ISSR 분자 마커를 이용한 왕대속 대나무의 유전적 다양성 및 계통 관계 (Genetic Diversity and Phylogenetic Relationship of Genus Phyllostachys by ISSR Markers)

  • ISSR분자 마커를 이용하여 왕대속 대나무의 유연관계를 분석 실시하였다. 전세계적으로 왕대속에 속하는 4종은 경제학적, 생태학적으로 중요한 식물 자원중 하나이다. 총 11개의 ISSR 프라이머 중 9개의 프라이머에서 증폭이 일어 났으며 총 64개의 밴드를 확인 할 수 있었다. ISSR분석결과 왕대속 4종의 대나무에서 70.49%인 43개의 다형현상이 나타났다. 4개의 분류군으로 분류된 왕대속 대나무의 집단내 다양성(Hs)은 0.092,집단간 다양성(Gst)은 0.499로 나타났다. 각 종에 대한 유전자 유동(Nm)은 0.559로 나타났다. 따라서 왕대속에 속하는 4종의 유전자 유동(Nm)은 아주 낮음을 알 수 있었다 4종의 분류군에 대한 유전자 다양성(Ht)은 0.291로 낮게 나타났으며 ISSR 마커로 명확하게 그들은 분류가 되었다. 또한 왕대속의 4종은 단일계통임을 확인할 수 있었다.

어류치사성 Cochlodinium polykrikoides 적조생물의 유전적 진화 및 특성 (Genetic Evolution and Characteristics of Ichthyotoxic Cochlodinium polykrikoides(Gymnodiniales, Dinophyceae))

  • 본 연구는 유해성 적조생물인 Cochlodinium polykrikoides의 유전적 계통진화를 설명하기 위하여 24 종의 개체에 대한 SSU을 대상으로 분석했다. C. polykrikoides는 와편모조류와 밀접한 단일 계통군을 형성하고 있다. Neighbor-joining 혹은 parsimony 분석에 의하면 C. polykrikoides는 Gymnodiniales 보다 Prorocentrals 목 (order)에 훨씬 근접한 100% 유연관계를 보이고 있으며, 과 (family)로 분석해 보면 Gymnodiniaceae에 속해 있고, 특히 Prorocentrum micans와는 매우 밀접한 관계를 보이고 있다. 형태적으로는 Gyrodinium속 (genus)에 가깝지만, 유전적으로는 Gymnodinium 속에 근접하고 있다. C. polykrikoides는 와편모조류 중에서 독립적인 계통군을 유지하고 있다. 따라서 p. micans는 Gymnodinium의 조상으로 추측되며, C. polykrikoides는 P. micans와 Gymnodinium 속의 중간단계인 것으로 보인다.

Molecular Cloning and Characterization of the Estrogen Receptor from the Slender Bitterling (Acheilognathus yamatsutae)

  • Objectives: In order to identify the possibility of slender bitterling (SB) (Acheilognathus yamatsutae) being used as a test species for estrogenic endocrine disrupting chemicals (EEDCs), we carried out the cloning and sequence characterization of the estrogen receptor (ER). Methods: The ER from a slender bitterling was obtained by reverse transcriptase-polymerase chain reaction (RT-PCR), 5'- and 3'-rapid amplification of cDNA ends (5'-RACE and 3'-RACE) and T-vector cloning. The expression of ER mRNA was also analyzed in six tissues (brain, liver, kidney, gill, gonad, and intestines) by real-time PCR. Results: We obtained an ER from the slender bitterling. The SB ER cDNA was 2189 base pairs (bp) in length and contained a 1707 bp open reading frame that encoded 568 amino acid residues. The SB ER amino acid sequence clustered in a monophyletic group with the $ER{\alpha}$ of other fish, and was more closely related to zebrafish $ER{\alpha}$(88% identity) than to the $ER{\alpha}$ of other fish. The SB ER cDNA was divided into A/B, C, D, E and F domains. The SB ER has conserved important sequences for ER functions, such as the DNA binding domain (D domain), which are consistent with those of other teleosts. Conclusions: The ER of the slender bitterling could provide basic information in toxicological studies of EEDCs in the slender bitterling.

