Microbial metal reduction influences the biogeochemical cycles of carbon and metals as well as plays an important role in the bioremediation of metals, radionuclides, and organic contaminants. The use of bacteria to facilitate the production of magnetite nanoparticles and the formation of carbonate minerals may provide new biotechnological processes for material synthesis and carbon sequestration. Metal-reducing bacteria were isolated from a variety of extreme environments, such as deep terrestrial subsurface, deep marine sediments, water near Hydrothemal vents, and alkaline ponds. Metal-reducing bacteria isolated from diverse extreme environments were able to reduce Fe(III), Mn(IV), Cr(VI), Co(III), and U(VI) using short chain fatty acids and/or hydrogen as the electron donors. These bacteria exhibited diverse mineral precipitation capabilities including the formation of magnetite ($Fe_3$$O_4$), siderite ($FeCO_3$), calcite ($CaCO_3$), rhodochrosite ($MnCO_3$), vivianite [$Fe_3$($PO_4$)$_2$ .$8H_2$O], and uraninite ($UO_2$). Geochemical and environmental factors such as atmospheres, chemical milieu, and species of bacteria affected the extent of Fe(III)-reduction as well as the mineralogy and morphology of the crystalline iron mineral phases. Thermophilic bacteria use amorphous Fe(III)-oxyhydroxide plus metals (Co, Cr, Ni) as an electron acceptor and organic carbon as an electron donor to synthesize metal-substituted magnetite. Metal reducing bacteria were capable of $CO_2$conversion Into sparingly soluble carbonate minerals, such as siderite and calcite using amorphous Fe(III)-oxyhydroxide or metal-rich fly ash. These results indicate that microbial Fe(III)-reduction may not only play important roles in iron and carbon biogeochemistry in natural environments, but also be potentially useful f3r the synthesis of submicron-sized ferromagnetic materials.
For microbial production of acetaminophen, a popular analgesic-antipyretic from aniline, we screened various fungi and bacteria. And we succeeded to some extents in acetaminophen production by successful protoplast fusion between S. lividans and S. globisporus and also between S. rimosus and S. aureofaciens. However, more fertile results might be brought via performing the cloning of acetanilide p-hydroxylation genes of Streptomyces in yeast. This study was initiated to determine whether the acetanilide p-hydroxylase of Streptomyces is cytochrome P-450 species or non-heme iron protein species. The p-hydroxylationactivity on acetanilide in S. aureofaciens ATCC 10762 was found to be unstable on exposing to the air. However, 100,000xg supernatant of the cell free extracts which were prepared in $N_2$ atmosphere showed the p-hydroxylation activity. Characteristic absorption peak of cytochrome P-450 after reduction with dithionite and addition of CO was not observed in the region of 450nm. Moreover, metyrapone and 2, 6-dichloroindophenol did not affect this enzyme activity, but sodium azide, sodium cyanide, cupric sulfate, cadmium chloride, ${\alpha}$, ${\alpha}'-dipyridyl$, and o-phenanthroline reduced p-hydroxylase activity considerably. S. fradiae NRRL 2702 was shown to have strong p-hydroxylation activity in intact cells. This activity disappeared in its cell free extracts. In its 100,000xg supernatant, however, characteristic absorption peak of cytochrome P-450 after reduction with dithionite and addition of CO was observed at 446nm. Thus, the results herein presented suggest that acetanilide p-hydroxylase of Streptomyces aureofaciens is not related to cytochrome P-450 and may include non-heme iron protein for its activity. However, it is not clear whether acetanilide p-hydroxylase in S. fradiae belongs to the same category of S. aureofaciens.
The marine sediment sustains from the anoxic condition due to increased nutrients of external sources. The nutrients are liberated from the sediment, which acts as an internal source. In hypoxic environments, anaerobic respiration results in the formation of several reduced matters, such as N2 and NH4+, N2O, Fe2+, H2S, etc. The experimental results have shown that nitrogen and sulfur played an influential, notable role in this biogeochemical cycle with expected chemical reductions and a 'diffusive' release of present nutrient components trapped in pore water inside sediment toward the bulk water. Nitate/ammonium, sulfate/sulfides, and ferrous/ferric irons are found to be the key players in these sediment-waters mutual interactions. Organonitrogen and nitrate in the sediment were likely to be converted to a form of ammonium. Reductive nitrogen is called dissimilatory nitrate reduction to ammonium and denitrification. The steady accumulation in the sediment and surplus increases in the overlying waters of ammonium strongly support this hypothesis as well as a diffusive action of the involved chemical species. Sulfate would serve as an essential electron acceptor so as to form acid volatile sulfides in present of Fe3+, which ended up as the Fe2+ positively with an aid of the residential microbial community.
