Polyvinyl Alcohol 분해 공생 균주에 의한 염색 폐수 중의 PVA 제거

  • The current processer of the textile wastewater treatment are mostly consisted of a combination of a physico-chemical and a biological treatment. The overall efficiency of these processes is, however, assessed to be fairly low. It is even worse during the summer season when temperature of the wastewater rises above 40$\circ $C. Therefore, a feasible process of the textile wastewater treatment which can work efficiently at higher temperatures was investigated in this work. We used a bench scale reactor consisted of one 4 liter anaerobic and one 8 liter aerobic tank, and the thermophilic symbiotic PVA degraders, Pasteruella hemolytica KMG1 and Pseudomonas sp. KMG6 that had been isolated in our laboratory. In the preliminary flask experiments, we observed that the thermophilic symbiotic PVA degraders could not grow in the wastewater substrate. Then, we isolated the mutant strains by acclimating the KMG1and KMG6 strains to the wastewater medium. The mutant symbionts (KMG1-1 and KMG6-1) were isolated through 6 times successive transfers into the fresh wastewater medium after 5 days culture for each. The mutant strains obtained grew well in the mixed medium composed of 75% wastewater and 25% synthetic medium, and supplemented with 0.5% PVA as a sole carbon source. During the culture for 14 days at pH 7.0 and 40$\CIRC $C, the bacteria assimilated about 89% of the added PVA. The symbionts degraded equally well all the PVA substrates of different molecular weight (nd=500~30000). In contrast to the flask experiments, in the reactor system the mutant strains showed very low levels of the PVA and COD removal rates. However, the new reactor system with an additional aerobic tank attained 82% removal rate of COD, 94% of PVA degradation and 71% of color index under the conditions of 5% inoculm on the tank 2, incubation temperature of 40$\circ $C, dissolved oxygen level of 2~3 mg/l and retention time of 30 hours. This result ensures that the process described above could be an efficient and feasible treatment for the PVA contained textile wastewater at higher temperatures.

Screening of zearalenone-producing strains by ELISA method (ELISA법에 의한 zearalenone 생성균주의 검색)

  • ELISA method was applied for the screening of zearalenone producing strains. The developed ELISA was as follow: $125\;{\mu}l$ of diluted solution (1 : 500) of antibody was added to each microtiter well and incubated overnight at $40^{\circ}C$. For direct competitive ELISA, samples and zearalenone-peroxidase conjugate were mixed in a 1 : 1 ratio, and a $100\;{\mu}l$ of aliquot was then added to antisera-coated wells. Plates were incubated for 30 minutes at $37^{\circ}C$, and wells washed 6 times, and $100\;{\mu}l$ of ABTS substrates was added. Plates were incubated for antother 15 minutes at $37^{\circ}C$, and $100\;{\mu}l$ of stopping reagent was added to the wells and absorbance was recorded at 410nm on ELISA Reader. Among 19 strains showed zearalenone-producing ability by ELISA, 3 strains (R-5, C-46, S-134) produced more than 50 ng/ml of zearalenone.

Degradation capability of macromolecular organic matters and antimicrobial activities of Bacillus species isolated from surf clam (Tresus keenae) (왕우럭(Tresus keenae)에서 분리된 Bacillus species의 고분자 유기물질 분해능력과 항균활성)

  • The production of enzymes that help digestion, assimilation of essential nutrients, and prevent pathogenic bacteria are important for probiotics used in aquaculture. The objective of this study was to investigate enzyme activities for macromolecular organic matters and antimicrobial properties of the selected potential probiotics isolated from gut of surf clam (Tresus keenae) against well-known shellfish-pathogenic bacteria. Among 65 isolates from guts of 60 surf clams, seven Bacillus strains with outstanding degradation capability of macromolecule organic matter were selected as potential probiotics as follows: TKI01 (B. vietnamensis), TKI02, TKI26 (B. thuringiensis), TKI14, TKI32, TKI42 (B. amyloliquefaciens), and TKI18 (B. stratosphericus). After in vitro antimicrobial activity test was performed against five shellfish-pathogenic bacteria including Listonella anguillarum, Vibrio parahaemolyticus, V. splendidus, V. harveyi, V. tubiashii, PCR assay was performed to detect bacteriocin-producing strain. PCR results revealed that the five Bacillus strains possessed diverse bacteriocin genes including ericinA, coagulin, surfactin, iturin, bacyllomicin, fengycin, bacylisin, subtilin, and lantibiotics. In the present study, the selected seven Bacillus strains showed different enzyme activities according to types of macromolecule organic matters. And their antimicrobial activities varied based on the species of pathogenic bacteria. In addition, at least five Bacillus strains had genetic potential to produce several natural lipopeptide antibiotics that may help biological control of surf clam aquaculture. Therefore, mixed use of probiotics might show co-operative effect and increase the efficiency of probiotics rather than separate use. To the best of our knowledge, it is the first report on antimicrobial properties of Bacillus species isolated from surf clam.

