The aim of this study was to investigate the epidermal moisturizing effects of artocarpin on normal human epidermal keratinocytes (NHEKs). To investigate the effects of artocarpin on NHEKs, cell viability and the expression of mRNAs related to skin hydration were measured. In addition, hyaluronic acid (HA)-ELISA assay was performed. Here, the effects of artocarpin on AQP3, HAS2, KRT1, and KRT10 mRNA expression, and on HA production, following UVA treatment were reported. The Quantitative real-time PCR results demonstrate that artocarpin increased AQP3, HAS2, KRT1, and KRT10 mRNA levels. The HA-ELISA assay revealed that artocarpin also increased HA production in NHEKs. Through these experiments, the epidermal moisturizing effects of artocarpin have been elucidated, providing evidence that artocarpin may be a potent cosmetic ingredient in skin anti-aging and moisturizing products. Based on these results, I anticipate that further research on the mechanisms of action of artocarpin may allow the development of not only cosmetics, but also medicines and healthcare foods.
This research was carried out to investigate the moisturizing effects of Agarum cribrosum extract on normal human epidermal keratinocytes (NHEKs). Moisturizing effects of A. cribrosum extract on NHEKs were measured by quantitative realtime RT-PCR to verify the gene expressions related to skin hydration, hyaluronic acid (HA)-ELISA to detect HA production, and cell viability assays. A. cribrosum extract increased the mRNA levels of the AQP3 and HAS2 genes and HA production in NHEKs. On the other hand, A. cribrosum extract decreased the mRNA level of the KRT1 and KRT10 genes known as differentiated keratinocyte marker in NHEKs. This research showed the moisturizing effects of A. cribrosum extract. The results indicate that A. cribrosum extract can be a potent functional ingredient for skin hydration and anti-aging products. Further study is warranted regarding the use of A. cribrosum extract to develop not only cosmetics but also food and medicine.
Purpose : In this study, we intended to observe the anti-wrinkle and moisturizing effects of dried pomegranate juice concentration powder (PCP) using in vitro test. Materials and methods : Antioxidant effects of PCP were determined by free radical scavenging capacity (DPPH assay) and the cytotoxicity of PCP was examined in human keratinocyte (HaCaT) and human primary dermal fibroblast-neonatal (HDF) cells. To investigate the moisturizing effect of PCP, hyaluronan synthesis was examined in HaCaT cells. Activity of procollagen production were assessed in HDF cells and elastase inhibition properties of PCP were evaluated in cell free condition, to determine their anti-wrinkle effects. Metalloproteinase 1 (MMP-1) activity was also assessed following UVB irradiation, in the current in vitro experiment. Results : No PCP treatment related significant cytotoxic effects were demonstrated against to the both HDF and HaCaT cells. PCP showed favorable free radical scavenging activities in dose-dependent manner. In PCP-treated HaCaT cells, hyaluronan synthesis was non-significantly but markedly increased, and pro-collagen productions were significantly increased in HDF cells, at all three different concentrations (0.25, 0.75 and 1 mg/ml), and elastase inhibitory activities were observed by PCP treatment. A significant decrease in UVB-induced MMP-1 activity was also observed in 1 mg/ml PCP-treated HDF cells as compared to those of UVB-exposed cells. Conclusions : Taken together, these results suggest that PCP has favorable antioxidant, anti-wrinkle and moisturizing effects without meaningful cytotoxicity on HDF and HaCaT cell lines.
Objectives: The aim of this study was to confirm the dose-dependent skin moisturizing effects of dried pomegranate concentrate powder (PCP) and pomegranate concentrate solution (PCS) in ICR mice. Materials and methods: To observe the in vivo skin moisturizing effects and possible underlying mechanisms of PCP and PCS, oral PCP (100, 200, and 400 mg/kg) and PCS (1, 2, and 4 mL/kg) were administered to normal ICR mice. Changes in body weight, skin water content, and skin type I collagen and hyaluronan contents were measured. Additionally, the mRNA expression levels of hyaluronan synthase (Has) 1, 2, and 3, and collagen type I alpha (COL1A) 1 and 2 were determined in the dorsal skin of mice by real-time reverse transcription polymerase chain reaction (RT-PCR). Results: Significant and dose-dependent increases in dorsal skin water content and type I collagen and hyaluronan contents were seen in PCP and PCS-treated mice. Moreover, the mRNA levels of Has 1, 2, and 3, involved in hyaluronan synthesis, and of COL1A1 and COL1A2, involved in collagen synthesis, were significantly and dose-dependently upregulated in PCS- and PCP-treated mice. Conclusions: In this study, PCP and PCS led to favorable skin moisturizing effects as indicated by increased skin water content and the upregulation of hyaluronan and collagen synthesis enzymes in mice treated with PCS (4 mL/kg) and PCP (200 mg/kg).
