한국산 비짜루목 및 백합목(백합강)에 대한 분류학적 재검토 (A taxonomic review of Korean Asparagales and Liliales (Liliopsida))

  • 분자생물학적 자료에 의한 한국산 백합강 식물에 대한 분류학적 재검토를 시도하였다. 2가지의 다른 자료, 즉 엽록체 DNA인 rbcL 및 atpB sequence 자료분석에서 대체적으로 일치하는 계통수를 얻었다. 즉, 한국산 백합강은 Dahlgren 등의 분류체계에 의한 비짜루목, 백합목, 마목의 3목으로 구분되었다. 이중 비짜루목과 백합목의 유연관계를 추론하는 분자생물학적 연구에서 마목의 분류군이 군외분류군으로 사용되었다. 두 엽록체 DNA인 rbcL 및 atpB sequence 분석에서 붓꽃과는 백합과 보다는 비짜루과에 더 가까운 유연관계를 보여주었다. 그러나, 넓은 의미의 Narthecaceae (종전의 Melanthiaceae [국명부재]에 속한 분류군)는 비짜루목이나 백합목에 속하는 분류군들과 가까운 유연관계를 보이지 않았다. 이전의 은방울꽃과 [Ruscaceae] 내에 취급되었던 애기나리속, 나도옥잠화속, 죽대아재비속 등은 각각 Colchiaceae, 백합과, Caloghortaceae로 이전 되어야 한다는 결론이 내려졌다. 마지막으로 한국산 백합강에 속하는 과의 체계에 대해서 토의하였다.

  • Rhizoctonia solani 분류를 위해 균사의 anastomosis group과 phylogenetic group간의 유전적 차이의 분석을 위해 AFLP를 실시하였다. 그 결과, 각 집단의 유사도는 크게 5집단으로 나뉘어졌다. 균주 9와 균주 20은 0.777을 나타내며 AG-5 group에 속하였고 균주 6은 균주 24와 0.670으로 AG-3 group에 속하였다. 균주 3은 균주 13과 0.844, 균주 10과 0.833의 높은 상동성을 보이며 AG-2-2 group에 속하였다. 균주 2는 균주 11과 0.744, 균주 3은 균주 13과 0.844, 균주 4는 균주 5와 0.870, 균주 5는 균주 16과 0.833을 나타내며 AG-1 group에 속하였다. 균주 7은 균주 8과 0.837의 높은 상동성을 나타내고, 균주 8은 AG-1( I B)와 0.751를 나타냈고, 균주 1과 균주 7,8은 AG-1( I B)에 속하였다.

새로운 동물계통 (New Animal Phylogeny)

  • 전통적인 동물의 계통수는 초기발생 형질들에 기초하여 몸 구조의 복잡성에 따라 동물문들을 단순한 동물에서 복잡한 동물 순으로 점진적으로 배열하는 것인데 현재 활발히 연구되는 분자계통 연구들이 이 경향을 재평가하고 새로운 계통과 의미를 제시하고 있다. 주로 18S rRNA 에 의한 분자계통에 의하면 무체강동물과 의체강동물과 같이 진체강동물로의 전이단계에 위치했던 무리들이 서로 나뉘어진 진체강동물 안에 속하게 된다. 이러한 새로운 분자계통은 좌우대칭동물의 공동조사에 대한 새로운 가정들을 세울 수 있는 뼈대와 비교발생학 및 비교유전체학 정보가 해석될 수 있는 토대를 제공한다.