First Description of Coleosporium plectranthi Causing Perilla Rust in Korea

  • Perilla rust is a damaging disease in perilla cultivation in Korea. Its causal agent was identified as Coleosporium plectranthi based on descriptions of morphological characteristics of spores and spore-producing fruiting structures(in uredinial and telial stages from perilla and in aecial stage from the alternate host pine) collected in 15 locations in Korea during the disease survey from 2004 to 2006. These characteristics were yellow or orange uredinium; globose or ellipsoid urediniospore of $20.8{\mu}m{\times}18{\mu}m$ in size; verruca of $0.3mm{\times}1.2mm$; orange telium; one-celled, oblong ellipsoid teliospore of $63.1{\mu}m{\times}19.7{\mu}m$ with one-layered crusts or four-celled(when mature), internal basidium of $64.2{\mu}m{\times}19.7{\mu}m$; ellipsoid to globoid basidiospore of $20.3{\mu}m{\times}12{\mu}m$; type 2 spermogonium; yellow, broadly ellipsoid peridial cell of $35.6{\mu}m{\times}23.1{\mu}m$; and broadly ellipsoidal or subglobose aeciospore of $25.9{\mu}m{\times}18.8{\mu}m$. Phylogenetic analysis of 28S rDNA sequences revealed the closest relatedness to those of the genus Coleosporium, a monophyletic group distinguished from other rust fungi and divided into two main lineages, one of which was C. plectranthi grouped with high bootstrap value(96%). In pathogenicity test, both aeciospores and urediniospores caused rust development on perilla leaves. This is the first description of C. plectranthi causing perilla rust with the first findings of its telial stage on perilla and the first rust disease on the aecial host in Pinus densiflora. These aspects would provide basic information for the development of control measures of the disease.

Developmental and Phylogenetic Characteristics of Stellantchasmus falcatus (Trematoda: Heterophyidae) from Thailand

  • This study aimed to investigate the infection status, worm development, and phylogenetic characteristics of the intestinal trematode, Stellantchasmus falcatus. The metacercariae of S. falcatus were detected only in the half-beak (Dermogenus pusillus) out of the 4 fish species examined. Their prevalence was 90.0%, and the intensity of infection was 919 metacercariae on average. Worms were recovered from 33 (97.1%) of 34 chicks that were experimentally infected with 200 S. falcatus metacercariae each, and the average recovery rate was 43.0%. The body size and inner organs of S. falcatus quickly increased in the experimental chicks over days 1-2 post-infection (PI). In addition, ITS2 sequence data of this parasite were analyzed to examine the phylogenetic relationships with other trematodes using the UPGMA method. The results indicated that the ITS2 sequence data recorded from trematodes in the family Heterophyidae appeared to be monophyletic. This study concluded that D. pusillus serves as a compatible second intermediate host of S. falcatus in Thailand and that S. falcatus can develop rapidly in the experimental chicks. Data collected from this study can help to close the gap in knowledge regarding the epidemiology, biology, and phylogenetic characteristics of S. falcatus in Thailand.

Phylogenetic position of eight Amphora sensu lato (Bacillariophyceae) species and comparative analysis of morphological characteristics

  • Amphora Ehrenberg ex Kutzing sensu lato is a common and widespread benthic diatom genus with a taxonomy that has been under continual revision, particularly based on molecular analyses. Although Amphora species have been studied using modern microscopy in recent years, there has not been much progress on molecular characterization of the species, especially in Asia. In this study of Amphora, sampling was carried out from September 2009 to August 2010 in Korean coastal waters. The morphological and molecular characteristics of eight Amphora sensu lato were examined: Amphora marina, A. proteus, Halamphora costata, H. coffeaeformis, H. eunotia, H. holsatica, H. terroris, and Halamphora sp. Based on previous accounts, morphology suggested that A. marina and A. proteus belong to the subgenus Amphora Cleve, which have smooth girdle bands and rather coarse and very distinct areolae on the valve. The other species, H. coffeaeformis, H. costata, H. eunotia, H. holsatica, H. terroris, and Halamphora sp. belong to the subgenus Halamphora Cleve, which was recently elevated to generic status by Levkov 2009, have plicate girdle bands, puncta which do not form straight longitudinal lines, valves which have a narrow ventral portion and apices that are generally rostrate-capitate and recurved. In agreement with analysis based on morphological characteristics, phylogenetic analysis based on small subunit rDNA suggested that the eight Amphora sensu lato species were not a monophyletic group as the morphological classification. Also, the results of molecular work and statistical analysis on all these Amphora sensu lato combined with phylogenic analysis on our geographically representative samples give strong evidence that Halamphora Levkov is independent of Amphora Cleve. Furthermore, in this study, Amphora terroris was transferred Halamphora as Halamphora terroris (Ehrenberg) Wang comb. nov. and Amphora marina was recorded for the first time in Korea.

형태학적 분계분석에 의한 한국산 꿩의다리속(Thalictrum L.) 식물의 계통학적 연구 (Systematics of Korean Thalictrum L. based on a morphological cladistic analysis)

  • 한국산 꿩의다리속 식물의 계통 및 유연관계를 밝히기 위해 39개의 형태 형질을 바탕으로 Maximum parsimony와 Neighbor-joining을 분석하였다. 식물재료는 한국 고유종을 포함해 내군 21분류군과 외군 1분류군이 사용되었다. 좀꿩의다리절과 금꿩의다리절은 각각 100% 부트츠랩 값과 83% 부트츠랩 값으로 단계통군을 형성하였고, 금꿩의다리절은 좀꿩의다리절의 자매군으로 나타났다. 꽃꿩의다리절과 산꿩의다리절은 측계통군을 형성하였다. 그늘꿩의다리를 제외한 산꿩의다리절은 방사상분지로 나타났다. 꿩의다리절과 발톱꿩의다리절은 각각 독립적인 분계군를 형성하였다. 꿩의다리속 식물에서 주요한 형질인 수술의 모양은 곤봉형이 가장 원시적이며, 원시적인 곤봉형에서 봉형, 그리고 실형으로 파생되는 것으로 나타났다. 수술의 형태는 수분방법(충매, 풍매), 개화시 꽃받침의 존재유무 등 꿩의다리속 식물의 꽃 진화에도 관련되어 있다고 판단된다.