In order to identify if bacteria surviving in soils and groundwater can change the oxidation/reduction potential of groundwater, Eh values of solution that contained bacteria were measured for 2 weeks. The Eh values of the solution reacted with sulfate-reducing bacteria decreased from -120 mV to -500 mV in 5 days, and $Desulfuricans$ was superior to $Vulgaris$ in reducing the solution. The Eh value was relatively higher for the solution containing $Shewanella$, iron-reducing bacteria, showing -400 mV. During the Eh decrease by the metal-reducing bacteria, a sulfide mineral such as mackinawite (FeS) started precipitating through the microbial reducing process for sulfate and ferric iron. These results show that the ORP of natrual groundwater may be sensitive to the geomicrobial respiration. In addition, a subsurface environment where groundwater is highly reduced and sulfide minerals are largely biogenerated may be a good place to retard the migration of oxidized radionu-clides by making them precipitated as reduced forms.

This study conducted a sediment culture experiment to investigate the effects of sediment oxygen demand (SOD) and environmental factors on sediment and water quality. We installed a leaching tank in the laboratory, cultured it for 20 days, and analyzed the relationship between P and Fe in the sediment. As a result, the dissolved oxygen of the water layer decreased with time, while the oxidation-reduction potential of the sediment progressed in the negative direction to form an anaerobic reducing environment. The SOD was measured to be 0.05 mg/g at the initial stage of cultivation and increased to 0.09 mg/g on the 20th day, indicating the tendency of increasing consumption of oxygen by the sediment. The change is likely to have caused by oxygen consumption from biological-SOD, which is the decomposition of organic matter accumulated on the sediment surface due to the increase of chl-a, and chemical-SOD consumed when the metal-reducing product produced by the reduction reaction is reoxidized. The correlation between SOD and causality for sediment-extracted sediments was positive for Ex-P and Org-P and negative for Fe-P. The analysis of the microbial community in the sediment on the 20th day showed that anaerobic iron-reducing bacteria (FeRB) were the dominant species. Therefore, when the phosphate bonded to the iron oxide is separated by the reduction reaction, the phosphate is eluted into the water to increase the primary productivity. The reduced substance is reoxidized and contributes to the oxygen consumption of the sediment. The results of this study would be useful as the reference information to improve oxygen resin.
Sediment works as a resource for electric cells. This paper was designed in order to verify how sediment cells work with anodic material such as metal and carbon fiber. As known quite well, sediment under sea, rivers or streams provides a furbished environment for generating electrons via some electron transfer mechanism within specific microbial population or corrosive oxidation on the metal surfaces in the presence of oxygen or water molecules. We experimented with one type of sediment cell using different anodic material so as to attain prolonged, maximum electric power. Iron, Zinc, aluminum, copper, zinc/copper, and graphite felt were tested for anodes. Also, combined type of anodes-metal embedded in the graphite fiber matrix-was experimented for better performances. The results show that the combined type of anodes exhibited sustainable electricity production for ca. 600 h with max. $0.57\;W/m^2$ Al/Graphite. Meanwhile, graphite-only electrodes produced max. $0.11\;W/m^2$ along with quite stationary electric output, and for a zinc electrode, in which the electricity generated was not stable with time, therefore resulting in relatively sharp drop in that after 100 h or so, the maximum power density was $0.64\;W/m^2$. It was observed that the corrosive reaction rates in the metal electrodes might be varied, so that strength and stability in the electric performances(voltage and current density) could be affected by them. In addition to that, COD(chemical oxygen demand) of the sediment of the cell system was reduced by 17.5~36.7% in 600 h, which implied that the organic matter in the sediment would be partially converted into non-COD substances, that is, would suggest a way for decontamination of the aged, anaerobic sediment as well. The pH reduction for all electrodes could be a sign of organic acid production due to complicated chemical changes in the sediment.

