Synthesis of Polyester Polyols by Using Double Metal Cyanide Catalyst and Physical Properties of Polyurethanes Produced by the Polyols (이중금속시안염 촉매에 의한 폴리에스테르 폴리올 제조 및 이를 이용한 폴리우레탄의 물성)

  • Copolymerizations of propylene oxide (PO) and phthalic anhydride (PA) have been performed in the presence of double metal cyanide (DMC) catalyst as a means of incorporating ester groups in the polyol backbone. DMC catalyst was effective for the copolymerization and the reactivity ratios measured by modified Kelen-$T{\ddot{u}}d{\ddot{o}}s$ equation were $r_1(PA)\;=\;0$, and $r^2(PO)\;=\;0.248$. Four different Polyol samples containing 1.0, 2.1, 7.52, and 11.42 mol% of PA unit were utilized for the synthesis of thermoplastic polyurethanes of their hard segments of 19 wt%. As the incorporated amount of PA increases, the elongation of the resulting polyurethane decreases and the tensile strength and the tensile modulus increase. The modulation of the incorporated amount of PA into polyol backbone was proven to be a feasible way to tune the physical properties of polyurethanes.

Properties of hydrolyzed α-lactalbumin, β-lactoglobulin and bovine serum albumin by the alcalase and its immune-modulation activity in Raw 264.7 cell

  • This study investigated the effects of the proteolytic hydrolysates of α-lactalbumin (LA), β-lactoglobulin (LG) and bovine serum albumin (BSA) by alcalase on inflammatory cytokines. The proteolytic hydrolysates were separated into two fraction of peptides, ≤ 10,000 Da and > 10,000 Da, respectively, because various low molecular weight peptides were generated during the hydrolysis reaction time. Among the hydrolysate peptides, BSA (all types), β-LG (> 10,000 Da), and α-LA (> 10,000 Da) showed an inhibitory activity against thymic stromal lymphopoietin (TSLP) mRNA expression in lipopolysaccharide-induced RAW264.7 murine macrophages. α-LA (> 10,000 Da), β-LG (hydrolysates), and BSA (> 10,000 Da) showed an inhibitory activity against tumor necrosis factor (TNF)-α expression. α-LA (all types), β-LG (hydrolysates, > 10,000 Da), and BSA (> 10,000 Da) showed an inhibitory activity against interleukin-6 (IL-6) expression. α-LA (> 10,000 Da), β-LG (> 10,000 Da), and BSA (all types) showed an inhibitory activity against inducible nitric oxide synthase (iNOS) expression. α-LA (> 10,000 Da), β-LG (> 10,000 Da), and BSA (all types) showed an inhibitory activity against cyclooxygenase (COX)-2 expression. The lowest level of TNF-α production was measured with α-LA (> 10,000 Da) and β-LG (> 10,000 Da) for all types, and a similar low level was measured for all types of BSA. The highest level of IL- 6 production was measured with α-LA (≤ 10,000 Da) among α-LA, β-LG, and IL-6. The low level of NO production was similar with α-LA, β-LG, and BSA but not with α-LA (≤ 10,000 Da). These potential peptides from whey protein hydrolysates could be used for food, medicinal, and industrial applications.

