The objective of this study was to find the way to prolong the storage time of sawdust-based oyster mushroom (Pleurotus osteratus) spent substrate (OMSS) by fermenting with potential probiotic microorganisms to recycle the otherwise waste of mushroom farms. To this purpose, lactic acid bacteria (LAB) were screened to select the best lactic acid-producing strains. Three strains of LAB (Lactobacillus plantarum Lp1', Pediococcus acidilacticii Pa193, L. plantarum Lp2M) were selected and in mixture they lowered the pH of the fermented OMSS to 3.81. fOMSS (fermented sawdust-based oyster mushroom spent substrate) could be stored at room temperature for at least 17 days without any deterioration of feed quality based on the pH, smell, and color. In dry matter disappearance rate in situ, commercial TMR (total mixed ration), OMSS and OMMM (oyster mushroom mycelium mass) showed no significant differences between the samples after 6, 12 and 24 h incubation except for 48 h. Two separate field studies were performed to test the effects of fOMSS supplement on the growth performance of postweaning Holstein calves. Field trials included groups of animals feeding calf starter supplemented with: Control (no supplement), AB (colistin 0.08% and oxyneo 110/110 0.1%), fOMSS (10% fOMSS) and fConc (10% fermented concentrate) and DFM (direct-fed microbials, average $10^9$ cfu for each of three LAB/d/head). Growth performance (average daily gain and feed efficiency) of the fOMSS supplement group was higher than that of AB followed by fConc and DFM even though there was no statistically significant difference. The Control group was lower than any other group. Various hematological values including IgG, IgA, RBC (red blood cell), hemoglobin, and hematocrit were measured every 10 days to check any unusual abnormality for all groups in trial I and II, and they were within a normal and safe range. Our results suggest that sawdust-based OMSS could be recycled after fermentation with three probiotic LAB strains as a feed supplement for post-weaning calves, and fOMSS has the beneficial effects of an alternative to antibiotics for a growth enhancer in dairy calves.
To search direct fed microbials, we isolated a Candida sp. from kefir grain. The isolated Candida sp. strain showed 99.8% of identity to the species of Candida kefyr by API 20C kit. Enzyme activity of Candida kefyr was higher in amylase (0.33±1.12μmol/min/mg) than that in phytase (0.052±0.98μmol/ min/mg) cellulase(0.051±μmol/min/mg) and xylanase (0.011±0.98mol/min/mg). The maximum numbers of Candida kefyr in growth curve were reached at 30 h fermentation. Candida kefyr showed high resistances to acidic environment, which was not perfectly extincted even at pH 2.0. And it showed high tolerance to bile salt which had almost 97.2% of survival in the presence of 1.0% bile salt.Especially, Candida kefyr showed high heat stability which remained 10% of initial microorganisms at 60℃. Candida kefyr was not generally inhibited by most of 11 antibiotic agent which contained tetracycline groups. These results suggest that the isolated Candida kefyr has a useful properties as probiotics.
Probiotics have many effects such as antihypertensive, prevention of cancer, antioxidation, reduction of dermatitis symptoms, improvement of mineral absorption, reduction of allergic symptoms, and decrease of cholesterol, However, the main role of probiotics is that they balance intestinal microbials proportion. L. acidophilus is one of probiotics and microflora in intestine. It has an acidification activity, aroma production, texture formation and probiotics properties. We studied on the roles of L. acidophilus in mice. In this study, body weights of mice were decreased when administration of L. acidophilus ($1{\times}10^{10}$ CFU) and swimming ability has been raised than a normal group after feeding on L. acidophilus ($1{\times}10^{10}$ CFU). After taking L. acidophilus ($1{\times}10^{10}$ CFU), total white cells were increased than a normal group; hemoglobin and thrombocytes were increased. The level of cholesterol and triglyceride were decreased in blood analysis. We knew L. acidophilus is related to innate immune system. We found out the secretion of cationic peptide was increased in the Lysoplate assays as a result of L. acidophilus ($1{\times}10^{10}$ CFU) administration. Appearance rate of lysozyme was also increased than the normal group on an immunohistochemistry stain. We confirmed L. acidophilus contributes to host health through innate immune system stimulation. L. acidophilus more than $1{\times}10^{10}$ CFU are thought to be beneficial for the host health and prevention of intestinal diseases in field condition.
