발달중인 Mongolian gerbil의 전뇌에서 TH 면역반응세포의 분포에 관한 연구 (The study on the TH(tyrosine hydroxylase) immunoreactive cells in forebrain of developing mongolian gerbil)

  • The immunocytochemical localization of tyrosine hydroxylase(TH) is examined in the developmental forebrain of mongolian gerbil in order to determine changes in the distribution and cytology of neurons. At each of the different developmental stage, including prenatal(E)14, E16, E18, E20, postnatal(P)0, P2, P4 days, mongolian gerbils were sacrificed. In E14, TH-IR cells were predominantly round or oval in shape and their processes were very short. In olfactory blub, TH-IR cells were begun to appear from E20. In the striatum, we observed only fibers of TH-IR at all ages. From E16, TH-IR perikarya and fiber were present in periventricular nucleus and paraventricular nucleus of hypothalamus. The changes from the early to the late prenatal stages of development appear to be the result of an increase in number of TH-IR perikarya and processes. These results were shown slight differences to other rodents.

  • TrkA is an essential component of the high affinity NGF receptor necessary to the mediate biological effects of the neurotrophins NGF. Here we report on the expression of TrkA in the olfactory bulb and basal nucleus of Mongolian gerbil brain during the postnatal development. The expressions of TrkA were identified in a immunohistochemical method. Higher levels of TrkA immunoreactivity were detected in septum than that in olfactory bulb and caudate putamen (CPu). But TrkA was not observed before postnatal days (PND6) in olfactory bulb and PND9 in CPu. No TrkA-positive cell was detectable in the olfactory fiber layer. Several regions, such as olfactory bulb and CPu, showed weak labeling. These data show that expression of TrkA is developmentally regulated during postnatal Mongolian gerbil brain development and suggest that high affinity neurotrophinreceptors mediate a transient response to neurotrophins in many regions during the brain ontogeny.

  • Mongolian gerbil (Meriones unguiculatus) has been as an model animal for studing the neurological disease such as stroke and epilepsy because of the congenital incompleteries in Willis circle, as well as the investigation of water metabolism because of the long time-survival in the condition of water-deprived desert condition, compared with other species animal. Aquaporin 2 (AQP2) expressed at the surface of principal cells in collecting duct results from an equilibrium between the AQP2 in intracellular vesicles and the AQP2 on the plasma membrane. Aquaporin 4 (AQP4), which is expressed in cell in a wide range of organ, is also present in the collecting duct principal cells where this is abundant in the basolateral plasma membranes and represent potential exit pathways from the cell for water entering via AQP2. In this research, we divide 3 groups of which each group include the 5 animals. In the study of 7 or 14 days water restricted condition, we investigated the AQP2 and AQP4 by using a quantitative immunohistochemistry in the kidney. The results obtained in this study were summarized as followings. AQP2 is abundant in the apical plasma membrane and apical vesicles in the collecting duct principal cell and at rare abundance in connecting tubules. In the water-deprived Mongolian gerbil kidney, expression of AQP2 was continuosly increased in the cortical collecting duct and inner medullary collecting duct. This increase was both the apical region and cytoplasm. AQP4 is mainly expressed in the inner medulla, although some expression is also noted in the more proximal segment. In the water-deprived Mongolian gerbil kidney, AQP4 was also increased in the inner medullary collecting duct. Immunoactivity was increased in entire inner medullary collecting duct and newly detected in cytoplasm of principal cell. These findings suggest that increased levels of AQP2 and AQP4 in the cortical and inner medulalry collecting duct may play a important role for maintain fluid balance in the water-deprived kidney.

Helicobacter pylori의 병원성 비교를 위한 gerbil의 수침구속스트레스 모델 (Water-Immersion-Restraint Stress model in Mongolian gerbil forcomparison of pathoaenicity of Helicobacter pylori strains)

  • Helicobacter pylori (H. pylori)-infection is an important pathogen of stomach cancer after chronic gastritis and ulceration in the stomach and duodenum. However, the virulences of H. pylori strains have not been well-defined between clinical isolates. This study was designed to establish water-immersion-restraint stress (WIRS) model in mongolian gerbil for comparison of pathogenicity of H. pylori strains. To determine an optimal duration time for WIRS model in gerbil, 5-week-old Mongolian gerbils were divided into different groups by WIRS duration time. After graded duration of WIRS, the macroscopic ulcer index (UI) was measured with the stomach and duodenum of sacrificed animal. There were no significant differences between male and female in same duration group. However, the UI increased significantly in a time-dependent fashion. The group of 6 hours-WIRS animals showed severe hemorrhage and ulceration in their stomach and duodenum. On the other hand, the very mild lesions induced in 2 hours-treated animals. Therefore, we determined an optimal duration time for WIRS model in gerbil as 4 hours. Thereafter, we evaluated whether this WIRS model in gerbil could be used as an useful tool for in vivo comparison of pathogenicity of H. pylori strains by enhancement of pathological severity in H. pylori-infected gerbils. Mongolian gerbils were divided into H. pyloriinfected and PBS-inoculated groups. Thereafter, they were divided again into 4 hours-WIRS and no WIRS subgroups. After treatment, the severity of pathological changes was evaluated in a same manner with previous duration-determining experiment. When the animals were exposed to WIRS, the UI was significantly higher in the infected group than in the uninfected group. These results suggested that the established gerbil-WIRS model in this study enhanced effectively the severity of pathogenic changes in the H. pylori-infected Mongolian gerbils and could be used as an useful tool for in vivo comparison of pathogenicity of H. pylori strains.