While sedimentological researches on Western coastal tidal flats of Korea have been much pelformed previously, mineralogical and biogeochemical studies are beginning to be studied. The objectives of this study were to investigate mineralogical characteritics of the inter-tidal flat sediments and to explore phase transformation of iron(oxyhydr)oxides and biomineralization by metal-reducing bacteria enriched from the inter-tidal flat sediments from Muan, Jeollanam-do, Korea. Inter-tidal flat sediment samples were collected in Chungkye-myun and Haeje-myun, Muan-gun, Jeollanam-do. Particle size analyses were performed using the pipette method and sedimentation method. The separates including sand, silt and clay fractions were examined by scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) analysis, transmission electron microscopy (TEM), and X-ray diffiaction (XRD). After enriching the metal-.educing bacteria from the into,-tidal flat sediments, the bacteria were used to study phase transformation of the synthesized iron (oxyhydr)oxides and iron biomineralization using lactate or glucose as the electron donors and Fe(III)-containing iron oxides as the electron accepters. Mineralogical studies showed that the sediments of tidal flats in Chung]rye-myun and Haeje-myun consist of quartz, plagioclase, microcline, biotite, kaolinite and illite. Biogeochemical researches showed that the metal-reducing bacteria enriched from the inter-tidal flat sediments reduced reddish brown akaganeite and mineralized nanometer-sized black magnetite. The bacteria also reduced the reddish brown ferrihydrite into black amorphous phases and reduced the yellowish goethite into greenish with formation of nm-sized phases. These results indicate that microbial Fe(III) reduction may play one of important roles in iron and carbon biogeochemistry as well as iron biomineralization in subsurface environments.

In the United States (U.S.), the monitored natural attenuation (MNA) approach has been used as an alternative remedial option for organic and inorganic compounds retained in soil and dissolved in groundwater. The U.S. Environmental Protection Agency (EPA) defines the MNA as“in-situ naturally-occurring processes include biodegradation, diffusion, dilution, sorption, volatilization, and/or chemical and biochemical stabilization of contaminants and reduce contaminant toxicity, mobility or volume to the levels that are protective of human health and the environment”. The Department of Soil Environment. National Institute Environmental Research (NIER) is in the process for demonstrating the MNA approach as a potential remedial option for the BTEX contaminated site in Uiwang City. The project is charactering the research site in terms of the nature and extend of contamination, biological degradation rate, and geochemical and hydrological properties. The microbial-degradation rate and effectiveness of nutrient and redox supplements will be determined through laboratory batch and column tests. The geochemical process will be monitored for determining the concentration changes of chemical species involved in the electron transfer processes that include methanogenesis, sulfate and iron reduction, denitrification, and aerobic respiration. Through field works, critical soil and hydrogeologic parameters will be acquired to simulate the effects of dispersion, advection, sorption, and biodegradation on the fate and transport of the dissolved-phase BTEX plume using Bioplume III model. The objectives of this multi-years research project are (1) to evaluate the MNA approach using the BTEX contaminated site in Uiwang City, (2) to establish a standard protocol for future application of the approach, (3) to investigate applicability of the passive approach as a secondary treatment remedy after active treatments. In this presentation, the overall picture and philosophy behind the MNA approach will be reviewed. Detailed discussions of the site characterization/monitoring plans and risk-based decision-making processes for the demonstration site will be included.
The smectite-illite (SI) reaction is a ubiquitous process in siliciclastic sedimentary environments. For the last 4 decades the importance of smectite to illite (S-I) reaction was described in research papers and reports, as the degree of the (S-I) reaction, termed "smectite illitization", is linked to the exploration of hydrocarbons, and geochemical/petrophysical indicators. The S-I transformation has been thought that the reaction, explained either by layer-by-layer mechanism in the solid state or dissolution/reprecipitation process, was entirely abiotic and to require burial, heat, and time to proceed, however few studies have taken into account the bacterial activity. Recent laboratory studies showed evidence suggesting that the structural ferric iron (Fe(III)) in clay minerals can be reduced by microbial activity and the role of microorganisms is to link organic matter oxidation to metal reduction, resulting in the S-I transformation. In abiotic systems, elevated temperatures are typically used in laboratory experiments to accelerate the smectite to illite reaction in order to compensate for a long geological time in nature. However, in biotic systems, bacteria may catalyze the reaction and elevated temperature or prolonged time may not be necessary. Despite the important role of microbe in S-I reaction, factors that control the reaction mechanism are not clearly addressed yet. This paper, therefore, overviews the current status of microbially mediated smectite-to-illite reaction studies and characterization techniques.

This study investigated the effects of elevated $CO_2$ and nitrogen addition on the anaerobic decomposition mediated by microorganisms to determine the microbial metabolic pathways in the degradation of organic matters of the sediments. There were statistically significant differences(P < 0.05) in the rates between denitrification and methanogenesis upon increased $CO_2$ concentration, nitrogen addition, in the presence of plants. Based on the assumption that anaerobic degradation of organic matter mainly occurs through denitrification, iron reduction, and methanogenesis, methanogenesis is the dominant pathways in the decomposition of organic matter under the condition of elevated $CO_2$ and nitrogen addition. In addition, the altered environment increased anaerobic carbon decomposition. Therefore, it can be concluded that freshwater wetland sediments have positive effects on the global warming by the increased methanogenesiss as well as increased anaerobic carbon decomposition.
