선식에서 분리한 Enterobacter sakazakii의 복합동정 및 RAPD를 이용한 genotyping (Multiple Confirmation and RAPD-genotyping of Enterobacter sakazakii Isolated from Sunsik)

  • 시판되고 있는 선식 원료를 수거하여 최근 새로운 식중독균으로 보고되고 있는 Enterobacter sakazakii 분리 실험을 실시하였다. 그 결과 총 23종의 선식 원료 중 8개의 선식 원료에서 E.sakazakii로 추정되는 콜로니를 분리할 수 있었으며 API 20E kit를 이용하여 1차적으로 동정한 결과, 다시마 분말, 멸치 분말, 현미 분말, 청국장 분말 및 멥쌀 분말에서 E. sakazakii를 분리할 수 있었다. 이후 3 종의 primer를 이용한 PCR을 실시하여 2차적으로 동정하였다. 또한, 분리된 균주에 대한 RAPD-PCR을 실시하여 최종적으로 8종의 분리균으로 molecular typing을 할 수 있었다.

기계적 합금화에 의한 Mg-Si계 열전화합물의 합성 및 평가 (Synthesis and characterization of Mg-Si thermoelectric compound subjected to mechanical alloying)

  • 본 연구에서는 나노결정립의 $Mg_2Si$ 열전화합물을 제조하기 위하여 기계적 합금화(MA)를 적용하였다. 단상의 초미세 $Mg_2Si$ 열전화합물을 얻기 위하여 최적 볼밀조건 및 열처리 조건을 X선 회절분석과 시차주사 열량분석을 이용하여 조사하였다. $Mg_{66.7}Si_{33.3}$ 혼합분말을 $20{\sim}180$시간까지 볼밀 처리한 경우 모든 시료에서 $220^{\circ}C$$570^{\circ}C$ 근방에 broad한 발열 반응이 관찰되었다. 한편 $Mg_{66.7}Si_{33.3}$ 혼합분말을 260시간 동안 볼밀 처리한 경우 $230^{\circ}C$에 예리한 발열피크를 보였다. 단상의 $Mg_2Si$ 화합물은 $Mg_{66.7}Si_{33.3}$ 혼합분말을 60시간 동안 MA처리 후 $620^{\circ}C$까지 열처리함으로써 얻을 수 있었다. MA분말시료의 치밀화는 50MPa, $800{\sim}900^{\circ}C$에서 흑연다이를 사용하여 SPS 소결을 실시하였다. Mg-Si계 MA 분말시료의 SPS 소결시 수축은 $200^{\circ}C$ 근방에서 현저하게 관찰되었다. SPS법으로 고화된 성형체의 밀도측정 결과, 모든 시료에서 이론밀도의 94% 이상 금속광택을 나타내는 치밀한 소결체임을 알 수 있었다.

현미 및 율무 함유 생식이 영양불균형이 유도된 흰쥐의 체내 지질농도, 항산화체계 및 면역기능에 미치는 영향 (Effect of Raw Broun Rice and Job식s Tear Supplemented Diet on Serum and Hepatic Lipid Concentrations, Antioxidative System, and Immune Function of Rats)

