagnosis of PVL with focal mild dysplasia, localized in the right maxillary gingiva. Polymerase chain reaction (PCR) was done for human papillomavirus (HPV) detection which revealed presence of HPV DNA, and the genotype revealed HPV 16 in the sample. The PVL in the right gingival area was treated on an outpatient basis by excision with a diode laser. This approach resulted in good clinical response and decreased morbidity over a 2-year follow-up period. Conclusions: This case illustrates the benefit of a conservative approach by diode laser treatment than wide surgical excision for management of the PVL lesions associated with mild dysplasia and HPV-16 infection.
Diabetes mellitus, the most common metabolic disorder, is divided into two types: type 1 and type 2. The essential treatment of type 1 diabetes, caused by immune-mediated destruction of ${\beta}-cells$, is transplantation of the pancreas; however, this treatment is limited by issues such as the lack of donors for islet transplantation and immune rejection. As an alternative approach, stem cell therapy has been used as a new tool. The present study revealed that bone marrowderived mesenchymal stromal cells (BM-MSCs) could be transdifferentiated into pancreatic cells by the insertion of a key gene for embryonic development of the pancreas, the pancreatic and duodenal homeobox factor 1 (PDX1). To avoid immune rejection associated with xenotransplantation and to develop a new cell-based treatment, BM-MSCs from ${\alpha}$-1,3-galactosyltransferase knockout (GalT KO) pigs were used as the source of the cells. Transfection of the EGFP-hPDX1 gene into GalT KO pig-derived BM-MSCs was performed by electroporation. Cells were evaluated for hPDX1 expression by immunofluorescence and RT-PCR. Transdifferentiation into pancreatic cells was confirmed by morphological transformation, immunofluorescence, and endogenous pPDX1 gene expression. At 3~4 weeks after transduction, cell morphology changed from spindle-like shape to round shape, similar to that observed in cuboidal epithelium expressing EGFP. Results of RT-PCR confirmed the expression of both exogenous hPDX1 and endogenous pPDX1. Therefore, GalT KO pig-derived BM-MSCs transdifferentiated into pancreatic cells by transfection of hPDX1. The present results are indicative of the therapeutic potential of PDX1-expressing GalT KO pig-derived BM-MSCs in ${\beta}-cell$ replacement. This potential needs to be explored further by using in vivo studies to confirm these findings.
In this study, (GaN)1-x(ZnO)x solid solution nanoparticles with a high zinc content are prepared by ultrasonic spray pyrolysis and subsequent nitridation. The structure and morphology of the samples are investigated by X-ray diffraction (XRD), field-emission scanning electron microscopy, and energy-dispersive X-ray spectroscopy. The characterization results show a phase transition from the Zn and Ga-based oxides (ZnO or ZnGa2O4) to a (GaN)1-x(ZnO)x solid solution under an NH3 atmosphere. The effect of the precursor solution concentration and nitridation temperature on the final products are systematically investigated to obtain (GaN)1-x(ZnO)x nanoparticles with a high Zn concentration. It is confirmed that the powder synthesized from the solution in which the ratio of Zn and Ga was set to 0.8:0.2, as the initial precursor composition was composed of about 0.8-mole fraction of Zn, similar to the initially set one, through nitriding treatment at 700℃. Besides, the synthesized nanoparticles exhibited the typical XRD pattern of (GaN)1-x(ZnO)x, and a strong absorption of visible light with a bandgap energy of approximately 2.78 eV, confirming their potential use as a hydrogen production photocatalyst.
