The objective of this study was investigated for the microbial community structure and treatment performance of domestic wastewater in lab-scale submerged membrane bioreactor operated with anoxic-oxic cycles. Respiratory quinone profiles were applied as tools for identifying different bacterial populations. The cycle time program of bioreactor was control under anoxic/oxic of 60/90 minutes with an hydraulic retention time of 8.4 hrs. The average $COD_{Cr}$ removal efficiency of domestic wastewater was as high as 93%. The results showed complete nitrification of $NH_4^+$-N generated during oxic period and up to 50% of the total nitrogen could be denitrified. The dominant quinone types of suspended microorganisms in bioreactor were ubiquinone (UQ)-8, -10, followed by menaquinone (MK)-6, and MK-7 for anoxic period, but those for oxic period were UQ-8, MK-6, followed by UQ-10 and MK-7. The microbial diversities of bioreactor at anoxic and oxic periods, calculated based on the composition of all quinones were 10.4 and 12.2-11.8, respectively. The experimental results showed that the microbial community structure in the submerged membrane bioreactor treating domestic wastewater was slightly affected by intermittent aeration.
The objective of this study was to investigate community structure of attached algae and microbes in sedimentation basin of water and wastewater treatment plants by using respiratory quinone profile. There was an evident difference, in microbial community structure and attached algae species, between inclination plate settler and drainage canal in the sedimentation basin. The algae was composed of species in following order; Chlorophyceae>Bascillariophyceae>Cyanophyceae. The dominant quinone types of attached microorganisms in the wastewater treatment plant were plastoquinone (PQ)-9, vitamin(V)K-1 followed by UQ-8, but those for water treatment plant were VK-1, PQ-9 followed by UQ-8. These results suggests that nutrients, seasons and material of sedimentation basin have notable influence on composition of attached algae and microbial community structure in water and wastewater treatment plants.

Permafrost is frozen soil below $0^{\circ}C$ for two or more years. Surface of permafrost is called as active layer that seasonally thaws during the summer. Although the thawing of permafrost may deepen the active layer and consequently increase the microbial activity, the microbial community structure in this habitat has not yet been well described. In this study, we presented bacterial and archaeal diversity in the active layer soil from Resolute, Canada using pyrosequencing analysis. The soil sample was collected from the surface of the marsh covered with moss and Carex. A total of 7,796 bacterial reads for 40 phyla and 245 archaeal reads for 4 phyla were collected, reflecting the high diversity of bacteria. Predominant bacterial groups were Proteobacteria (37.7%) and Bacteroidetes (30.0%) in this study. Major groups in Archaea were Euryarchaeota (51.4%) and Thaumarchaeota (46.1%). Both methane producing archaea and consuming bacteria were detected in this study. Although it might be difficult to characterize microbial community with only one sample, it could be used for the basis of assessing the relative importance of the specific groups with a high resolution on the bacterial and archaeal community in this habitat.
Understanding soil microbial community structure in the Arctic is essential for predicting the impact of climate change on interactions between organisms living in polar environments. The hypothesis of the present study was that soil microbial communities and soil chemical characteristics would vary depending on their associated plant species and local environments in Arctic mature soils. We analyzed soil bacterial communities and soil chemical characteristics from soil without vegetation (bare soil) and rhizosphere soil of three Arctic plants (Cassiope tetragona [L.] D. Don, Dryas octopetala L. and Silene acaulis [L.] Jacq.) in different local environments (coal-mined site and seashore-adjacent site). We did not observe any clear differences in microbial community structure in samples belonging to different plant rhizospheres; however, samples from different environmental sites had distinct microbial community structure. The samples from coal-mined site had a relatively higher abundance of Bacteroidetes and Firmicutes. On the other hand, Acidobacteria was more prevalent in seashore-adjacent samples. The relative abundance of Proteobacteria and Acidobacteria decreased toward higher soil pH, whereas that of Bacteroidetes and Firmicutes was positively correlated with soil pH. Our results suggest that soil bacterial community dissimilarity can be driven by spatial heterogeneity in deglaciated mature soil. Furthermore, these results indicate that soil microbial composition and relative abundance are more affected by soil pH, an abiotic factor, than plant species, a biotic factor.
Thermophilic anaerobic digestion (TAD) is characterized by higher biogas production rates as a result of assumedly faster microbial metabolic conversion rates compared to mesophilic AD. It was hypothesized that the thermophilic microbiome with its lower diversity than the mesophilic one is more susceptible to disturbances introduced by alterations in the operating factors, as an example, the supply of nitrogen-rich feedstock such as poultry manure (PM). Laboratory scaled TAD experiments using cattle slurry and increasing amounts of PM were carried out to investigate the (in-) stability of the process performance caused by the accumulation of ammonium and ammonia with special emphasis on the microbial community structure and its dynamic variation. The results revealed that the moderate PM addition, i.e., 25% (vol/vol based on volatile substances) PM, resulted in a reorganization of the microbial community structure which was still working sufficiently. With 50% PM application, the microbial community was further stepwise re-organized and was able to compensate for the high cytotoxic ammonia contents only for a short time resulting in consequent process disturbance and final process failure. This study demonstrated the ability of the acclimated thermophilic microbial community to tolerate a certain amount of nitrogen-rich substrate.
We compared the composition of phospholipid fatty acids (PLFA) to assess the microbial community structure in the soil and rhizosphere community of non-transgenic watermelons and transgenic watermelons in Miryang farmlands in Korea during the spring and summer of 2005. The PLFA data were seasonally examined for the number of PLFA to determine whether there is any difference in the microbial community in soils from two types of watermelons, non-transgenic and transgenic. We identified 78 PLFAs from the rhizosphere samples of the two types of watermelons. We found eight different PLFAs for the type of plants and sixteen PLFAs for the interaction of plant type and season. The PLFA data were analyzed by analysis of variance separated by plant type (P<0.0085), season (P<0.0154), and the plant type${\times}$season interaction (P<0.1595). Non-parametric multidimensional scaling (NMS showed a small apparent difference but multi-response permutation procedures (MRPP) confirmed that there was no difference in microbial community structure for soils of both plant types. Conclusively, there was no significant adverse effect of transgenic watermelon on bacterial and fungal relative abundance as measured by PLFA. We could reject our hypothesis that there might be an adverse effect from transgenic watermelon with our statistical results. Therefore, we can suggest the use of this PLFA methodology to examine the adverse effects of transgenic plants on the soil microbial community.
Effects of elevated $CO_2$ on soil microorganisms are known to be mediated by various interactions with plants, for which such effects are relatively poorly documented. In this review, we summarize and synthesize results from studies assessing impacts of elevated $CO_2$ on soil ecosystems, focusing primarily on plants and a variety the of microbial processes. The processes considered include changes in microbial biomass of C and N, microbial number, respiration rates, organic matter decomposition, soil enzyme activities, microbial community composition, and functional groups of bacteria mediating trace gas emission such as methane and nitrous oxide. Elevated $CO_2$ in atmosphere may enhance certain microbial processes such as $CH_4$ emission from wetlands due to enhanced carbon supply from plants. However, responses of extracellular enzyme activities and microbial community structure are still controversy, because interferences with other factors such as the types of plants, nutrient availabilitial in soil, soil types, analysis methods, and types of $CO_2$ fumigation systems are not fully understood.
We assayed the effects of simulated acid rain on the mass loss, $CO_2$ evolution, dehydrogenase activity, and microbial biomass-C of decomposing Sorbus alnifolia leaf litter at the microcosm. The dilute sulfuric acid solution composed the simulated acid rain, and the microcosm decomposition experiment was performed at 23$^{\circ}C$ and 40% humidity. During the early decomposition stage, decomposition rate of S. alnifolia leaf litter, and microbial biomass, $CO_2$ evolution and dehydrogenase activity were inhibited at a lower pH; however, during the late decomposition stage, these characteristics were not affected by pH level. The fungal component of the microbial community was conspicuous at lower pH levels and at the late decomposition stage. Conversely, the bacterial community was most evident during the initial decomposition phase and was especially dominant at higher pH levels. These changes in microbial community structure resulting from changes in microcosm acidity suggest that pH is an important aspect in the maintenance of the decomposition process. Litter decomposition exhibited a positive, linear relationship with both microbial respiration and microbial biomass. Fungal biomass exhibited a significant, positive relationship with $CO_2$ evolution from the decaying litter. Acid rain had a significant effect on microbial biomass and microbial community structure according to acid tolerance of each microbial species. Fungal biomass and decomposition activities were not only more important at a low pH than at a high pH but also fungal activity, such as $CO_2$ evolution, was closely related with litter decomposition rate.