Localization of Quantitative Trait Loci for Bone Mineral Density on Chromosome 13 in the Mongolian Population

  • Although the genetic basis for bone mineral density (BMD) has been studied by many groups so far, genes responsible for this complex trait has not been completely revealed. In order to localize quantitative trait loci (QTLs) for BMD variation in Asian population, the study was designed using a group of Mongolian population, a genetically closed population with a homogeneous lifestyle. BMD was measured at the left and right wrists and ankles using DEXA in 1,082 participants from 142 families. Genotyping of 13 polymorphic microsatellite markers on chromosome 13 (average spacing 8-9 cM) and two-point and multipoint linkage analysis were performed. In two-point linkage analysis, we identified two markers, D13S175 (6.03 cM) and D13S265 (68.73 cM) that had LOD scores greater than 1 for left ankle (LOD=2.09, LOD=1.49, respectively). We also found a marker D13S175 (6.03 cM) with a high LOD for left wrist (LOD=1.49) and the markers D13S265 (68.73 cM) and D13S217 (17.21 cM) for the right wrist (LOD= 1.82, LOD= 1.62, respectively). Among these significant marker regions, only two regions at 17 cM (13p11) and 65 cM (13q21) for the right wrist overlapped with major QTLs reported in following multipoint linkage analysis (LOD= 1.7549, LOD=1.4462, respectively). This study provides the possible evidence of the presence of QTLs affecting right wrist BMD in Mongolian populations on 13p11 and 13q21. Modest evidence was also found for genes affecting left ankle and left wrist BMD on 13p13.

Hepatitis B, C, and D Virus Infections and AFP Tumor Marker Prevalence Among the Elderly Population in Mongolia: A Nationwide Survey

  • Objectives: Infections with hepatitis B, C, and D virus (HBV, HCV, and HDV) are a major public health problem and lead to serious complications such as cirrhosis and hepatocellular carcinoma. We aimed to determine the seroprevalence of hepatitis B surface antigen (HBsAg), anti-HCV, anti-HDV immunoglobulin G, alpha-fetoprotein (AFP), and dual and triple hepatitis virus infections in Mongolia. Methods: A total of 2313 participants from urban and rural regions were randomly recruited for this cross-sectional study. A questionnaire was used to identify the risk factors for hepatitis virus infections, and the seromarkers were measured using immunoassay kits. Results: Among all participants, the prevalence of HBV, HCV, and HDV was 15.6%, 36.6%, and 14.3%, respectively. The infection rates were significantly higher in females and participants with a lower education level, rural residence, older age, and a history of blood transfusion. HBV and HCV co-infection was found in 120 (5.2%) participants and HBV, HCV, and HDV triple infection was detected in 67 (2.9%) participants. The prevalence of elevated AFP was 2.7%, 5.5%, and 2.6% higher in participants who were seropositive for HBsAg (p=0.01), anti-HCV (p<0.001), and anti-HDV (p=0.022), respectively. Elevated AFP was more prevalent in participants co-infected with HBV and HCV (5.8%, p=0.023), HBV and HDV (6.0%, p<0.001), and triple-infected with HBV, HCV, and HDV (7.5%) than in uninfected individuals. Conclusions: Nearly half (49.8%) of the study population aged ≥40 years were infected with HBV, HCV, or HDV, and 22.4% had dual or triple infections.

