  • In the course of screening for new bioactive compounds from oligotrophs in soil, a microorganism, designated as SA-246 and now identified as Micromonospora sp., has been shown to produce a strong antibacterial compound. The active compound was purified from broth filtrate by ethylacetate extraction, silica gel column chromatography, preparative TLC and HPLC, and was identified as crisamicin A based on mass and NMR spectral data. The compound SA- 246 exhibited not only strong antibacterial activity against Gram-positive bacteria but also cytotoxicity against cancer cell lines such as A549 (lung), SK-OV-3 (ovarian), SK-MEL-2 (melanoma), XF498 (central nervous system) and HCT15 (colon).

  • For the isolation of Actinomycetes showing nematocidal activity against Pine wood nematode, Bursaphelenchus xylophilus, about 2000 culture broth of Actinomycetes were tested and their activity were compared with that of Streptomyces avermitilis resulting a selected strain AW050027. The cultural, morphological and physiological analysis was performed for the identification of a selected strain. Phylogenetic analyses based on 165 rDNA gene sequences showed that the selected strain AW050027 belonged to the genus Micromonospora and M. corioriae $NAR01^T$ was the closest neighbors, sharing 98.9% 165 rDNA gene sequence similarity.

Control Effects of Micromonospora sp. AW050027 by Media Optimization and Microbial Treatment Against Pine Wood Nematode (Micromonospora sp. AW050027 균주의 배지최적화 및 미생물제제 처리에 의한 소나무재선충 방제효과)

  • Nematicidal activity against pine wood nematode, Bursaphelenchus xylophilus, was tested in the pot and field by the treatment of microbial formulation after media optimization. The optimized media composition was glycerol 10 g, soybean meal 10 g, NaCl 1 g, $CaCO_3$ 2 g, $K_2HPO_4$ 0.125 g per liter and microbial complex formulation was made with liquid and powder type. Most effective antibiotics against symbiotic microorganism with nematode, kanamycin, was added to the formulation. The control effects against pine wood nematodes were checked by pot test and field test. In the result of treatment by trunk injection, five times treatment was more effective than one time and the treatment with the formulation of concentrated culture supernatant was the most effective in the nematicidal activity showing below 10% mortality in pine tree.

  • The anti-melanogenic activity of 259 actinomycete strains was tested, and based on the results for the inhibition of mushroom tyrosinase activity and the reduction in melanin content, Micromonospora sp. JCS1 and JCS7 were selected as the strains with the highest anti-melanogenic potential. The activity-guided fractionation of extracts from JCS1 and JCS7 led to the isolation of the dipeptides cyclo(ʟ-Phenyl alanine (Phe)-ʟ-Proline (Pro)) (1) and cyclo(ʟ-Tryptophan (Trp)-ʟ-Proline (Pro)) (2). These two compounds were tested for their inhibition of mushroom tyrosinase by monitoring ʟ-DOPA levels and melanin production. Cyclo(ʟ-Phe-ʟ-Pro) (1) and cyclo(ʟ-Trp-ʟ-Pro) (2) were thus confirmed to have the potential for use in functional whitening cosmetics containing actinomycete-derived secondary metabolites.

  • This study was conducted to evaluate the effects of fowl dzopping. saw dust and rice hall on the soil microflora in vitro. The experiment was designed in seven treatments with the various organic materials and they were only soil (control). soil + fowl dropping (S+F), soil+fowl dropping+rice hull (S+F+R) soil+fowl dropping*saw dust (S+F+S). soil+chemical fertilizer (S+C.F), fowl dropping+rice hull (F+R) and fowl dropping+saw dust (F+S). All the samples of treatment were incubated in $28{\pm}2^{\circ}C$ condition and tested the activity of soil microflora for 84 days The activity of fungi, total bacteria, gram-negative bacteria and actinomycetes showed the highest values at, twenty-first day and the spore-forming bacteria was at forty-second day after incubation. The number of fungi and gram-negative bacteria showed the highest values in the treatment of F+S, the spore-forming bacteria and the actinomycetes were in the S+F+S. and the number of total bacteria was in the F+C.F., but in the treatment of F+R. all the microorganism except fungi showed the lowest values in their numbers. The composition ratio of dead bacteria was higher in the treatments of S+F+R and F+R than in those of others as 70% and 40% respectively. Actinomycetes isolated from the treatments of S+F and S+F+S were identified as Streptomyces sp.. Nocardia sp., Micromonospora sp. Actinomadura sp. and Saccharomonospora sp.
