Hyperparameter Optimization and Data Augmentation of Artificial Neural Networks for Prediction of Ammonia Emission Amount from Field-applied Manure (토양에 살포된 축산 분뇨로부터 암모니아 방출량 예측을 위한 인공신경망의 초매개변수 최적화와 데이터 증식)

  • A sufficient amount of data with quality is needed for training artificial neural networks (ANNs). However, developing ANN models with a small amount of data often appears in engineering fields. This paper presented an ANN model to improve prediction performance of the ammonia emission amount with 83 data. The ammonia emission rate included eleven inputs and two outputs (maximum ammonia loss, Nmax and time to reach half of Nmax, Km). Categorical input variables were transformed into multi-dimensional equal-distance variables, and 13 data were added into 66 training data using a generative adversarial network. Hyperparameters (number of layers, number of neurons, and activation function) of ANN were optimized using Gaussian process. Using 17 test data, the previous ANN model (Lim et al., 2007) showed the mean absolute error (MAE) of Km and Nmax to 0.0668 and 0.1860, respectively. The present ANN outperformed the previous model, reducing MAE by 38% and 56%.

Studies on the Denitrification in the Submerged Paddy Soil -IV. Influences of soil organic matter contents, soil temperature, pH values, kinds and levels of N-fertilizer on the evolution of N2O gas (논토양(土壤)의 탈질작용(脫窒作用)에 관(關)한 연구(硏究) -제(第)4보(報) 토양유기물함량(土壤有機物含量), 온도(溫度), pH, 질소비종(窒素肥種) 및 시비량(施肥量)이 탈질작용(脫窒作用)에 미치는 영향(影響))

  • A series of laboratory experiments were carried out to find the effects of soil organic matter contents, soil temperature, pH values, kinds and amount of nitrogen fertilizers on the denitrification-$N_2O$ gas evolution-. The results obtained were summarized as follows: 1. Denitrification rate, amount of $N_2O$ gas evolution, was influenced the order of organic matter contents>soil temperature>pH values>kinds of N-fertilizer>levels of N-fertilizer. 2. The highest dentrification rate was observed in organic matter content of 3.0%, pH values at 6.0 with application of $KNO_3$ at levels of 20 mgN/100g soil. 3. For the evolution of I mole $N_2O$ gas, averaged carbon consumption was obtained as 0.5 mole in all these experiment condition. However, the highest carbon consumption rate was obtained in organic matter contents for 1.0% with application of $(NH_4)_2SO_4$ at levels of 10 mgN/100g soil (1.06 mole) while lowest carbon consumption rate was obtained in organic matter contents for 3.0% with application of $KNO_3$ at levels of 20 mgN/100g soil (0.13 mole). 4. According to Michaelis-Menten's equation, the V/2 values for evolution of $N_2O$ gas was estimated by progress curve. The results obtained was as 550 ug for $(NH_2)_2CO$ and 1100 ug $N_2O/100g$ soil by application of $KNO_3$ in organic matter contents of 1.0% soil. On the other hand, when the application $(NH_4)_2SO_4$ the V/2 values of $N_2O$ gas was obtained as the amount of 490 ug/100g soil while V/2 values of $N_2O$ gas by application of $KNO_3$ was on the linear line in soil organic matter contents of 3.0%.

Application of Earthworm Casting-derived Biofilter Media for Hydrogen Sulfide Removal (지렁이 분변토를 이용한 생물담체가 충전된 바이오필터에 의한 황화수소 제거)

  • Earthworm casting was the natural fertilizer that contained high concentrations of nutrients such as nitrogen, phosphate and potassium and of over $10^8$ CFU/ml of microorganisms. Greater than 80% of feed was excreted through the fermentation by the intestinal enzyme, after worm had eaten feeds such as fallen leaves and rotten roots under the ground. Also, the soil structure of casting was known to be very efficient in the aspects of the porosity, the water permeability, and deodorizing activities. In this research, the biofilter packed with a biomedia made of casting and waste polyurethane foam, a binder, which helped to improve the durability and perpetuity of casting, was investigated to degrade malodorous hydrogen sulfide gas. The biomedia had no need of extra supply of nutrients and of microbial inoculations. On the beginning of the operations, it showed 100% removal of hydrogen sulfide gas without lag phase. At SV of 50 $h^{-1}$, hydrogen sulfide gas from the outlet of the biofilter was not detected, when inlet concentration increased to 450 ppmv. After that, removal efficiency decreased as increasing inlet hydrogen sulfide concentration. Hydrogen sulfide removal was maintained at almost 93% until inlet concentration was increased up to 950 ppmv, at which the elimination capacity of $H_2S$ was 61.2 g $S{\cdot}m^{-3}{\cdot}h^{-1}$. Maximum elimination capacity guaranteing 90% removal was 61.2, 65.9, 84.7, 89.4 g $S{\cdot}m^{-3}{\cdot}h^{-1}$ at SV ranging from 50 $h^{-1}$ to 300 $h^{-1}$, but was 59.3 g $S{\cdot}m^{-3}{\cdot}h^{-1}$ at SV of 400 $h^{-1}$. The results calculated from Michaelis-Menten equation revealed that $V_m$ increased from 66.04, 88.96, 117.35, 224.15, to 227.54 g $S{\cdot}m^{-3}{\cdot}h^{-1}$ with increasing space velocity in the range of 50 $h^{-1}$ to 400 $h^{-1}$. However, saturation constant$(K_s)$ decreased from 79.97 ppmv to 64.95 and 65.37 ppmv, and then increased to 127.72 and 157.43 ppmv.

