Anti-allergic effect of Cortex Mori (상백피(桑白皮)의 항알레르기성 효과)

  • Moraceae comprise a large family of sixty genera and neary 1,400 specieses, including important groups such as Artocarpus, Morus, and Ficus. In particular, Morus(mulberry) is a small genus of tree and shrubs found in temperate and subtropical regions of the Northern hemishere and has been widely cultivated in China and Korea, In addition, the root bark of mulberry tree have been used as an antiphlogic, diuretic, and expectorant in white medicine, and the crude drug is known as "Sangbaikpi" in Korea. Recently, some papers have been published reporting the hypotensive effect, antiviral effect, antifungal effect, inhibitory effect of cAMP-phosphodiesterase, and anticancer effect of this extract. Little is known about that Cortex mori could have been an antiallergic effect. The purpose of this study was the development of an antiallergic agent with an antiallergic effect from Cortex mori. For this, several in vivo and in vitro experimental models were used. Results are 1) Cortex mori inhibited the compound 48/80-induced degranu-lation, histamine release and calcium uptake of rat peritoneal mast cells, 2) compound 48/80-induced anaphylactic shock and cutaneous reaction were significantly inhibited by pretreatment of Cortex mori, and 3) Cortex mori inhibited the ovalbumin-induced late astmatic reaction. From the above results it is suggested that Cortex mori has some substances with an antiallergic activity. Our final purpose of this study is to develope the new drug with an antiallergic activity from Cortex mori

Composition and Diversity of Tree Species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh

  • Information on plant diversity and community structure are required to chalk out necessary actions for conservation management. The present study assessed the composition and diversity of tree species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh, during April 2010 to November 2011. A total of 107 tree species belonging to 72 genera and 37 families were recorded, where Moraceae family was represented by maximum (11) species. Density, Basal area and volume of tree species were $418{\pm}20.09stem/ha$, $21.10{\pm}2.62m^2/ha$ and $417.4{\pm}79.8m^3/ha$ respectively. Diameter and height class distribution of tree species revealed an almost reverse J-shaped curve. Both the number of species and percentage of tree individuals were maximum in the lower DBH and height ranges. Anthropogenic disturbances like illegal tree cutting, over extraction, settlement inside forest area etc. were noticed during the study, which are supposed to cause gradual decrease of both tree species and individuals in the higher DBH and height classes. However, Artocarpus chama was found dominant showing maximum IVI followed by Schima wallichii, Aporosa wallichii, and Lithocarpus acuminata. The quantitative structure of the tree species of Kamalachari natural forest is comparable to other tree species rich tropical natural forests. The findings of the study may help in monitoring future plant population changes of the identified species and adopting species specific conservation programs in Kamalachari natural forest.

Anti-oxidant Compounds of Cudrania tricuspidata Leaves (구지뽕나무 잎의 항산화 성분)

  • Cudrania tricuspidata Bereau (Moraceae) have been used for anti-inflammatory, anti-hepatotoxic, anti-hyper­tensive and anti-diabetic activities. In order to investigate the efficacy of antioxidant activity, the bio-activity guided fraction and isolation of Biologically active substance were performed. $H_{2}O,\;30\%,\;60\%,\;100%$ MeOH and acetone fractions were examined on the antioxidant activity by DPPH method. It was shown that $30\%,\;60\%,\;100\%$ MeOH fractions have sig­nificantly antioxidant activity. From $30\%$ MeOH fraction, two dihydroflavonoid glycosides dihydroquercetin 7-O-$\beta$-D-glu­copyranoside (I), dihydrokaempferol 7-O-$\beta$-D-glucopyranoside (V) were isolated and $60\%$ MeOH fraction, six flavonoids including quercetin 3-O-$\alpha$-L-rhamnopyranosyl($1\rightarrow6$)-$\beta$-D-glucopyranoside (II), quercetin 3-O-$\beta$-D-glucopyranoside (III), quercetin 7-O-$\beta$-D-glucopyranoside (IV), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl($1\rightarrow6$)-$\beta$-D-glucopyranoside (VI), kaempferol 3-O-$\beta$-D-glucopyranoside (VII), kaempferol 7-O-$\beta$-D-glucopyranoside (VIII) were isolated. To investigate the antioxidant activities of each compounds, we measured radical scavening activity with DPPH method and anti-lipid per­oxidative efficacy on low density lipoprotein (LDL) with TBARS assay. Four compounds of quercetin glycosides (I, II, III, IV) showed significant antioxidant activity.

