Endosomes lower their internal pH by an ATP-driven proton pump, which is critical to dissociation of many receptor-ligand complexes, the first step in the intracellular sorting of internalized receptors and ligands. Endosomes are known to exhibit n great range of pH values that can vary between 5.0 and 7.0 within a single cell although the factors that regulate endosomal pH remain uncertain. To evaluate the morphological and topological differences of endosomes in the different stages, confocal microscopy was used. The early endosomes labeled with fluorescein isothiocyanate-dextran for 10 min at $37^{\circ}C$ were identifiable at the peripheral and tubule-vesicular endosome compartment. In contrast, the late endosomes formed by 10 min pulse and 20 min trace were located deeper in the cytoplasm and showed more vesicular features than early endosomes. For the purpose of determining whether ATP-dependent acidification was heterogeneous and whether the differences in acidification were attributed to differences in the activity of $Na^{+}-K^{+}$-ATPase and/or $Cl^{-}$ channel, endocytic compartments were fractionated into subpopulation using percoll gradient and measured ATP-dependent acidification. While all fractions exhibited ATP-dependent acidification activity, both the initial rate of acidification and extent of proton translocation were lower in early endosomes and gradually increased in late endosomes. Phosphorylation by PKA and ATP enhanced ATP-dependent acidification in both early and late endosomes, hut there was no difference in the degree of enhancement by phosphorylation between two subpopulations. When ATP-dependent acidification was determined in the presence or absence of vanadate ($Na_{3}VO_{4}$) or ouabain, only early endosomes exhibited the vanadate or ouabain dependent stimulation of acidification activity, suggesting the inhibition of $Na^{+}-K^{+}$-ATPase. Therefore, it seems probable that the inhibition of early endosome acidification by $Na^{+}-K^{+}$-ATPase observed in vitro at least in part plays a physiological role in controlling the acidification of early endosomes in vivo.
Black rot disease in orchids is caused by the water mold Phytophthora palmivora. To gain better biocontrol performance, several factors affecting growth and antifungal substance production by Pseudomonas aeruginosa RS1 were verified. These factors include type and pH of media, temperature, and time for antifungal production. The results showed that the best conditions for P. aeruginosa RS1 to produce the active compounds was cultivating the bacteria in Luria-Bertani medium at pH 7.0 for 21 h at $37^{\circ}C$. The culture filtrate was subjected to stepwise ammonium sulfate precipitation. The precipitated proteins from the 40% to 80% fraction showed antifungal activity and were further purified by column chromatography. The eluted proteins from fractions 9-10 and 33-34 had the highest antifungal activity at about 75% and 82% inhibition, respectively. SDS-PAGE revealed that the 9-10 fraction contained mixed proteins with molecular weights of 54 kDa, 32 kDa, and 20 kDa, while the 33-34 fraction contained mixed proteins with molecular weights of 40 kDa, 32 kDa, and 29 kDa. Each band of the proteins was analyzed by LC/MS to identify the protein. The result from Spectrum Modeler indicated that these proteins were closed similarly to three groups of the following proteins; catalase, chitin binding protein, and protease. Morphological study under scanning electron microscopy demonstrated that the partially purified proteins from P. aeruginosa RS1 caused abnormal growth and hypha elongation in P. palmivora. The bacteria and/or these proteins may be useful for controlling black rot disease caused by P. palmivora in orchid orchards.
This study was carried out to investigate the effects of Salvia plebeia R. Br. ethanolic extract with different aspects (stem/leaf and whole plant) on differentiation and lipid accumulation in 3T3-L1 preadipocytes. The morphological changes and the degrees of lipid accumulation in 3T3-L1 cells were measured by Oil Red O staining and intra-cellular triglyceride (TG) assay. The mRNA expressions of special peroxisome proliferation activated receptor- genes (PPAR), CCAAT/ enhancer-binding protein (C/$EBP{\alpha}$), fatty acid synthase (FAS) and lipoprotein lipase (LPL) were detected by reverse transcriptase polymerase chain reaction (RT-PCR). The 50% ethanolic extracts ($100{\mu}g/mL$) of stem and leaf (SALE) and 30% ethanolic extracts (100 g/mL) of whole plant (SAE) from Salvia plebeia R. Br. were significantly attenuated lipid accumulation during adipogenesis in 3T3-L1 cells. Ethyl acetate-soluble fractions ($50{\mu}g/mL$) significantly inhibited lipid droplet accumulation in 3T3-L1 cells. In addition, SALE induced down-regulation of specific adipogenic transcriptional factors (C/$EBP{\alpha}$ and $PPAR{\gamma}$) and target genes (FAS and LPL) during adipogenesis. Salvia plebeia R. Br. may be used as a safe and efficient natural substance to manage obesity.
