Effect of Ruminal NH3-N Levels on Ruminal Fermentation, Purine Derivatives, Digestibility and Rice Straw Intake in Swamp Buffaloes

  • The experiment was aimed at studying the effect of ruminal $NH_3-N$ levels on ruminal fermentation, microbial population, urinary purine derivative excretion, digestibility and rice straw intake in swamp buffaloes. Five, 3 to 4 years old, rumen fistulated swamp buffaloes were randomly assigned according to a $5{\times}5$ Latin square design to rceive five different intraruminal infusions of $NH_4HCO_3$ (0, 150, 300, 450 and 600 g/d) on a continuous daily basis. Rice straw as a roughage was offered ad libitum while concentrate was given at 0.8% BW daily. The results were that as levels of $NH_4HCO_3$ increased, ruminal $NH_3-N$ concentrations increased from 7.1 to 34.4 mg%. The highest digestibility and voluntary straw intakes were found at 13.6 to 17.6 mg% ruminal $NH_3-N$ levels; straw intake was highest at 13.6 mg%. Total bacterial and protozoal counts linearly increased as the ruminal $NH_3-N$ increased and were highest at 17.6 mg%. Total urinary purine derivatives and allantoin excretion were highest (44.0, 37.4 mM/d) at 17.6 mg% ruminal $NH_3-N$. Highest total VFAs (115 mM) were obtained a 13.6 mg% ruminal $NH_3-N$ while blood urea nitrogen significantly increased as ruminal $NH_3-N$ increased. The results from this experiment suggest that optimum ruminal $NH_3-N$ in swamp buffaloes is higher than 13.6 mg%, for improving rumen ecology, microbial protein synthesis, digestibility and straw intake.

Effects of various weaning times on growth performance, rumen fermentation and microbial population of yellow cattle calves

  • Objective: This study was conducted to investigate the effects of weaning times on the growth performance, rumen fermentation and microbial communities of yellow cattle calves. Methods: Eighteen calves were assigned to a conventional management group that was normally weaned (NW, n = 3) or to early weaned (EW) group where calves were weaned when the feed intake of solid feed (starter) reached 500 g ($EW_{500}$, n = 5), 750 g ($EW_{750}$, n = 5), or 1,000 g ($EW_{1,000}$, n = 5). Results: Compared with NW, the EW treatments increased average daily gain (p<0.05). The calves in $EW_{750}$ had a higher (p<0.05) starter intake than those in $EW_{1,000}$ from wk 9 to the end of the trial. The concentrations of total volatile fatty acids in $EW_{750}$ were greater than in NW and $EW_{1,000}$ (p<0.05). The EW treatments decreased the percentage of acetate (p<0.05). The endogenous enzyme activities of the rumen were increased by EW (p<0.05). EW had no effect on the number of total bacteria (p>0.05), but changes in bacterial composition were found. Conclusion: From the present study, it is inferred that EW is beneficial for rumen fermentation, and weaning when the feed intake of the starter reached 750 g showed much better results.

  • The soil of the reclaimed tidal land, located in Chogi-ri, Is. Kanghwa, Korea was used in this experiment. The experimented soil samples were collected from 18 sites with its time elapsed after the shore-protection works, soil-depth and the vegetation of saline plants, and at each site samplings were conducted monthly from March through October, 1968, for the purposes of examining the changes of microbial populations for the microbes such as bacteria, actinomycetes and fungi, by using the dilution plate method. The numbers of the microbes in these soils generally showed lower levels comparing with those of other soils. The more time elapsed after the reclamation, the higher numbers of the microbes inhibited the soils. Higher populations were there in the surface soils than in the lower part of the area. The surface soils included comparatively better conditions in aeration and contents of organic matter than in the lower part, and this fact was. same as in general soils. However, not so was this in the case of March, April and October due to the higher soil temperatures in the lows. At the experimental sites where the halophytes such as Salicorniu were grown vigourously, the more densly the plants grew, the higher populations of actinomycetes and fungi were, but not in the case of bacterial population. This means, in this soil with dense Salicornia, it is difficult to obtain good-natured soils in short time without a higher population of bacteria. For the rapid utilization of the land soil, in this view of point, the methods increasing the number of bacteria in the soil are needed as well as the cultivation and harvesting Salicorniu which indicated in the privious paper(Hong, et al., 1969a). According to the results of this experiment, the changes of soil-microbial populations in the reclaimed tidal land soil containing high salinity depend deeply upon the interrelations of many environmental factors such as soil-salinity, soil-components and contents, concentration of organic matters, pH, aeration, and air and soil temperatures, as in the general soils.

