Quantitative Visualization of Oxygen Transfer in Micro-channel using Micro-LIF Technique (마이크로 레이저 형광 여기법을 이용한 미세채널 내부에서의 산소 확산에 대한 정량적 가시화)

  • In the present study, oxygen transfer process across gas-liquid interface in a Y-shape micro-channel is quantitatively visualized using the micro laser induced fluorescence (${\mu}$-LIF) technique. Diffusion coefficient of Oxygen ($D_L$) is estimated based on the experimental results and compared to its theoretical value. Tris ruthenium (II) chloride hexahydrate was used as the oxygen quenchable fluorescent dye. A light-emitting diode (LED) with wavelength of 450 nm was used as the light source and phosphorescence images of fluorescent dye were captured by a CMOS high speed camera installed on the microscope system. Water having dissolved oxygen (DO) value of 0% and pure oxygen gas were injected into the Y-shaped microchannel by using a double loading syringe pump. In-situ pixel-by-pixel calibration was carried out to obtain Stern-Volmer plots over whole flow field. Instantaneous DO concentration fields were successfully mapped according to Stern-Volmer plots and DL was calculated as $2.0675{\times}10^{-9}\;m^2/s$.

  • In order to investigate flow characteristics of blood flow in a micro tube ($100{\mu}m$ in diameter) according to hematocrit, in-vitro experiments were carried out using a micro-PIV technique. The micro-PIV system consists of a microscope, a 2 head Nd:YAG laser, a 12 bit cooled CCD camera and a delay generator. Blood was supplied into the micro tube using a syringe pump. Hematocrit of blood was controlled to be 20%, 30% and 40%. The blood flow has a cell free layer near the tube wall and its thickness was changed with increasing the flow rate and hematocrit. The hemorheological characteristics such as shear rate and viscosity were evaluated using the velocity field data measured. As the flow rate increased, the blunt velocity profile in the tube center was sharpened. The viscosity of blood was rapidly increased with decreasing shear rate, especially in the region of low shear rate, changing RBC rheological properties. The variation of velocity profile and blood viscosity shows typical characteristics of Non-Newtonian fluids. On the basis of inflection points, the cell free layer and two-phase flow consisting of plasma and suspensions including RBCs were clearly discriminated.

PIV Measurements of the Pressure Driven Flow Inside a T-Shaped Microchannel (T형 마이크로채널 내부 압력구동 유동의 PIV 계측)

  • A custom micro-PIV optics assembly has been used to measure the flow field inside a T-shaped microchannel. The micro-PIV system consists of microscope objectives of various magnifications, a dichroic cube, and an 8-bit CCD camera. Fluorescent particles of diameters 620nm have been used with a Nd:YAG laser and color filters. A programmable syringe pump with Teflon tubings were used to inject particle-seeded distilled water into the channel at flow rates of $420,\;40,\;60{\mu}L/hr$. The microchannels are fabricated with PDMS with a silicon mold, then $O_2-ion$ bonded onto a slide glass. Results show differences in flow characteristics and resolution according to fluid injection rates, and magnifications, respectively. The results show PIV results with vector-to-vector distances of $2{\mu}m$ with 32 pixel-square interrogation windows at $50{\%}$ overlap.

  • A microlens connected to microfluidic channel is fabricated. The microlens is sealed with an elastomeric membrane which deforms by pressure of fluid driven by a syringe pump resulting in the shape change of the microlens. The optical properties of the microlens could be controlled by changing the microlens shape. The microlens system were made of an elastomer, PDMS, using molding from a photoplastic master patterned by UV photolithography. The test results show the optical property of the lens could be made into convex and concave type by applying the fluidic pressure positive and negative.

Effect of Hexane Extract of Galla Rhois on Inflammatory Alveolar Bone Loss

  • One of functions of Galla Rhois (GR) is reportedly an anti-inflammatory effect on the several inflammatory diseases. However, an effect of GR related to periodontitis has not been investigated. In the present study, we examined the effect of the hexane extract of Galla Rhois (GR-H) on periodontitis. Cytotoxicity was assessed by MTS analysis using human gingival fibroblast (hGF) cells. Experimental periodontitis was induced by injecting E.coli LPS into the palatal gingiva maxillary molar thrice weekly for 3 weeks (LPS group). GR-H diluted in 1xPBS was orally administrated using a syringe at 30 mg/kg body weight and 100 mg/kg body weight once a day (GR-H group). GR-H effect on the alveolar bone loss (ABL) was digitized with a micro-CT. GR-H treatment at concentrations exceeding 0.5 mg/ml showed cytotoxic effect in hGF cells. The micro-CT among groups were presented for the different distances from cemento-enamel junction (CEJ) to alveolar bone crest (ABC). The results indicated an inhibitory effect on alveolar bone loss for orally administered GR-H in a model of LPS-induced periodontitis.