Effect of Supplmental Lactobacillus on Laying Performance, Intestinal Microflora and Egg Quality (유산균의 첨가 급여가 산란 생산성, 소화기관 미생물 변화 및 계란 품질에 미치는 영향)

  • A feeding trial was carried out effect of supplemental Lactobacillus on productivity, egg quality and intestinal microflora in 320 21 weeks - old laying hens for 12 week. Supplemented Lactobacillus strains were Lactobacillus amylovorus LLA7(LA), Lactobacillus crispatus LLA9(LC) and Lactobacillus vaginalis LLA11(LV). Three strains mixed to basal diet which containing 2,800㎉/kg ME, 16% CP with none, LA, LC, LV, LA+LC, LA+LV, LC+LV and LC+LC+LV. Supplemental level was 10(sup)7 cfu/g diet. Egg production was tended to increase with adding Lactobacilus, but not difference significantly. Average egg weight was heavier in adding Lactobacillrs compared to the none, and heaviest in LA+LV, LC+LV(P〈0.05). In periodic observation, the gap of egg weight with adding Lactobacillus or not was severe persisting laying periods. The diet containg MC or LV was better than LA, which means the difference by Lactobacillus strains for egg weight. Daily egg mass also increased in adding Lactobacillus about 1.1 to 2.3 g/hen, but not difference significantly. Feed intake and feed conversion were not difference regardless Lactobacillus strains and laying periods. Haugh unit improved with adding Lactobacillus. Cecal Lactobacillus spp. was increased with adding Lactobacillus(P〈0.05), didn't observed E. coli depression. In summary, supplemental Lactobacillus could improve for egg production, egg weight, egg mass and egg white. And those of effect expect much beneficial with mixing Lactobacillus which established well as single strain.

Improvement of Degrading Activity of Poly(butylene succinateco-butylene adipate)-Degrading Strains Isolated from Soils (토양에서 분리한 Poly(butylene succinate-co-butylene adipate) 분해균의 분해활성 증진)

  • From leaf mold and reclamation site soil of the Capital area of Korea, 3 poly(butylene succinate-co-butylene adipate: PBSA)-degrading strains were isolated through the clear zone test. The PBSA-degrading activities of the strains were assessed by means of a modified Sturm test using 0.01% of PBSA film as a sole carbon source. After the modified Sturm tests for 40 days at the respective isolation temperatures, the 3 strains degraded 30%, 55% and 43% of PBSA, respectively. The isolated strains were identified to be Burkholderia cepacia PBSA-4, Bacillus licheniformisPBSA-5 and Burkholderia sp. PBSA-6 through the 16S rDNA gene sequence analysis. Among them, PBSA-5 degraded both PBSA and Poly(vinyl alcohol). The degradation activity of the PBSA degrading strains appeared to be high at moderate temperatures such as $27^{\circ}C$ and $37^{\circ}C$, and initial inoculum size of $10^{10}cfu\;mL^{-1}$ degraded PBSA 1.2~1.3 more times than that $10^9cfu\;mL^{-1}$. Addition of 0.1 or 0.5% (w/w) of gelatin, yeast extract and ammonium sulfate raised the PBSA degrading activity, and especially addition of 0.1% (w/w) of gelatin enhanced the PBSA degrading activity by more than 33%. The mixed strains degraded PBSA faster than the single strain.

On the Modification of a Classical Higher-order Shear Deformation Theory to Improve the Stress Prediction of Laminated Composite Plates (적층평판의 응력해석 향상을 위한 고전적 고차전단변형이론의 개선)

  • In this paper, an systematic approach is presented, in which the mixed variational theorem is employed to incorporate independent transverse shear stresses into a classical higher-order shear deformation theory(HSDT). The HSDT displacement field is taken to amplify the benefits of using a classical shear deformation theory such as simple and straightforward calculation and numerical efficiency. Those independent transverse shear stresses are taken from the fifth-order polynomial-based zig-zag theory where the fourth-order transverse shear strains can be obtained. The classical displacement field and independent transverse shear stresses are systematically blended via the mixed variational theorem. Resulting strain energy expressions are named as an enhanced higher-order shear deformation theory via mixed variational theorem(EHSDTM). The EHSDTM possess the same computational advantage as the classical HSDT while allowing for improved through-the-thickness stress and displacement variations via the post-processing procedure. Displacement and stress distributions obtained herein are compared to those of the classical HSDT, three-dimensional elasticity, and available data in literature.