IMPLICATION OF STELLAR PROPER MOTION OBSERVATIONS ON RADIO EMISSION OF SAGITTARIUS A

  • It is suggested that a flying-by star in a hot accretion disk may cool the hot accretion disk by the Comptonization of the stellar emission. Such a stellar cooling can be observed in the radio frequency regime since synchrotron luminosity depends strongly on the electron temperature of the accretion flow. If a bright star orbiting around the supermassive black hole cools the hot disk, one should expect a quasi-periodic modulation in radio, or even possible an anti-correlation of luminosities in radio and X-rays. Recently, the unprecedentedly accurate infrared imaging of the Sagittarius A$\ast$ for about ten years enables us to resolve stars around it and thus determine orbital parameters of the currently closest star S2. We explore the possibility of using such kind of observation to distinguish two quite different physical models for the central engine of the Sagittarius A$\ast$, that is, a hot accretion disk model and a jet model. We have attempted to estimate the observables using the observed parameters of the star S2. The relative difference in the electron temperature is a few parts of a thousand at the epoch when the star S2 is near at the pericenter. The relative radio luminosity difference with and without the stellar cooling is also small of order $10^{-4}$, particularly even when the star S2 is near at the pericenter. On the basis of our findings we tentatively conclude that even the currently closest pass of the star S2 is insufficiently close enough to meaningfully constrain the nature of the Sagittarius A$\ast$ and distinguish two competing models. This implies that even though Bower et al. (2002)have found no periodic radio flux variations in their data set from 1981 to 1998, which is naturally expected from the presence of a hot disk, a hot disk model cannot be conclusively ruled out. This is simply because the energy bands they have studied are too high to observe the effect of the star S2 even if it indeed interacts with the hot disk. In other words, even if there is a hot accretion disk the star like S2 has imprints in the frequency range at v $\le$ 100 MHz.

Influence of the Epithelium on the Contraction of Guinea Pig Isolated Tracheal Smooth Muscle (Guinea Pig 기도상피세포가 기도 평활근 수축에 미치는 영향)

  • It has been well known that the integrity of airway epithelium is important in development of bronchial hyperreactivity and bronchial asthma. But the mechanisms involved are still unclear. To evaluate that airway epithelium is able to modulate the contraction of guinea pig tracheal smooth muscle, we investigated the responsiveness of intact and epithelium-denuded tracheal strips to histamine and acetylcholine. And to evaluate whether cyclooxgenase products play a role in this modulatory mechanism, we also investigated the effect of indomethacin pretreatment on the tracheal responsiveness to histamine. Results were as follows: 1) In guinea pig tracheal smooth muscle the presence of airway epithelium significantly reduced the response to histamine. 2) In the presence of indomethacin dose-response curves and $EC_{50}$ values were similar between intact and epithelium-denuded tracheal strips, that is, indomethacin abolished the influence of epithelium on the contracion of tracheal smooth muscle. 3) The response of tracheal smooth muscle to acetylcholine was similar both in the presence and absence of epithelium. These results suggest that airway epithelium of guinea pig may generate an inhibitory signal to decrease the response of tracheal smooth muscle to histamine and cyclooxygenase products may contrbute to the modulation of airway epithelium on the contracion of tracheal smooth muscle.

Modulatory Effect of Four Azulene Derivatives from the Fruiting Bodies of Lactarius hatsudake on Interferon-$\gamma$ Production (젖버섯아재비 자실체로부터 분리한 Azulene계 화합물이 Interferon-$\gamma$ 생성에 미치는 영향)

  • Investigation of secondary bioactive metabolites from the fruiting bodies of Lactarius hatsudake led to the isolation of four azulene derivatives by means of repeated column chromatography and preparative HPLC, and they were identified as 1-formyl-4-methyl-7-isopropyl azulene (1), lactaroviolin (2), 4-methyl-7-isopropyl-azulene-1-carboxylic acid (3), and 1-formyl-4-methyl-7-(1-hydroxy-1-methylethyl) azulene (4) by their physico-chemical properties and spectroscopic analysis. The isolated compounds were evaluated for the effects on modulation of cytokines in natural killer cell line (NK92 cells). Compounds 1 and 4 strongly inhibited IFN-$\gamma$ production in a dose-dependent manner, corresponding to 101.3 % and 92.7 % inhibition at 400 ${\mu}M$, and 11.9 % and 24.1 % at 100 ${\mu}M$, respectively, whereas compounds 2 and 3 showed weak inhibitory effect on INF-$\gamma$ production, corresponding to 45.9 % and 18.0 % inhibition at 400 ${\mu}M$.

