ADVANTAGES OF USING ARTIFICIAL NEURAL NETWORKS CALIBRATION TECHNIQUES TO NEAR-INFRARED AGRICULTURAL DATA

  • Artificial Neural Network (ANN) calibration techniques have been used commercially for agricultural applications since the mid-nineties. Global models, based on transmission data from 850 to 1050 nm, are used routinely to measure protein and moisture in wheat and barley and also moisture in triticale, rye, and oats. These models are currently used commercially in approx. 15 countries throughout the world. Results concerning earlier European ANN models are being published elsewhere. Some of the findings from that study will be discussed here. ANN models have also been developed for coarsely ground samples of compound feed and feed ingredients, again measured in transmission mode from 850 to 1050 nm. The performance of models for pig- and poultry feed will be discussed briefly. These models were developed from a very large data set (more than 20,000 records), and cover a very broad range of finished products. The prediction curves are linear over the entire range for protein, fat moisture, fibre, and starch (measured only on poultry feed), and accuracy is in line with the performance of smaller models based on Partial Least Squares (PLS). A simple bias adjustment is sufficient for calibration transfer across instruments. Recently, we have investigated the possible use of ANN for a different type of NIR spectrometer, based on reflectance data from 1100 to 2500 nm. In one study, based on data for protein, fat, and moisture measured on unground compound feed samples, dedicated ANN models for specific product classes (cattle feed, pig feed, broiler feed, and layers feed) gave moderately better Standard Errors of Prediction (SEP) compared to modified PLS (MPLS). However, if the four product classes were combined into one general calibration model, the performance of the ANN model deteriorated only slightly compared to the class-specific models, while the SEP values for the MPLS predictions doubled. Brix value in molasses is a measure of sugar content. Even with a huge dataset, PLS models were not sufficiently accurate for commercial use. In contrast an ANN model based on the same data improved the accuracy considerably and straightened out non-linearity in the prediction plot. The work of Mr. David Funk (GIPSA, U. S. Department of Agriculture) who has studied the influence of various types of spectral distortions on ANN- and PLS models, thereby providing comparative information on the robustness of these models towards instrument differences, will be discussed. This study was based on data from different classes of North American wheat measured in transmission from 850 to 1050 nm. The distortions studied included the effect of absorbance offset pathlength variation, presence of stray light bandwidth, and wavelength stretch and offset (either individually or combined). It was shown that a global ANN model was much less sensitive to most perturbations than class-specific GIPSA PLS calibrations. It is concluded that ANN models based on large data sets offer substantial advantages over PLS models with respect to accuracy, range of materials that can be handled by a single calibration, stability, transferability, and sensitivity to perturbations.

황-이용 독립영양 탈질시 알칼리도 저감을 위한 종속영양 탈질의 이용방안 (A Use of Heterotrophic Denitrification for the Supply of Alkalinity during Sulfur-utilizing Autotrophic Denitrification)

  • 황-이용 탈질시 요구되는 알칼리도를 공급하기 위해 종속영양탈질을 동시에 이용하는 방안(종속/독립 동시탈질)의 가능성을 검토하였다. 이를 위해 $35^{\circ}C$에서 운전된 황 충전상 반응조에 $NO_3{^-}-N$$COD_{Cr}$이 각각 700~900 및 900~2500mg/L인 질산화된 침출수를 주입하며 종속/독립 동시탈질시 첨가된 외부탄소원의 종류 및 농도, 그리고 HRT가 탈질효율에 미치는 영향을 파악하였다. 종속/독립 동시탈질시 알칼리도 공급량을 0으로 유지하기 위해 종속영양탈질에 의해 제거되어야 하는 $NO_3{^-}-N$분율($HDNR_{fraction}$)은 탄소원별로 차이를 나타내었다. 메탄올과 아세트산 나트륨의 경우 이론적인 $HDNR_{fraction}$ 값에서 100%의 탈질효율을 얻었으나 글루코스와 당밀의 경우 이론치보다 높은 $HDNR_{fraction}$ 값이 필요하였다. 대부분의 탈질반응은 반응조 하단 11.5cm에서 완료되었으며, 이를 기준으로 계산한 종속/독립 동시탈질시 100% 탈질효율을 얻을 수 있는 EBCT와 용적부하는 각각 6.76 h와 $2.84kg-NO_3{^-}-N/m^3{\cdot}d$이었고, 최대 탈질율은 처리효율 89%에서 $5.05kg-NO_3{^-}-N/m^3{\cdot}d$이었다. 그러나 짧은 EBCT에서는 종속 영양균의 과도한 성장에 의한 반응조의 막힘현상이 발생하였는 바, 이는 역세척 또는 종속영양탈질과 황-이용 탈질의 분리운전 등의 대안으로 해결할 수 있을 것이다.

