Dual Role of Acidic Diacetate Sophorolipid as Biostabilizer for ZnO Nanoparticle Synthesis and Biofunctionalizing Agent Against Salmonella enterica and Candida albicans

  • In the present study, a yeast species isolated from CETP, Vellore, Tamilnadu was identified as Cryptococcus sp. VITGBN2 based on molecular techniques and was found to be a potent producer of acidic diacetate sophorolipid in mineral salt media containing vegetable oil as additional carbon source. The chemical structure of the purified biosurfactant was identified as acidic diacetate sophorolipid through GC-MS analysis. This sophorolipid was used as a stabilizer for synthesis of zinc oxide nanoparticles (ZON). The formation of biofunctionalized ZON was characterized using UV-visible spectroscopy, XRD, scanning electron microscopy (SEM), and Fourier transform infrared spectroscopy. The antimicrobial activities of naked ZON and sophorolipid functionalized ZON were tested based on the diameter of inhibition zone in agar well diffusion assay, microbial growth rate determination, protein leakage analysis, and lactate dehydrogenase assay. Bacterial pathogen Salmonella enterica and fungal pathogen Candida albicans showed more sensitivity to sophorolipid biofunctionalized ZON compared with naked ZON. Among the two pathogens, S. enterica showed higher sensitivity towards sophorolipid biofunctionalized ZON. SEM analysis showed that cell damage occurred through cell elongation in the case of S. enterica, whereas cell rupture was found to occur predominantly in the case of C. albicans. This is the first report on the dual role of yeast-mediated sophorolipid used as a biostabilizer for ZON synthesis as well as a novel functionalizing agent showing antimicrobial property.

전통누룩으로 빚은 발효주의 품질 평가 (Quality Assessment of Yakju Brewed with Conventional Nuruk)

  • To evaluate of quality of Yakju brewed with different types of fermenters(Nuruk) and brewed by a method described in Sallymkyungjae, blucoamylase and protease and protease activities of Nuruk were measured, and proximate composition , coloring degree, acetaldehyde , alcohol , , fusel oils , amino acid content , mineral content and sugar composition of Yakju samples were also determined. It was foung that the lower the pH, the lower the glucoamylase acitvity in Nuruk samples A to E . In A, B, D and H, protease activity washigher at ph 5.5 than at pH 5.0 . Yakju sampels 1 to 5 during fermentation , total acidity and amino acid content were high at the first step and were getting lower gradually at the second step. In Yakju samples 6 to 11 , ethanol content was high in 6b, 7b, 8a, 8b and 11b. At the second step, residual sugar content was getting higher gradually in 7 a, 11 a and 11b. The coloring degree of Yakju wasinfluenced by not only color of Nuruk but also Fe content in Yakju . Of the Yakju examined , only 9a and 9b contained acetaldehyde in trace amounts. Thanol content was the highest in 8a and 8b. Fusel oil content was high in 8a and 8b. In samples 6 to 9 , aspartic acid content was higher in treatment a than treatment a than treatment b, but tyrosine , histidine and proline contents were higher in treatment a than treatment b. The levels of frucose, melibiose, sorbitol, and arabinose in Yakju brewed from unsteamed rice were higher than in Yakju brewed from steamed rice. K content was the lowest in 9a and 9b. Na content was higher in treatment a than in treatment a than in treatment b. In only 6a, Ca/P ratio was more than one.

심해 시추와 관련된 국내 해양 환경 및 안전 관련법 개선에 대한 입법론적 연구 (A Study on Legislation for the Improvement of the Marine Environment and Safety Act for Deep Sea Drilling)

  • 이 연구는 국내 대륙붕 주변에서 진행되는 심해 시추의 개요 및 현황에 대하여 시추의 개념 및 종류, 대륙붕 개발의 연혁 및 현황을 중심으로 검토하였고, 시추와 관련된 해양 환경, 안전법상 적용의 범위, 문제점 등에 대하여 법적으로 분석하였다. 그리고 이 연구는 해저광물자원의 체계적인 개발을 위하여 영국의 '해양구조물(안전관리절차)규정' 기초로 별도의 법률 신설 방안 및 해양환경관리법, 해사안전법, 석유광산안전규칙에 대한 개선안을 입법론적으로 제안하였다. 특히 해외 해양플랜트와 관련된 사고 사례를 기초로 국내 인력에 대한 국제인증교육에 준하는 교육훈련 및 평가 제도의 구축의 필요성을 강조하였다.