완속여과 여층 내 종속영양세균의 분포 특성 (Distribution characteristics of heterotropic bacteria population in slow sand filters)

  • Slow sand filtration (SSF) was the first engineered/mechanical filtration process used in drinking water treatment. In SSF, untreated water slowly percolate through a bed of porous sand. Biological activity within the sand bed have the strongest influence on removal efficiency of pollutants by slow sand filtration. In this study, the microbial population distributions in slow sand filters operated at the various operation conditions was evaluated. The concentrations of $10^4$ to $10^5$ CFU per g dry wt. were observed. No significant differences were seen between the number of filter-covered materials. The data indicate that the temperature has affect on population distribution. Also, the light exposure was influenced on microorganism in slow sand filtration according to the heterotropic plate counts. The role of microorganism within the sand media requires further study.

  • Dynamics of certain $N_2$fixing bacteria such as Rhizobium, Azospirillum and Azotobactor, nodule number in dominant legume, Atylosia trinervia, P-solubilizing bacteria, actinomycetes and fungi were studied in unburned and burned site of natural grassland, southern India. Population of $N_2$- fixing bacteria, P-solubilizing bacteria, fungi and nodule number in legume increased significantly in burned sites. On the other hand, the actinomycetes population remained unchanged. Thirty six species of fungi with tricalcium phosphate solubilizing ability were recorded. The most efficient P-solubilizing fungi recognised in the soils of the study sites are Absidia ramosa, Gongronella butlerii, Mortieralla spinosa, Mucor racemosus, Rhizopus nigricans, R. stolonifer, R. oryzae, Aspergillus fumigatus, A. nidulans, A. niger Theilavia terricola and Cheatomium lunasporium.

  • Influence of the moisture content in soils was examined on population variations of soil microbes, including Cylindrocarpon destructans causing root rot of ginseng, in vivo and under the field condition. Fungal populations decreased in soils treated with various moisture contents in vivo as days after the treatment in creased, but there was not a significant difference in the population among other treatments except 135% moisture content (flooding) at 15 weeks after the treatment. In flooded soils populations of total fungi and C. destructans were reduced to 1/10 and 1/50 of initial populations, respectively. There was, however, a little difference in the population of total bacteria or Actinomycetes between before and at 15 weeks after flooding. On the other hand, population variations of bacteria and Actinomycetes were much greater than those of fungi at different intervals after the moisture treatment. Variations of microbial populations in flooded soils under the field condition were similar to those in vivo. Especially, populations of Fusarium and pectolytic bacteria in flooded soils were reduced to 1/100 of populations in nonflooded soils at 170 days after treatment.

배추김치의 자동화 제조 공정 중 미생물 분석 (Microbial Analysis of Baechu-kimchi during Automatic Production Process)

