  • For the purpose of external and internal morphological study on the origin of Folium, writing on herbolagy have been thoroughly reviewed. The object herb medicine(9 kinds) is Lophatheri Folium(Herba), Isatidis Folium, Eoriobotryae Folium, Mori Folium, Perillae Folium, Artemisiae argi Folium, Phyllostachys Folium, Clerodendri Folium, Biotae Folium(Cacumen). After this study, additional experimental verification is required as a matter of fact. As comparison materials, this thesis will be utilized.

Effect of Foilum mori on Adipocyte Differentiation

  • Several natural products were tested to control the differentiation of cultured human mesenchymal stem cell into adipocyte. The extent of the inhibitory effect on the conversion of adipose was measured using Oil red O staining, which stains accumulated lipid droplets in the cytoplasm of adipocyte. Of the various natural product extracts, the adipocyte differentiation was inhibited by an extract from Folium mori in the concentration range 1${\times}$10$\^$-4/∼5${\times}$l0$\^$-2/ g/mL. These results suggest that Folium mori has an inhibitory activity toward the adipose conversion, which is a major cause of obesity.

  • Mori Folium(MF) methanol extract and its water soluble fraction showed significant blood glucose lowering effects alloxan-induced hyperglycemic mice. Their hypoglycemic activities seemed to nothing to do with the stimulation of insulin release or insulin-like action, according to our experiments. On the other hand, MF prevents the hyperglycemic responses from an oral load of starch and glucose in vivo. Since complex carbohydrates present in a diet must be degraded to monosaccharides by $\alpha$-glucohydrolase before being absorbed in the gastrointestinal tract, it is thought that blood glucose lowering effects of MF may be related to the inhibition of $\alpha$-glucohydrolase catalyzed enzymatic reaction. In addition, experiments that examined an effect of MF water soluble fraction on gastrointestinal movement showed no significant GI movement inhibitory effect. In conclusion, MF water soluble fraction may possess active component which is a potential candidate as an orally active agent for the treatment of diabetes mellitus.

  • The effect of prolonged treatment with Mori Folium column fraction(MFCF) on intestinal ${\alpha}$-glycosidase activity has been studied in mice made hyperglycemic and hyperinsulinemic by feeding a high carbohydrate-containing diet. Mice were treated for 10 week with or without MFCF, added to the high carbohydrate-containing diet at 50mg/100g food. While MFCF had no effect on body weight, it prevented the rise in glycemia and insulinemia. Maltase, sucrase and lactase activities were measured in intestinal homogenates of proximal, middle and distal segments of jejunoileum. Following 10 week of MFCF administration, MFCF significantly induced all three enzyme activities especially at middle and distal segments. The mechanisms responsible for these changes and their potential biochemical implications remain to be determined.

Anti-oxidative effects of Mori Folium, Mori Ramulus and Mori Cortex Radidus (상엽(桑葉), 상지(桑枝) 및 상백피(桑白皮)의 항산화 활성 연구)

  • Objective : This study was designed to investigate the anti-oxidative potentials to wide the usage of Morus alba L. derived from medicinal herbs on cosmoceutical fields. Methods : Anti-oxidative potentials were investigated by using several kinds of assays including electron donating ability, and the total contents of phenolic compounds were also measured. In some cases TLC (Thin Layer Chromatography) method were used to identify the patterns of phenolics in herbal extracts. Results : Aurones which are sub-types of flavonoids were observed in methanol extract of Mori Cortex Radidus, and total phenolic contents in Mori Cortex Radidus were more than in methanol extracts of Mori Folium and Mori Ramulus. Various kinds of phenolic compounds were observed in chloroform fraction of Mori Cortex Radidus. Conclusion : According to these results, the chloroform fraction of Mori Cortex Radidus could be the most possible resource that has potentials on anti-oxidant agents on cosmoceutical fields.

Anti-fibrotic Effect of Mori Folium Extract in Hepatic Stellate Cells (간성상세포에서 상엽(桑葉) 추출물의 섬유화 억제 효과)

  • Objectives : Mori Folium was popularly used as one of the traditional medicinal herbs. Although M. Folium has been cultivated for rearing silkworm historically, it's use has been expanded as natural therapeutic agent for the treatment of filariasis, diabetes and dropsy in East Asia. However, little has been known about the effect of M. Folium on liver fibrosis. Therefore, we would like to explore an anti-fibrogenic potential of M. Folium extract (MFE) using immortalized human hepatic stellate cell line, LX-2 cells. Methods : We examined the effects of MFE on the transforming growth factor ${\beta}1$ ($TGF{\beta}1$)-induced liver fibrosis in LX-2 cells. Cell viability, Smad binding element-driven luciferase activity, phosphorylations level of Smad 2/3, and expression level of $TGF{\beta}1$-dependent target genes were monitored in the MFE-treated LX-2 cells. Results : Up to 30 ${\mu}g/ml$ MFE treatment did not show any possible toxic effect in LX-2 cells. MFE inhibited $TGF{\beta}1$-inducible Smad binding element-driven luciferase activity and decreased the $TGF{\beta}1$-inducible phosphorylations of Smad 2 and Smad 3 in hepatic stellate cell in a dose dependent manner. Furthermore, increases of plasminogen activator inhibitor type 1, $TGF{\beta}1$ and matrix metalloproteinases 2 genes by $TGF{\beta}1$ were also attenuated by MFE treatment. Conclusions : These findings suggested that MFE would be used as a potential therapeutic agent for the treatment liver fibrosis, which might be mediated by the inhibition of $TGF{\beta}1$-inducible Smad 2/3 transactivation and target genes expression.

