Myosin X and Cytoskeletal Reorganization

  • Myosin X is one of myosin superfamily members having unique cellular functions on cytoskeletal reorganization. One of the most important cellular functions of myosin X is to facilitate the formation of membrane protrusions. Since membrane protrusions are important factors for diverse cellular motile processes including cell migration, cell invasion, path-finding of the cells, intercellular communications and so on, it has been thought that myosin X plays an important role in various processes that involve cytoskeletal reorganization including cancer progression and development of neuronal diseases. Recent studies have revealed that the unique cellular function of myosin X is closely correlated with its unique structural characteristics and motor properties. Moreover, it is found that the molecular and cellular activities of myosin X are controlled by its specific binding partner. Since recent studies have revealed the presence of various specific binding partners of myosin X, it is anticipated that the structural, biochemical and cell biological understanding of the binding partner dependent regulation of myosin X function can uncover the role of myosin X in diverse cell biological processes and diseases.

키토산 처리에 의한 위생용 부직포의 기능성 개질화 (제1보) -표면구조 및 태 변화- (Functional Modification of Sanitary Nonwoven Fabrics by Chitosan Treatment (Part I) -Change of Surface Structure and Hand-)

  • 위생용 탑시트로 쓰이는 폴리프로필렌 부직포에 천연고분자이며, 인체친화력이 우수한 가공제인 키토산을 사용하여, 키토산의 분자량과 처리농도를 변화시켜 처리하므로써 기능성 개질화된 부직포의 표면구조와 역학적 성질 및 태의 변화를 살펴보았다. 저분자량의 키토산 처리포가 더 균일하게 코팅되었으며, 분자량과 처리농도가 클수록 부착률이 증가하였다. 키토산 처리포는 굽힘특성과 전단특성이 증가하여 다소 강경해지고, 형태안정성은 고분자량의 키토산 처리 시 더 향상되었다. 표면 촉감은 매끄러워졌으며, 부피감이 다소 향상되었다. 굽힘특성의 증가에 비해 Koshi의 증가가 크지 않으므로 지나치게 뻣뻣해지지는 않았고, 키토산 처리농도가 증가할수록 저분자량의 키토산 처리포가 더 매끄러워졌으며, 벌키성이 부여되었다. 키토산 처리에 의한 위생용 부직포의 태 변화는 미처리포에 비해 전반적으로 THV가 향상되었으며, 키토산 처리농도가 0.5$\%$일 경우 고분자량의 키토산 처리포의 THV가 더 좋았으나, 처리농도가 1.0$\%$일 경우는 저분자량의 키토산 처리포의 THV가 더 우수하였다.

Oligomer Model of PB1 Domain of p62/SQSTM1 Based on Crystal Structure of Homo-Dimer and Calculation of Helical Characteristics

  • Autophagy is an important process for protein recycling. Oligomerization of p62/SQSTM1 is an essential step in this process and is achieved in two steps. Phox and Bem1p (PB1) domains can oligomerize through both basic and acidic surfaces in each molecule. The ZZ-type zinc finger (ZZ) domain binds to target proteins and promotes higher-oligomerization of p62. This mechanism is an important step in routing target proteins to the autophagosome. Here, we determined the crystal structure of the PB1 homo-dimer and modeled the p62 PB1 oligomers. These oligomer models were represented by a cylindrical helix and were compared with the previously determined electron microscopic map of a PB1 oligomer. To accurately compare, we mathematically calculated the lead length and radius of the helical oligomers. Our PB1 oligomer model fits the electron microscopy map and is both bendable and stretchable as a flexible helical filament.

Structure and catalytic mechanism of human protein tyrosine phosphatome

  • Together with protein tyrosine kinases (PTKs), protein tyrosine phosphatases (PTPs) serve as hallmarks in cellular signal transduction by controlling the reversible phosphorylation of their substrates. The human genome is estimated to encode more than 100 PTPs, which can be divided into eleven sub-groups according to their structural and functional characteristics. All the crystal structures of catalytic domains of sub-groups have been elucidated, enabling us to understand their precise catalytic mechanism and to compare their structures across all sub-groups. In this review, I describe the structure and mechanism of catalytic domains of PTPs in the structural context.

Crystal Structure of TTC0263, a Thermophilic TPR Protein from Thermus thermophilus HB27

  • The hypothetical protein TTC0263 of Thermus thermophilus HB27 is a thermophilic tetratricopeptide repeat (TPR)-containing protein. In the present study, the TPR region (residues 26-230) was resolved at $2.5{\AA}$ with R-factors of $R/R_{free}$ = 23.6%/28.6% $R/R_{free}=23.6%/28.6%$. TTC0263 consists of 11 helices that form five TPR units. Uniquely, it contains one atypical "extended" TPR (eTPR) unit. This comprises extended helical residues near the loop region of TTC0263, such that the helical length of eTPR is longer than that of the canonical TPR sequence. In addition, the hybrid TPR domain of TTC0263 possesses oligomer-forming characteristics. TPR domains are generally involved in forming multi-subunit complexes by interacting with each other or with other subunit proteins. The dynamic structure of TTC0263 described here goes some way to explaining how TPR domains mediate the formation of multi-subunit complexes.

