Partial Characterization of Physicochemical and Kinetic Properties of $Ca^{++}-ATPase$ System in Luteal Membranes (황체막에서의 $Ca^{++}-ATPase$의 특성)

  • It has been reported that the luteal function may be regulated by the intracellular calcium in luteal cells (Higuchi et al, 1976; Dorflinger et at, 1984; Gore and Behrman, 1984) which is adjusted partially by $Ca^{++}-ATPase$ activities in luteal cell membranes (Verma and Pennistion, 1981). However, the physicochemical and kinetic properties of $Ca^{++}-ATPase$ in luteal membranes were not fully characterized. This study was, therefore, undertaken to partially characterize the physicochemical and kinetic properties of $Ca^{++}-ATPase$ system in luteal membranes and microsomal fractions, known as an one of the major $Ca^{++}$ storge sites (Moore and Pastan, 1978), from the highly luteinized ovary Highly luteinized ovaries were obtained from PMSG-hCG injected immautre female rats. Light membrane and heavy membrane fractions and microsomal fractions were prepared by the differential and discontinuous sucrose density gradient centrifugation method desribed by Bramley and Ryan (1980). Light membrane and heavy membrane fractions and microsomal fractions from highly luteinized ovaries are composed of the two different kinds of $Ca^{++}-ATPase$ system. One is the high affinity $Ca^{++}-ATPase$ which is activated in low $Ca^{++}$ concentration (Km, 10-30 nM), the other is low affinity $Ca^{++}-ATPase$ activated in higher $Ca^{++}$ concentration $(K_{1/2},\;40\;{\mu}M)$. At certain $Ca^{++}$ concentrations, activities of high and low affinity $Ca^{++}-ATPase$ are the highest in light membrane fractions and are the lowest in microsomal fractions. It appeares that high affinity $Ca^{++}-ATPase$ system have 2 binding sites for ATP (Hill's coefficient; around 2 in all membrane fractions measured) and the positive cooperativity of ATP bindings obviously existed in each membrane fractions. The optimum pH for high affinity $Ca^{++}-ATPase$ activation is around S in all membrane fractions measured. The lipid phase transition temperature measured by Arrhenius plots of high affinity $Ca^{++}-ATPase$ activity is around $25^{\circ}C$. The activation energies of high affinity $Ca^{++}-ATPase$ below the transition temperature are similar in each membrane fractions, but at the above transition temperature, it is the hightest in heavy membrane fractions and the lowest in microsomal fractions. According to the above results, it is suggested that intracellular $Ca^{++}$ level, which may regulate the luteal function, may be adjusted primarily by the high affinity $Ca^{++}-ATPase$ system activated in intracellular $Ca^{++}$ concentration range $(below\;0.1\;{\mu}M)$.

  • Malassezia furfur is an important causal factor for seborrheic dermatitis. Nowadays, the drugs available to treat this fungal infection are few. Several studies have documented the biological activity of essential oils. However, its antifungal properties are not completely understood, especially its anti-Malassezia activity. The aim of this study were to evaluate the effect of the plant essential oils on the growth of M. furfur using disk diffusion method and analyze by Gas chromatography-mass spectrometry (GC-MS) most active essential oils. In first screening, the 17 plant essential oils have possesses inhibitory activity against M. furfur at 2 mg/mL. Among the plant essential oils, oil of Citrus auranifoli was most active against M. furfur and its activity showed dose dependency. This anti-malassezial activity was high than that of itraconazole at 2 mg/mL. Oil of Citrus auranifolia also was phytochemically examined by GC-MS analysis, its main constituents were identified as limonene, ${\gamma}$-terpinene and terpinolene. It can be concluded that essential oils of Citrus auranifolia may have interesting applications to control fungal-derived diseases.