Anti-Diabetic Effects of Mori Folium Extract on High-Fat Diet and Streptozotocin-Induced Type II Diabetes Mellitus in Mice (고지방식이와 STZ 유도 제2형 당뇨병 마우스에서 상엽 추출물의 항당뇨 효과)

  • Objectives : The present study was designed to investigate the anti-diabetic effects of Mori Folium (Morus alba L. of Moraceae) extract (MFE) on high fat diet (HFD) and streptozotocin (STZ)-induced type II diabetes mellitus in mice. Methods : The mice (C57BL/6J) were fed HFD for 8 weeks and then was induced with a single injection of STZ (75 mg/kg). The diabetic mice were divided into four groups [(STD, HFD, HFD + MFE and HFD + quercetin (QUR)] and administered with MFE or OUR for 4 weeks. Fasting blood glucose, lipid profile (triglycerides and cholesterol etc.), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), insulin and leptin were measured every 2 weeks. Results : Body weight gain was lower in the MFE and QUR groups than HFD group. The fasting blood glucose was lower in the MFE and QUR groups. Oral glucose and insulin tolerance were decreased in the MFE and QUR groups. The levels of serum total cholesterol, triglycerides, and LDL cholesterol were reduced in the MFE and QUR groups. The HDL cholesterol was much higher in the MFE and QUR groups than HFD group. The levels of GOT, GPT and atherogenic index were decreased in the MFE and QUR groups. The serum insulin and leptin concentrations were reduced in the MFE and QUR groups. Conclusions : These results showed that MFE could decrease blood glucose level and lead to an amelioration in dyslipidemia states on HFD/STZ-induced type II diabetes mellitus in mice.

Melanin-concentrating Hormone-1 Receptor (MCH-1) Antagonism of the Leaves Extract from Morus alba

  • The present study was performed to investigate the binding affinity of the ethanol extract from the leaves of Morus alba (EMA) and some EMA related plant materials (EMA-D, EMA-DM) for melanin-concentrating hormone-1 receptor (MCH-1) and also to examine the antagonistic effect of them for the recombinant MCH-1 receptor expressed in CHO cells. EMA, dichloromethane fraction (EMA-D) and EMA-DM exhibited high affinity for mammalian MCH receptor in receptor binding assays ($IC_{50}$ value: 2.3, 1.6 and $1.0{\mu}g/ml$, respectively). Other plant materials (MMA-D, MMA-DM) obtained from methanol extracts from the leaves of Morus alba (MMA) also exhibited high affinity for mammalian MCH receptor, even though the $IC_{50}$ values of them were lower than those of EMA-D and EMA-DM. In Chinese hamster ovary (CHO) cells expressing human MCH-1, EMA-DM and EMA-D significantly inhibited MCH-induced intracellular $Ca^{2+}$ increase ($IC_{50}$ values: 16.5 and $22.7{\mu}g/ml$, respectively). These results clearly indicate that the ethanol extract from the leaves of Morus alba (EMA) and some EMA related plant materials (EMA-D, EMA-DM) are novel selective MCH-1 receptor antagonist, respectively.

Tumor cell growth inhibition and antioxydative activity of flavonoids from the stem bark of Cudrania tricuspidata (꾸지 뽕나무로부터 분리한 flavonoid계 화합물의 암세포성장 저해 및 항산화 활성)

  • Five cytotoxic and antioxidative flavonoids were isolated from the stem bark of Cudrania tricuspidata by consecutive purification using HP-20, silicagel and prep-HPLC. They were identified as taxifolin, orobol, eriodictyol, dihydrokaempferol and steppogenin by means of spectral studies. The antioxidative activities $(IC_{50})$ assayed by TBA method of these compound $1{\sim}5$ to were 6, 3, 3, >50, and $10\;{\mu}g/ml$, respectively. The effect on the growth inhibition $(IC_{50})$ of these compounds against P388 cell line were found to be 0.18, 3.3, 15 and $6.2\;{\mu}g/ml$, respectively in the order of compound 2 to 5.

  • Four known prenylated flavonoids, artonins E (1) and O (2), artobiloxanthone (3), and cycloartobiloxanthone (4), were isolated from the stem bark of Artocarpus kemando by bioassayguided fractionation using the DNA strand-scission and the KB cytotoxicity assays as monitors. Compounds 1 and 3 exhibited strong DNA strand-scission activity, and all four compounds were found to be cytotoxic.