Contactless Electroreflectance Spectroscopy of In0.5(Ga1-xAlx)0.5P/GaAs Double Heterostructures (In0.5(Ga1-xAlx)0.5P/GaAs 이중 이종접합 구조의 Contactless Electroreflectance에 관한 연구)

  • We have investigated the contactless electroreflectance (CER) properties of $In_{0.5}(Ga_{1-x}Al_x)_{0.5}P$/GaAs double heterostructures grown by metal-organic chemical vapour deposition (MOCVD). The CER measurements on the sample were studied as a function of temperature, modulation voltage ($V_{ac}$), and dc bias voltage ($V_{bias}$). Five signals observed at room temperature are related to the GaAs, $In_{0.5}Ga_{0.5}P$, $In_{0.5}(Ga_{0.73}Al_{0.27})_{0.5}P$, $In_{0.5}(Ga_{0.5}Al_{0.5})_{0.5}P$, and $In_{0.5}(Ga_{0.2}Al_{0.8})_{0.5}P$ transitions, respectively. From the temperature dependence of CER spectrum, the Varshni coefficients and broadening parameters were determined and discussed. In addition, we found that the behavior of the CER amplitude for the reverse bias is larger than that of the forward.

An analysis on Flicker Phenomenon of a Fluorescent lights for the commercial operating EMU (영업운행 전동차 객실형광등의 플리커(Flicker) 현상에 관한 분석 연구)

  • Generally, there are two types of main factors to affect output power quality of a auxiliary power supply an EMU(electric multiple unit). One is a voltage flicker by amplitude modulation of short time and air compressors. The other is repetitive motion of large capacity motor such as air compressors, HVAC unit etc. in main factors. This paper compared two kinds of fluorescent lamp, 32W (after remodeling interior) and 40W(before remodeling interior) and measured the light output varying input power(AC220V) for a flicker phenomenon related power supply of lamps in EMU. Also, we analyzed a flicker considering EMU operating time and density in order to grasp main factors of a load change to cause a voltage change. As a results of test, a 40W fluorescent lamp was more insensitive with 20.26% degree an eye recognition degree sides about changes of the input power and lower with 19.9% voltage side generating flicker compare with fluorescent lamp 32W. Also, we confirmed the fact which the fluorescent lamp flicker was generated by varying fluorescent lamp output voltage when the commercial EMU was in high driving density and at the busy time. Additionally, we confirmed the frequency band which an EMU passenger could feel sensitively blinking of a fluorescent lamp was visually $8Hz{\sim}15Hz$.

Modulation of Cell Cycle Regulators by Sulforaphane in Human Mepatocarcinoma HepG2 Cells (HepG2 인체간암세포의 세포주기조절인자 발현에 미치는 sulforaphane의 영향)

  • Sulforaphane, an isothiocyanate derived from hydrolysis of glucoraphanin in broccoli and other cruciferous vegetables, was shown to induce phase II detoxification enzymes and inhibit chemically induced mammary tumors in rodents. Recently, sulforaphane is known to induce cell cycle arrest and apoptosis in human canter cells, however its molecular mechanisms are poorly understood. In tile present study, we demonstrated that sulforaphane acted to inhibit proliferation and induce morphological changes of human hepatocarcinoma HepG2 cells. Treatment of HepG2 cells with $10{\mu}M\;or\;15{\mu}M$ sulforaphane resulted in significant G2/M cell cycle arrest as determined by DNA flow cytometry. Moreover, $20{\mu}M$ sulforaphane significantly induced the population of sub-G1 cells suggesting that sulforaphane induced apoptosis. This anti-proliferative effect of sulforaphane was accompanied by a marked inhibition of ryclin A, cyclin 31 and Cdc2 protein. However, the levels of tumor suppressor p53 and Cdk inhibitor p21 mRNA and protein expression were significantly increased by sulforaphane treatment in a concentration-dependent manner. Although further studies are needed, the present work suggests that sulforaphane may be a potential rhemoprevetiveichemotherapeucc agent for the treatment of human cancer cells.