  • Purpose : When the pliability of the Lumbar is degraded, the kinesiology function undergoes an influence and it receives an overload. Finally, it is connected with a Lumbar muscular injury. When it does the regular exercise treatment the lumbago patient the balance and function of changeover are improved. The purpose of this study is to review of effect of the PNF application for lumbago patient Methods : By using internet, we research the PubMed, Science Direct, KISS, DBpia and the article on the effect of PNF. We selected the article between 1970 and 2007. Key words were low back pain, lumbago, PNF. Results : PNF is a therapeutic exercise for central nervous system(CNS) patient and the peripheral nervous system(PNS) patient and it is widely applied from sports medicine in the last. According to research of the Jung Young Jo(2007) and Han Kou Soo(2002), PNF technique improves the pain and a condition of the lumbago patient. And the affection balanced change and dynamic balance change and spontaneous movable scope are improved. And it improved to daily life activity. According to research of Moore and Hutton(1980), Lee Kyoung Hye(1999), PNF stretching where it is one of muscular relaxation is good in lumbar muscle fascia ache solutions, and PNF stretching that muscle is relaxed and increases ROM by Reflective system of the vertebra. These researches is relation all each other. it have appeared many report of research about Pliability, muscular power, balance, mobility improve ADL movement of attendance ability and functional action improved excise therapy put effect about lumbago patient recently, it have appeared research about therapy of lumbago patient through PNF. But PNF technique is not application about lumbago patient in clinic, therefore also it is not enough for case study about this therapy. Conclusion : PNF improve combination operation of muscle, inconvenience decrease from daily life activity, ROM(range of movement) of lumbar is increase and Lumbago reduce. So we suggest that PNF will be applies on the lumbago patient and must do a many research.

The Specific Plant Species and Conservation of Juwangsan National Park, Korea (주왕산국립공원의 특정식물과 보전방안)

  • The flora of Juwangsan National Park was surveyed. The field works was done for seven times from March to October 2008. Endangered species designated by The Ministry of Environment in Juwangsan National Park were found to be Hylotelephium ussuriense (KOM.) H. OHBA, Berchemia berchemiaefolia (MAK.) KOIDZ, Jeffersonia dubia BENTH., Iris odaesanensis Y.N.LEE and Acanthopanax senticosus (RUPR. et MAX.) HARMS, Lilium cernum KOM., Thalictrum coreanum LEV.(7 taxa). The floristic special plants were recorded to a total of 88 taxa; that is, degree V species (8 family 9 taxa) were containing as Jeffersonia dubia BENTH., Berchemia berchemiaefolia (MAK.) KOIDZ, Lilium cernum KOM., etc, degree IV species (4 family 5 taxa) were Cimicifuga heracleifolia KOM., Anemone reflexa STEPH. et WILLD., Patrinia rupestris JUSS., etc, degree III species (16 family 18 taxa) were containing; Pyrrosia petiolosa (CHRIST.et BARONI) CHING, Exochorda serratifolia S. MOORE., Aristolochia manshuriensis KOM., etc, degree II species (14 family 18 taxa) were Paeonia japonica MIYABE et TAKEDA, Isopyrum raddeanum (REGEL) MAX., Cypripedium macramthum SW., etc and degree I species (30 family 38 taxa) were Botrychium virginianum (L.) SW., Chloranthus japonicus SIEB., Dictamnus dasycarpus TURCZ., etc. 15 taxa that included Salix hulteni FLODERUS, Celtis choseniana NAKAI, Clematis trichotoma NAKAI, Vicia venosissima NAKAI, etc. were found to be endemic to korea and 21 taxa that included ; Bilderdykia dumetora (L.) DUM., Bilderdykia convolvulus DUM., Lepidium apetalum WILLD., Amorpha fruticosa L. and Symphytum offcinale L., Hibiscus trionum L. etc. were identified as naturalized plants.

