Repeated Dose 90-Day Oral Toxicity Study of Modified Samjung-Hwan in Sprague-Dawley Rats (삼정환의 랫드를 이용한 90일 반복 경구투여 독성시험)

  • Objectives: The study is aimed at evaluating the possible toxicity in 90-day repeated oral administration of modified Samjung-hwan (mSJH) in Sprague-Dawley (SD) rats. This study was conducted to detect the no-observed adverse effect level (NOAEL). Methods: Modified SJH extract was administered orally in male and female SD rats at dose of 0, 1,000, 2,000, 4,000 mg/kg. Each group consisted of 10 rats of each gender. The modified SJH extract was given once a day for 90 days. We monitored the changes of mortalities, clinical signs, body weight changes, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers of all animals treated with modified SJH extract during the study period. Results: There were no toxicologically significant changes in mortalities, clinical signs, body weight gains, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers in any of rats tested. Conclusions: The NOAEL of the modified SJH extract in male rats and no observed effect level (NOEL) in female rats are considered 4,000 mg/kg.

Safety Evaluation of Genetically Modified Organisms (GMO) for a 90-day Exposure in Rats (랫드에서 유전자 재조합 식품(GMO)의 90일간 노출에 대한 안전성 평가)

  • We performed to evaluate the safety of GMOs for a long term exposure in Sprague-Dawley (SD) rats. In this study, groups often or fifteen SD rats were fed one of the following four diets for 90 days: (1) AIN-76A rodent diet only; (2) AIN-76A rodent diet containing 5% genetically modified soybean from USA; (3) AIN-76A rodent diet containing 5% genetically non-modified soybean from USA; (4) AIN-76A rodent diet containing 5% genetically non-modified soybean from Korea. The effects of AIN-76A rodent diet containing genetically modified soybean on body weights, food uptake, water consumption, hematology, serum bio-chemistry, urinalysis, organ weights, gross findings and histopathological findings were not significantly different, compared with others. Taken together, these results suggested that genetically modified soybean did not induce any toxic effects in rats treated for 90 days.

Modified Organs of Air Breathing Fishes in Korea (어류의 공기호흡 기관)

  • Modified organs for air respiration in Korean fish was reviewed in the following 6 Korean fishes: three mudskippers (Boleophthalmus pectinirostris, Periophthalmus modestus and P. magnuspinnatus), two mud loaches (Misgurnus mizolepis and M. anguillicaudatus), and a torrent catfish (Liobagrus mediadiposalis). Three mudskippers and a torrent catfish have a modified epidermis to in order to make up for the deficient oxygen supply. Their epidermis has abundant intraepithelial blood capillaries except dermal capillaries situated just beneath the stratum germinativum of the epidermis in B. pectinirostris. The epidermis was thick due to component of the following cells: two kinds of glands as a small mucous cells and a large club cells in L. mediadiposalis, voluminous cells (swollen cells) swollen by epidermal cells and a small mucous cells in B. pectinirostris, and only voluminous cells having no any glandular cells in P. modestus and P. magnuspinnatus. In Particular, the epidermis of the mudskippers appears to be a web-like structure due to the swollen epithelial cells. The dermal bulges are found in B. pectinirostris and they are situated at the skin covering the body, not appendage of all the fins and the sucking disc. Another modified organ in M. mizolepis and M. anguillicaudatus occurs in intestine and its mucosal epithelium has abundant blood capillaries.

  • Recently, polymer-clay hybrid materials have received considerable attention from both a fundamental research and application point of view. This organ-inorganic hybrid, which contains a nanoscale dispersion of the layered silicates, is a material with greatly improved thermal and mechanical characteristics. Two classes of nanocomposites were synthesized using an unsaturated polyester resin as the matrix and sodium montmorillonite as well as an organically modified montmorillonite as the reinforcing agents. X -ray diffraction pattern of the composites showed that the interlayer spacing of the modified montmorillonite were exfoliated in polymer matrix. The mechanical properties also supported these findings, since in general, tensile strength, modulus with modified montmorillonite were higher than the corresponding properties of the composites with unmodified montmorillonite. Adding organically modified clay improved the tensile strength of unsaturated polyester by $22\%$ and the tensile modulus of unsaturated polyester was also improved by $34\%$.