  • 본 연구에서 는 sub-optimal 수준의 비타민과 무기질을 섭취하며, 고콜레스테롤 식이에 알코올 섭취를 병행하여 영양 불균형이 유도된 흰쥐를 대상으로 현미와 율무로 구성된 생식 또는 열처리생식을 식이에 첨가하여 5주간 사육시킨 후 체내 지질농도, 항산화체계 및 면역기능에 미치는 영향을 평가하여 다음과 같은 결과를 얻었다. 영양불균형대조군의 식이효율은 일반대조군에 비해 더 낮았고, 현미 및 율무 함유생식 또는 열처리생식을 첨가한 결과 영양불균형대조군에 비해 식이효율이 유의적으로 증가하였다(p<0.05).혈청 총콜레스테롤 농도는 일반생식군 또는 열처리생식군에서 영양불균형대조군에 비 해 각각 24% 또는 16% 유의적으로 감소하였고(p<0.05) LUL+VLDL-콜레스테롤 농도 역시 일반생식군에서 영양불균형대조군에 비해 26% 유의적으로 감소하였으나(p<0.05), atherogenic index에는 유의적인 차이가 없었다. 일반생식군의 간조직 콜레스테롤 농도는 영양불균형대조군에 비해 16% 유의적으로 감소하였다(p<0.05). 간조직의 지질과산화물 농도는 일반생식군 또는 열처리 생식 군에서 영양불균형대조군에 비해 각각 38%또는 59% 유의적으로 감소하였다(p<0.05). 한편, 영양불균형대조식이에 일반생식 또는 열처리생식을 첨가한 결과 간조직의 항산화효소계 활성에 유의적인 영향을 미치지 못하였다 장간막임파절의 CD4$^{+}$T-세포 분포 및 CD4$^{+}$/CD8$^{+}$비율은 일반생식군에서 영양불균형대조군에 비해 유의적으로 증가한 반면(P<0.05), 열처리생식군과 영양불균형대조군 간에는 유의적 차이가 없었다. 이상의 결과를 종합해 보면 열처리를 하지 않은 현미 및 율무 함유 생식은 열처리생식에 비해 영양불균형 흰쥐의 체내 콜레스테롤 농도 저하효과 및 장간막임파절의 면역활성도 증가효과가 더 우수한 것으로 나타냈다.

17세기 이전 조선시대 과정류의 문헌적 고찰 (An Investigation on 'Kwa-Jung';Traditional Korean Confectionery Items, Found in Korean Literatures Prior to the 17th Century)

  • In this study, we investigated the names and various types of "kwa-jung" along with their recipes and ingredients occurring in Korean cookbooks published before the 17th century. The kwa-jungs were classified into 7 groups including Yoomilkwa, Yookwa, Dasik, Junkwa, Kwapyun, Yutganjung and Dang. A total of 72 kinds of kwa-jung were found. 26 kinds of Yoomilkwa, 18 kinds of Yookwa, 7 kinds of Dasik, 10 kinds of Junkwa, 1 kind of Kwapyun, 3 kinds of Yutganjung and 8 kinds of Dang. Among the types of Yoomilkwa, Yackwa was recognized as the best food according to the references. Also the cooking methods for items had a tendency to become simpler as society became modernized. Original and rare Ingredients were often substituted with other items that could be found more easily where people lived As recorded, the Yookwa group included kangyung, sanja, and bingsakwa. And as society became modernized, people tended to buy Yookwa in the store rather than making it at home due to its complicated and often difficult cooking process. The Dasik items were a kneaded mixture of flour, or the flour of chestnuts, with honey. These were formed into various patterns like birds, animals, butterflies, tree leaves and flowers, or as ki-wha by using printing cooking utensils Honey was used as a sweetener and as a combining material. The Junkwa consisted of roots or fruits that could be easily obtained these cooked or raw foodstuffs were then mixed with sugar and simmered. The Kwapyun used sour fruit juices as a main ingredient. These were then combined with sugar and simmered and allowed to harden. Then they were cut into square shapes after cooling. Yutkangjung was a mixture of yut, chochung, honey or syrup and pine nuts. which was combined over low heat. After mixing and stirring it was cut into square shapes. Finally, the Yut was typically cooked with grains and powdered malt and stirred until thickened.

전자빔 증착 열차폐 코팅용 란타늄-가돌리늄 지르코네이트(La2O3-Gd2O3-ZrO2계) 세라믹 잉곳의 제조공정에 따른 열충격 저항성 (Thermal Shock Resistance According to the Manufacturing Process of Lanthanum Gadolinium Zirconate Ceramic Igot for Thermal Barrier Coating by Electron Beam in the La2O3-Gd2O3-ZrO2 System)