Effects of fermentation on protein profile of coffee by-products and its relationship with internal protein structure measured by vibrational spectroscopy

  • Objective: To our knowledge, there are few studies on the correlation between internal structure of fermented products and nutrient delivery from by-products from coffee processing in the ruminant system. The objective of this project was to use advanced mid-infrared vibrational spectroscopic technique (ATR-FT/IR) to reveal interactive correlation between protein internal structure and ruminant-relevant protein and energy metabolic profiles of by-products from coffee processing affected by added-microorganism fermentation duration. Methods: The by-products from coffee processing were fermented using commercial fermentation product, called Saus Burger Pakan, consisting of various microorganisms: cellulolytic, lactic acid, amylolytic, proteolytic, and xylanolytic microbes, for 0, 7, 14, 21, and 28 days. Protein chemical profiles, Cornell Net Carbohydrate and Protein System crude protein and CHO subfractions, and ruminal degradation and intestinal digestion of protein were evaluated. The attenuated total reflectance-Ft/IR (ATR-FTIR) spectroscopy was used to study protein structural features of spectra that were affected by added microorganism fermentation duration. The molecular spectral analyses were carried using OMNIC software. Molecular spectral analysis parameters in fermented and non-fermented by-products from coffee processing included: Amide I area (AIA), Amide II (AIIA) area, Amide I heigh (AIH), Amide II height (AIIH), α-helix height (αH), β-sheet height (βH), AIA to AIIA ratio, AIH to AIIH ratio, and αH to βH ratio. The relationship between protein structure spectral profiles of by-products from coffee processing and protein related metabolic features in ruminant were also investigated. Results: Fermentation decreased rumen degradable protein and increased rumen undegradable protein of by-products from coffee processing (p<0.05), indicating more protein entering from rumen to the small intestine for animal use. The fermentation duration significantly impacted (p<0.05) protein structure spectral features. Fermentation tended to increase (p<0.10) AIA and AIH as well as β-sheet height which all are significantly related to the protein level. Conclusion: Protein structure spectral profiles of by-product form coffee processing could be utilized as potential evaluators to estimate protein related chemical profile and protein metabolic characteristics in ruminant system.

고강도 초음파에 의한 PEO의 분해특성에 관한 연구 (Effect of High Intensity Ultrasonic Wave on the Degradation Characteristics of PEO)

  • 본 연구에서는 고강도 초음파를 이용하여 분자량이 다른 poly(ethylene oxide) (PEO)를 증류수에 분산시켜 초음파 가진(加振) 시간이 PEO의 성질에 미치는 영향을 조사하였다. 고강도 초음파는 PEO 수용액 상에서 동공 형성 및 파괴를 일으켜서 PEO 거대 라디칼을 형성하였으며 이로인하여 PEO 자체의 유변학적 성질, 화학 구조 그리고 용융 거동이 현저하게 변화되었다. 초음파 가진에 의하여 PEO의 용융 점도는 감소되었고, 물과 PEO 사슬에서 비롯된 여러 종류의 라디칼들의 상호 작용으로 말미암아 새로운 말단기들이 생성되었다. 아울러 분자량에 따라 상이한 용융 거동을 나타내었는데, 상대적으로 분자량이 큰 경우에는 가진되지 않은 PEO에 비하여 결정화 속도가 느려지고 용융 피크의 강도가 감소되는 특성을 나타내었다.

A Study on the Electrical Physical Properties of Organic Thin Films for Manufacture in Power Device

  • Monolayers of lipids on a water surface have attracted much interest as models of biological membranes, but also as precursors of multilayer systems promising many technical applications. Until now, many methodologies have been developed in order to gain a better understanding of the relationship between the structure and function of the monolayers. Maxwell displacement current (MDC) measurement has been employed to study the dielectric property of Langmuirfilms. MDC flowing across monolayers is analyzed using a rod-like molecular model. A linear relationship between the monolayer compression speed a and the molecular area Am. Compression speed a was about 30, 40, and 50 mm/min. Langmuir-Blodgett(LB) layers of Arachidic acid deposited by LB method were deposited onto slide glass as Y-type film. The structure of manufactured device is Aul Arachidic acid! AI, the number of accumulated layers are $9{\sim}21$. Also, we then examined of the Metal-Insulator-Metal(MIM) device by means of I-V. The I-V characteristics of the device are measured from -3 to+3 V. The insulation property of a thin film is better as the distance between electrodes is larger.

Characterization of Exopolysaccharides Produced by Submerged Culture of an Entomopathogenic Fungus Paecilomyces sinclairii by Using a Multi-Angle Laser Light Scattering System

  • Three groups of exopolysaccharides (EPSs) (designated as Fr-I, Fr-II, and Fr-III) were isolated from the culture filtrates of Paecilomyces sinclairii by gel filtration chromatography on Sepharose CL-4B. Their molecular characteristics were examined by multi-angle laser light scattering (MALLS) connected online to a size exclusion chromatography (SEC) and refractive index (RI) detector system. The weight-average molar mass of Fr-I, Fr-II, and Fr-III of EPSs were determined to be $1.540{\times}10^6,\;6.302{\times}10^4$, and $9.389{\times}10^4\;g/mol$, respectively. All three EPSs showed a fairly low polydispersity indice, ranging from 1.008 to 1.059 (nearly mono disperse behavior), and showed different carbohydrates and amino acids compositions; all fractions of EPSs consisted of mainly cystine, valine, and arginine in the protein moiety, and mainly ribose, galactose, and glucose in the carbohydrate moiety. The determination of gyration radii of the EPSs in SEC/MALLS analysis revealed the molecular shape of the Fr-I to be a rod-like structure, whereas the Fr-II and Fr-III had a random-coil structure in an aqueous solution.