배지최적화를 통한 재조합 바실러스 서브틸리스에서 바실러스 아밀로리퀴파시엔스 유래 γ-글루타밀펩타이드전달효소의 대량생산 (Overproduction of a γ-glutamyltranspeptidase from Bacillus amyloliquefaciens in Bacillus subtilis through medium optimization)

  • 본 연구를 통해 BAGGT를 재조합 B. subtilis를 이용하여 대량 생산하기 위하여 유전자 클로닝, 발현 시스템 구축 및 배지 최적화를 진행하였다. 이중프로모터 시스템을 이용하여 야생형 균주에 비해 42배 효소 생산성이 향상된 발현 시스템을 구축하였다. 또한 PBD 분석을 통해 당밀과 CSL이 재조합 B. subtilis 시스템에서 BAGGT의 생산성에 큰 영향을 주는 인자임을 확인하였으며, 염류의 첨가에 의한 효소 생산성 증대 효과는 미비하거나 부정적이었다. 탄소원으로 당밀을 선택하고 고가의 질소원인 트립톤을 저가의 CSL로 교체한 후 CCD 분석을 통해서 결정된 최적배지 사용 시 최적화 이전의 LB 배지 대비 4.3배의 생산성 증대를 이루었으며, 이는 LB 배지에서 야생형 균주의 BAGGT 생산성 대비 180배의 효소 생산성 개선에 해당하였다. 본 연구를 통해 식품용 효소로서 BAGGT의 대량생산을 위한 공정을 구축하였으며, 이후 정미성 소재 생산에 활용할 수 있을 것으로 기대한다.

Different Sources and Levels of Copper Supplementation on Performance and Nutrient Utilization of Castrated Black Bengal (Capra hircus) Kids Diet

  • Twenty eight 3-4 month old castrated Black Bengal kids (Capra hircus) were used to determine the effects of source and level of dietary copper (Cu) concentration on their performance and nutrient utilization. Cu was supplemented (0, 10, 20 and 30 mg/kg diet DM) as copper sulfate ($CuSO_4$, $5H_2O$) or copper proteinate (Cu-P). Kids were fed a basal diet containing maize (19.5%), soybean (17.0%), deoiled rice bran (56.5%), molasses (4.0%), di-calcium phosphate and salt (1.0% each) and mineral and vitamin mixture (0.5% each) supplements at 3.5% of body weight to meet NRC (1981) requirements for protein, energy, macro minerals and micro minerals, excluding Cu. The basal diet contained 5.7 mg Cu/kg, 122.5 mg Fe/kg, 110 mg Zn/kg, 0.26 mg Mo/kg and 0.32% S. $CuSO_4$ or Cu-P was added to the basal diet at the rate of 10, 20 and 30 mg/kg. Kids were housed in a well ventilated shed with facilities for individual feeding in aluminum plated metabolic cages. Blood samples were collected from the jugular vein on d 0, 30, 60 and 90 to determine hemoglobin (Hb), packed cell volume (PCV), total erythrocyte count (TEC), total leukocyte count (TLC) and serum enzymes (alkaline phosphatase, alanine transferase and aspertate transferase). A metabolism trial of 6 days duration was conducted after 90 days of experimental feeding. Statistical analysis revealed that source and level of Cu supplementation improved live weight gain (p<0.04) and average daily gain (p<0.01). No significant contribution of source and level of Cu to alter serum serum enzymes was evident. Goats fed Cu-P tended to have higher Hb, PCV and TEC than with $CuSO_4$ supplementation. Cu-P increased digestibility of ether extract (EE, p<0.02) and crude fiber (p<0.05) and showed an increasing trend (p<0.09) for digested crude protein (CP) and crude fiber (CF). Supplemental dose of Cu linearly improved (p<0.02) digestibilities of dry matter (DM), organic matter (OM), EE and nitrogen free extract (NFE). Though the absorption of nitrogen (N) was not affected (p>0.10) by both source and dose of Cu, N retention was affected (p<0.04) and there was a significant $Source{\times}Dose$ interaction (p<0.05). Final body weight (BW) was not influenced (p>0.10) by the source of Cu but increasing dose of Cu increased (p<0.04) the BW of kids. TDN intake (g/kg $W^{0.75}$) was higher (p<0.05) with the increased dose of Cu and there was a significant $Source{\times}Dose$ interaction. It was concluded that supplementation of Cu from different sources and varying dose level in a concentrate based diet may improve performance, nutrient utilization and plane of nutrition in castrated Black Bengal kids. The effects on performance and nutrient utilization are more pronounced with Cu-P than $CuSO_4$ supplementation. Higher dose of Cu showed better result than lower dose.