We investigated the effects of triterpene acids (TAs), ursolic acid (UA) and oleanolic acid (OA), on the induction of proliferation and differentiation of normal rat mammary epithelial cells (RMEC) or organoids cultured in Matrigel or primary culture system. To elucidate the effects, we tested their differentiation inducing activities with intercellular communication ability, cell cycle patterns, induction of apoptosis, and morphological differentiation in the three dimensional extracellular culture system. To study the changes of RMEC subpopulation in culture, the cultured cells were isolated, immunostained with peanut lectin (PNA) and anti-Thy-1.1 antibody and then analyzed with flow cytometry. Four different subpopulations, such as PNA and Thy-1.1 negative cells (B-), PNA positive cells (PNA+), Thy-1.1 positive cells (Thy-1.1+), PNA and Thy-1.1 positive cells (B+), were obtained and the size of each subpopulation was changed in culture with time in the presence of TAs. Intercellular communication was observed in culture for 7 days in TAs-treated cells, but not in culture for 4 days with scrape-loading dye transfer technique. $G_2$/M phase cells and the number of apoptotic population were increased in TAs-treated groups in cell cycle analyses. S phase fractions were reduced and the change of $G_1$ phase cells was not observed. The colonies with distinct multicelfular structures, such as stellate, ductal, webbed, squamous, lobulo-ductal colonies, were observed in Matrigel culture and the frequencies of each colony were changed in the presence of TAs. These results suggest that UA and OA have differentiation inducing effects on rat mammary epithelial cells in primary or in Matrigel culture.
약용식물로 이용되는 한약재 21 종에서 깍두기의 보존성을 증가시킬수 있는 약재를 선별하고 효과가 우수한 한약재를 용매분획하여 김치 유산균 및 효모에 대한 항균활성을 조사하였다. 그 중 포공영을 최종 선정하였으며, 클로로포름 분획추출물은 5 균주 중 Leu. mesenteroides와 S. faecalis에서 항균효과가 나타났으며, 농도별 항균활성은 E. faecalis는 80 mg/mL의 농도에서 배양 24시간까지 균증식이 억제되었으며, Leu. mesenteroides는 배양 24시간, L. plantarum는 배양 12시간까지 억제되었다. L. brevis는 포공영 클로로포름 분획 추출물을 함유한 배지에서는 별 영향을 받지 않았다. 효모인 S. cerevisiae는 20 mg/mL 이하의 저농도에서는 오히려 촉진되어 세균에 비해 저해활성이 약하게 나타났다. 포공영 클로로포름 분획추출물로 처리한 Leu. mesenteroides와 E. faecalis 두 균주를 전자현미경으로 관찰한 결과, 두 균주 모두 형태학적인 변화가 있음이 관찰되었다.