Effect of the Mixed Culture of Bacillus subtilis and Lactobacillus plantarum on the Quality of Cheonggukjang (Bacillus subtilis와 Lactobacillus plantarum의 혼합배양이 청국장의 품질에 미치는 영향)

  • The goal of this study was to improve the quality of cheonggukjang by the optimization of the inoculation methods of the Bacillus subtilis (B. subtilis) and Lactobacillus plantarum (L. plantarum) strains. In order to optimize the mixed cultivation of B. subtilis and L. plantarum, the B. subtilis strain was inoculated into steamed soybeans after cultivation of L. plantarum. Inoculation size of B. subtilis was changed to the simultaneous inoculation method in order to stimulate the growth of the L. plantarum in cheonggukjang. The viable cell count of L. plantarum increased from $2{\times}10^7$ CFU/g to $2-6{\times}10^8$ CFU/g and B. subtilis grew to $9{\times}10^8$ CFU/g. These results showed that 2 strains were successfully able to grow in the steamed soybean for good quality of cheonggukjang by optimization of the inoculation methods. The sensory evaluation indicated that a favorable aroma and overall acceptance of cheonggukjang by the optimized mixed cultivation of B. subtilis and L. plantarum, which was relatively higher than those of cheonggukjang by single strain inoculation of B. subtilis.

Production and Fermentation Characteristics of Mukeunji with a Mixed Starter (혼합 스타터를 이용한 묵은지의 제조 및 발효 특성)

  • To develop a starter culture system for the fermentation of mukeunji, we introduced lactic acid bacteria and yeast isolated from mukeunji into kimchi fermentation as a single or a mixed culture. On evaluating mukeunji flavor, we found that the mixed starter kimchi prepared with two strains, ML17 and MY7, gave the best sensory score. These strains were identified as Lactobacillus (Lb.) curvatus ML17 and Saccharomyces (S.) servazzii MY7 by molecular identification method. The fermentative characteristics of starter kimchi were investigated by measuring changes in the physicochemical and microfloral characteristics during the fermentation. The decrease in pH and increase in acidity in the starter kimchi were faster compared to respective values of control kimchi. There was a gradual decrease in hardness of starter kimchi, which was still slow compared to hardness decrease in control kimchi. Microbial analysis of starter kimchi revealed that Lb. curvatus ML17 and S. servazzii MY7 were the dominant organisms during the entire fermentation period. The lactic acid and citric acid contents of starter kimchi were higher than those of the control kimchi after 90 days of fermentation. By sensory evaluation, the starter kimchi scored higher in appearance, mukeunji flavor, sourness, carbonated flavor, savory taste, texture, and overall acceptability, but lower in off-flavor than the control kimchi.

Effects of Seeding on the Microbial Changes During Thermophilic Composting of Food Waste (종균 첨가가 음식물 쓰레기 퇴비화 미생물에 미치는 영향)

  • The effect of seeding on the thermophilic composting of food waste was investigated with selected strains of yeast, thermophilic bacteria and the mixed strains of yeast and bacteria in the lab-scale composter at $50^{\circ}C$. The results demonstrated that in all case the growth of yeasts preceded the growth of thermophilic bacteria by one or two days. The seeding of bacteria alone was ineffective for thermophilic bacterial development and the rapid increase of total thermophilic bacterial count was obtained by mixed seeding of yeasts and bacteria. Moreover, the bacterial growth was accelerated by the addition of yeasts alone. It was revealed that the yeasts removed organic acids from the food waste at the initial period and consequently induced the growth of thermophilic bacteria. The results presented in this paper indicate the indispensable role of yeasts for thermophilic process of food waste composting.

Optimal Conditions of Co-Immobilized Mixed Culture System with Aspergillus awamori and Zymomonas mobilis (Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 최적 조건)

  • Co-immobilized mixed culture system(A-Z system) composed of two different oxygen-demanding strains, aerobic(Aspergillus awamori) and anaerobic(Zymomonas mobilis) strains, in a Ca-alginate gel beads was developed to increase ethanol production from raw starch as a carbon source. Optimal mixture ratio of A. awamori and Z. mobilis was $1.25{\times}10^{9}\;spores/L-gel$ and 0.5g cells/L-gel, respectively. After 120 hours of cultivation, gel beads distinguished oxygen-rich surface for A. awamori from oxygen-deficient central part for Z. mobilis. At A-Z culture system, yield of ethanol on glucose, $Y_{p/s}=0.18$, was very low and there was high leakage of cells from surface of gel beads. At A-Z 36 cultrue system with changing silicon check valve for cotton plug at 36 hours in A-Z culture system, there was no cell leakage from gel beads, pH was maintained at around 4.3 during cultivation, and yield of ethanol on glucose, $Y_{p/s}=0.36$, showed 2 times higher than that of control culture system(cotton plug culture).