Upregulation of STK15 in Esophageal Squamous Cell Carcinomas in a Mongolian Population

  • Background: The STK15 gene located on chromosome 20q13.2 encodes a centrosome-associated kinase critical for regulated chromosome segregation and cytokinesis. Recent studies have demonstrated STK15 to be significantly associated with many tumors, with aberrant expression obseved in many human malignancies. The purpose of this study was to investigate expression of STK15 in esophageal squamous cell carcinomas (ESCCs) in a Mongolian population. Methods: Two non-synonymous single nucleotide polymorphisms in the coding region of STK15, rs2273535 (Phe31Ile) and rs1047972 (Val57Ile) were assessed in 380 ESCC patients and 380 healthy controls. We also detected STK15 mRNA expression in 39 esophageal squamous cell carcinomas and corresponding adjacent tissues by real time PCR. Results: rs2273535 showed a significant association with ESCC in our Mongolian population (rs227353, P allele = 0.0447, OR (95%CI) = 1.259 (1.005~1.578)). Real time PCR analysis of ESCC tissues showed that expression of STK15 mRNA in cancer tissues was higher than in normal tissues (p = 0.013). Conclusions: Our study showed that functional SNPs in the STK15 gene are associated with ESCC in a Mongolian population and up-regulation of STK15 mRNAoccurs in ESCC tumors compared adjacent normal tissues. STK15 may thus have an important role in the prognosis of ESCC and be a potential therapeutic target.

Linkage and association scan for tanning ability in an isolated Mongolian population

  • Tanning ability is important, because it represents the ability of the skin to protect itself against ultraviolet (UV) radiation. Here, we sought to determine genetic regions associated with tanning ability. Skin pigmentation was measured at the outer forearm and buttock areas to represent facultative and constitutive skin color, respectively. In our study population consisting of isolated Mongolian subjects, with common histories of environmental UV exposure during their nomadic life, facultative skin color adjusted by constitutive skin color was used to indicate tanning ability. Through linkage analysis and family-based association tests of 345 Mongolian subjects, we identified 2 potential linkage regions regulating tanning ability on 5q35.3 and 12q13.2, having 6 and 7 significant single nucleotide polymorphisms (SNPs), respectively. Those significant SNPs were located in or adjacent to potential candidate genes related to tanning ability: GRM6, ATF1, WNT1, and SILV/Pmel17.

발생중인 Mongolian gerbil의 전뇌에서 NGF, GFAP 및 CNTF의 분포 (Immunohistochemical Localization of Nerve Growth Factor, Glial Fibrillary Acidic Protein and Ciliary Neurotrophic Factor in the Forebrain of the Developing Mongolian Gerbil)

  • 중추신경계는 신경원(neuron)과 이를 지지해주는 신경아교세포(neuroglia)로 이루어져 있다. 발생과정중 신경원의 발달은 NGF(neruv growht factor)에 의해 관찰이 가능하고, 아교세포들중 별모양아교세포는 GFAP(Glial fibrillary acidic protein)항체로 밝혀낼 수 있다. CNTF(Cillary neurotrophic factor)는 이전에는 운동신경원의 발생 및 유지에 있어 중요한 역할을 하는 인자로 알려져 왔으며, 최근에는 발생과정 중 NGF와 GFAP의 역할에 도움을 주는 것으로 알려졌다. 본 연구에서는 이러한 NGF, GFAP, CNTF를 발생과정 중(임신 15,17,19,21일, 출생 1,2,3,일, 1,2,3주)의 몽골리안 저빌의 전뇌에서 시간에 따른 분포를 광학현미경, 형광염색을 통한 공초점현미경 및 전자현미경을 통해 알아보고자 하였다. 전뇌에서 NGF는 발생 19일령에서부터 대뇌피질에서 관찰되기 시작해서 후각망울, 해마체 및 diagonal band에 관찰되었고, 출생 3일령에서 가장 강한 염색성을 보였으며 그 반응은 출생 3주까지도 관찰되었다. GFAP는 출생17일령에서 뇌실로부터 아교세포가 관찰되는 형태가 보였으며, 외측뇌실과 제3뇌실에서부터 피질로 이동하는 형태로 관찰되었다. 또한 후각망울의 과립층, 대뇌피질 및 해마체에서 관찰되었으며, 출생 2일령에서 가장 강한 반응을 나타내었다. CNTF는 신경원과 신경아교세포에서 관찰되었으며, NGF와 GFAP와는 달리 출생 전에서는 관찰되지 않다가 출생 1일령부터 후각망울 및 대뇌피질에서 약하게 관찰되기 시작되었으며, 이러한 반응은 출생 2주령에서 잘 관찰되었다. 전자현미경상에서는 신경원과 신경아교세포에서 특징적인 구조는 관찰되지 않았으나, 각각의 항체에 대한 반응이 나타난 세포에서는 사립체와 형질내세망과 같은 세포소기관들이 많이 관찰되었다. 이러한 Mongolian gerbil 전뇌에서의 NGF, GFAP 및 CNTF의 분포는 비슷한 임신일령의 설치류와 거의 유사하게 관찰되었으며, 배아과정 및 출생 후 발달에 따른 전뇌에서의 신경원과 신경아교세포에서의 분포를 볼 수 있었다.