폭염 브로일러 닭의 혈액지질 및 짧은 사슬지방산에 대한 폭염사료와 역전점등 효과 (Effects of inverse lighting and extreme heat diet on short chain fatty acid and blood lipid profile in extreme heat stress-exposed broilers)

  • This study was carried out to investigate the effects of feeding the broilers that are exposed to extreme heat stress by control of inverse lighting times with night restricted feeding of extreme heat diet(EHD1, 2: extreme heat diet) containing different amount of soy oil, molasses, amino acids and vitamin C on short chain fatty acid and blood lipid profile. 300 broiler chickens(Abaica strain) were randomized into four dietary treatment groups according to a randomized block design on the day they were hatched. The four dietary treatment groups were: T1(EHD 1, 10:00~19:00 Dark, 19:00~10:00 Light), T2(EHD 2, 10:00~19:00 Dark, 19:00~10:00 Light), T3(EHD 1, 09:00~18:00 Dark, 18:00~09:00 Light), T4(EHD 2, 09:00~18:00 Dark, 18:00~09:00 Light). The body weight gain of the broilers was highest in T2, and high in order T1, T4, T3(p<0.05). Weights of the lymphoid organ, thymus and bursa of Fabricius were high in T1, T2 as compared to T3, T4 but spleen was lower in T4 than T1, T2, T3(p<0.05). Blood triglyceride, total cholesterol and glucose were higher in T1, T2 than T3, T4(p<0.05). LDL-C was high in orderT4, T3, T2, T1 but HDL-C showed the opposite trend(p<0.05). Blood concentrations of IgG, IgG and IgM were higher in T1, T2 than inT3, T4, but the corticosterone concentration decreased significantly in them. In T1 and T2, Lactobacillus in the feces increased, but total aerobic bacteria, E.coli, coliform bacteria was decreased rather significantly, compared with those in T3 and T4(p<0.05). Concentrations of acetic acid, propionic acid and total SCFA in cecum were high in order T2, T1, T3, T4, but butyric acid, isobutyric acid, valeric acid, isovaleric acid were lower in T1, T2 than in T3, T4 (p<0.05).

수산물 가공부산물의 이용에 관한 연구 I -가다랭이 내장 발효 silage 제조를 위한 유산균주의 배양특성 (Studies on the Utilization of Wastes from Fish Processing I - Characteristics of Lactic Acid Bacteria for Preparing Skipjack Tuna Viscera Silage)

  • 가다랭이 통조림을 제조할 때 대량 폐기되는 내장을 유효하게 이용할 목적으로 어류 내장 silage 제조를 위한 유산균주의 배양특성 및 저장기간에 따른 원료의 성상변화를 조사한 결과, MRS 배지에서 배양한 L. bulgaricus 및 L. plantarum 균주는 $pH\;5.5\~6.5$, 배양온도 $35^{\circ}C$ 부근에서 산생성 및 증식이 양호하였으며, 가다랭이 내장 자숙액에 사용된 당질원인 당밀 dextrose의 적정 첨가농도는 가다랭이 내장 중량 당 각각 $10\%$$7\%$ 정도인 것으로 나타났다. 저장기간에 따라 산첨가 silage의 pH는 초기 4.0에서 저장 42일째 pH 4.5로 다소 증가하였으며, 유산균 발효 silage는 초기 pH 5.9에서 점차 감소하여 pH 약 4.0까지 저하되었다. silage의 초기 VBN 함량은 약 $62\~65\;mg/100g$에서 저장기간에 따라 점차 증가하여 저장 42일후 산첨가 silage는 112.6\;mg/100g이었고, L. bulgarius 및 L. plantarum 발효 silage는 각각 139.9 mg/100g와 155.0 mg/100g 이었다. L. plantarum 발효 silage의 저장중 경시별 아미노질소 함량은 저장 4일째 1391.3 mg/100g으로서 약 $81\%$의 가수분해율을 나타내었다. 또한 L. plantarum을 이용하여 제조한 발효 silage의 초기 생균수는 $2.7\times10^8/g$에서 저장 35일째는 $2.3\times10^7/g$으로 나타났다.