Mitochondrial Genome of Spirometra theileri Compared with Other Spirometra Species

  • This study was carried out to provide information on the taxonomic classification and analysis of mitochondrial genomes of Spirometra theileri. One strobila of S. theileri was collected from the intestine of an African leopard (Panthera pardus) in the Maswa Game Reserve, Tanzania. The complete mtDNA sequence of S. theileri was 13,685 bp encoding 36 genes including 12 protein genes, 22 tRNAs and 2 rRNAs with absence of atp8. Divergences of 12 protein-coding genes were as follow: 14.9% between S. theileri and S. erinaceieuropaei, 14.7% between S. theileri and S. decipiens, and 14.5% between S. theileri with S. ranarum. Divergences of 12 proteins of S. theileri and S. erinaceieuropaei ranged from 2.3% in cox1 to 15.7% in nad5, while S. theileri varied from S. decipiens and S. ranarum by 1.3% in cox1 to 15.7% in nad3. Phylogenetic relationship of S. theileri with eucestodes inferred using the maximum likelihood and Bayesian inferences exhibited identical tree topologies. A clade composed of S. decipiens and S. ranarum formed a sister species to S. erinaceieuropaei, and S. theileri formed a sister species to all species in this clade. Within the diphyllobothridean clade, Dibothriocephalus, Diphyllobothrium and Spirometra formed a monophyletic group, and sister genera were well supported.

Asia and Europe: So Distant So Close? The Case of Lipoptena fortisetosa in Italy

  • In Europe, 5 Lipoptena species have been recorded, including Lipoptena fortisetosa. This species, native to Asian countries, was described as a parasite of sika deer and its appearance in Europe dates back to more than 50 years ago. Lipoptena fortisetosa has been recently reported in Italy, sharing its hosts with Lipoptena cervi. A morpho-molecular approach was developed to determine the phylogenetic interrelationship of Italian and Asian CO1 haplotypes sequenced from Lipoptena fly individuals collected in Italy, and their DNA sequences were compared with conspecifics available in GenBank; morphological key-characters (terminalia) of L. fortisetosa were compared with the original description. Two haplotypes were recorded from Italy and assigned to L. cervi and L. fortisetosa, respectively. The latter was part of the monophyletic clade L. fortisetosa, along with 2 Central European and 2 Korean haplotypes (100% identical to one of the Korean haplotypes); moreover, Italian L. fortisetosa female terminalia were consistent with the original description of Asian individuals. Pending more in-depth investigations, this study provides a first answer to the hypothesis of the recent colonization of Italy by L. fortisetosa from Asia as we did not detect any obvious and stable morphological and molecular differences in specimens from the 2 geographical areas. The presence of the sika deer in Europe was retraced and the possible route traveled by the parasite from Asia and the eco-biological factors that may have enhanced its settlement are discussed.

Phylogenetic analysis of Neottia japonica (Orchidaceae) based on ITS and matK regions

  • To elucidate the molecular phylogeny of Neottia japonica, which is a terrestrial orchid distributed in East Asia, the internal transcribed spacer (ITS) of nuclear DNA and the matK of chloroplast DNA were used. A total 22 species of 69 accessions for ITS and 21 species of 114 accessions for matK phylogeny were analyzed with the maximum parsimony and Bayesian methods. In addition, we sought to establish a correlation between the distribution, morphology of the auricles and genetic association of N. japonica with phylogenetic data. The phylogenetic results suggest that N. japonica is monophyletic and a sister to N. suzukii in terms of the ITS phylogeny, while it is paraphyletic with N. suzukii in terms of the matK phylogeny. N. japonica and N. suzukii show similar morphologies of the lip and column, they both flower in April, and they are both distributed sympatrically in Taiwan. Therefore, it appears to be clear that N. japonica and N. suzukii are close taxa within Neottia, although there is incongruence between the nrDNA and cpDNA phylogenies of N. japonica. The incongruence between the two datasets may have various causes, meaning that further studies are needed to confirm the evolutionary process of N. japonica. The phylogenetic status of N. kiusiana, which was not included in previous studies, was as a sister to N. nidus-avis. Meanwhile, the ITS and matK phylogenies are unsuitable for identifying genetic associations with the characteristic of auricles. The phylogenetic topologies of Korean, Taiwanese and mainland Chinese individuals suggest that the populations of N. japonica in Korea originated from China's mainland and island areas. The characterization of regional gene differences could provide useful preliminary data for future studies.
