Effect of Growth Inhibition and Quinone Reductase Activity Stimulation of Makgeoly Fractions in Various Cancer Cells (막걸리 분획물에 의한 암세포 성장 억제 및 Quinone Reductase 활성 증가 효과)

  • In this study, we investigated the anticancer activity of Makgeoly (MG). MG was fractionated into four fractions by using solvent partition method, affording hexane (MGH), methanol (MGM), butanol (MGB) and aquous (MGA) soluble fractions. We determined the cytotoxicity of these four fractions in four kinds of cancer cell lines, such as HepG2, MCF-7, B16-F10 and HT29 by MTT assay. Among the various fractions, the MGM showed the strongest cytotoxic effects on all cancer cell lines. The morphological changes such as membrane shrinking and blebbing of cells were also observed by MGM treatment in HepG2 cell. In addition, we observed quinone reductase (QR) activity stimulating effects in all fraction layers of MG on HepG2 cells. QR activity increased approximately 2.6 and 2.1 times in MGM and MGH treated HepG2 cell at $100{\mu}g/mL$, respectively, compared to that in control value. Although further studies are needed, the present work could suggest that the fin of MG has a potential to be used as a chemopreventive agent against cancer.

  • The strain producing bioemulsifier was isolated from soil samples. The isolated strain was identified as the genus Acinetobacter through its morphological, cultural and physiological characteristics. The highest emulsification activity and stability by Acinetobacter sp. BE-254 was observed after 5 days of cultivation in the culture medium containing n-hexadecane 4%, NaNO$_{3}$ 0.2%, KH$_{2}$PO$_{4}$ 0.01%, MgSO$_{4}$-7H$_{2}$O 0.01%, CaCl$_{2}$ 0.01%, and yeast extract 0.01%. The optimum pH and temperature for bioemulsifier production were pH 7.0 and 30$\circ$C, respectively. Furthermore the most of bioemulsifier was produced during the exponential growth phase, and this suggested that the bioemulsifier production was growth-associated. The bioemulsifier showed good emulsification activity on various emulsifying substrates such as hydrocarbons, edible oils, and petroleum fractions.

  • Diversities of lipid compositions according to the morphological differences of the Panax ginseng root were studied by means of column, thin layer, gas-liquid chromate-graphies and histochemical stainings. Purified lipids from various parts were 1.08-2.23% of dry weight, of which 64.2-73.5% were neutral lipids, 15.4-17.4% were glycolipids and 10.4-19.2% were phospholipids. Especially the contents of neutral lipids were highest in cortex, suggesting to be the presence of lipid ducts only in cortex. Triglycerides, sterol esters and hydrocarbons were abundant in the neutral lipid fractions. Twelve components were identified in the periderm and cortex, but unidentified II, IV and V components were not present in the medulla. The major components of glycolipid freactions were sterol glycoside, digalactosyl diglyceride and esterified sterol glucoside. Phosphatidyl glycerol, phosphatidyl choline and phosphatidyl ethanolamine were major components of phospholipid fractions, And phosphatidyl choline was extreamly much in the periderm and medulla, but phosphatidyl glycerol was largest in quantity in the cortex. Eighteen kinds of fatty acids were identified in the neutral lipid, glycolipid and phospholipid fractions. Linoleic, palmitic, oleic and linolnic acids were the main components of fatty acids. The contents of saturated fatty acids, unsaturated fatty acids and essential fatty acids of each three fractions were different one another regardless of the Periderm, cortex and medulla.