Flowers of kiwifruit are morphologically hermaphroditic and survivable binucleate pollen is produced by the male flowers. In this study, we investigated microbial diversity in kiwifruit pollens by analyzing amplicon sequences of 16S rRNA. Four pollen samples were collected: 'NZ' was imported from New Zealand, 'CN' from China in year of 2014, respectively. 'KR13' and 'KR14' were collected in 2013' and 2014' in South Korea. Most of the identified bacterial phyla in the four different pollens were Proteobacteria, Actinobacteria and Firmicutes. However, the imported and the domestic pollen samples showed different aspects of microbial community structures. The domestic pollens had more diverse in diversity than the imported samples. Among top 20 OTUs, Pseudomonas spp. was the most dominant specie. Interestingly, a bacterial pathogen of kiwifruit canker, Pseudomonas syringae pv. actinidiae was detected in 'NZ' by the specific PCR. This study provides insights microbial distribution and community structure information in kiwifruit pollen.

Pseudomonas fluorescens pc78 is an effective biocontrol agent for soil-borne fungal diseases. We previously constructed a P43-gfp tagged biocontrol bacteria P. fluorescens pc78-48 to investigate bacterial traits in natural ecosystem and the environmental risk of genetically modified biocontrol bacteria in tomato rhizosphere. Fluctuation of culturable bacteria profile, microbial community structure, and potential horizontal gene transfer was investigated over time after the bacteria treatment to the tomato rhizosphere. Tagged gene transfer to other organisms such as tomato plants and bacteria cultured on various media was examined by polymerase chain reaction, using gene specific primers. Transfer of chromosomally integrated P43-gfp from pc78 to other organisms was not apparent. Population and colony types of culturable bacteria were not significantly affected by the introduction of P. fluorescens pc78 or pc78-48 into tomato rhizosphere. Additionally, terminal restriction fragment length polymorphism profiles were investigated to estimate the influence on the microbial community structure in tomato rhizosphere between non-treated and pc78-48-treated samples. Interestingly, rhizosphere soil treated with strain pc78-48 exhibited a significantly different bacterial community structure compared to that of non-treated rhizosphere soil. Our results suggest that biocontrol bacteria treatment influences microbial community in tomato rhizosphere, while the chromosomally modified biocontrol bacteria may not pose any specific environmental risk in terms of gene transfer.