EXPERIMENTAL STUDIES ON THE EFFECT OF RLN ANASTOMOSIS REMOVAL ON PHONATION (반회신경 문합과 후윤상피열근 절제가 발성기능에 미치는 영향에 대한 실험적 연구)

  • 편측 성대마비의 음성개선을 위하여 현재까지 알려진 치료방법은 teflon 또는 silicone 주입술, 외과적 성대 내전술, 갑상연골 성형술, 신경재식술, 등이 있으며, 근래까지는 Teflon 또는 Gelform 주입술이 제일 효과적인 방법이라 알려져 있으나, 이는 음의 높낮이 조절능력의 한계가 있고, 신경재식술의 결과 역시 만족스럽지 못한 경우가 많다. 따라서 저자는 발성기능의 생리학적인 측면에서, 보다 효과적인 음성개선의 방법을 찾기 위하여, 사람의 후두와 비교적 유사한 특성을 가진 개를 사용하여 실험을 하였다. 반회신경을 절단하여 편측 성대 마비를 유발한 뒤, 동측의 유일한 외전근인 후윤상피열근을 절제후 절단된 신경을 문합하여, 성대의 외전근 작용의 소실과, 발성시에 주로 작용하는 내전근만의 작용을 유도하여, 공동운동의 차단과, 내전운동의 향상을 기대하였다. 본 실험에 앞서, 임의적으로 신경을 자극시킬 수 있는 신경자극기와, 주위조직으로부터 신경만을 분리하여 자극할 수 있는 전극을 개발하여 사용하였으며, 또한 성대의 발성음을 임의대로 유발시킬 수 있는 생체 내 후두발성 모형(in vivo laryngeal phonation model)을 Moore(1987, 1988)등이 발표한 모형을 토대로 자체 개발하여 사용하였다. 실험은 13마리의 개를, 반회신경 절단군(3 마리), 반회신경 절단 및 후윤상피 열근 절제군(3 마리), 후윤상피열근 절제군(3 마리), 반회신경 문합 및 후윤상피 열근 절제군(4 마리)등으로 나누어 실험하였으며, 3 개월 후, 현수후두경, 후두 내시경, videolaryngoscopy, 신경자극 검사 등으로 성대의 내전운동 및 위치 변화의 관찰, 발성음의 음향분석 및 갑상피열근의 형태학적 변화를 관찰하여 다음과 같은 결과를 얻었다. 1. 반회신경 절단군과 반회 신경 절단 및 후윤상피열근 절제군은, 수술후 3 개월에 성대의 움직임이나 각도의 변화가 없었으며, 음향분석 결과 발성음이 아닌 잡음만이 포착되었다. 즉, 절단된 신경의 문합 없이 후윤상피열근 절제 만으로는 음성개선은 기대할 수 없었다. 2. 후윤상피열근 절제군에서는 수술 후 3 개월에 정상적인 성대의 내전운동이 관찰되었으며, 음향분석상 기본주파수 동요율 및 발성음의 강도는 약간의 증가를 보였다. 즉, 반회신경이 절단되지 않은 상태에서 후윤상피 열근 만을 절제하면, 발성음의 변화는 거의 없었다. 3. 반회신경을 절단후 문합하고 후윤상피열근을 절제한군 에서는 수술후 3 개월에, 성대의 정상적인 내전운동을 보였고, 음향분석상 수술직후에는 잡음만이 포착되었으나, 3 개월 후에는 잡음이 아닌 발성음을 들을 수 있었으며, 이 발성음의 기본주파수 및 강도는 수술전보다 감소되어 있었으나, 기본주파수 동요율은 증가되어 있었다. 즉 이 술식으로 인한 뚜렷한 발성음의 향상이 있었다. 이상의 결과로 미루어, 편측 신경 절단으로 인한 성대마비시, 절단된 신경을 봉합하고 성대의 유일한 외전근인 후윤상피열근을 절제하면, 공동운동의 차단과, 내향운동의 향상으로, 음성개선에 효과적이라고 사료되었으며, 이 방법이 편측 성대마비 환자의 효과적인 음성개선의 치료방법의 하나로 응용될 수 있으리라 생각된다.