Effect of Chlorella vulgaris intake on cadmium detoxification in rats fed cadmium

  • The aim of this study was to investigate if dietary Chlorella vulgaris(chlorella) intake would be effective on cadmium(Cd) detoxification in rats fed dietary Cd. Fourteen-week old male Sprague-Dawley(SD) rats weighing $415.0{\pm}1.6\;g$ were randomly divided into two groups and fed slightly modified American Institute of Nutrition-93 Growing(AIN-93G) diet without(n=10) or with(n=40) dietary Cd(200 ppm) for 8 weeks. To confirm alteration by dietary Cd intake, twenty rats fed AIN-93G diet without(n=10) and with(n=10) dietary Cd were sacrificed and compared. Other thirty rats were randomly blocked into three groups and fed slightly modified AIN-93G diets replacing 0 (n=10), 5 (n=10) or 10% (n=10) chlorella of total kg diet for 4 weeks. Daily food intake, body weight change, body weight gain/calorie intake, organ weight (liver, spleen, and kidney), perirenal fat pad and epididymal fat pad weights were measured. To examine Cd detoxification, urinary Cd excretion and metallothonein (MT) concentrations in kidney and intestine were measured. Food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weight and fat pad weights were decreased by dietary Cd intake. Urinary Cd excretion and MT concentrations in kidney and small intestine were increased by dietary Cd. After given Cd containing diet, food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weights and fat pad weights were not influenced by dietary chlorella intake. Renal MT synthesis tended to be higher in a dose-dependent manner, but not significantly. And chlorella intake did not significantly facilitate renal and intestinal MT synthesis and urinary Cd excretion. These findings suggest that, after stopping cadmium supply, chlorella supplementation, regardless of its percentage, might not improve cadmium detoxification from the body in growing rats.

Effects of Modified Montmorillonite Nanocomposite on Growing/Finishing Pigs during Aflatoxicosis

  • Experiments were conducted to determine the efficacy of modified montmorillonite nanocomposite (MMN) to reduce the toxic effects of aflatoxin (AF) in growing/finishing pigs. 96 weaned pigs were assigned to four dietary treatment groups (0 g of MMN and 0 mg of AF/kg feed, 3 g of MMN/kg feed, 0.1 mg of AF/kg feed, and 3 g of MMN plus 0.1 mg of AF/kg feed). Body weight gain (BW gain), feed/gain ratio, serum biochemical values and enzyme activities were evaluated. Compared with the control, AF alone markedly reduced BW gain and resulted in a significantly higher feed/gain ratio. There were no differences in BW gain and feed/gain ratio between 0.3% MMN or 0.3% MMN plus AF and the control. These results suggested that the deleterious effects of AF were ameliorated by MMN addition. AF intake markedly increased relative organ weights of liver, kidney, spleen and pancreas, and resulted in significant alterations of serum parameters. However, these parameters for pigs fed diets containing MMN and AF returned to normal values, indicating that MMN had the ability to recover the AF-decreased performance, organ damage and to correct aberrations in serum parameters. These findings in our study suggested that MMN can effectively modulate the toxicity of AF in growing/finishing pigs and may offer a novel approach to the preventive management of aflatoxicosis in animals.