비정질 $SiO_2$와 $Al(OH)_3{\cdot}xH_{2}O$를 출발물질로 하여 $250^{\circ}C$, $30\;kg/cm^2$의 조건에서 반응시간을 2~36시간까지 변화시키면서 캐올리나이트를 수열합성하였다. 합성된 캐올리나이트에 대하여 XRD, IR, TG-DTA, TEM 및 EDS를 사용하여 특성분석을 실시하였으며, 이로부터 합성된 캐올리나이트의 반응 시간별 광물학적 특성변화를 규명하고자 하였다. X선 회절 분석결과, 반응시간 2시간에 비정질 출발물질에서부터 뵈마이트가 결정화되었으며 2시간 이상 반응시키면, 뵈마이트 피크가 소멸되거나 혹은 감소하였다. 캐올리나이트는 반응 10시간 이후 생성되기 시작하였으며 반응시간의 증가에 따라 결정도가 점차 증가하였다. TGA 분석결과, 반응시간 36시간 이후에 약 13 wt%의 총중량손실이 관찰되었으며 DTA 분석결과 $258^{\circ}C$에서 관찰된 뵈마이트에 의한 발열피크는 서서히 감소되어 반응시간 10시간 이후에 관찰되지 않았고 스피넬상으로의 전이를 나타내는 발열피크는 반응시간 5시간 이후에 관찰되기 시작하여 반응시간 증가에 따라 피크강도가 증가되었다. FT-IR 분석 결과, 합성 캐올리나이트의 Al-OH 신축진동 밴드인 $3696\;cm^{-1}$과 $3620\;cm^{-1}$사이에서 $3670\;cm^{-1}$과 $3653\;cm^{-1}$의 두개의 작은 피크 분리 및 $940\;cm^{-1}$과 $915\;cm^{-1}$에서 관찰된 캐올리나이트의 특징적인 흡수띠의 관찰은 비교적 질서도가 우수한 캐올리나이트가 합성된 것을 지시하였다. TEM 분석결과, 반응시간 15시간 후에서야 비로소 (001)면의 성장으로 판상형의 캐올리나이트가 형성되었다. 반응시간 36시간 후 합성 캐올리나이트는 단순 판상형에서 육각외형의 판상형으로 변화됨을 확인되었다.
본 연구는 경남 화훼연구소에서 개발 육성된 스프레이형 국화 18 품종들에 대한 재분화율을 각각 조사하고, 그 중에서 '무랑루즈'가 가장 우수한 재분화 능력이 있음을 확인하였다. 특히 BA($0.5mg{\cdot}L^{-1}$)와 NAA($1.0mg{\cdot}L^{-1}$)를 함유한 MS배지를 신초재분화 배지로 이용함으로써 스탠다드형 국화인 신마와 함께 스프레이형 국화인 '무랑루즈'의 잎 절편체부터 효율적으로 신초 형성을 유도하고 완전한 식물체로 재분화시킬 수 있었다. 이러한 신초재분화 조건에서 '무랑루즈'잎 절편에 아그로박테리움을 감염하고 10일 동안 함께 배양한 후에, 선발항생제 kanamycin 농도를 저농도($10mg{\cdot}L^{-1}$)에서 고농도($30mg{\cdot}L^{-1}$)로 단계적으로 선발과정을 강화함으로써 효율적인 국화 형질전환체 제작이 가능하였다. 조사한 kanamycin 저항성 식물체 모두에서 전이유전자 $AtSICKLE$가 존재함을 genomic PCR 방법으로 확인하였다. 비록, 국화에서 그 기능이 비효율적인 CaMV 35S 프로모터를 사용했기 때문에 RT-PCR로 확인한 형질전환체에서의 전이유전자 $AtSICKLE$의 발현율은 상대적으로 낮았으나, 그 발현이 상대적으로 높은 개체에선 잎의 상편생장에 의한 엽형의 변화가 나타났다. 이러한 연구의 결과는 모델 식물체에서 분리되고 연구된 많은 유전자들이 가지는 화훼작물의 분자육종학적 가치를 국화에서 대량으로 조사할 수 있는 기반을 제공 할 것으로 기대된다.