Localization of the Neurons Projecting to the Gallbladder Meridian (족소양담경(足少陽膽經)에서 투사(投射)되는 신경원(神經元)의 표지부위(標識部位)에 대한 연구(硏究))

  • The purpose of this morphological studies was to investigate the relation to the meridian, acupoint and nerve. The common locations of the spinal cord and brain projecting to the the gallbladder, GB34 and common peroneal nerve were observed following injection of transsynaptic neurotropic virus, pseudorabies virus(PRV), into the gallbladder, GB34 and common peroneal nerve of the rabbit. After survival times of 96 hours following injection of PRV, the thirty rabbits were perfused, and their spinal cord and brain were frozen sectioned($30{\mu}m$). These sections were stained by PRV immunohistochemical staining method, and observed with light microscope. The results were as follows: 1. In spinal cord, PRV labeled neurons projecting to the gallbladder, GB34 and common peroneal nerve were founded in thoracic, lumbar and sacral spinal segments. Densely labeled areas of each spinal cord segment were founded in lamina V, VII, X, intermediolateral nucleus and dorsal nucleus. 2. In medulla oblongata, The PRV labeled neurons projecting to the gallbladder, GB34 and common peroneal nerve were founded in the A1 noradrenalin cells/C1 adrenalin cells/caudoventrolateral reticular nucleus, rostroventrolateral reticular nucleus, medullary reticular nucleus, dorsal motor nucleus of vagus nerve, nucleus tractus solitarius, raphe obscurus nucleus, raphe pallidus nucleus, raphe magnus nucleus, gigantocellular nucleus, lateral paragigantocellular nucleus, principal sensory trigeminal nucleus and spinal trigeminal nucleus. 3. In Pons, PRV labeled neurons were parabrachial nucleus, Kolliker-Fuse nucleus and cochlear nucleus. 4. In midbrain, PRV labeled neurons were founded in central gray matter and substantia nigra. 5. In diencephalon, PRV labeled neurons were founded in lateral hypothalamic nucleus, suprachiasmatic nucleus and paraventricular hypothalamic nucleus. 6. In cerebral cortex, PRV labeled neuron were founded in hind limb area.This results suggest that PRV labeled common areas of the spinal cord projecting to the gallbladder, GB34 and common peroneal nerve may be first-order neurons related to the somatic sensory, viscero-somatic sensory and symapathetic preganglionic neurons, and PRV labeled common area of the brain may be first, second and third-order neurons response to the movement of smooth muscle in gallbladder and blood vessels.These PRV labeled neurons may be central autonomic center related to the integration and modulation of reflex control linked to the sensory system monitoring the internal environment, including both visceral sensation and various chemical and physical qualities of the bloodstream. The present morphological results provide that gallbladder meridian and acupoint may be related to the central autonomic pathways.

Synergistic antitumor activity of ST1571 and camptothecin in human cancer cells (Camptothecin 에 의한 ST1571 의 항암 활성 증강)

  • The in vitro activity of ST1571, an inhibitor of the Abl group of protein-tyrosine kinases, alone or in combination with camptothecin (CPT), a specific topoisomerase I inhibitor, was evaluated against human cancer cells with different metastatic capacity and drug resistance potency. These cell lines showed different sensitivity to ST157 on growth inhibition, and the expression of DNA-dependent protein kinase (DNA-PK), which interacts constitutively with c-Abl, was significantly decreased in drug sensitive CEM and MCF-7 cells and poorly metastatic PC3 and KMl2 cells as compared with that of multidrug resistant CEM/MDR and MCF-7/MDR cells and highly metastatic PC3-MM2 and KM/L4a cells, respectively. These results suggest differential modulation of DNA-PK by ST1571 treatment in drug resistance and metastatic degree dependent manner. We showed that CPT as well as ST1571 significantly inhibits the expression of DNA-PK. The combined treatment with ST15fl and CPT revealed synergistic effect, and the effect was accompanied by inhibition of cell proliferation due to significant reduced expression of DNA-PK components, which resulted in CPT sensitizes human cancer cells resistant to ST1571. Therefore, the results of our study suggested that the suppression of DNA-PK using combination of ST1571 and CPT could be a novel molecular target for against drugresistant and metastatic cancer cells.
