몽골산 약용식물 수종의 항비만 활성 탐색을 위한 기초 연구 (Basic research for exploring anti-obesity activity of several medicinal plants from Mongolia)

  • Objective : Obesity in modern society has a significant impact on pathological, psychological and social problems. Therefore, many studies on obesity treatment using herbal extracts with fewer side effects have been conducted. This study was designed to investigate the effect of inhibiting fat accumulation in vitro in order to Mongolian medicinal plants find anti-obesity candidate among. Methods : We measured pancreatic lipase inhibitory activity in Mongolian medicinal plants extract. Cytotoxicity of these extracts was monitored in 3T3-L1 cells by MTT assay. In addition, the anti-obesity effects was confirmed at concentrations of 0.2, 0.1 and 0.05 mg/㎖ through Oil red O staining. Results : Among Mongolian medicinal plants, Rheum undulatum roots in September (RURS), Paeonia anomala L. (PAL), and Fragaria orientalis (FO) showed the highest pancreatic lipase inhibitory activity. As a result of the MTT assay, more than 80% was judged to be non-toxic, and the concentration was determined, and as a result of evaluating the lipid accumulation inhibitory effect, 6 types were selected as candidates. Conclusion : Based on these results, the top 7 species expected to be used as anti-obesity functional materials were selected. However, additional efficacy verification and mechanism of action need to be established in the future. So, it is expected that the medicinal plants verified through this will be used as functional materials for the prevention and treatment of obesity.

  • Mongolian gerbil (Meriones unguiculatus) has been as an animal model for studing the neurological diseases such as stroke and epilepsy because of the congenital incompleteries in Willis circle, as well as the investigation of water metabolism because of the long time-survival in the condition of water-deprived desert condition, compared with other species animals. In order to accomplish the this research, first of all another divided the laboratory animals 5 groups of which each group include the 5 animals. In this study were investigated the histological structure in the kidney, measured the plasma osmolalities at the time of sacrifice of indivisual animals, and the body weights every day during water-deprived. The results obtained in this study were summarized as followings: 1. The body weights and decreasing rates of the body weight in water-deprived mongolian gerbil groups were continuosly decreased. 2. The plasma osmolalities were increased from the 5th water-deprived day, after then the gradually increasing reached nearly its equilibrium state at the 10th water-deprived day. 3. The urine volumes were abruptly decreased from the 2th water-deprived day, after then the gradually decreasing patterns were reached nearly its equilibrium state at the 10th day, and stopped the 11th day. 4. In the light microscopical observation of the kidney, glomerular capillary loop thickening, mesangial matrix increasing, sclerosis, glomerular cystic atrophy, interstitial fibrosis, tubular dilatation, mononuclear interstitial inflammation, interstitial mineralization, and hyperplasia of the collecting duct epithelium in the cortex area, were observed from the 10th water deprived day, and the lesions were gradually severe changed as the time lapse. 5. In the electron microscopical findings of the kidney, the degenerative changes of endothelial cell, podocyte and mesangial cell in glomeruli were initially observed on the 10th water-deprived day as well as the degeneration of microvilli and intracellular organelle in the renal tubules.

거풍지보단(祛風至寶丹)이 Mongolian Gerbil의 가역성 전뇌허혈 모델에 미치는 영향 (The Effect of Geupoongjibo-dan Extracts on Reversible Forebrain Ischemia in Mongolian Gerbil)