Objects of Research This research is purposed to find Dong-mu's thought on Pharmacology, through summarizing sentences related to pharmacology proposed in the chief writings on Sasang Constitutional Medicine. 2. Methods of Research It was researched bibliologically with Dong-mu's chief medical writings such as "Dongyi Suse Bowon Sasang Chobongyun(東醫壽世保元四象草本卷)", "Dong-mu's posthumos work(東武遺藁)", "Dongyi Suse Bowon(東醫壽世保元)". 3. Results and Conclusions 1. The grouping criteria according to the constitution proposed in "Dongyi Suse Bowon Sasang Chobongyun" is 'Spreading Outside' 'Hardening Inside' 'Cooling Organ' 'Warming Inside', but the grouping of medical herbs according to the constitution is yet not accomplished, and medical herbs is comprehended on base of effectiveness as like existing oriental medicine. 2. The grouping criteria according to the constitution proposed in "Dong-mu's posthumos work"is 'Spreading Choking Raising Lowering' and 'Perfume Smell Fluid Taste', and medical herbs is comprehended on base of effectiveness as like "Chobongyun", and the example on mixing of the herb classified according to the constitution is showed. 3. The medical herbs are comprehended on the basis of how to effect on treatment of constitutional disease, and the principle that it's not to mix of the herb classified according to the constitution is proposed in "Dongyi Suse Bowon". 4. The pharmacology of Sasang Constitutional Medicine is accomplished after it is modified many ti mes through clinic experience for a long time. 5. The pharmacology of Sasang Constitutional Medicine in the early period, is to comprehend herbs on the basis of how to complement 'Spreading Gathering Raising Lowering' of 'Small-Organ(偏小之臟)' according 'Qi-Mei(氣味)' and effectiveness, but that in the late period is to comprehend herbs on the basis of how to complement or to control 'Small-Organ(偏小之臟)' and 'Large Organ(偏大之臟)'.

  • Ventricular septal defect complicating myocardial infarction is rare but fatal condition which requires early surgical intervention before end-organ failure ensues from cardiogenic shock. Since the first successful repair by Cooley et al in 1956, surgical skills and strategies were developed and modified to a great extent, and we adopted the new repair technique in our case which stresses that minimal or no part of the infarcted septum and left ventricular wall be resected. This technique obviates the need to resect the infarcted part of the septum and prevents recurrence of an even larger VSD, and provides adequate size and shape of the left ventricle after of transinfarction left ventriculotomy.

Changes of Tissue Factor Activity on Inflammatory Stimulus and Aging in Rat

  • Tissue factor (TF), a principal initiator of the veertebrate coagulation cascade, is expressed in organ tissues, cells and blood. TF is konwn to be induced in endothelial cells, monocytes and macrophages by inflammatory stimuli and in many pathologic conditions. By using the modified method for in vido TF activity assay, we found that turpentine oil injection as an inflamatory stimulus also induced the TF activity in lung and brain tissues of rats. And the age-related increase in Tf activity was observed in healthy rat brain tissue.

Thirteen-week repeated-dose oral toxicity study of the Modified Wenpitang-Hab-Wulingsan (WHW$^{(R)}$) in Sprague-Dawley rats (WHW$^{(R)}$의 랫드에서의 반복경구투여 독성에 관한 연구)

  • Objectives : We investigated the repeated-dose toxicity of Wenpitang-Hab-Wulingsan(WHW), a Korean traditional medicine prescribed with twelve herbs, which has been used for the treatment of renal disease. Methods : WHW extract prepared by GLP company. WHW was supplemented by gavage at 0, 100, 500 and 1000 mg/kg/day for 13-week consecutive days. We recorded the clinical signs of toxicity, body weight, organ weights, hematology, gross and histological changes in target organs rats and clinical chemistry analysis for all rats. Results : WHW extract at all doses was shown no mortality or abnormal clinical signs in rats during at the observation period. Furthermore, there was no difference in body weight and food-take consumption, organ weight, gross pathological findings, and urine analysis among the groups of rats treated with different doses of WHW extract. The hematological analysis and clinical blood chemistry data were revealed no toxic effects from WHW-treated rats. Conclusions : The results suggest that WHW extract in rats is a wide margin of safety on a acute toxicity.