  • 본 연구는 자동화된 상업용 배추김치 제조공정 중에서 원료와 공정별로 미생물의 변화를 조사하고자 수행하였다. 김치제조에 사용되는 여러가지 재료별로 미생물 분포를 조사한 결과, 마늘, 생강, 고추가루에서 많은 수의 호기성 박테리아, 젖산균, Leuconostoc, 그리고 효모가 검출되었으며, 이로써 위 부원료들이 김치에 존재하는 미생물들의 주요 공급원임을 알 수 있었다. 김치를 제조하는 공정 별로 미생물 변화를 모니터링 한 결과, 배추절임 공정에서 총 균수가 증가하였고 그 이후 세척공정에서는 단지 미생물수를 1 log CFU/g 줄이는데 그쳐 미생물 수 감소효과는 적은 것으로 판명되었다. 효모 또한 다양한 부재료와 최종 김치에서 검출되었다. 따라서, 김치 제품에 최종적으로 존재하는 미생물 수를 줄이고자 한다면 위의 원료 중에서는 위의 부재료에 대해, 그리고 제조공정에서는 절임에 사용되는 천일염과 절임탱크에 대해 적절한 살균대책이 강구되어야 하겠다. 또한 차염소산 살균 공정은 절임과 세척공정 사이에 위치해야만 가장 큰 효과를 기대할 수 있을 것으로 판단되었다. 또한 검출된 효모에 대한 저해 수단으로 적절한 저해제의 첨가도 고려되어야 하겠다. 본 연구의 결과는 공장김치의 HACCP 구축과 개선에 활용될 것으로 기대된다.

Effect of Dietary Supplementation of Red Ginseng By-product on Laying Performance, Blood Biochemistry, Serum Immunoglobulin and Microbial Population in Laying Hens

  • This study was carried out to investigate the effect of dietary supplementation of red ginseng by-product (RGB) on the laying performance, blood biochemistry, and microbial population in laying hens. A total of 120 Hy-Line Brown laying hens (75 weeks old) were randomly allotted to 1 of 3 dietary treatments with 4 replicates per treatment. A commercial-type basal diet was prepared, and 2 additional diets were prepared by supplementing 5.0 or 10.0 g/kg of RGB to the basal diet at the expense of corn. The diets were fed to hens on an ad libitum basis for 4 weeks. There were no differences in feed intake, egg weight, and feed conversion ratio during 4 weeks of the feeding trial. However, hen-day egg production was significantly greater (p<0.05) for the RGB treatment groups than that for the basal treatment group. There were no differences in triglyceride, aspartate aminotransferase, and alanine aminotransferase during the 4-week feeding trial. However, RGB supplementation increased (p<0.05) the serum immunoglobulin G (IgG) and IgM content compared with basal treatment group. The total cholesterol was lower (p<0.05) in the RGB treatments groups than that in the basal treatment group. The intestinal Lactobacillus population was greater (p<0.05) for the RGB treatments groups than that for the basal treatment group. However, the numbers of Salmonella and Escherichia coli were not different among dietary treatments. During the entire experiment, there was no significant difference in egg quality among all the treatments. In conclusion, in addition to improving hen-day production, there were positive effects of dietary RGB supplementation on serum immunoglobulin and cholesterol levels in laying hens.

Community of natural lactic acid bacteria and silage fermentation of corn stover and sugarcane tops in Africa

  • Objective: To effectively utilize crop by-product resources to address the shortage of animal feed during the dry season in Africa, the community of natural lactic acid bacteria (LAB) of corn stover and sugarcane tops and fermentation characteristics of silage were studied in Mozambique. Methods: Corn stover and sugarcane tops were obtained from agricultural field in Mozambique. Silage was prepared with LAB inoculant and cellulase enzyme and their fermentation quality and microbial population were analyzed. Results: Aerobic bacteria were the dominant population with 107 colony-forming unit/g of fresh matter in both crops prior to ensiling, while 104 to 107 LAB became the dominant bacteria during ensiling. Lactobacillus plantarum was more than 76.30% of total isolates which dominated silage fermentation in the LAB-treated sugarcane top silages or all corn stover silages. Fresh corn stover and sugarcane tops contain 65.05% to 76.10% neutral detergent fiber (NDF) and 6.52% to 6.77% crude protein (CP) on a dry matter basis, and these nutrients did not change greatly during ensiling. Corn stover exhibits higher LAB counts and water-soluble carbohydrates content than sugarcane top, which are naturally suited for ensiling. Meanwhile, sugarcane tops require LAB or cellulase additives for high quality of silage making. Conclusion: This study confirms that both crop by-products contain certain nutrients of CP and NDF that could be well-preserved in silage, and that they are potential roughage resources that could cover livestock feed shortages during the dry season in Africa.