백운모광의 품위향상에 관한 연구 (A study on the Beneficiation of Muscovite Ore)

  • A general study on the upgrading of mica minerals which are mainly found in pegmatite deposits was carried out for the purpose of developing a technique for recovering mica in form of comercial grade products. By the way the grade one of about 5~6% $K_2O$ still is not developed. The target of this research work is to be establish a process for the efficient concentration of muscovite, containing more than 10% $K_2O$. The tests are applied to incraese the recovery and grade of concentrates in term of variations of conditions. The test sample consists of mainly muscovite and gangue mineral such as quartz, pyrite and chlorite. Decantation and shaking table tests were ineffective to up-grade this low grade one, but flotation method gave satisfactory result. By means of grade one, but flotation method gave satisfactory result. By means of grindability tests, an optimum result could be obtained from the sample ground to -48mesh feed size. The flotation result indicates that the dodecyl ammonium chloride used as a cationic collector is effective on the negatively charged surface, while the sodium dodecyl sulfate as an anionic collector is effective on the positively charged surface. Muscovite was floated by petroleum sulfonate as well as amine type collector, it also floated by MIBC as well as pine oil frother under well condition. Fine muscovite concentrates of about 10.68% $K_2O$ was obtained with 22.4% yield, by decantation, the muscovite concentrates of 10.10% $K_2O$ was obtained with 23.54% yield, by table concentration, the muscovite concentrates of 11.51% $K_2O$ was obtained with 23.0% yield by flotation.

  • 진공 배기 시스템에 위험한 환경을 초래할 수 있는 모든 가능성을 찾아 낼 수는 없지만 누적된 현장 경험과 연구 결과에 맞추어 최대한 필요한 안전 조치들을 취해야 한다. 진공 배기 시스템이나 그 구성품들에 대한 심각한 파손을 유발하는 공통적인 요인들은 발화성 물질의 점화나 진공 배기 시스템의 배기구 막힘에 의해 발생한다. 따라서, 진공 펌프와 진공 시스템의 안전한 가동과 사용을 위해서는 다음과 같은 것들을 반드시 준수하여야 한다. ${\blacksquare}$ 발화성, 폭발성 공정 물질을 사용하는 진공 배기 시스템은 정규 유지 보수 작업(PM) 후 첫 번째 배기 과정은 매우 천천히 진행하여 진공 배기 시스템 내부에 급격한 난류가 형성되지 않도록 해 주어야 한다. ${\blacksquare}$ 진공 배기 시스템 내에서 발화성 물질들의 농도가 발화 영역(flammable zone, potentially explosive atmosphere)에 들어가지 않도록 하여야 한다. 이를 위해서는 불활성 가스를 이용하여 진공 펌프와 진공 배기 시스템의 가동 예상 조건이나 고장 환경하에서 안전한 농도 이하로 희석시켜야 한다. ${\blacksquare}$ 진공 펌프와 진공 배기 시스템에 장착되어 사용되는 밸브 등의 기계적 부품들이나 공정에 사용되는 물질과 공정 부산물들(by-products)로 인하여 배관, 필터 배기구 등이 막히지 않도록 하여야 한다. ${\blacksquare}$ 공정에 사용되는 물질들, 특히 산소($O_2$), 오존 ($O_3$) 등의 산화제 농도가 높을 때는 오일 회전 배인 진공 펌프(Oil rotary vane vacuum pump)에 미네랄(mineral) 오일을 사용하지 말아야 하며, PFPE(Perfluoropolyether) 오일을 사용하여야 한다. 시판되는 진공 펌프 오일 중 비발화성(non-flammable)으로 표기된 오일이라고 하더라도 산화제(oxidant)의 농도가 체적비로 30 % 넘는 공정 환경에는 사용하지 말아야 한다. ${\blacksquare}$ 진공 펌프와 진공 배기 시스템에 의해 배기되는 물질들이 물($H_2O$)과 격렬하게 반응하는 경우는 물이 아닌 다른 냉각제를 사용하여야 한다. ${\blacksquare}$ 안전하지 않다고 판단되는 상황에서는 해당 전문가의 조언이나 해당 전문가의 직접적인 현장 도움을 통해 문제를 해결하여야 한다.

수용성 키토산으로 제조한 미세구의 분해성과 약물 방출 거동 (Drug Release Behavior and Degradability of Microspheres Prepared using Water-Soluble Chitosan)

  • 수용성 키토산의 농도와 가교제 (글루타알데히드)의 양을 변화시키면서 미네랄오일 내에서 키토산용액의 유화법에 의해 수용성 키토산 미세구를 제조하였다. 그러고 약물이 봉입되어진 수용성 키토산 미세구의 분해에 따른 형태의 변화, 약물의 봉입효율, 약물 방출 거동과 같은 물리화학적 특성을 규명하였다. Norfloxacion 이 봉입된 수용성 키토산 미세구는 표면의 약물에 의해 나타나는 과량의 약물 방출이 없는 높은 약물 봉입 함량을 보였다. 표면에 약물이 존재하지 않음을 선 회절 분석으로 확인하였다. 수용성 키토산 미세구의 분해 특성과 약물방출 거동을 관찰한 결과 가교제의 양이 약물의 봉입량, 방출, 그리고 분해에 중요한 역할을 하는 것을 확인하였다. 수용성 키토산 미세구는 가교제의 양이 증가함에 따라 분해속도가 느렸으며, 이와 동시에 약물이 천천히 방출되었음을 확인하였다.