밤 발효사료 제조과정에서 미생물 첨가수준이 영양소 함량과 반추위 내 발효특성에 미치는 영향 (Effects of Inoculant Application Level on Chemical Compositions of Fermented Chestnut Meal and Its Rumen Fermentation Indices)

  • This study aimed to estimate the effect of inoculant application level on chemical composition and bacterial count of fermented chestnut meal (FCM), and its rumen fermentation characteristics. The inoculant contained Lactobacillus acidophilus ($1.2{\times}10^{10}cfu/g$), Bacillus subtilis ($2.1{\times}10^{10}cfu/g$), and Saccharomyces cerevisiae ($2.3{\times}10^{10}cfu/g$). The chestnut meal mixed with molasses, double distilled water, and inoculant at 1 kg, 3 g, 480 mL, and 20 mL ratio for the basal chestnut meal diet. The double distilled water from basal chestnut meal diet was substituted with bacterial inoculant at a level of 0 (Control), 20 (Medium), and 40 mL (High) in the experimental diets. The mixed experimental diets were incubated at $39^{\circ}C$ for 7, 14, and 21 days, respectively. On 7 days of FCM incubation, the contents of crude protein (CP) (quadratic, P=0.043) and neutral detergent fiber (quadratic, P=0.071) decreased by increases of inoculant application levels, whereas bacterial count (quadratic, P=0.065) and rumen $NH_3-N$ (linear, P=0.063) increased. By increases of inoculant application levels on 14 days of FCM incubation, the increases were found on dry matter (DM) (quadratic, P=0.085), CP (quadratic, P=0.059), acid detergent fiber (quadratic, P=0.056), in vitro DM digestibility (linear, P=0.002), rumen total volatile fatty acid (VFA) (linear, P=0.057), and rumen iso-butyrate (linear, P=0.054). However, the decreases were found on bacterial count (linear, P=0.002), propionate (linear, P=0.099), and butyrate (quadratic, P=0.082). On 21 days of FCM incubation, in vitro DM digestibility (linear, P=0.002) and total VFA (linear, P=0.001) increased by increases of inoculant application levels, whereas the contents of CP (quadratic, P=0.034) and neutral detergent fiber (quadratic, P=0.047) decreased. These results indicate that the FCM with a medium level of inoculant application and 14 of fermentation had beneficial effects by increasing DM digestibility and rumen total VFA content, without altering bacterial count.

  • This study aimed to estimate the effects of replacing Mushroom By-Product (MBP) with Tofu By-Product (TBP) on the chemical composition, microbes, and rumen fermentation indices of Fermented Diets (FDs). The basal diet was formulated using MBP, TBP, rice bran, molasses, and inoculants. The MBP in the basal diet was replaced with TBP at 0, 5, and 10% on Dry Matter (DM) basis for the experimental diets. The experimental diets were fermented at $39^{\circ}C$ for 144 h. Chemical composition, pH, microbes, and rumen fermentation indices of the FDs were analyzed. With increasing TBP replacement, crude protein content of FDs increased (L, P < 0.001), whereas crude ash content decreased (L, P = 0.002). Lactic acid bacteria and Bacillus subtilis contents in the TBP-replaced FDs were higher than those in the control (P < 0.05), whereas pH level and mold count were lower (P < 0.05). With increasing TBP replacement, in vitro rumen digestibility of DM (L, P = 0.053) and neutral detergent fiber (L, P = 0.024) increased, wheres rumen pH changed (P = 0.026) quadratically. Rumen total volatile fatty acid (L, P = 0.001) and iso-butyrate contents (Q, P = 0.003) increased with increasing TBP replacement. In conclusion, this study indicates that the replacement of MBP with TBP could improve the quality of FD.

