In Vitro Plant Regeneration from Stolen Node Explant in Eremochloa Ophiuroides (Munro) Hack

  • In vitro shoot regeneration and multiple shoot induction has been obtained from the stolen node explants in Eremochloa ophiuroides (Munro) Hack. The highest number of shoots ($10.66{\pm}0.21$) was observed from initial explants after one month culture duration on Murashige and Skoog (MS) medium containing 6-benzyladenine (BA: 0.5 mg/l). First generation shoot was excised and sub-cultured on the same fresh media for further multiplication of shoots. An enhanced number of second round shoots ($15.33{\pm}0.21$) was obtained compared to the initial culture media containing BA (0.5 mg/l). The number of shoots/stolon node was higher among all the concentrations of BA than kinetin (KN). In vitro regenerated shoots were successfully rooted in the phytohormone free MS medium. Plantlets generated with roots were transferred to pots containing compound mixture of soil and kept in green house conditions. Acclimatized plants showed 100% survival rate with normal morphology in green house conditions. The present study demonstrates the effect of explant and different plant growth regulators towards in vitro response in E. ophiuroides. Moreover, the study reveals the effect of cytokinin on induction of shoot number per stolen node explant in E. ophiuroides.

Numerical Analysis of Rocket Exhaust Plume with Equilibrium Chemistry and Thermal Radiation (화학 평형과 열복사를 포함한 로켓 플룸 유동 해석)

  • Numerical study is carried out to investigate the effects of chemistry and thermal radiation on the rocket plume flow field at various altitudes. Navier-Stokes equations for compressible flows were solved by a fully-implicit TVD code based on the finite volume method. An infinitely fast chemistry module for hydrocarbon mixture with detailed thermo-chemical properties and a thermal radiation module for optically thick media were incorporated with the fluid dynamics code. The plume flow fields of a kerosene-fueled rocket flying at Mach number zero at sea-level, 1.16 at altitude of 5.06 km and 2.90 at 17.34 km were numerically analyzed. Results showed the plume structures at different altitude conditions with the effects of chemistry and radiation. It is understood that the excess temperature by the chemical reactions in the exhaust gas may not be ignored in the view point of propulsion performance and thermal protection of the rocket base, especially at higher altitude conditions.

  • Synthesis diamond films have been deposited on the molybdenum substrates using an microwave plasma enhanced chemical vapor deposition method. The effects of deposition time, surface morphology, infrared transmittance and Raman scattering have been studied. Diamond deposited on molybdenum substrate for 100 hours by MW plasma CVD from $CH_4-H_2-O_2$ gas mixture had good crystallity with $100[{\mu}m]$ thickness needed for radiation detector. Diamond radiation detector of M-I-M type was made and the current of radiation detector was increased by increasing X-ray dose.

  • A new method for immobilization of glucose oxidate by the aerobic gamma radiation of synthetic monomers was developed. The radiocopolymerization was conducted aerobically at -70 to -8$0^{\circ}C$ with the mixture of several polyfunctional esters, acrylates and native enzyme. The retained activity of immobilized glucose oxidase was about 50 to 55% when a NK 23G ester, acrylamide-bis and water mixture (1:1:2) in cold toluene treated with 450 krad of gam-ma radiation. The radiation dose did not influence significantly to the enzyme activity. The solvents used to prepare the beads of glucose oxidase and monomers were toluene, n-hexane, petoleum ether and chloroform. 0.05M tris-gycerol (pH 7.0) was a more suitable bugger solution for immobilizing the enzyme than was 0.02M phosphate. Immobilization of glucose oxidase shifted the optimum pH for its reaction from 6.0 to 6.5. The pH profile for the immobilized enzyme showed a broad range of optimum activity while the native enzyme gave a sharp pick for its optimum pH value. The immobilized enzyme reaction temperature was at the range of 30~4$0^{\circ}C$.

Environmental Control in the Plant Factory System Influences Year-Round Production of Allium hookeri Leaves (삼채(Allium hookeri) 잎 연중생산을 위한 식물공장 환경제어 효과)

  • The demand for the fresh leaf of hooker chive, which is mainly used as functional roots and contains dietary sulfur or saponin, is increasing, but the leaves are only harvested 3-4 times per year under conventional field conditions. A plant factory system with different light qualities or intensities was applied for year-round production of the fresh leaves. Hooker chive (Allium hookeri) roots were hydroponically cultured under the plant factory with a mixture of blue plus red LEDs (Light-Emitting Diodes) and fluorescent lights for 50 weeks. Maximum leaf growth was attained with the 1.5 dS/m EC in the culture medium under the plant factory. The average leaf and shoot numbers of hooker chive grown hydroponically under a mixture of 200 µmol/m2/s LEDs increased by 147% and 140%, respectively compared to those under 100 µmol/m2/s LEDs at the 10th harvest. The leaf length of hooker chive grown under the LEDs treatment with the lowest light intensity significantly increased by 27% compared with the natural light treatment at the 10th harvest. However, there was no significant difference in leaf pigmentation between natural and 200 µmol/m2/s LEDs treatments. Plant factory with the mixture LEDs of blue and red lights can be applied for year-round production of hooker chive fresh leaves to ensure a stable supply of leafy vegetable throughout the year.