Floristic study of Songnisan National Park in Korea (속리산 국립공원의 관속식물상)

  • The flora in the region of Songnisan National Park ($N36^{\circ}28^{\prime}05^{{\prime}{\prime}}{\sim}36^{\circ}46^{\prime}38^{{\prime}{\prime}}$, $E\;127^{\circ}47^{\prime}06^{{\prime}{\prime}}{\sim}127^{\circ}57^{\prime}50^{{\prime}{\prime}}$) was surveyed from April 2000 to July 2006. Vascular plants in 26 collections included a total of 723 taxa: 115 families, 392 genera, 629 species, 4 subspecies, 72 varieties and 18 forms. Ninety-three of these taxa were first records for the region. In the flora of Songnisan National Park, 27 taxa were Korean endemic plants, 19 taxa were rare and endangered plants as designated by the Korea Forest Service. Forty-five of the taxa were with the levels III, IV, V in the floristic regional indicator plants which was designated by Korean Ministry of Environment, and 29 taxa were alien plants now naturalized in Korea. Owing to their unusual phytogeography and low number of populations, the taxa Crypsinus hastatus (Thunb.) Copel., Paeonia japonica (Makino) Miyabe & Takeda, Jeffersonia dubia (Maxim.) Benth. & Hook.f. ex Baker & S.Moore, Bupleurum euphorbioides Nakai, Phacellanthus tubiflorus Siebold & Zucc., Lilium cernuum Kom., Gastrodia elata Blume and Cephalanthera erecta var. subaphylla (Miyabe & Kud ) Ohwi were considered the most important. Based on these results, it is clear that appropriate plans to conserve the natural habitat of Songnisan National Park must be made immediately.

Inhibitory Effect of Stephanniae Tetrandrae Radix Extract on $TNF-{\alpha}$, $IL-1{\beta}$, IL-6 and Nitric Oxide Production in Lipopolysaccharide - Activated RAW 264.7 Cells (방기(防己) 추출물이 LPS로 유도된 Raw 264.7 cell에서의 $TNF-{\alpha}$, $IL-1{\beta}$, IL-6 및 Nitric Oxide Production에 미치는 영향)

  • Tetrandra is the root of Stephania tetrandra 5. Moore (family Menispermaceae), or of Aristolochia frangchi Wu (family Aristolochiaceae). It is a Differ-flavored and cold-property herb acting on the urinary bladder, kidney and spleen meridiands. Known biological effects of this herb are expelling wind to relieve pain and inducing diuresis to alleviate edema. This herb also has anti-inflammatory and anti-hypersensitivity actions. Recent studies have shown that Stephanniae Tetrandrae Radix has antimicrobial effects, namely, a protective effect on acute renal failure induce by gentamicin sulfate and a suppressive effect against clostridium perfringes. However, there is a lack of studies concerning the immunological activities of this herb. The present study was conducted to evaluate the immunological activities of Stephanniae Tetrandrae Radix on the regulatory mechanisms of cytokines and nitric oxide (NO) in Raw 264.7 cells. Cell viability was measured by MTT assay after the treatment of Stephanniae Tetrandrae Radix extract (STRE) and NO production was monitored by measuring the nitrite content in culture medium. COX-2 and iNOS were determined by immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results provided evidences that STRE inhibited the production of nitrite and nitrate (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, $interleukin-1{\beta}(IL-1{\beta})$ and interleukin-6 (IL-6) in Raw 264.7 cells activated with lipopolysaccharide (LPS). These findings showed that STRE could produce some anti-inflammatory effects which might play a role in adjunctive therapy in Gram-negative bacterial infections.