  • Objectives : Mori Folium is one of the traditional medicinal herb. It was commonly used for sericulture in the world and has been traditionally administered as natural therapeutic agent for the treatment of filariasis, diabetes and dropsy in East Asia. This study investigated an anti-inflammatory potential of Mori Folium ethanol extract (MFE). Methods : We examined the effects of MFE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in a murine macrophage cell line, RAW 264.7. Results : MFE inhibited production of NO and $PGE_2$ in a dose dependent manner and also decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2, interleukin (IL)-1, IL-6 and tumor necrosis factor-${\alpha}$. As a plausible molecular mechanism, increased degradation of I-${\kappa}B{\alpha}$ and phosphorylation of I-${\kappa}B{\alpha}$, NF-${\kappa}B$ and MAP kinases by LPS were partly blocked by MFE treatment. Conclusions : These results suggest that MFE has an anti-inflammatory therapeutic potential, which may result from inhibition of NF-${\kappa}B$ activation and MAPK phosphorylation, thereby decreasing the expression of pro-inflammatory genes.

The Immunomodulatory Activity of Mori folium, the Leaf of Morus alba L., in RAW 264.7 Macrophages in Vitro

  • Background: Immunoregulatory elements have emerged as useful immunotherapeutic agents against cancer. In traditional medicine, Mori folium, the leaf of Morus alba L. (Moraceae), has been used for various medicinal purposes; however, the immunomodulatory effects have not been fully identified. We evaluated the immunoenhancing potential of water extract of Mori folium (WEMF) in murine RAW264.7 macrophages. Methods: RAW264.7 cells were treated with WEMF for 24 hours and cell viability was detected by an MTT method. Nitric oxide (NO) levels in the culture supernatants were assayed using Griess reagent. The productions of prostaglandin $E_2$ ($PGE_2$) and immune-related cytokines was measured using ELISA detection kits. The mRNA and protein expression levels of Inducible NO synthase, COX-2, and cytokines were assayed by reverse transcription-PCR and Western blotting, respectively. The effect of WEMF on phagocytic activity was measured using a Phagocytosis Assay Kit. Results: WEMF significantly stimulated the production of NO and $PGE_2$ as immune response parameters at noncytotoxic concentrations, which was associated with the increased expression of inducible NO synthase and COX-2. The release and expression of cytokines, such as $TNF-{\alpha}$, interleukin $(IL)-1{\beta}$, IL-6, and IL-10, were also significantly increased in response to treatment with WEMF. Moreover, WEMF promoted the macrophagic differentiation of RAW264.7 cells and the resulting phagocytosis activity. Conclusions: WEMF has the potential to modulate the immune function by regulating immunological parameters. Further studies are needed to identify the active compounds and to support the use of WEMF as an immune stimulant.

Antidiabetic Activity of Mori Folium Ethanol Soluble Fraction in db/db mice (db/db 마우스에서 상엽 에탄올가용분획의 항당뇨활성)

  • Antidiabetic activity of Mori folium ethanol soluble fraction (MFESF) was examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model . 500 and 1000mg/kg dose for MFFSF (designated by SY 500 and SY 1000, respectively) and 5mg/kg dose for acarbose were administered for 6 weeks. Body weight gain, fasting and non-fasting serum glucose, glycated hemoglobin and triglyceride were all reduced dose dependently when compared between db/db control group and MFESF treated group. At 11th and 13th week after birth, MFESF increased an insulin secretion which may result in lowering serum glucose level. Total activities of sucrase and maltase in SY 500 treated group were decreased when compared to db/db control. On the other hand, those in SY 1000 and acarbose treated groups were increased. This result may suggest that proteins for sucrase and maltase were compensatorily induced due to significant inhibition of glycosidase-catalyzed reaction at doses administered in this study.

Contents of Nicotinamide and GABA in Several Sericultural Products (수 종 잠상산물의 Nicotinamide 및 GABA함량)

  • Nicotinamide and GABA(${\gamma}$-aminobutyric acid) contents in sericultural products, Mori Folium, Mori Fructus, Mori Cortex Radicis, silkworm podwer, Bombycis Exerementum and Bombycis Corpus were analyzed by HPLC. Nicotinamide content was higher in silkworm derived-products than in mulberry derived-products. Nicotinamide contained Bombycis Excrementum was the highest by 0.31 mg/g DW among the sericultural products. GABA content showed the highest by 1.68 mg/g DW in Mori Cortex Radicis among the sericultural products.