Synergistic Antibacterial Activity of Fig (Ficus carica) Leaves Extract Against Clinical Isolates of Methicillin-resistant Staphylococcus aureus (병원내에서 분리된 메티실린내성 황색포도상구균에 대한 무화과잎 추출물의 항미생물효과)

  • Fig (Ficus carica L.) belongs to the mulberry tree (Moraceae) which is one of the oldest fruits in the world. It has been used as a digestion promoter and a cure for ulcerative inflammation and eruption in Korea. The present study investigated the antimicrobial activity of methanol (MeOH) extract of fig leaves against methicillin-resistant Staphylococcus aureus (MRSA) isolated in clinic. The MeOH extract (MICs, 2.5 to 20 mg/mL; MBCs, 5 to 20 mg/mL) was demonstrated as antibacterial activity in isolates MRSA 1-20. The administration of the MeOH extract in combination with oxacillin or ampicillin induced a reduction of ${\geq}$4-8-fold in all tested bacteria, which was considered to be synergistic based on a FICI of ${\leq}$0.375-0.5. Furthermore, time-kill study was found that a combination of MeOH extract with oxacillin or ampicillin produced a more rapid decrease in the concentration of bacteria CFU/mL than MeOH extract alone. The results suggest that fig leaves could be employed as a natural antibacterial agent in MRSA infection care products.

The Immunomodulatory Activity of Mori folium, the Leaf of Morus alba L., in RAW 264.7 Macrophages in Vitro

  • Background: Immunoregulatory elements have emerged as useful immunotherapeutic agents against cancer. In traditional medicine, Mori folium, the leaf of Morus alba L. (Moraceae), has been used for various medicinal purposes; however, the immunomodulatory effects have not been fully identified. We evaluated the immunoenhancing potential of water extract of Mori folium (WEMF) in murine RAW264.7 macrophages. Methods: RAW264.7 cells were treated with WEMF for 24 hours and cell viability was detected by an MTT method. Nitric oxide (NO) levels in the culture supernatants were assayed using Griess reagent. The productions of prostaglandin $E_2$ ($PGE_2$) and immune-related cytokines was measured using ELISA detection kits. The mRNA and protein expression levels of Inducible NO synthase, COX-2, and cytokines were assayed by reverse transcription-PCR and Western blotting, respectively. The effect of WEMF on phagocytic activity was measured using a Phagocytosis Assay Kit. Results: WEMF significantly stimulated the production of NO and $PGE_2$ as immune response parameters at noncytotoxic concentrations, which was associated with the increased expression of inducible NO synthase and COX-2. The release and expression of cytokines, such as $TNF-{\alpha}$, interleukin $(IL)-1{\beta}$, IL-6, and IL-10, were also significantly increased in response to treatment with WEMF. Moreover, WEMF promoted the macrophagic differentiation of RAW264.7 cells and the resulting phagocytosis activity. Conclusions: WEMF has the potential to modulate the immune function by regulating immunological parameters. Further studies are needed to identify the active compounds and to support the use of WEMF as an immune stimulant.

A Study on the Bioactive active substance of Cudrania tricuspidata Leaf and Fruit Using Aspergillus oryae Period of fermentation (Aspergillus oryae를 이용한 발효시간별 꾸지뽕나무 잎, 열매의 생리활성 비교)

  • 꾸지뽕나무(Cudrania tricuspidata)는 뽕나무과(Moraceae)에 속하는 낙엽활엽 소교목이다. 우리나라와 중국, 일본과 같은 동아시아에 주로 분포하며 척박한 땅에서도 잘자라고 병충해에 강하다고 알려져 있다. 예로부터 꾸지뽕나무는 항암, 간보호, 눈을 밝게하는 작용이 있다고 동의보감과 신농본초경에 기록되어 있다. 우리나라는 오랫동안 된장 등 발효식품을 자주 접하고 섭취하여 왔다는 점을 고려하여 본 연구를 수행하였다. 발효균을 접종하면 항암활성, 면역체계 개선 등 다양한 생리활성 물질이 증가한다고 알려져 있다. 본 연구는 가시가 없고 잎이 커 작업성이 용이한 대품 품종을 2018년 9월에 전남 신안군에서 채취하여 분석 시료로 사용하였다. 항산화활성 측정은 프리라디칼(DPPH, ABTS) 소거능을 측정하여 농도(EC50)별 측정결과 $100{\mu}g/mL$ ext. 이하로 항산화 활성이 열매보다 잎이 높다는 것을 확인하였다. 황국균(Aspergillus oryae)을 꾸지뽕나무 잎과 열매에 접종시켜 페놀성화합물을 스크리닝 한 결과 기존에 발견되지 않은 Salicylic acid, Naringenin, Vanilic acid, Oxyresveratrol 등 기능성 물질이 발견되었고, 잎의 경우 36시간 발효물(355mg/g)은 무처리군(179mg/g)에 비해 2배정도 상승하였다. 열매의 경우 48시간 발효시켰을 경우(472mg/g)으로 무처리군(156mg/g)보다 3배정도 상승하였다. 발효를 통해 꾸지뽕나무 잎과 열매의 최적의 추출조건을 확립하고 생리활성 물질 분석을 이용한 효능탐색 등을 진행하였다. 향후 꾸지뽕나무를 활용한 식품 소재개발 등 사업화에 기초적인 자료를 제공하여 임업인의 새로운 소득품목 육성에 기어코자 한다.