This study focuses on the effects of the clothes worn by kisaeng; courtesans trained in singing and dancing, on changes in female psychology as reflected in general women's fashions during the later years of the Chosun dynasty. During this period, the social order had broken down considerable, due in part to the introduction of Roman Catholicism, and in part to the actions of Sil-hak, who emphasized open-ness and practicality in the organization of social affairs. This freer social environment disrup-ted the established social hierarchies. The kisaeng were among the first to respond to the new social mores by adopting more colorful, sensual, and individualized fashions. Their social position allowed them to reflect the new aesthetics of the time right away. Those aesthetics seemed to lay great emphasis on the artistic effects of contrast. The kisaeng would adorn their heads with large Kache (an elaborate wig or hairdo typically reserved for use by women in full formal dress). In contrast to this conspicuous hairstyle, they typically wore very tight-fitting Jogori (short-cropped Korean traditional jackets for women) around their upper torsos. The long skirts emerging from beneath these short jackets would typically flare out dramatically, with the aid of petticoats. However, these skirts would be bound at the waist with a sash, increasing the sexual suggestiveness of the clothing by drawing at-tention to the hips, and by exposing the bottom frills of the petticoats, or the wide pantal-oons and other undergarments the kisaeng wore to add volume to their skirts. The relative freedom enjoyed by the kisaeng to experiment with new fashions was not widely shared by most women. This generated envy from women of the noble classes, who were more bound by convention, and restrained from adopting such a mode of dress. It also generated envy from women of the humble classes, who saw the kisaeng as working little for their wealth, and yet dressing every day in finery that the average women would only ever be able to afford on her wedding day. This envy directed at the relative freedom/wealth of the kisaeng by women who faced greater socioeconomic constraints was given cultural expression through the adoption of elements of the kisaeng's fashion in the fashions of both noblewomen and humble women in old korea. The luxurious Kache sported by the kisaeng had in fact been borrowed from the habitual attire of upper-class women. So to distinguish themeselves from the kisaeng, they began to abandon these elaborate hairstyles in favor of traditional ceremonial hoods (Nel-ul-a thin black women's hood) and coronets (Suegaechima). This supposed reaction to the abuse of the Kache by the kisaeng still remained influenced by the kisaeng still remained influence by the kisaeng, however, as these headdresses became adorned with many more jewels and decorations, in imitation of the kisaeng's adaptations of the coronet. At the same time, noblewomen began sporting the Jangwue ; a headdress previously worn only by kisaeng and lower class women, and lower class women were then permitted to wear the Kache at weddings. All women behan to wear shorter, tighter Jogori jackets, and to add volume to their skirts. They also attached frills to their under-garments in imitation of the kisaeng's exposed petticoats and pantaloons. The impact of kisaeng fashions was thus deep and widespread, and can be understood as an expression of women's longing for freedom from socioeconomic constraints in the late Chosun dynasty. This study adopts an interdisciplinary ap-proach to the understanding of historical changes in women's fashions. Such interdisciplinary work can greatly enrich the study of fashion, often narrowly focused on clothing morphology and broad generalizations about society. For this reason, specific dynamics of feminine psychology in the late Chosun dynasty were elaborated in this study, to provide a deeper under-standing of the changes in fashion underpinned by them. If more such detailed analyses are undertaken, a whole new understanding of changes in fashion can be generated, and perhaps a transformation of the field of fashion history can be ultimately achieved.
국내 옥수수 3품종 (두메찰, 미백찰, 흑점찰)과 5 계통(HW1, KL103, HW3, HW4, KW7)의 미숙배를 1 mg/L 2,4-D, 25 mM proline, 100 mg/L casamino acid, 3 mM MES, 1.7 mg/L $AgNO_3$, Eriksson's vitamin 및 20 g/L sucrose가 첨가된 MS배지 (SEM)에 5주 동안 배양하였다. 각 계통 및 품종의 체세포배 발생 빈도는 HW1 (45.20%), KL103 (5.75%), HW3 (37.20%), HW4 (30.10%), KW70 (55.20%), 미백찰 (18.74%), 흑점찰 (22.41%), 두메찰 (36.72%)이었으며, 모델 품종인 Hi II type에서는 10% 이하였다. Hi II계통에서 형성되는 type II캘러스와 같은 노란색의 부드러운 배발생캘러스 (yellowish friable embryogenic callus, YFEC)는 오직 HW3와 흑점찰 품종에서 형성되었으며, 반면 다른 품종 및 계통에서는 late-embryo단계의 단단한 체세포배가 직접 발생되거나 형태적으로 비정상 모양을 갖는 단단하고 팽창된 체세포배가 발생 되었다. 노란색의 부드러운 배발생캘러스 (yellowish friable embryogenic callus, YFEC)는 동일배지에서 빠르게 증식 되었고 6개월 이상 배발생 능이 유지 되었으며, 1차 재분화 배지와 2차 재분화배지에 옮겼을 때 모두 식물체로 전환 되었다. 그러나 단단한 노란색의 배발생캘러스와 비정상 형태의 체세포배는 동일배지에서 매우 느리게 증식되었고, 재분화 배지에서 낮은 식물체 전환율 (25%)을 보였다. 따라서 본 연구로부터 선발된 HW3와 흑점찰은 향후 국내 옥수수 유전자원을 이용한 형질전환 시스템 개발연구에 이용 될 수 있을 것으로 예상된다.