The antimicrobial substances from Rhodiola sachalinensis were extncted, isolated and identified. The highest level of antimicrobial activity and its yield were obtained in methanol extract. The minimum inhibition concentrations of the methanol extract were 500 $\mu\textrm{g}$mL on agar plate and 100 $\mu\textrm{g}$mL in broth media for four gram positive and four gram negative microbials. The methanol extract was fractionated by n-hexane, chloroform, ethyl ether, ethyl acetate, and butanol, orderly. The separate was developed on the TLC plate with different solvent system ratio of chloroform and methanol. Nine substances were isolated from chloroform and methanol mixture(9:1, v/v). Among them, three isolates showed antimicrobial activity. Three substances separated by HPLC were identified by GC/MS(EI) spectrum and $^1$H, /sup13/C-NMR spectrum. They were gallic acid, (-)-epicatechin and kaempferol. The antimicrobial activities of each substances were shown gallic acid, (-)-epicatechin, kaempferol orderly.

This study investigated the characteristics of nitrogen removal and microbial community in a lab-scale A$_2$O activated sludge process filled with the fluidized media at an aerebic basin. The change of microbial community was monitored based on quinone profiles of activated sludge according to feeding sewage with/without external carbon source. Low C/N ratio(COD$_{Cr}$/T-N of 1.24) sewage was fed. The obtained results from this study were as follows; Ubiquinone(UQ) in the influent was in the descending order of UQ-8, UQ-10 and UQ-9. Menaquinone(MK) was simpler and much less than UQ. The ratio of UQ/MK was less than 0.41 and the dissimilarity was below 0.26. Without an external carbon source, MK-8 was the dominant species and there were 3 kinds of quinone species and low DQ and EQ values in an anaerobic basin. The ratio of UQ/MK increased to 2.3 in an anoxic basin. In an oxic basin, UQ-7 and UQ-8 were the dominant species. UQ-7 was dominating in suspended microorganisms, while UQ-8 was in attached microorganisms. With an external carbon source addition, MK-8 decreased but UQ-8 increased in an anaerobic basin. So did quinone species, DQ and EQ values. There was also a change in an anoxic basin with the improvement of denitrification. UQ-8 decreased instead, MK-7 and MK-8 increased. UQ/MK ratio decreased 2.3 to 1.4. It means that the dominant species change from Pseudomonas sp. to Bacillus and Micrococcus species. etc. In an oxic basin, UQ-8 replaced UQ-7 in suspended microorganisms and UQ-10 replaced UQ-8 in attached microbials. This seemed related with the growth of Nitrosomonas and Nitrobactor species.

Background: The appropriate empirical antimicrobial choice in the treatment of community-acquired pneumonia (CAP) should be advocated by community-based information on the etiologic pathogens, their susceptibility to antimicrobials, clinical characteristics and outcomes. Jeju is a geographically isolated and identical region in Korea. However, there is no regional reference on adult CAP available. This study investigated the etiologic agents and clinical outcomes of adult patients diagnosed with CAP in Jeju, Korea, to help guide the empirical antimicrobial choice. Methods: A prospective observational study for one year in a referral hospital in Jeju, Korea. Patients diagnosed with CAP were enrolled with their clinical characteristics. Microbiological evaluations to identify the etiologic agents in the adult patients with CAP were performed with blood culture, expectorated sputum smear and culture, antibody tests for mycoplasma, chlamydophila, and antigen tests for legionella and pneumococcus. The clinical outcomes of the initial empirical treatment were analyzed. Results: Two hundred and three patients with mean age of 64 and 79 females were enrolled. Ten microbials from 90 cases (44.3%) were isolated and multiple isolates were confirmed in 30. Among the microbial isolates, S. pneumoniae (36.3%) was the most common, followed by M. pneumoniae (23.0%), C. pneumoniae (17.0%), S. aureus (9.6%) and P. aeruginosa (5.9%). The initial treatment failure (23.8%) was related to the isolation of polymicrobial pathogens, elevated inflammatory markers, and the presence of pleural effusion. Among the 30 isolates of S. pneumoniae, 16 (53.3%) were not susceptible to penicillin, and 19 isolates (63.3%) to erythromycin and clarithromycin. However, 29 isolates (96.7%) were susceptible to levofloxacin and ceftriaxone. Conclusion: S. pneumoniae, M. pneumoniae, S. aureus, and P. aeruginosa are frequent etiologic agents of adult CAP in Jeju, Korea. The clinical characteristics and antibiotic resistance should be considered when determining the initial empirical antimicrobial choice. Respiratory quinolone or ceftriaxone is recommended as an empirical antimicrobiotic in the treatment of adult CAP in Jeju, Korea.