Molecular systematics of Poaceae based on eight chloroplast markers, emphasizing the phylogenetic positions of Korean taxa

  • This study was conducted to clarify the phylogenetic position and relationships of Korean Poaceae taxa. A total of 438 taxa including 155 accessions of Korean Poaceae (representing 92% and 72% of Korean Poaceous genera and species, respectively) were employed for phylogeny reconstruction. Sequence data of eight chloroplast DNA markers were used for molecular phylogenetic analyses. The resulted phylogeny was mostly concordant with previous phylogenetic hypotheses, especially in terms of subfamilial and tribal relationships. Several taxa-specific indels were detected in the molecular phylogeny, including a 45 bp deletion in rps3 (PACMAD [Panicoideae, Arundinoideae, Chloridoideae, Micrairoideae, Aristidoideae, Danthonioideae] clade), a 15 bp deletion in ndhF (Oryzeae + Phyllorachideae), a 6 bp deletion in trnLF (Poeae s.l.), and two (17 bp and 378 bp) deletions in atpF-H (Pooideae). The Korean Poaceae members were classified into 23 tribes, representing eight subfamilies. The subfamilial and tribal classifications of the Korean taxa were generally congruent with a recently published system, whereas some subtribes and genera were found to be non-monophyletic. The taxa included in the PACMAD clade (especially Andropogoneae) showed very weak and uncertain phylogenetic relationships, presumably to be due to evolutionary radiation and polyploidization. The reconstructed phylogeny can be utilized to update the taxonomic positions of the newly examined grass accessions.

First Record of Box Jellyfish, Carybdea brevipedalia (Cnidaria: Cubozoa: Carybdeidae) from Korean Coastal Waters: Morphology and Molecular Descriptions

  • We firstly described a box jellyfish, Carybdea brevipedalia collected from the southern coasts of Korea. It is morphologically characterized by gastric phacellae, a special digestive system of cubozoan jellyfish, and velarium, the thin muscle flap forming the opening of the subumbrellar cavity. The phacellae are linear-shaped, comprising numerous cirri branched from multiple roots. Each root also has multiple numbers of cirrus bundles. Basis of velarial cannels parts into two branches in each octant of velarium. Its geographic distributions were limited to Jeju-do and the middle-southern coasts where the organisms are seriously hazardous to bathers in summer. Numerous individuals and/or large-sized populations were observed specifically from beaches at Jeju-do, Namhae-do and a small fishing port of Namildae, while only a small amount of the individuals was observed in Gamak Bay, Jaran-Goseung Bay, and Geoje-do. We confirmed molecular identity of the Korean C. brevipedalia with comparison of nuclear ribosomal DNA sequences. Until now, Carybdea brevipedalia is the only cubozoan species reported to be distributed in Korean waters.

Molecular phylogeny of parasitic Platyhelminthes based on sequences of partial 28S rDNA D1 and mitochondrial cytochrome c oxidase subunit I

  • The phylogenie relationships existing among 14 parasitic Platyhelminthes in the Republic of Korea were investigated via the use of the partial 28S ribosomal DNA (rDNA) D1 region and the partial mitochondrial cytochrome c oxidase subunit 1 (mCOI) DNA sequences. The nucleotide sequences were analyzed by length, G + C %, nucleotide differences and gaps in order to determine the analyzed phylogenie relationships. The phylogenie patterns of the 28S rDNA D1 and mCOI regions were closely related within the same class and order as analyzed by the PAUP 4.0 program, with the exception of a few species. These findings indicate that the 28S rDNA gene sequence is more highly conserved than are the mCOI gene sequences. The 28S rDNA gene may prove useful in studies of the systematics and population genetic structures of parasitic Platyhelminthes.

Extensive Chromosomal Polymorphism Revealed by Ribosomal DNA and Satellite DNA Loci in 13 Citrus Species

  • Little is known about the chromosomal variability and polymorphism existing in mitotic chromosomes of Citrus, mainly due to lack of reliable chromosomal markers and small chromosome size. To test the hypothesis of chromosomal polymorphism and provide the foundation of the genome organization in the Citrus cultivars, we have developed molecular cytogenetic markers for 13 Citrus species collected from Jeju island, Korea. In this study, we demonstrated that the chromosomal locations of cytogenetic markers are quite variable and extremely polymorphic, in contrast to the previous studies. The data obtained in this study will be of utmost importance in cytological systematics and karyotyping of the Citrus species.