반연속식(半連續式) 배양(培養)에 의(依)한 Rhizobium meliloti M14의 균체생산(菌體生産) (Production of Rhizobium meliloti M14 Inoculum by Semi-continuous Cultivation)

  • 전보(前報)에서 분리(分離)한 Rhizobium meliloti M14를 공시(供試)하여 그 종균생산(種菌生産)을 위한 기초적(基礎的) 연구(硏究)로서, 탄소원(炭素源) 및 질소원(窒素源), chemostat의 반련속식(半連續式) 운용(運用)에 의한 액체배양(液體培養)을 시도(試圖)하여 아래와 같은 결과(結果)를 얻었다. 1. 탄소원(炭素源) 비교(比較)한 단당류(單糖類), 이당류(二糖類) 및 당(糖)알콜류(類) 가운데 당(糖)알콜류(類)에서 가장 생육(生育)이 높았고 단당류중(單糖類中) 오탄당(五炭糖)에서의 생육(生育)은 육탄당(六炭糖)보다 저조(低調)하였다. 2. 천연(天然) 탄소원중(炭素源中) malt extract 및 molasses에서의 생육(生育)은 mannitol에서의 생육수준(生育水準) 이상(以上)이었다. 3. 질소원(窒素源)으로서 yeast water에 대(對)한 편향성(偏向性)이 인정(認定)되었으며 그 10%에서의 생육수준(生育水準)은 yeast extract 0.24%와 상응(相應)하였다. 4. Chemostat (MicroFerm 28L, New Brunswick model CMF-128S) 의 단식배양(單式培養)에 의(依)하여 36시간(時間) 배양(培養)으로 생균수(生菌數) $5{\sim}7{\times}10^9cells/ml$ 수준(水推)의 stationary phase에 도달(到達)하였고, 반련속식(半連續式) 운용(運用)으로 매(每) 24시간(時間)마다 동일수준(同一水準)의 생균(生菌)을 가진 배양액(培養液)을 그 운용량(運用量)에 대(對)하여 5분(分)의 4를 회수(回收)할 수 있었다.

Effect of Graded Levels of Rice Mill Feed (RMF) Supplementation on Intake, Nutrient Digestibility, Microbial N Yield and Growth Rate of Native (Bos Indicus) Bulls Fed Rice Straw Alone

  • Rice bran commonly available in Bangladesh is a mixture of rice hulls (60%), bran (35%) and polishing (5%), referred here as rice mill feed (RMF). Dose response effect of RMF supplementation to a straw diet including a zero level was measured on the intake, digestibility, nitrogen balance, microbial N yield and growth rate of growing native (Bos indicus) bulls. Twelve bulls of 33 months old and $272{\pm}31.5kg$ weight were randomly allocated to diets having 0 (T1), 1 (T2) and 2 (T3) kg RMF in addition to 200 g wheat bran, 200 g molasses, 60 g salt and 30 g oyestershe\l powder. Concentrate intake was 5.5, 19.2 and 29.5% of the dietary intake for the T1, T2 and T3 treatment respectively. RMF supplementation had no significant effect on the straw DM intake. However, with the increasing levels of RMF supplementation, total DM & digestible OM intake and the whole gut digestibilities of DM, OM, N & ADF increased but in deminishig return. Total microbial N yield estimated from the urinary purine excretion were 15.35, 26.56 and 38.44 g/d for the treatment T1, T2 and T3 respectively. Both the N intake and the N balance increased linearly in response to increasing level of RMF. Supplementation of RMF linearly increased the energy intake and dietary energy concentration. Growth rate in the T1, T2 and T3 treatments were 112, 125 and 250 g/d respctively. The basal N excretion and the maintenance energy requirement of the experimental animals were estimated to be 615 mg/kg $W^{0.75}/d$ and 447 kJ/kg $W^{0.75}/d$ respectively. The estimated efficiency on N utilization was 0.83 mg/mg of N intake ($r^2=0.997$) while the efficiency of metabolizable energy utilization for growth was 0.15. Since animal refused higher levels of RMF, inclusion up to 2 kg level (about 25% of the total DM intake) appears to have no depressing effect on the performances of animal. However, RMF itself fail to meet the critical nutrient need of the rumen microbes. Therefore response of supplementing RMF after correcting the critical nutrient deficiency need to be studied.
