The effect of moisture content an the reductive dechlorination of polychlorinated biphenyls and population dynamics of dechlorinating microorganisms was investigated in sediments spiked with Aroclor 1248. In sediment slurry with an overlying water layer, dechlorination ensued after a 4-week lag period and reduced the average number of chlorines per biphenyl from 3.91 to 3.15 after 48 weeks. In the sediments of reduced moisture content, however, dechlorination occurred after a lag period of 12 weeks and decreased the average number of chlorines per biphenyl to only 3.62, and the dechlorination rate was also slower. When the population size of dechlorinators, methanogens, and sulfate-reducing bacteria was determined by the most probable number techniques, however, no difference was found between the slurry and the low-moisture sediments, except for methanogens. The growth of dechlorinating populations coincided with the end of the lag period and they then increased by 3 orders of magnitude in two conditions. Specific growth rate of dechlorinators showed little difference between the slurry and the low-moisture sediments; however, growth yield was high in the sediments of reduced moisture content. The reduction of sediment moisture decreased the dechlorination rate and extent of PCBs but did not inhibit the growth of PCB dechlorinators.
We gamma-irradiated to natural casings from lamb and pork to improve sanitary quality and microbiological shelf stability of sausage stuffed in animal intestine-induced natural casing from lamb and pork. The microbial changes of irradiated casings and the microbial growth of the sausages with gamma-irradiated natural casings were observed. Population of total aerobic bacteria was decreased by gamma irradiation, dependent upon the irradiation dose. The number of microflora in lamb and pork casings was reduced by about 5.5 log cycle at 5 kGy. Population of total aerobic bacteria of sausages stuffed in non-irradiated lamb and pork casing was about 3.6 log CFU/g after just manufacture and was about 7 log after 10 day storage. However, population of total aerobic bacteria of sausages with 3 kGy-irradiated and 5 kGy-irradiated casings was 2.48 and 0.32 log in lamb, and 2.78 and 0.60 log in pork, respectively. Gamma irradiation to animal intestine- induced natural casings was effective and thus, can be used for enhancing shelf life of final products due to reduction of the number of contaminated microflora.
In order to study the effects of cropping system and fungicide (Dachigaren) on soil microbes, the seasonal fluctuations of soil microbes in the fields of radish and Chinese cabbage including soil pH, Soil moisture content and soil temperature were investigated on every 15 day interval from the begining of March to late October in 1981. The population of total fungus peaked at the begining of July, while that of total bacteria, at the begining of August. They were affected by soil temperature, however pathogenic microbes seemed to be more related with host plants than the soil temperature, because pathogens showed high density through the whole cultivation period. The pathogenic microbes showed the density of order ; Xanthomonas, Erwinia, Pseudomonas, Agrobacterium and Corynebacterium. Xanthomonas, Erwinia and Pseudomonas, which induced radish and Chinese cabbage diseases were higher than Agrobacterium and Corynebacterium in population densigy. Bacterial soft rot occured at the density of Erwinia $5.9{\sim}6.6{\times}10^5/dry$ soil 1 gram. The density of microbes on continuous fields were higher than that of rotating fields, but there were no significant difference between treated fungicide plot and non treated in the density of microbes, also no difference between Chinese cabbage and radish growing fields.

1986년 8월부터 12월까지 낙동간 하구의 3정점에서 Aeromonas의 분포 및 이에 영향을 미치는 환경요인을 조사하였다. 조사기간 중 Aeroamonas spp. 개체수 변화는 $4.3\times10^{2}-4.6\times 10^{4}$ MPN/100ml로 기록되었다.분산분석의 결과 Aeromonas spp. 의 분포는 정점간에 유의한 차이가 이쓴 것으로 나타났으며 정점 2에서 가장 높은 개체수를, 정점 3-B에서 가장 작은 개체수를 보였다. Aeromonas spp.의 분포에 영향을 미치는 환경요인을 알아보기 위하여 중회귀 분석 및 주요인 분석(principal component analysis)을 한 결과 Aeromonas spp. 의 분포는 담수의 유출과 무기영양염류의 유입에 의해 크게 영향을 받는 것으로 나타났으며 종속영양세균, 이용가능한 질소원, 분변성대장균 및 수온고 밀접한 상관관계를 나타내었다.

Two kinds of organic materials, powders of Salicornia and Oryza sativa L. wre added as a source of organic matter to the suspensions of saline soils(soil : water = 1:3) to be 4 per cent of dried saline soil grams. And then, the samples were incubated at $28^{\circ}C$ to improve the decomposition of organic materials by soil microflora. Resutls of this experiment are summarized as followings : 1) The pH of soil suspension showed its highest value on the second or third week after the treatments, which were similar to those of the soil microflora. Both of the bacteria and actinomycytes population showed in the third weak after the tratment. In general, 30-years old of saline soil contained microbial population much more than those of 5-year old of saline soil. Salicornia powder favored the increase of both the bacteria and fungal population, and Oryza sativa L. seemed to have been a great role in increase of actinomycetes. Especially, fungal population of the untreated soil suspension contained higher microbial populations more than those of the soils treated with both of the organic materials. 4) Sugar contents of soil suspesions decreased remarkably on the first week after the treatments. 5) The amount of nitrate had similar tendency to those of population changes of soil microflora. Total acidity decreased continuously and the amount of potassium showed its highest value on the third week treatment. However, the amount of phosphorus was determined to be insignificant.
The intent of this study was to scrutinize the consequence of salt type [sundried salt, refined salt, baked salt, or nitrate pickling salt (NP)] on the physicochemical and microbiological features of salami formulated by soaking with Aspergillus spp. before ripening. The effects of nitrate-free salts added were not significant. Nitrate pickling salt samples were significantly higher in protein level, whereas those were lower in fat level during ripening (p<0.05). The pH of salamis treated with NP was higher than that of other salt treatments, while weight losses of those was lower (p<0.05). During the ripening and drying, NP produced lower extent of volatile basic nitrogen and lipid oxidation than those with other salts (p<0.05). The total aerobic population counts of NP samples revealed lower than that of other samples over the ripening time. The addition of NP in salamis produced redder sausages. The salamis containing NP found to be better physicochemical and microbiological quality attributes than the other salt types.
There are a number of ways in which environmental microbiology and microbial ecology will benefit from DNA micro array technology. These include community genome arrays, SSU rDNA arrays, environmental functional gene arrays, population biology arrays, and there are clearly more different applications of microarray technology that can be applied to relevant problems in environmental microbiology. Two types of the applications, bacterial identification chip and functional gene detection chip, will be presented. For the bacterial identification chip, a new approach employing random genome fragments that eliminates the disadvantages of traditional DNA-DNA hybridization is proposed to identify and type bacteria based on genomic DNA-DNA similarity. Bacterial genomes are fragmented randomly, and representative fragments are spotted on a glass slide and then hybridized to test genomes. Resulting hybridization profiles are used in statistical procedures to identify test strains. Second, the direct binding version of microarray with a different array design and hybridization scheme is proposed to quantify target genes in environmental samples. Reference DNA was employed to normalize variations in spot size and hybridization. The approach for designing quantitative microarrays and the inferred equation from this study provide a simple and convenient way to estimate the target gene concentration from the hybridization signal ratio.