MODFLOW or FEFLOW: A Case Study of Groundwater Model Selection for the Upper Waikato Catchment, New Zealand

  • Groundwater in the Waikatoregion is a valuable resource for agriculture, water supply, forestry and industries. The 434,000 ha study area comprises the upper Waikato River catchment from the outflow of Lake Taupo (New Zealand's largest lake) through to Lake Karapiro (a man-made hydro lake with high recreational value) (Figure 1). Water quality in the area is naturally high. However, there are indications that this quality is deteriorating as a result of land use intensification and deforestation. Compounding this concern for decision makers is the lag time between land use changes and the realisation of effects on groundwater and surface water quality. It is expected that the effects of land use changes have not yet fully manifested, and additional intensification may take decadesto fully develop, further compounding the deterioration. Consequently, Environment Waikato (EW) have proposed a programme of work to develop a groundwater model to assist managing water quality and appropriate policy development within the catchment. One of the most important and critical decisions of any modelling exercise is the choice of the modelling platform to be used. It must not inhibit future decision making and scenario exploration and needs to allow as accurate representation of reality as feasible. With this in mind, EW requested that two modelling platforms, MODFLOW/MT3DMS and FEFLOW, be assessed for their ability to deliver the long-term modelling objectives for this project. The two platforms were compared alongside various selection criteria including complexity of model set-up and development, computational burden, ease and accuracy of representing surface water-groundwater interactions, precision in predictive scenarios and ease with which the model input and output files could be interrogated. This latter criteria is essential for the thorough assessment of predictive uncertainty with third-party software, such as PEST. This paper will focus on the attributes of each modelling platform and the comparison of the two approaches against the key criteria in the selection process. Primarily due to the ease of handling and developing input files and interrogating output files, MODFLOW/MT3DMS was selected as the preferred platform. Other advantages and disadvantages of the two modelling platforms were somewhat balanced. A preliminary regional groundwater numerical model of the study area was subsequently constructed. The model simulates steady state groundwater and surface water flows using MODFLOW and transient contaminant transport with MT3DMS, focussing on nitrate nitrogen (as a conservative solute). Floristic study of Mt. Baekasan (백아산의 식물상)

  • This study presents a discussion of vascular plants found on Mt. Baekasan, Hwasun-gun, Jeollanam-do, based on specimens collected during 32 separate field trips amounting to a total of 36 days conducted between May of 2011 and October of 2016. There are 651 taxa on Mt. Baekasan, consisting of 109 families, 366 genera, 576 species, 5 subspecies, and 70 varieties. Twelve plants endemic to Korea were found, including Cirsium setidens (Dunn) Nakai and Scutellaria insignis Nakai. There are a total of 61 taxa of floristic regional indicator plants specially designated by the Ministry of the Environment. Rare plants such as Jeffersonia dubia (Maxim.) Benth. & Hook. ex Baker & S. Moore in Mt. Baekasan are 17 taxa in total, and red list plants number 8 taxa, including Cremastra variabilis (Blume) Nakai ex Shibata. Nine calciferous plants were found, including Asplenium ruprechtii Sa. Kurata, Ulmus davidiana var. japonica (Rehder) Nakai and Morus cathayana Hemsl. Thirty-three naturalized plants were also found, including Ambrosia artemisiifolia and Rumex acetosella.

Environmental Characteristics of Exochorda serratifolia S.Moore Habitats (가침박달 자생지의 환경특성)

  • The habitats characteristics of Exochorda serratifolia were investigated to compile basic data for conservation and restoration. Natural habitats were located at an altitude of 1 m to 624 m above sea level with angles of inclination ranging from 2 degrees to 81 degrees. A total of 248 vascular plant taxa were identified from 37 quadrats in 16 habitats. The importance value of Exochorda serratifolia is 18.70%, and 5 highly ranked species such as Rhododendron mucronulatum (3.79%), Abelia tyaihyoni (3.27%), Euonymus alatus for. ciliatodentatus, Quercus mongolica (3.23%), and Indigofera kirilowii (2.97%) are considered to be an affinity with Exochorda serratifolia in their habitats. Species diversity was 1.03, dominance and evenness were found to be 0.15 and 0.85, respectively. Average field capacity was 24.19%, the organic matter was 8.21%, pH was 5.52, and available phosphorus was $7.71{\mu}g/g$. Correlation coefficients analysis based on environmental factors, vegetation, and soil analysis shows that the coverage of Exochorda serratifolia is correlated with field capacity and available phosphorus.