  • The ingot fabrication conditions related with the thermal shock bearing phase and microstructure have investigated for the rare earth zirconate ceramic material, lanthanum gadolinium zirconate, as a thermal barrier coating using electron beam evaporation method. The thermal shock resistance of the prepared ingot was evaluated by high energy electron beam irradiation. The rare earth zirconate ceramic powder was prepared by controlling the raw material powder composition of $La_2O_3$, $Gd_2O_3$ and $ZrO_2$ so as to have a composition of $(La_{0.3}Gd_{0.7})_2Zr_2O_7$ which was selected from the former study. Ingot samples were prepared under two conditions. The first condition is prepared by sintering the prepared powder mixture to form an ingot. The second condition is prepared by calcining the prepared powder mixture to form a composite phase and then sintering to form an ingot. X-ray diffraction(XRD) and Scanning Electron Microscope(SEM) were used to analyze phase forming behavior and microstructure of ingot samples. Nanoindentation method used to obtain elastic modulus and hardness of each ingot specimen. Also the stress distribution of ingot was simulated by using FEM method assuming the ingot surface was exposed to electron beam. As a results, in the case of an ingot having a network-shaped microstructure in which relatively coarse pores are included, it seems that the thermal shock resistance was higher than in the case of an ingot having a microstructure composed of relatively fine grains only or particles with the similar level size when the high energy electron beam irradiation.

고연색 백색 광원용 BaSi2O2N2:Eu 형광체의 광학·구조 특성 분석 (Optical and Structural Analysis of BaSi2O2N2:Eu Green Phosphor for High-Color-Rendering Lighting)

  • Green $BaSi_2O_2N_2:0.02Eu^{2+}$ phosphor is synthesized through a two-step solid state reaction method. The first firing is for crystallization, and the second firing is for reduction of $Eu^{3+}$ into $Eu^{2+}$ and growth of crystal grains. By thermal analysis, the three-time endothermic reaction is confirmed: pyrolysis reaction of $BaCO_3$ at $900^{\circ}C$ and phase transitions at $1,300^{\circ}C$ and $1,400^{\circ}C$. By structural analysis, it is confirmed that single phase [$BaSi_2O_2N_2$] is obtained with Cmcm space group of orthorhombic structure. After the first firing the morphology is rod-like type and, after the second firing, the morphology becomes round. Our phosphor shows a green emission with a peak position of 495 nm and a peak width of 32 nm due to the $4f^65d^1{\rightarrow}4f^7$ transition of $Eu^{2+}$ ion. An LED package (chip size $5.6{\times}3.0mm$) is fabricated with a mixture of our green $BaSi_2O_2N_2$, and yellow $Y_3Al_5O_{12}$ and red $Sr_2Si_5N_8$ phosphors. The color rendering index (90) is higher than that of the mixture without our green phosphor (82), which indicates that this is an excellent green candidate for white LEDs with a deluxe color rendering index.

Influence of grain size ratio and silt content on the liquefaction potentials of silty sands

  • Soil liquefaction has been one of the most important concerns in geotechnical earthquake engineering in recent years, due to its damages to structures and its destructive effects. The cyclic liquefaction of silty sands, in particular, remains of great interest for both research and application. Although many factors are known that affect the liquefaction resistance of sands, the effect of fine grain content is perhaps one of the most studied and still controversial. In this study, 48 deformation-controlled cyclic simple shear tests were performed on BS and CS silt samples mixed with 5%, 15% and 30% by weight of Krk085, Krk042 and Krk025 sands in constant-volume conditions to determine the liquefaction potential of silty sands. The tests were carried out at 30% and 50% relative density and under 100 kPa effective stress. The results revealed that the liquefaction potential of silty sand increases with increasing average particle size ratio (D50sand / d50silt) of the mixture for a fixed silt content. Furthermore, for identical base sand, the liquefaction potentials of coarse grained sands increase with increasing silt content, while the respective potentials of fine grained sands generally decrease. However, this situation may vary depending on the silt grain structure and is affected by the nature of the fine grains. In addition, the variation of the void ratio interval was shown to provide a good intuition in determining the liquefaction potentials of silty sands, while the intergranular void ratio alone does not constitute a criterion for determining the liquefaction potentials of silty sands.