Characterization of Lactate Dehydrogenase in Acanthogobius hasta (풀망둑(Acanthogobius hasta) 젖산탈수소효소의 특성)

  • The lactate dehydrogenase (EC 1.1.1.27, LDH) isozymes in tissues from Acanthogobius hasta were characterized by biochemical, immunochemical and kinetic methods. The activities of LDH in skeletal muscle and eye tissues were 65.30 and 53.25 units, but LDH activities in heart and liver tissues were very low. LDH/CS (EC 4.1.3.7, citrate synthase) in skeletal muscle was the highest as 22.29. Specific activities of LDH in brain, eye and skeletal muscle were 56.45, 38.04 and 11.0 units/mg, respectively. The LDH isozymes in tissues were separated by polyacrylamide gel electrophoresis after immunoprecipitation with antiserum against $A_4,\;B_4$ eye-specific $C_4$ and liver-specific $C_4$. LDH $AC_4$ isozymes were detected predominantly in skeletal muscle, brain and eye tissues, and $B_4$ isozyme was detected in heart. Anodal eye-specific $C_4$ and cathodal liver-specific $C_4$ were coexpressed in A. hasta. The eye-specific $C_4$ isozyme showed higher activity in eye tissue, but liver-specific $C_4$ isozyme showed lower activity in liver. As a result, one part of molecular structures in $A_4\;and\;C_4,\;A_4\;and\;B_4$, and eye-specific $C_4$ and liver-specific $C_4$ were similar, but in $B_4\;and\;C_4$ were different with each other. Therefore the subunit A may be conservative in evolution, and the evolution of subunit B seems to be faster than that of subunit A. The LDH $A_4$ isozyme of skeletal muscle was purified in the fraction from elution with NAD+ containing buffer of affinity chromatography and eye-specific $C_4$ isozyme was eluted right after $A_4$, so the structure of eye-specific $C_4$ isozyme is similar to $A_4$. And LDH activity remained 35.22-43.47% as a result of the inhibition by pyruvate, the Michaelis-Menten constant values for pyruvate was 0.080-0.098 mM, and Vmax were 153.85 units, 35.09 units in skeletal muscle and eye, respectively. Also the $B_4$ isozyme was the thermo-stablest and $C_4$ was stabler than $A_4$ isozyme. The optimum pH of LDH was 6.5. The results mentioned above indicate that isozymes in tissues showed the properties between LDH $A_4\;and\;B_4$ isozyme as A. hasta was adapted to hypoxic conditions. Also LDH seems to function more effectively under anaerobic condition because LDH in skeletal muscle and eye tissues have high affinity for pyruvate.

Comparison of Kinetic Parameters, pH and Thermal Properties of Soluble and Immobilized Transglucosidase from Aspergillus niger (Aspergillus niger 유래의 Soluble과 고정화 Transglucosidase의 속도상수, pH 및 열 특성 비교)

  • Enzymatic properties of immobilized transglucosidase (TG) from Aspergillus niger was characterized and compared with soluble TG. Michaelis-Menten constant $(K_m)$ and maximum velocity $(V_{max})$ of immobilized TG were $122\;mM,\;3.9{\times}10^{-2}\;mM/min$ and in case of soluble TG, 21 mM, 0.4 mM/min. The optimum pH of immobilized TG was pH 5.0 like soluble TG but immobilized TG showed 16% and 45% higher activity than soluble TG at pH 5.0 and pH 6.0. Both of immobilized TG and soluble TG were stable from pH 2.0 to pH 9.0, and therefore their activities in these pH ranges were remained more than 90%. The temperature was optimal at $60{\sim}70^{\circ}C\;and\;70{\sim}80^{\circ}C$ for soluble TG and immobilized TG, respectively. The thermal stability of immobilized TG was significantly improved than that of soluble TG, and immobilized TG retained $32{\sim}40%$ higher activity than soluble TG. D-values from thermal inactivation of immobilized TG were 7690 sec at $65^{\circ}C$, 83 sec at $75^{\circ}C$, 7.2 sec at $80^{\circ}C$. Z-values of soluble and immobilized TG were $6.4^{\circ}C\;and\;5.3^{\circ}C$, respectively. The little difference of activation energies of soluble TG and immobilized TG supposed that there was little difference in mass transfer limitation during the reaction of soluble TG and immobilized TG.

Optimization of Microbial Production of Ethanol form Carbon Monoxide (미생물을 이용한 일산화탄소로부터 에탄올 생산공정 최적화)

  • The method to optimize the microbial production of ethanol from CO using Clostridium ljungdahlii was developed. The kinetic parameter study on CO conversion with Clostridium ljungdahlii was carried out and maximum CO conversion rate of 37.14 mmol/L-hr-O.D. and $K_{m}$ / of 0.9516 atm were obtained. It was observed that method of two stage fermentation, which consists of cell growth stage and ethanol production stage, was effective to produce ethanol. When pH was shifted from 5.5 to 4.5 and ammonium solution was supplied to culture media as nitrogen source at ethanol production stage, the concentration of ethanol produced was increased 20 times higher than that without shift. Ethanol production from CO in a fermenter with Clostridium ljungdahlii was optimized and the concentration of ethanol produced was 45 g/L and maximun ethanol productivity was 0.75 g ethanol/L-hr.