Objectives : The purpose of this study is to confirm the moisturizing effects of Herbal cosmetics containing Sinhyotakrisan(SHTL) extracts on facial skin.Methods : A total of 23 women who visited Semyung Oriental Medical Center from January 10th, 2014 to Feburary 12th, 2014 were included in the study. Provided both cosmetics containing Sinhyotakrisan extract and control products, all the subjects applied one of them to half face and another to remaining half for 4 weeks. We observed change of skin hydration, skin elasticity, skin scale in 0 week, 2 weeks, and 4weeks. Statistical anyalysis was performed by using SPSS, and statistical significance was achieved if the proability was less than 5%(p<0.05).Results : 1. After 4 weeks of using SHTL, skin hydration showed a statistically significant increase compared with using control product(p=0.011). And improvement rate also did(p=0.031). 2. After 4 weeks of using SHTL, skin elasticity showed statistically significant increase compared with using control product(p=0.025). But improvement rate didn't showed statistically significant increase(p=0.068). 3. After 4 weeks of using SHTL, skin scale showed a statistically significant decrease compared with using control product(p=0.029). And improvement rate also did(p=0.047).Conclusions : Considering the above results, herbal cosmetics containing Sinhyotakrisan extracts were effective for skin moisturizing.
본 연구에서는 해파리로부터 콜라겐 추출물을 제조하였고, 이에 대한 보습 효과를 측정하였다. 보습 효과는 caspase 14 발현, filaggrin, hyaluronan synthase-3 (HAS- 3), aquaporin-3 (AQP-3) 및 desmocollin (DSC)을 측정하였다. 그 결과, caspase 14 mRNA 발현은 비교물질인 retinoic acid (RA)와 유사한 효과를 나타냈다. 그리고 콜라겐 추출물은 20 mg/mL의 농도에서 각각 211.7%, 139.9%, 212.5% 및 116.8%의 filaggrin, HAS-3, AQP-3 및 DSC 발현 증가를 나타냈다. 따라서, 본 연구에서 해파리 콜라겐 추출물은 보습 효과를 갖는 화장품 소재로서의 개발 가능성이 클 것으로 기대할 수 있다.
In this study, we studied the anti-inflammatory and physiological activities Abelmoschus esculentus extracts by hot water (AEW) and 70% ethanol (AEE) and confirm the possibility to use it as a natural ingredient. For this study measure the antioxidative activity, total polyphenol content was measured, and DPPH and ABTS scavenging activity assays were conducted. Total polyphenol content of AEW and AEE was measured, and the results showed that they were 126.76 mg TAE/100 g and 144.21 mg TAE/100 g, respectively. DPPH and ABTS radical scavenging activities were measured to determine the antioxidative activity, and the results indicated that DPPH and ABTS radical scavenging activities increased in both extracts concentration-dependently. The moisturizing effect was measured by measuring the amount of hyaluronic acid (HA) produced within HaCaT cell using the ELISA kit. AEW and AEE increased the amount of HA production in a concentration-dependent manner. In order to determine the anti-inflammatory activity of AEW and AEE, the NO assay was conducted, and the inhibitory effects were found to be 11.46% and 25.28%, respectively in 100 ㎍/ml. In order to measure the anti-inflammatory activity, nitric oxide (NO) inhibitory activity was measured, and the inhibition of expression of iNOS, COX-2 proteins was measured and shown. Furthermore, inhibition of expression was found in inflammatory inducing factors iNOS and COX-2 proteins showing concentration-dependent inhibition. This study found the excellent effects of antioxidative, moisturizing effect, anti-inflammatory activity in Abelmoschus esculentus extracts, which indicates that can be used as functional materials for aesthetics, food and functional cosmetics.