Phylogenetic Contributions of Partial 26S rDNA Sequences to the Tribe Helleboreae (Ranunculaceae)

  • Monophyly and intergeneric relationships of the tribe Helleboreae, sensu Tamura, and related genera were studied using a 1,100-bp segment at the 5'end of the 26S ribosomal RNA gene. Forty-one OTUs, including eight species of the Helleboreae, were either directly sequenced or obtained from previous publications. Data were analyzed using distance and discrete character methods to infer phylogenetic relationships among the included taxa. The inferred phylogeny did not support monophyly of either Helleboreae or Cimicifugeae whose members were intermixed in our inferred phylogeny. This result is congruent with our previous study, which recommended against finely subdividing, suprageneric higher taxa within the R-chromosome group (subfamily Ranuncluloideae, sensu lato) until more molecular data were accumulated. Our data convincingly suggest the presence of the following three monophyletic groups: the Cimicifuga group (the clade of Actaea, Cimicifuga, Souliea, Eranthis, Anemonopsis, and Beesia), the Trollius group (the clade of Trollius, Megaleranthis, Adonis), and a clade including Anemonopsis and Beesia. Our data also suggest that Trollius and Megaleranthis might be congeners and Eranthis a paraphyletic group.

Morphological Description, DNA Barcoding, and Taxonomic Review of Five Nudibranch Species (Gastropoda) from South Korea

  • The nudibranch is one of the most colorful gastropod species found in oceans worldwide. Unlike many other gastropod groups, the nudibranch loses an external shell in the adult stage, but instead develops various chemical defense systems. More than 2,500 nudibranch species have been reported worldwide, and 73 species are currently recorded in Korean waters. In this study, we present morphological descriptions, DNA barcode information of mtDNA cox1 sequence, and taxonomic review for five nudibranch species: Apata pricei (MacFarland, 1966), Doto rosacea Baba, 1949, Janolus toyamensis Baba and Abe, 1970, Polycera abei (Baba, 1960), and Trinchesia sibogae (Bergh, 1905). Of these, we also provide in-depth discussion of taxonomic issue of A. pricei that was previously subdivided into two subspecies, A. pricei pricei and A. pricei komandorica. Our morphological examination and molecular analyses of the mtDNA cox1 sequences indicate that these two subspecies are not taxonomically warranted. The phylogenetic information for the other nudibranch species from mtDNA cox1 sequence analysis is also included, providing a molecular basis for species identification and inferring their local phylogenies within each of the species groups discussed herein.

한국산 십각류의 18S 리보솜 RNA의 염기분석과 분자진화에 관한 연구(II) (A Study on the Nucleotide Analysis of 18S rRNA and the Molecular Evolution of the Korean Decapods(II))

  • 중합효소연쇄반응(PCR)을 이용한 클로닝 기법과 Taq 염기서열분석법을 사용하여 갑각류에 속하는 뿔물맞이게(Pugettia quadridens)(십각 목, 범배 아목, 게 하목)에 대한 18S 리보솜 RNA 유전자의 1차염기서열을 밝혔다. 본 종의 18S 리보솜 RNA 유전자는 십각류에 속하는 또 다른 종인 두드러기어리게(Oedignathus inermis)보다 46개가 짧은 1837개의 염기로 이루어져 있었다. 염기가 삽입되거나 결실된 부분을 고려하지 않았을때에 두 종간에 염기서열 유사도는 90.8%이었다. 염기서열이 가장 보존적인 부위는 1137-1206(70개) 분위로 이 부위에서는 두 종이 완전히 동일한 염기서열을 보이고 있었고, 변이가 연속적으로 가장 큰 부위는 46-55 부위였고 399-407 부위가 그 다음으로 많은 연속적 변이를 가지고 있었다. 18S 리보솜 RNA 유전자의 1차구조에 있어서 염기서열의 변이는 이 유전자 전체를 통해 고르게 분포하지 않았다.