목 적: 항암요법에 의한 부작용 중 방사선섬유증(radiation fibrosis)은 방사선치료 후 빈번하게 일어나지만 발생빈도 및 심각성에 비해 치료법이 제한적이다. 본 연구에서는 방사선섬유증의 기전 연구 및 치료제 개발을 위해 방사선섬유증 동물모델계를 확립하고자 하였다. 대상 및 방법: 마우스의 대퇴부에 20 Gy의 방사선을 1주 간격으로 2회 조사한 후 날짜 별로 육안적 평가와 haematoxylin & eosin (H&E) 및 Masson’s Trichrome 염색을 통한 조직학적 변화를 평가하였다. 연부조직 수축도를 평가하기 위하여 특별히 고안된 틀을 이용하여 양쪽 하지의 길이 차이를 측정하였고, transforming growth factor (TGF)-${\beta}$의 발현 정도를 면역조직염색 및 중합효소연쇄반응(polymerase chain reaction)을 통해 측정하였다. 결 과: 본 연구에서 20 Gy씩 2회의 방사선조사를 시행하고 14, 28, 42, 97일 후 양측 하지의 수축도를 비교한 결과 42일 이후에 유의한 길이 차이가 나타났고 조직검사 결과 방사선이 조사된 피부 및 연부조직에서 아교질(collagen)과 세포외바탕질(extracellular matrix) 섬유가 불규칙한 배열을 나타내었으며 근육섬유모세포(myofibroblast)의 증가가 관찰되었다. TGF-${\beta}$ 발현 정도를 면역염색으로 조사한 결과 시간이 지남에 따라 증가하였고, 중합효소연쇄반응상 mRNA 발현량도 대조군에 비해 유의하게 증가하였다. 결 론: BALB/c 마우스의 대퇴부에 20 Gy의 방사선을 2회 조사하였을 때 방사선섬유증과 관련된 주요 인자인 TGF-${\beta}$의 발현량이 증가하였을 뿐만 아니라 외관적으로도 뚜렷한 방사선섬유증이 관찰되어 앞으로의 연구를 위한 효과적인 동물모델계로 사료된다.
차가버섯(Inonotus obliquus)의 추출물과 분획들은 항돌연변이, 항암, 항산화 및 면역자극 효과 등을 비롯하여 다양한 생물학적 활성을 갖는 것으로 알려져 있다. 본 연구는 나노분쇄 기법으로 제작된 10% 차가사료를 C57BL/6 생쥐에게 식이한 후 항암효과를 확인하기 위해 시행되었다. 부피평균에 의한 입도분석에 따르면 1 ${\mu}m$ 내에 포함되는 초미세 차가버섯 입자는 전체의 40%인 것으로 나타났다. 생쥐에게 B16BL6 흑색종세포를 피하로 주사한 결과, 종양의 부피(p<0.001)와 무게(p<0.01)는 초미세 차가버섯을 식이한 실험군(Nch)이 일반사료를 먹인 대조군(C)에 비해 현저히 감소하였고, 종양성장 억제율은 29.2%를 나타내었다. 흑색종 세포를 복강 내로 주사한 후 생존율을 관찰한 결과, 마리 당 평균 생존기간은 대조군과 실험군이 각각 17.7일과 26.0일이었다. 대조군이 모두 사망한 날인 35일 째 실험군의 생존율은 40%를 보여주었다. 초미세 차가입자가 소화기관에 문제를 유발시킬 수 있는 가능성 때문에 안전성을 확인하기 위해 소장과 대장을 조직학적으로 관찰한 결과 두 군 모두 형태학적 또는 병리학적 변화는 관찰되지 않았다.
Acanthnmoeba sp. YM-4의 영양형은 사상의 침상위족을 내어 전형적인 각canthamoeba 속의 특징이 관찰되었으며, 포낭(씨스트)은 외벽이 등근혐이었고 내벽이 단순한 구형이거나 다소 불규칙한 2개의 세포벽을 갖고 있었다. SDS-PAGE 결과, Acanthamoeha sp. Yhf-4는 약 16개의 주요 단백질 분획이 관찰되었는데, 2차원 전기영동 결과와 함께 Acanthamoeba sp. YM-4의 단백질 분획은 A. culbertsoni와 거의 같은 양상이 관찰되었다. Acanthamoeba sp. YM-4로 면역시킨 마우스의 비장세포와 myeloma 세포를 융합한 결과, 항체를 분비하는 17개 clone을 얻을 수 있었다. 생성된 단세포를 항체 중 isotyping을 실시하였는데 실험에 사용된 McAY 6, McAY 7, McAY 8, McAY 13, McAY 16 단세포군은 IgGl 항체를, McAY 10, McAY 11 단세포군은 19M 항체를 분비하였다. 생성된 단세포군 항체는 대부분이 아메바의 세포막에 있는 항원과 반응하였으며, EITB 실험결과, McAY 7 단세포군 항체는 43 kD에서 반응대가 관찰되었고, McAY 10 단세포군 항체는 55 kD과 105 kD에서 반응대가 관찰되었다. ELISA 방법을 이용한 단세포를 항체들과 여러 아메바 간의 교차반응실험 결과, McAY 7 단세포군 항체는 Acanthamoeba sp. YM-4에만 반응하였고, McAY 6과 McAY 10은 A. culbertsoni와도 반응하였다. 또한 McAY 11은 모든 아매바와 교차반응을 보였다.