  • Some physical and chemical properties of wood-plastic combinations(W.P.C.) made of domestic soft woods such as pinus densiflora, pinus rigida and poplus deltoides were measured. The rates of improvement in properties were roughly proportional to the contents of polymer or polymer mixtures in W.P.C. For the W.P.C. obtained by means radiation curing and containing 80% of polymer or polymer mixture, the hardness and water absorptirity were improved 2.2 times and 4 times those of the original wood, respectively. The improvement of hardness was especially remarkable in the W.P.C. made of pinus densiflora and polystyrene(120%) to show 7 times increased hardness. For the W.P.C. obtained by means of thermal curing and containing 80% of polymer or polymer mixture, the hardness and water absorptivity were improved 2.4 times and 3.4 times those of the non-treated woods, respectively. These data indicate that the properties of W.P.C. prepared by means of radiation curing are not much different from those of W. p. C. prepared by means of thermal curing. Both of acid resistancy and alkali resistancy of the W.P.C. were also improved remarkably in comparison with the non-treated wood.

  • It is possible to count and perform quench correction on two ${\beta}$ -label samples so long as the maximum ${\beta}$-energies are sufficiently different. However, when 4he conventional technique is applied to the radioassay of a mixture of more than three nuclides, the reliability of the activities determined is considerably reduced, resulting from the large overlapping of liquid scintillation pulse height distributions of each nuclide. A technique that allows the activities of multiple ${\beta}$-labeled samples to be radioassayed was proposed by using the least square method. The technique was applied to mixture samples of $^3H,\;^{14}C,\;^{36}Cl$, and $^{90}Sr$. The analytical values were in good agreement with the reference values within 7% relative error.

New polyester composites synthesized with additions of different sized ZnO to study their shielding efficiency

  • This investigation developed a novel polyester composite based on the addition of zinc oxide (ZnO) of different sizes. We prepared nine samples Containing different percentages and sizes of ZnO as well as the control sample (Pol-ZnO0). The attenuation factors of Pol-micro ZnO were estimated using Phy-x software, while the HPGe detector and various gamma sources were used to experimentally measure the all-prepared composites. In terms of the two methods for micro composites, good agreement was observed. The linear attenuation coefficient (LAC) of Pol-ZnO20, Pol-ZnO40, and Pol-ZnO60, two more samples one with ZnO nanoparticles instead of microparticles, and the other with half microparticles and half nanoparticles (referenced as 0.5 M + 0.5 N) were determined. For all the Polyester composites and energies, the mixture of microparticles and nanoparticles had greater LAC values than each of the particles on their own. For example, the LAC values for the Pol-ZnO20 polymer at 1.330 MeV are 0.0836, 0.0888, and 0.0903 cm-1 for the microparticles, nanoparticles, and mixture, respectively. The values of the prepared polymer samples' radiation protection efficiency (RPE) against energy with a thickness of 2 cm was determined experimentally. The Pol-ZnO60 0.5 M + 0.5 N sample has the highest RPE out of all the samples, followed by its nanoparticle counterpart, and then its microparticle counterpart. On the other hand, the Pol-ZnO0 sample, the polymer with no ZnO content, at all energies has the lowest RPE, followed by the three Pol-ZnO20 samples.

Preparation of Epoxy-Methacrylate Prepolymer and Electron Beam Curing of Its Mixture with Monomers (Epoxy-Methacrylate Prepolymer의 合成 및 그의 單位體 混合物의 電子線硬化)

  • Epoxy-methacrylate prepolymer was synthesized from methacrylic acid and a diglycidyl ether type low molecular weight epoxy resin, and the electron beam curing of the prepolymer was studied using styrene and polyethyleneglycol dimethacrylates as comonomers. The esterification was carried out quantitatively without any side reaction adding more than 250∼300ppm hydroquinone and less than $1{\%}$ (wt) triethanolamine by wt. of methacrylate acid, respectively. In the radiation curing under air atmosphere, the maximum gel fraction was observed at 10∼$30{\%}$ monomer contents. The rate of gel formation was decreased by the presence of unreacted epoxy group and acid in the prepolymer and increased with increasing degree of polymerization of polyethyleneglycol block in the dimethacrylates. Considerable oxygen effect in the curing, particularly when polyethyleneglycol dimethacrylate was used as a comonomer, was recognized through the differences of the product properties, though no remarkable differences were found in the rate of gel formation in air and nitrogen atmosphere.

Development of an Organic Scintillator Sensor for Radiation Dosimetry using Transparent Epoxy Resin and Optical Fiber (투명 에폭시와 광섬유를 이용한 방사선량 측정용 유기섬광체 센서 개발)

  • Remote detecting system for a radiation contamination using a plastic scintillator and an optical fiber was developed. Using a commercially available silica optical fiber and a plastic scintillator, we tested then for a real possibility as a remote monitoring detector. Also, a plastic scintillator was developed by itself, and evaluated as a radiation sensor. The plastic scintillator was made of epoxy resin, a hardener and an organic scintillation material. The mixture rate of the epoxy resin, hardener and organic scintillator was fixed by using their emission spectrum, transmittance, intensity etc. In this study, in order to decrease the light loss of an incomplete connection between an optical fiber and a scintillator, the optical fiber was inserted into the scintillator during the fabrication process. The senor used a plastic optical fiber and was estimated for its detection efficiency by an optic fiber's geometric factor.