Preparation and their humidity-sensitive properties of crosslinked polymer containing phosphonium salts (포스포늄염을 포함하는 가교 고분자의 제조 및 그들의 감습특성)

  • 고습도 또는 결로시 내수성을 지니는 저항형 습도센서를 제조하기 위하여 vinylbenzyl tributyl phosphonium chloride를 포함하는 4원 공중합체를 합성하여 저습도와 고습도 범위에서 측정이 가능한 습도센서용 감습막을 제작하였다. 1,5-dibrompentane과 4차 염화 가교 반응을 통하여 최종 저항형 습도센서를 제작하였다. 감습막은 알루미나 기판에 한 쌍의 금 paste가 인쇄된 전극을 사용하였으며, 감습막은 다양한 비율의 vinylbenzyl tributyl phosphonium chloride, 2-(dimethlamino)ethyl methacrylate, 2-ethylhexyl acrylate, 그리고 [2-(methacryloyloxy)ethyl]trimethyl ammonium chloride 등을 이용하여 제조하였다. 감습막은 micro-syringe를 이용하여 도포하였으며 건조 oven에서 50℃에서 6시간 동안 가교반응을 진행하였다. 습도센서의 임피던스 특성은 2-(dimethylamino)ethyl methacrylate 또는 [2-(methacryloyloxy)ethyl]trimethyl ammonium chloride의 함량이 증가할수록 감소하였다. vinylbenzyl tributyl phosphonium chloride / 2-(dimethylamino)ethyl methacrylate / 2-ethylhexyl acrylate / [2-(methacryloyloxy)ethyl]trimethyl ammonium chloride의 조성비가 4/4/1/1인 공중합체의 경우 30-90%RH 범위에서 2.64 ㏁ - 3.11 ㏀의 임피던스 특성을 보였으며 우수한 직선성과 낮은 히스테리시스 나타내었다.

Improved Radiochemical Yields, Reliability and Improvement of Domestic $^{18}F$-FDG Auto Synthesizer (국산 $^{18}F$-FDG Auto Sysnthesizer의 수율 향상과 성능 개선)

  • Purpose: 2-[$^{18}F$]Fluoro-2-deoxy-D-glucose ([$^{18}F$]FDG) particularly plays as a important role in Positron Emission Tomography (PET) imaging in nuclear medicine. Domestic [$^{18}F$]FDG auto synthesizers are installed in Seoul National University Bundang Hospital (SNUBH) at June 2008, these modules were known that it's synthetic yields were guaranteed in average $45{\pm}5%$ so far. To improve yields and convenience of domestic [$^{18}F$]FDG auto synthesizer, numerous trials in reaction time, base concentration, pressure and temperature were performed to increase [$^{18}F$]FDG yields. Materials and Methods: Several synthetic factors (temperature, time and pressure) and shortcoming were corrected based on many evaporation test. Syringe dispensing of tetra-butylammonium bicarbonate (TBAB) was replaced with micro pipette to prepare tetrabutyl ammonium fluoride salt ([$^{18}F$]TBAF). Troublesome refill of liquid nitrogen every 2 hours which was used to protect vacuum system was changed to charcoal cartridge, base guard filter. To monitor the volume of delivered $[^{18}O]OH_2$ from cyclotron by surveillance camera, we set up the volumetric vial on the cover of the module. In addition to, the recovery vial was added in [$^{18}F$]FDG production system to recover [$^{18}F$]FDG loss due to the leak of valve ($V_{13,14}$) in [$^{18}F$]FDG purification process. Results: When we used micro pipette for adding TBAB ($30\;{\mu}L$ in 12% $H_2O$ in acetonitrile), this quantitative dispensation has enabled to improve $5.5{\pm}1.7%$ residual fluorine-18 activity in fluorine separation cartridge compared to syringe adding. Besides, the synthetic yields of [$^{18}F$]FDG has increased $58{\pm}2.6%$ (n=19), $58{\pm}2.9%$ (n=14), $60%{\pm}2.5%$ (n=17) for 3 months. The life cycle of charcoal cartridge and base vacuum was 3 months prior to filling liquid nitrogen every 2 hours and additional side separator can prevent pump corrosion by organic solvent. After setting of volumetric indicator vial, the operator can easily monitor the total volume of irradiated $[^{18}O]OH_2$ from cyclotron. The recovery vial can be used for the stabilizer when an irregular [$^{18}F$]FDG loss was generated by the leak of valves ($V_{13,14}$). Conclusions: We has optimized appropriate synthetic conditions (temperature, time, pressure) in domestic [$^{18}F$]FDG auto synthesizer. In addition to, the remodeling with several accessories improve yields of domestic [$^{18}F$]FDG auto synthesizer with reliable reproducibility.