요 약: 정액 동결 과정은 활성산소종의 생성을 유발하며, 생성된 활성산소종은 정자의 손상을 일으키는 것으로 알려져 있다. 따라서 본 연구의 목적은 동결 과정 중 항산화 효소 중 하나인 catalase (CAT)를 첨가함으로써 융해 후 정자의 기능과 활성산소종의 수준에 미치는 효과를 알아보고자 하였다. 5마리 돼지에서 채취한 정액은 0 (대조군), 200, 400 U/mL CAT가 첨가되어 있는 동결 희석액으로 각각 동결하였다. 융해 후, 정자 운동성, 생존성, 정상 형태율, 형질막 온전성, 미토콘드리아 기능, 세포내 ROS를 평가하였다. CAT는 400 U/mL의 농도에서 전체 정자 운동성을 향상시켰지만 (P < 0.05), 전진 운동성, 생존성, 기형율, 형질막 온전성, 미토콘드리아 기능의 향상을 나타내지 않았다. 활성산소종의 평가에서, CAT는 융해된 생존 정자의 ${\cdot}O_2$의 감소에는 효과를 나타내지 않은 반면 $H_2O_2$를 감소시켰다(P < 0.05). 결론으로 CAT는 동결 및 융해된 정자의 질을 향상시키는 데 큰 효과를 나타내진 않았지만 생존 정자에서 $H_2O_2$을 제거함로써 생존정자의 산화적 손상을 감소시킬 수 있으리라 판단된다.
천연 항균 소재를 탐색하기 위한 목적으로 은행잎, 은행종실 그리고 은행 외종피를 methanol, ethanol, water 추출물을 얻은 후 8종의 세균에 대하여 항균효과를 검색하였다. 은행잎, 은행 외종피의 methanol 추출물과 ethanol 추출물에서는 항균활성이 검색된 반면, 은행잎과 은행 외종피의 물추출물, 은행 종실의 모든 추출물에서는 항균활성이 검색되지 않았다. 은행잎의 경우 모든 균에 있어 methanol 추출물이 ethanol 추출물보다 강한 항균활성을 나타냈으며, methanol 추출물은 K. pneumoniae와 B. cereus에서 강한 항균 효과를 보였다. 은행 외종피의 경우 methanol 추출물은 B. cereus, S. aureus, B. subtilis, L. monocytogenes 균에, ethanol 추출물은 B. cereus, S. aureus, B. subtilis, K. pneumoniae균에서 강한 항균활성을 보였다. 항균성이 우수했던 은행잎과 은행 외종피의 methanol 추출물을 용매별로 순차 분획하여 항균효과를 검색한 결과 각각의 균주에 대해 hexane, chloroform, ethyl acetate 분획추출물에서 항균효과가 검색되었으며, 특히 은행 외종피의 chloroform 분획추출물에서 우수한 항균효과를 보였다. 또한 은행잎과 은행 외종피의 methanol 추출물 용매분획물의 최소저해농도(MIC)를 측정한 결과, 각각의 chloroform 분획에서 가장 낮은 MIC 값을 보였으며, 특히 은행 외종피의 chloroform 분획물은 B. cereus에 대해 $62.5{\mu}g$/mL로 본 실험 내에서 가장 낮은 MIC를 보였다. 은행잎과 은행 외종피 methanol 추출물의 chloroform 분획물($2,000{\mu}g$/mL)을 각 균주의 생육배지에 첨가하여 배양한 결과 세포가 팽윤되거나 일부 세포벽이 완전히 파괴된 형태를 관찰할 수 있었다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