  • Objectives : The purpose of this investigation is to evaluate the effect of Geupoongjibo-dan Extracts on Reversible Forebrain Ischemia in Mongolian Gerbils. Methods : The change rate of water content in cerebral tissues, the numercal change of the CA1 pyramidal neuron in the hippocampus, the change of delayed neuronal death(necrosis apoptosis) through light microscopy, the reactivity change of glycoprotein in neuronal membrane and the ultrastructural change of pyramidal neuron through electron microscopy caused by dalayed neuronal death were investigated. Results : 1. The change rate of water content in the normal group showed 78.90% on the third day, and 79.12% on the seventh day after an attack of ischemia. The rate in the control group showed 82.25% and 85.13%, respectively. The rate in the sample group showed a significant decrease: 81.72% and 83.66%. 2. Light microscopy revealed that the cells, continuous and systematic forms in the pyramidal cells of hippocampus, changed into discontinuous and unsystematic forms in the normal group when compared with the control group. The cells were less damaged in the sample group. 3. The mean of the numerical change of the CA1 pyramidal neurons in the hippocampus was 104 in the normal group. The mean of the control group was decreased to 27. The mean of the sample group was 44. 4. TUNEL staining examination reveals that the whole part of the hippocampus of the normal group had negative reactivity. As far as CA1 pyramidal neurons in the hippocampus, the control group had positive reactivity. The sample group was more positive than the control group. 5. Electron microscopy reveals that the ischemic injury of the control group had both necrotic and apoptotic morphology. The sample group was less necrotic, and more apoptotic morphology than the control group. 6. Lectin histochemisrical examination reveals that the normal group had positive reactivity to PNA and SBA in interneuron, and weak positive reactivity to WGA Con A LCA in intercelluar space. The reactivity to PNA and WGA decreased in the control group. The reactivity to PNA and WGA tended to increase in the sample group. Conclusions : The data shows that the effect of Geupoongjibo-dan Extracts on Reversible Forebrain Ischemia in MG is a significant result.

Discovery of Parvatrema duboisi and Parvatrema homoeotecnum (Digenea: Gymnophallidae) from Migratory Birds in Korea

  • Adult worms of Parvatrema spp. (Digenea: Gymnophallidae) were found in the intestines of 2 species of migratory birds, i.e., a great knot, Calidris tenuirostris, and 2 Mongolian plovers, Charadrius mongolus, in the coastal area of Gunsan-si, Jeollabuk-do in October 2009. The recovered Parvatrema worms were 79 in total number and composed of 2 species. The worms from a great knot were $289{\mu}m$ in length with the oral and ventral sucker ratio of 2:1. They had a single vitellarium, and their intrauterine eggs were $25.0{\times}17.5{\mu}m$ in size. These findings were compatible with P. duboisi (Dollfus, 1923) Bartoli, 1974 (syn. P. timondavidi Bartoli, 1963). The worms recovered from the Mongolian plovers were smaller in length than P. duboisi and had 2 vitellaria. The oral and ventral sucker ratio was 2.5 : 1, and the eggs were $17.5{\times}8.8{\mu}m$ in size. These worms were assigned to be P. homoeotecnum James, 1964. This is the first report on the natural final hosts of Parvatrema spp. in Korea.

Chemical Composition of Mongolian Sea-buckthorn (Hippophae rhamnoides L.) Fruits

  • Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.

타락(駝酪)의 한반도 수용과 의미 변천 (Changed Conception of Korean Tarag)

  • Objective : Tarag refers to fermented milk, which has been recognized as daily food of summer in nomadic culture. Also, tarag is cleary defined as a fermented milk product in most of east Asian medical texts. When it comes to tarag-juk described in Korean medical texts, however, there is no definite distinction between milk porridge and tarag-juk. This paper is aimed at finding out whether tarag was merely meaning milk in Joseon. Method : Historical documents of related historical stage, Tibetan and East Asian medical texts, and some cookbooks are mainly consulted, as well as other sources that contains the perception of tarag in Korean history. Result : Tarag is documented as fermented milk in the medical texts of herbs, while tarag-juk is defined as milk porridge in some medical texts in Korea. In one of the Mongolian-Korean dictionary of 18C, milk tea is explained as tarag tea. Conclusion : Although there is not much evidence to back up this conclusion with satisfaction, it would not be to much to say that conception of tarag met some changes from yogurt to boiled milk during Joseon dynasty.

Early Diagnosis for Mucopolysaccharidosis I - A 6-month-old Female Infant Presenting with Gibbus, Hirsutism and Mongolian Spots in a Well Baby Clinic

  • Mucopolysaccharidosis (MPS) I is a rare, progressive and multisystemic disease with insidious initial signs and symptoms, and making an early diagnosis can be a challenge for the first-line general medical practitioner. We report a 6-month-old girl who was brought to our well baby clinic for regular immunization with the manifestations of lumbar gibbus, hirsutism, large Mongolian spots over back and buttock, and mild bilateral legs spasticity noticed by the general pediatrician, and then newly diagnosed with MPS I after referral to the geneticist in time. Her surgical history included inguinal hernia repair at 1 month old, $CO_2$ laser supraglottoplasty for laryngomalacia and tracheostomy due to chronic respiratory failure with ventilator dependence at 2 months old. Understanding and identification of the early signs and symptoms of this disease have the potential to early diagnosis and timely appropriate treatment, which could contribute to a better clinical outcome.