The potential of encapsulated inuloprebiotics from domestic Jerusalem artichokes (Helianthustuberosus) as natural antibacterial growth promotor for an antibiotic replacement in broiler chickens was presently assessed through assays of growth performance, serum immunoglobulin production and influence on caecal microflora. Two hundred-forty, 1-day-old, male broilers (Ross 308) were randomly allotted to four treatments (T1-T4), with three replicate pens per treatment and 20 chicks per pen. Broiler chicks were fed a basal diet (T1: control) or basal diet plus antibiotics (T2: Chlorotetracycline, 0.10%), 300 ppm of the inuloprebiotics (T3), or 450 ppm of the inuloprebiotics (T4) for 35 days. Body weight, dressing percentage or weight of breast and thigh muscles relative to carcass weight of T3 and T4 broiler chickens was significantly (P<0.05) higher than T1 and T2 broiler chickens. The weight of abdominal fat from T3 and T4 broiler chickens were significantly (P<0.05) lower than that of T1 and T2 chickens. Serum immunoglobulins in the T3 and T4 groups were significantly (P<0.05) elevated compared to the T1 and T2 groups. The weight of immune organs, thymus and Bursa of Fabricius relative to live body weight in the T3 and T4 groups were significantly (P<0.05) higher than the T1 and T2 groups. Bifidobacteria and Lactobacillus, which are beneficial bacteria, were present in greater numbers in the caecum of T3 and T4 birds than T1 and T2 groups, whereas potentially harmful Escherichiacoli and Salmonella were present in lower numbers, with differences being significant (P<0.05). These results suggest that a diet supplemented with 300 ppm of inuloprebiotics has potential as an antibiotic replacement for organic livestock feed supplement intended to improve production of broiler chicken.
This study was conducted to investigate the effects of dietary direct-fed microbials(DFM) on the growth performance and nutrient digestibility in broiler chickens. A total of two hundred eighty eight broiler chickens were randomly allocated into six treaments with four replications and fed for five weeks. Dietary treatments included 1) CON (basal diet), 2) DFM-1(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri), 3) DFM-2(basal diet + 0.2% Enterococcus sp. & Lactobacillus plantarum), 4) DFM-3(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum), 5) DFM-4(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum & Yeast), 6) DFM-5(basal diet + 0.2% Enterococcus sp. & Lactobacillus reuteri & Lactobacillus plantarum & Bacillus subtilis). During the period of 1∼3weeks, average daily gain (ADG) and Feed/Gain were not significant different among treatments. In the later(3∼5weeks) and overall period(l∼ 5weeks), ADG and Feed/Gain tended to be improved in DFM treatments compared to the control. However, there were no significant differences among DFM complexes and between control and DFM treatments. The broilers fed DFM-3 diet was showd poor growth performance compared to the broilers fed DFM-l diet and similar to the broilers fed DFM-2 diet but there was no significant differences. The treatments of DFM-4 diet added Yeast and DFM-5 added Bacillus subtilis were improved in ADG and Feed/gain but no significant differences were found and also there was no specific DFM treatments in experiment period. In digestibility, the broiler chicks fed DFM treatments tended to improve N digestibility compared to control treatment. However, they were not significantly different among the treatments. In conclusion, DFM tended to improve growth performance in later stage of broiler.

Effect of commercial Direct Fed Microbials (DFM) or protease treated feed (PTF) supplementation on growth rate and biogenic substances such as BUN, glucose, IgG, GOT, GPT and Vitamin A, C, E from Holstein steers was studied for 7 months. Thirty two steers aged 2~3 months were separated with 4 groups for control, DFM (PS), protease (ES) and their mix (PS + ES) supplementally fed 0, 100, 100 and 50 + 50 g/day respectively. Weight gain was averagely higher in PS than any others, although there were no differences significantly. All treatments enhanced to 3~8% of control in dry matter, crude protein and total digestible nutrient (P>0.05). Metabolic diseases with veterinary cure had not shown in this study. Plasma GOT and GPT were lower in the PS and ES than control. Plasma glucose concentration was also lower in PS than the others. Total cholesterol of ES was higher than the others but that of PS is the lowest. Plasma vitamin C was higher in PS than the others. It was shown that dietary PS affected change from glucose to vitamin C with not overloading liver. Conclusionally, PS and ES were shown to enhance metabolic health of steers during growing period.