대형주강의 고온파단형태에 미치는 [Mn/S]비의 영향 (The Influence of [Mn/S] Ratios on the Fracture Morphology of a Heavy-section Steel Castings at Elevated Temperature)

  • Using the Gleeble test, the effects of [Mn/S] ratios and the presence of sulfides on the high-temperature fracture morphology of heavy-section steel castings were analysed via the observations of the microstructures. The specimens for which the [Mn/S] ratio was in the range of 60~80 showed a ductile fracture morphology with an area reduction of more than 60%, while some specimens with similar [Mn/S] ratios showed a brittle fracture morphology with an area reduction of 0.0% due to the liquidation of sulfides at the grain boundary. The fracture morphology was classified into three types in the Gleeble high-temperature tensile test specimens. The first type showed dimple formation at the grain boundary, the formation of globular MnS sulfides, and plastic deformation of sulfides at an elevated temperature, indicating a needle-point type of ductile fracture with area reductions of 96.0~97.8%. The second type was a knife-edge type brittle fracture with an area reduction of 0.0% due to the film-type liquidation of sulfides at the grain boundary, band-type liquidation, and the liquidation of a terraced nipple pattern. The third type was the typical ductile fracture with an area reduction of 31.3~81.0%, in accordance with the mixture of dimples with in the grains and terraced nipple pattern at the grain boundary.

Preparation of Highly Tough Ethylene Vinyl Acetate (EVA) Heterogeneous Cation Exchange Membranes and Their Properties of Desalination

  • A manufacturing method has been devised to prepare novel heterogeneous cation exchange membranes by mixing ethylene vinyl acetate (EVA) copolymers with a commercial cation exchange resin. Optimum material characteristics, mixture ratios and manufacturing conditions have been worked out for achieving favorable membrane performance. Ion exchange capacity, electrical resistance, water uptake, swelling ratio and tensile strength properties were measured. SEM analysis was used to monitor morphology. Effects of vinyl acetate (VA) content, melt index (MI) and ion exchange resin content on properties of heterogeneous cation exchange membranes have been discussed. An application test was carried out by mounting a selected membrane in a membrane capacitive deionization (MCDI) system to investigate its desalination capability. 0.92 meq/g of ion exchange capacity, $8.7{\Omega}.cm^2$ of electrical resistance, $40kgf/cm^2$ of tensile strength, 19% of swelling ratio, 42% of water uptake, and 56.4% salt removal rate were achieved at best. VA content plays a leading role on the extent of physical properties and performance; however, MI is important for having uniform distribution of resin grains and achieving better ionic conductivity. Overall, manufacturing cost has been suppressed to 5-10% of that of homogeneous ion exchange membranes.

Optimization of One-step Extraction/Methylation Method for Analysis of Fatty Acid Composition in Brown Rice

  • Traditionally fatty acid composition used to be analysed by a GC and the sample preparation process includes lipid extraction from sample and subsequent methyl esters preparation, which are time-consuming and cumbersome. As an alternative, simultaneous extraction/methylation methods are being developed for rapid and simplified sample preparation. To optimize one-step extraction/methylation method for analysis of fatty acid composition in brown rice, various reaction factors such as sample to reaction solution ratio, reaction time and temperature, shaking intensity were changed and resultant fatty acid composition data were evaluated in comparison with previous reports. The ratio of sample weight to reaction solution volume was the most critical factor in that higher sample to reaction solution ratio caused overestimation of palmitic acid and linoleic acid composition, resulting in underestimation of oleic acid. Lower reaction temperature also induced overestimation of linoleic acid and underestimation of oleic acid. Reaction duration and the intensity of shaking prior to and during the reaction, however, caused no significant changes in analysis results. In conclusion, the optimum condition was mixing 5 grains (about 0.2 g) of brown rice with $680{\mu}L$ of extraction/methylation mixture and $400{\mu}L$ of heptane, followed by reaction at $80^{\circ}C$ for 2 hours.