Helicobacter pylori의 감수성과 숙주 연령과의 상관성 연구 (Elder ages decreases the susceptibility for Helicobacter pylori infection in an animal model)

  • Helicobacter pylori (H. pylori) is an important bacterial pathogen that causes chronic gastritisand is associated with gastroduodenal ulcer disease, adenocarcinoma of the distal stomach, and gastricH. pylori infection associated with host agehave not been well-defined in human. To evaluate the difference in host susceptibility to infection in relationto age of acquisition of H. pylori infection, we designed an experiment involving inoculation of H. pyloriATC 43504 at different ages of Mongolian gerbils. H. pylori was inoculated at 5 weeks and 18 monthsof age, as representatives of early and late infection, respectively. Animals were sacrificed 1 week and 4weeks after challenge, and the stomach was removed from each animal for bacterial culture, histologicalexamination, and polymerase chain reaction test. 5 week-old gerbils revealed infection andmaintained continuously its infection until 4 weeks. However, old gerbils did not maintained H. pyloriinfection. These data suggest the insusceptibility of H. pylori in old Mongolian gerbils and the importanceof animal ages for successful animal experimental infection. Also, the results demonstrated that earlyinfection of H. pylori increases its host susceptibility, as compared to the case with later infection, possiblybecause of differences in host gastric mucosal factors and imunologic responses.

Vacor 유발당뇨 모래쥐의 부신수질의 크롬친화성세포와 신경절세포의 미세구조 (Ultrastructural Changes in the Adrenal Chromaffin Cells and Ganglion Cells in the Adrenal Gland of Vacor-Induced Diabetic Mongolian Gerbil)

  • The ultrastructural changes of adrenal chromaffin cells and ganglion cells in the adrenal gland of vacor-induced diabetic Mongolian gerbils were studied by electron microscopy. After one month of vacor-induced diabetes, some chromaffin cells were filled with dense bodies and large cytosomes with formy contents. Most of degenerating axon terminals were observed on chromaffin cells. A few macrophages were found among chromaffin cells at one month after induction. Several of these macrophages were filled with numerous phagosomes. After one month of vacor-induced diabetes, the ganglion cells showed increase in numbers of dense bodies and degenerating dendrites compared with the normal ganglion cells. Both electron dense and lucent types of degenerating axon terminals were found in interstitial space of the ganglion cells. Degenerating unmyelinated and myelinted axons contained dense and lamellar bodies. The satellite cells and macrophages with engulfed degenerated axon terminals were observed. After three months of vacor-induced diabetes, the unmyelinated and myelinated axons showed degenerative changes, whereas no structural changes could be demonstrated in adrenal ganglion and chromaffin cells. The satellite cells and macrophages containing partially digested debris were still commonly observed in the interstitial space of adrenal medulla. These results suggest that the degenerative changes occur in the adrenal ganglion cells as well as adrenal chromaffin cells of vacor-induced diabetic Mongolian gerbils.

Studies on the Genetic Relationships of Sheep Populations from East and South of Central Asia

  • Hu sheep was sampled randomly from Huzhou city, Zhejiang province, China. Of the 11 genetic markers from the blood examined by starch-gel and cellulose acetate electrophoresis, polymorphisms in Hu sheep were found for 10 loci, i.e. post-albumin (Po), transferring (Tf), alkaline phosphatase (Alp), leucine aminopeptidase (Lap), arylesterase (Ary-Es), hemoglobin-$\beta$ (Hb-$\beta$)、Xprotein(X-p), carbonic anhydrase (CA), catalase (Cat) and lysine (Ly). The same data except for Po locus were collected from another 14 sheep breeds from China and other countries, in order to ascertain their genetic relationships with one another and with the Hu sheep. The sheep populations from the east and south of Central Asia can be classified into three genetic groups: 'Mongolian sheep', 'South Asian sheep' and 'European sheep'. The Hu sheep belong to the 'Mongolian sheep' group.