Microfluidic System Based High Throughput Drug Screening System for Curcumin/TRAIL Combinational Chemotherapy in Human Prostate Cancer PC3 Cells

  • We have developed a fully automated high throughput drug screening (HTDS) system based on the microfluidic cell culture array to perform combinational chemotherapy. This system has 64 individually addressable cell culture chambers where the sequential combinatorial concentrations of two different drugs can be generated by two microfluidic diffusive mixers. Each diffusive mixer has two integrated micropumps connected to the media and the drug reservoirs respectively for generating the desired combination without the need for any extra equipment to perfuse the solution such as syringe pumps. The cell array is periodically exposed to the drug combination with the programmed LabVIEW system during a couple of days without extra handling after seeding the cells into the microfluidic device and also, this device does not require the continuous generation of solutions compared to the previous systems. Therefore, the total amount of drug being consumed per experiment is less than a few hundred micro liters in each reservoir. The utility of this system is demonstrated through investigating the viability of the prostate cancer PC3 cell line with the combinational treatments of curcumin and tumor necrosis factor-alpha related apoptosis inducing ligand (TRAIL). Our results suggest that the system can be used for screening and optimizing drug combination with a small amount of reagent for combinatorial chemotherapy against cancer cells.

pH Measurements with a Microcantilever Array-Based Biosensor System

  • In this paper, we present a pH measurement method that uses a microcantilever-array-based biosensor system. It is composed of microcantilever array, liquid cell, micro syringe pump, laser diode array, position sensitive detector, data acquisition device, and data processing software. Four microcantilevers are functionalized with pH-sensitive MHA(mercaptohexadecanoic acid) as a probe, while three microcantilevers are functionalized with HDT(hexadecane thiol) as reference. We prepare PBS(phosphate buffered saline) solutions of different pH and inject them into the liquid cell with a predefined volumetric speed at regular time intervals. The functionalized mircocantilevers in the liquid cell deflect as a self-assembled monolayer on the microcantilever binds with probe molecules in the solution. The difference in deflection between the MHA-covered probe microcantilever and the HDT-covered reference microcantilever was used to compensate for thermal drift. The deflection difference clearly increases with increasing pH in the solution. It was shown that when the pH values of the PBS solutions are high, there were large variations in the deflection of microcantilevers, whereas there were small variations for low pH value. The experimental results show that the microcantilever array functionalized with MHA and HDT can detect pH value with good repeatability.

Multichannel Liquid Phase Microextraction System (다채널 액상 미세 추출 시스템 설계 및 제작)

  • In this study, a multichannel gas-liquid microextraction system is designed by integrating the automatic elution of extraction line and multichannel gas-purging liquid phase microextraction. The system uses an injection pump and inert gas to push the extraction solvent to a sample bottle of a gas-phase color autosampler and then implements multichannel gas-liquid microextraction and gas chromatography-mass spectrometry. The system also employs a three-way integrated micro-high-temperature heater, syringe pump, and microflow controller to realize the simultaneous processing of multiple groups of samples, thus improving the sample pretreatment speed and reproducibility and reducing human error. Autoinjection experiments were implemented with polycyclic aromatic hydrocarbon standard samples. The experiments show that the average recovery rate of the system exceeds 70%, and the relative standard among the channels is less than 15%.