말의 그물막구멍포착 교정을 위한 소장 절제 및 문합 1례 (Small Intestine Resection and Anastomosis for Epiploic Foramen Entrapment of a Horse)

  • A 10-year-old gelding Warmblood weighing 560 kg was referred to J&C Equine Hospital with the history of hyperpnea, depression, pawing, and rolling for 7 hours. According to the results of clinical and ultrasound examination, it was considered that intestines were distended with thickened wall. The horse had been treated with lactated Ringers' solution (14 L, IV), flunixin meglumine (1.1 mg/kg, IV), and mineral oil (1 L, PO), but he did not show any responses to those treatments. Exploratory laparotomy was performed and identified incarcerated small intestine through the epiploic foramen. The horse received resection and anastomosis of the entrapped small intestine. After surgery, the horse was treated with intensive postoperative care of fluid therapy (5 L with 20 mEq/L KCl, every 2 hours), flunixin meglumine (1.1 mg/kg, IV, sid), antibiotics (penicillin 22,000 IU/kg, IV, qid and gentamicin 6.6 mg/kg, IV, sid), lidocaine constant rate infusion (bolus 1.3 mg/kg over 15 minutes then 0.05 mg/kg/minute), common nutritional supplements, nasogastric intubation every 2 hours and trunk bandage. Postoperative feeding program had started with small amount of hay every 4 hours and gradually increased to normal amount till 5 days. At 77 days after surgery, he showed sudden outbreak of colic and was euthanized. The causes of colic were small intestinal strangulation by passing through the mesenteric rents and postoperative adhesion between small intestines. According to the results, it is recommended to perform perioperative intensive care of horse with colic and to use several methods to prevent adhesions during abdominal surgery of horses.

유유아(乳幼兒) 및 성장기 아동을 위한 영양식품 개발에 관한 연구 -흰쥐 성장에 미치는 영향- (The Protein rich Food Mixtures for Korean Infants)

  • Korea is one of the typical rice eating countries where availability of animal protein is limited even for the growing generation. Nutritive food products for infants and children are not available commercially in large scale at the present time, although a limited amount of expensive milk products are produced. The present study deals with a pioneering attempt on a new food mixture to meet this demand. Several food mixtures, possibly produced in the country, consisting of rice, soybean, FPC, vitamin, minerals and other food additives are developed in this work Sixty female and male rats aged $30{\sim}40$ days were divided into five groups, twelve rats each. The rats had been maintained with the six different diets, as follow. F-P-4 group tended to remain the heaviest and F-R-1 the lightest in the body weight among three groups throughout the experimental period. In terms of FER (Feed Efficiency Ratio) and PER (Prortein E. R.) value, F-P-4 group kept the highest record throughout. F-P-4 group showed the highest value of nitrogen retention in the body. In comparision between F-P-4 groups and casein 20% group (standard) in all respects of this experiment, F-P-4 group which is the most superior among experimental group, revealed statisfically no significant inferior than that of standard group. This fact could be interpreted that most limiting a. a., methionine in the soy protein produced little inferiority of the experimental groups in the study.

증발기와 응축기 온도변화에 따른 R22 대체냉매 R290 및 R1270의 성능평가 (Performance of R290 and R1270 as Alternative Refrigerants of R22 According to Temperature Variations of Evaporator and Condenser)

  • In this study, performance of 2 pure hydrocarbons R290 and R1270 was measured in an attempt to substitute R22 under 3 different temperature conditions. They were tested in a refrigerating bench tester with a hermetic rotary compressor. The test bench provided about 3.5 kW capacity and water and water/glycol mixture were employed as the secondary heat transfer fluids. All tests were conducted under the same external conditions resulting in the average saturation temperatures of $7/45^{\circ}C$ and $-7/41^{\circ}C$ and $-21/28^{\circ}C$ in the evaporator and condenser, respectively. Test results show that the coefficient of performance (COP) of these refrigerants is up to 11.54% higher than that of R22 in all temperature conditions. Compressor discharge temperatures were reduced by $14{\sim}31^{\circ}C$ with these fluids. There was no problem with mineral oil since the mixtures were mainly composed of hydrocarbons. The amount of charge was reduced up to 58% as compared to R22. Overall, these fluids provide good performance with reasonable energy savings without any environmental problem and thus can be used as long term alternatives for residential air-conditioning and heat pumping application.