On the Identification of Strains and the Cultural Characteristics of Flammulina velutipes in Korea (한국산 팽나무 버섯의 계통류별과 그 배양적 특성에 관한 연구)

  • In order to investigate morphological and cultural characteristics of strains in species, Flammulina velutipes, the author collected isolates of Flammultina velutipes at 49 locations in Korea and cultivated these isolates on the various kinds of solid media. After investigating the cultural characteristics, appeared on the various media, he obtained the following results : 1. The variation of colors in the fruit bodies is connected with the variation of climatic environments(composite effects between mean temperature in January and number of rain days of 1mm and over precipitation). The author, therfore, can find out the trend that brown type is distributed in the midland climatic region and yellow type in the southern climatic regoin. 2. Two types can be classified into several strains respectively : the strain of abundant or insufficient productivity, and strain of selectivity or non-selectivity of media. 3. According to the results of mutual comparison of soluble mycelial proteins by disc electrophoresis using polyacrylamide gels, each type has special common protein fractions(brown type : band located at 26..5mm position from surface of gel, yellow type : band located at 24.5mm position from surface of gel), and each strain has special protein fractions too. Therefore this phenomenon seems to support the results obtained by the above-stated morphological and cultural studies. 4. In the adaptability of strains to the temperature, every strain has the nature of growing in lower temperature(the optimum temperature of 20.deg.C to 25.deg.C) except that YI-1 strain has the optimum temperatue of $25^{\circ}C$-26^{\circ}C. And mycelial growth of every strain is discontinued at $35^{\circ}C.$ 5. In the adaptability of strains to the H-ion concentration, every strain has wide adaptable range of H-ion concentration, and has optimum range of pH 5.5 to 6.6 in mycelial growth excepting YA01, BI-2 and YI-1. 6. In the utility of carbon sources, the mycelial growth of every strain is very poor on the media containing xylose(average diameter of mycelial growth : 18mm), and most strains utilize favorably sucrose(39mm), maltose(37mm) and dextrose(35mm) in mycelial growth. In the utility of nitrogen sources, every strain utilizes favorably organic nitrogens(36 mm)more than inorganic nitrogens (25 mm), and utilizes fully peptone nad asparagine in organic nitrogens. Especially BA-1, BIK-2 and YA-1 strains grow vigorously on each media containing various carbon and nitrogen sources. 7. The characteristic tests of the number of days required for mycelial growth, the number of days requried for sprout of young bodies, the length of stipe and the number of fruit bodies formation seem to be useful methods in the early selection of the strain of the abundant productivity.

  • To develope food preservative, antimicrobial activities of Pinus densiflora (PD) ethanol extract against Listeria monocytogenes Scott A. Listeria monocytogenes Brie I and Listeria monocytogenes ATCC 19111 were investigated. The ethanol extracts of PD showed strong antimicrobial activities on Listeria monocytogenes. The crude ethanol extracts of PD were further fractionated by ether, ethyl acetate and butanol. The ether fraction from ethanol extract showed the strongest antimicrobial effects on Listeria monocytogenes in tryptic soy broth containing 40 mg/mL ether fractions compared with other fractions. The effect of ethanol extract of pinus densiflora against Listeria monocytogenes culture for growth stage in tryptic soy broth at 35$^{\circ}C$ showed the strongest antimicrobial activites for lag phase. The morphological changes of the cells were observed with transmission electron microscope (TEM) and scanning electron microscope (SEM) and the cells were injured by treatment of 40 mg/mL ethanol extract of Pinus densiflora.

  • This Study was carried out develop antitumor effect of the n-butanol soluble of fraction of Perilla frutescens on human skin melanoma cells. The antitumor activity of various fractions obtained form n-butanol soluble fraction of Perilla frutescens was evaluated in human skin melanoma cells. The antitumor activity of the n-butanol soluble fraction in human skin melanoma cells was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, neutral red (NR) assay and sulforhordamine B protein (SRB) assay of colorimetic assay methods. The light microscopic study was carried out to observe morphological changes of cultured human skin melanoma cells. These results were obtained follows; The fractions 5 and 6 of the n-butanol soluble fraction of P frutescens were shown significant antitumor activities. The number of human skin melanoma cells were decreased and tend to form cell cluster by treatment with actions 5 and 7 of the n-butanol soluble fraction of P. frutescens. The fraction 6 of the the n-butanol soluble fraction showed the highest antitumor activity on P. frutescens. These results suggest that the fraction 6 of the n-butanol soluble fraction of P. frutescens may be a valuable choice for the studies on the treatment of human skin tumors.