편백나무(Chamaecyparis obtusa)는 측백나무과에 속하며 우리나라 남쪽 지역에서 주로 잘 자생한다. 편백나무의 목재는 재질이 우수하여 가구로서의 활용이 높으며 남은 목재, 가지, 잎은 정유 추출에 사용되고 있다. 편백 정유는 항염, 향균, 탈취, 진정 효능이 탁월한 것으로 알려져 있다. 본 연구는 이러한 편백나무의 잎에서 추출한 오일이 주름 개선, 피부 장벽 및 보습능에 미치는 영향을 평가하고자 하였다. 먼저, DPPH assay와 ABTS assay를 통해 항산화 활성을 평가한 결과, 편백 오일은 두 가지 라디칼을 농도 의존적으로 유의하게 소거하였다. 그리고 주름 생성 억제 효능 평가를 위해 elastase 활성을 측정한 결과, 편백 오일은 elastase 활성을 직접적으로 억제시킴을 확인하였다. 다음으로는 real-time PCR을 통해 유전자 발현을 확인한 결과, 편백 오일은 인간섬유아세포에서 MMP-1 유전자 발현을 유의하게 감소시켰다. 또한, 편백 오일에 의해 각질형성세포에서 filaggrin과 HAS-2 유전자 발현이 유의하게 증가함을 확인하였다. 이러한 결과를 종합해보면, 편백 오일은 주름 개선과 피부 장벽 및 보습능 강화에 도움을 주어 항노화 화장품의 기능성 원료로서 활용될 수 있을 것으로 기대된다.
본 연구에서는 황금 70% 에탄올 및 열수 추출물의 항산화, 항염, 미백, 보습, 세포 재생 및 자외선 및 블루라이트에 대한 세포 보호 효능을 확인하였다. 항산화 실험 결과, 70% 에탄올 및 열수 추출물 모두 DPPH 라디칼 소거 활성을 나타내었으며, 항염 실험 결과, 70% 에탄올 및 열수 추출물 모두 NO, 전염증성 cytokine(IL-6) 및 PGE2의 생성을 억제하는 것을 확인하였다. 미백 실험 결과, 70% 에탄올 추출물과 열수 추출물 모두 멜라닌 생성을 저해하였으며, 보습 실험 결과, 70% 에탄올 및 열수 추출물 모두 보습 인자인 HA의 생성량이 증가시키는 것을 확인하였다. UVB 및 블루라이트에 대한 세포 보호 효과를 분석한 결과, 70% 에탄올 추출물과 열수 추출물 모두 UVB에 대한 세포 보호 효능을 나타내었으며, 70% 에탄올 추출물은 블루라이트에 대한 세포 보호 효능 또한 나타내었다. 또한, 세포 이동 및 증식 효능을 분석한 결과, 70% 에탄올 추출물에서 세포 성장이 촉진되는 것을 확인하였다. 이러한 결과를 바탕으로 황금 70% 에탄올 추출물, 열수 추출물은 항산화, 항염, 미백, 피부재생, 자외선 및 블루라이트에 대한 세포 보호 효과가 있는 화장품 관련 천연소재로써의 활용가능성이 있는 것으로 사료된다.
바이오계면활성제는 생물학적으로 박테리아나 효모 등에 의해 많은 기질로 부터 생산된다. 바이오계면활성제는 많은 다른 합성 계면활성제에 비해서 환경친화적이고 보다 효과적인 것으로 알려져 있다. 본 연구에서는 Candida bombiocola로부터 생성된 소포로리피드의 항균효과를 다양한 균에 대해서 시험하였다. 소포로리피드의 치아우식 원인균(S. mutans), 액취균(C. xerosis) 및 여드름균(P. acnes)에 대한 최소억제농도(MIC)는 각각 0.005, 0.05, 0.005% (w/v)이였다. 이러한 항균효과는 합성 계면활성제인 SLS나 APG에 비해 우수한 항균효과를 보였다. 소포로리피드를 함유한 제형에서의 각질박리와 보습 효과를 측정하였다. 0.25% 수용액의 소포로리피드는 5.0% 젖산 수용액과 동등한 수준의 각질박리효과를 보였으며 보습효과는 소포로리피드를 함유한 에멀젼 제형에서 농도 의존적인 경향을 보였다. 이러한 결과는 소포로리피드을 항균제나 보습 또는 각질박리제로서 화장품에 대한 사용 가능성을 제시한다.