Studies on hemolytic reaction of normal chicken serum and sheep erythrocytes (닭혈청(血淸)에 의한 면양적혈구(緬羊赤血球)의 용혈(溶血)에 관한 연구(硏究))

  • Hemolytic reaction of normal fresh chicken serum on sheep erythrocytes was studied and the following experimental results were obtained and summarized. 1. Chicken sera, 258 (78%) out of 344 samples showed hemolytic activity on sheep erythrocytes. 2. Distribution of a different hemolytic titer of chicken sera was not dependent to sex and age difference of test chicken. 3. Hemolytic activity of serum component obtained from normal fresh chicken was heat inactivated at $56^{\circ}C$. 30 minutes heating. 4. The most enhanced hemolytic activity of chicken serum on sheep erythrocytes was observed at the incubation temperature of $46^{\circ}C$. 5. The most effective pH for the hemolytic reaction of chicken serum on sheep erythrocytes was observed at 7.0, and pH 6.0 or 8.5 resulted less or no hemolysis. 6. Hemolytic reaction of chicken serum and sheep erythrocytes required Mg⧻ and Ca⧻ ions as, co-factor, and the former was required more compared to the latter. 7. Hemolytic activity of chicken serum was observed in ChC 2, 4 fraction but not in ChC 1, 3, ChC 3, 4, ChC 1, 2, 4 and ChC 1, 2, 3 fractions. 8. In electron micrography, morphological changes of sheep erythrocyte membrane by normal chicken serum was similar to that of immune hemolysis: that was, the hemolytic hole was circular and it was surrounded with a white ring. 9. Electron micrography of morphological changes on sheep erythrocyte membrane indicated that the size of hemolytic hole and white ring were functional to the chicken serum concentration used and reaction time.

Morphological characterization of Vibrio alginolyticus specific bacteriophage isolated from fish farms on west coast of Korea (서해안 양식장에서 분리한 Vibrio alginolyticus의 특이 bacteriophage에 대한 구조적 특성)

  • Vibrio alginolyticus (V. alginolyticus), which is one of bacterial pathogens evoking severe infection in fish and shellfish as well as in human has been found at high frequency around all coast areas in Korea. Both V. alginolyticus and V. alginolyticus specific bacteriophage were isolated from sea water and various fishes from fish farms on west coast in Korea. In a morphological study based on electron microscope, the purified phage appeared to be composed of hexagon head of 60 nm and short tail of 20 nm. In the denatured SDS-PAGE analysis, the structural proteins of the phage were found to be 7 different protein fractions ranging from 37.8 to 198 kda. The kind of nucleic acid of the phage was ascertained to a double stranded DNA.

  • We investigated the growth inhibitory effect of internal organs of Todarodes pacificus (TP) on proliferation in human cancer cell lines in vitro. The internal organs of TP were extracted with methanol (TPM), which was then further fractionated into four subfractions by using a solvent partition method, resulting in hexane (TPMH), methanol (TPMM), butanol (TPMB), and aqueous (TPMA) soluble fractions. We determined the cytotoxic effect of these four fractions in three kinds of cancer cell lines - HepG2, MCF-7 and HT-29 - by MTT assay. Among the four subfractions of TPM, TPMH showed the strongest cytotoxic effects at a concentration of $300{\mu}g$/ml, displaying 91.56% on the HepG2 cell line and 85.93% on the MCF-7 cell line. Morphological changes such as membrane shirinking and blebbing of cells were also observed during TPMH treatment of HepG2 cells. In addition, we also observed quinone reductase (QR) induced effect in the methanol (TPMM) layer of HepG2 cells. TPMM showed the highest induction activity of quinone reductase on HepG2 cells among the other partition layers. The QR induced effect of TPMM was determined to be 2.7 at a level of $360{\mu}g$/ml with a control value of 1.0. Although further studies are needed, the present work suggests that internal organs of Todarodes pacificus (TP) may be a chemopreventive agent for the treatment of human cells.
