Caffeine 및 Phenobarbital 장기투여가 흰쥐 각종 장기에 미치는 영향 (The Influence of Long Term Treatment with Caffeine and Phenobarbital on Various Organs in Rats)

  • The present study is concerned with the demonstration of the influence of long term treatment with caffeine and phenobarbital on pentobarbital sleeping time, gastric secretion, increase rate of body weight and brain and liver weight in rats. The experimental subjects were rats weighing about 140 to 150 g, each of them was isolated in a separate cage. Each group was given 1 ml normal saline solution as control, caffeine 10 mg/kg and phenobarbital 30 mg/kg as experimental groups. All drugs were injected intraperitoneally, daily for 4 weeks. The results obtained are summarized as follows; 1. There was significant difference between before and after injection of drugs (caffeine citrate 10 mg/kg and phenobarbital 30 mg/kg) on pentobarbital sleeping time. The sleeping time of caffeine treated group was delayed (22.4%, p<0.01) significantly compared with that of before injection. The sleeping time of phenobarbital treated group was markedly shortened (93.6%, p<0.001) compared with that of before injection of drugs. 2. The volume, free and total acidity and pH of gastric juice determined five hours after pyloric ligation in fasting rats were not significantly changed in experimental groups compared with control group. However the volume of gastric juice was increased 25% in both caffeine and phenobardital treated group. 3. The increased ratio of body weight revealed no remarkable difference compared with intial body weight. However, caffeine treated group showed markedly increased body weight after first and second week of injection. 4. The brain and liver weight in experimental group showed no significant difference compared with control group (as percentage of body weight).

메탄자화균에 의한 코발트의 생물흡착 (Biosorption of Cobalt by Methanotrophic Biomass)

  • 메탄자화균에 의한 코발트 제거의 최적 pH 영역은 6.0~12.0이었으나 메탄자화균을 넣지 않은 blank는 10.5~11.5이었다. 코발트의 제거능은 pH에 크게 의존하였으나 blank보다는 민감하지 않았다. 초기 pH 6.0에서 1.0 g/L의 메탄자화균을 투입했을 때 170 mg Co/g biomass가 제거되었다. SEM 분석 결과에 의하면 코발트는 메탄자확균의 표면이나 세포의 분비 고분자에 흡착되어 제거된 것으로 사료된다. 초기 pH 6.0, 400 mg Co/L에서 메탄자화균의 최적의 투입량은 1.0 g/L이었다. 2.0 M NaCl과 $NaNO_3$의 높은 이온강도 하에서도 코발트 제거능은 그다지 영향을 받지 않았다.

추출조건이 장뇌삼추출물의 화학성분 조성에 미치는 영향 (Effect of Extracting Conditions on Chemical Compositions of Korean Mountain Ginseng Extract)

  • 국내산 장뇌삼을 $-70^{\circ}C$쉐서 동결건조 분말시료에 대한 추출용매와 농도에 따른 화학성분을 분석, 비교하였다. 장뇌삼추출액의 고형분함량은 $50\%$ 에탄올추출액과 메탄올추출액이 $11.16\%$$13.45\%$로 가장 높은 함량을 보였다. 당도는 $90\%$ 에탄올추출액과 $100\%$ 증류수추출액이 각각 $19.98\%$$19.65\%$로 가장 높았고, pH는 $5.82\~6.60$수준이었고, 갈색도는 흡광도가 $50\%$ 에탄올추출액이 1.458로 가장 높은 값을 나타내었다 색도의 경우 L값은 $100\%$ 증류수추출액이 21.28로 에탄올추출액 $17.18\~21.02$보다 높은 값을 나타내었고, a값은 $100\%$ 증류수추출액이 -0.12로 가장 높은 수준이었고, b값은 $90\%$ 메탄올추출액이 1.34로 가장 높은 수준이었다 유리당 함량은 에탄을 농도가 높을수록 함량이 높게 나타나 $90\%$ 에탄올추출액은 6,159 mg/100 g으로 높은 함량을 나타내었다. 유리당 조성 중 가장 많은 함량을 차지 하는 sucrose는 에탄을 농도가 높아짐에 따라 매우 높은 증가 현상을 보였는데 $90\%$ 에탄올추출액과 메탄올추출액에서 6,159 mg/100 g과 5,238 mg/100 g으로 매우 높은 함유량을 나타내었다 유기산 조성은 citric acid와 malic acid가 주된 유기산이었다. 주요유리아미노산으로는 arginine, proline, $\gamma$ -amino-n-butyric acid, alanine 및 aspartic acid가 매우 높은 함량을 보였다. 총사포닌 함량은 $90\%$ 메탄올추출액에 $10.50\%$로 높은 함유량을 나타내었으며, P, K, Na, Mg 및 Ca 등이 주된 무기질로 확인되었다.

세포 밖 2가 양이온이 과분극에 의해 활성화되는 전류($I_h$)에 미치는 영향 (Effects of Extacellular Divalent Cations on the Hyperpolarization-activated Currents in Rat Dorsal Root Ganglion Neurons)

  • The hyperpolarization-activated current ($I_h$) is an inward cation current activated by hyperpolarization of the membrane potential and plays a role as an important modulator of action potential firing frequency in many excitable cells. In the present study we investigated the effects of extracellular divalent cations on $I_h$ in dorsal root ganglion (DRG) neurons using whole-cell voltage clamp technique. $I_h$ was slightly increased in $Ca^{2+}$-free bath solution. BAPTA-AM did not change the amplitudes of $I_h$. Amplitudes of $I_h$ were decreased by $Ca^{2+}$, $Mg^{2+}$ and $Ba^{2+}$ dose-dependently and voltage-independently. Inhibition magnitudes of $I_h$ by external divalent cations were partly reversed by the concomitant increase of extracellular $K^+$ concentration. Reversal potential of $I_h$ was significantly shifted by $Ba^{2+}$ and $V_{1/2}$ was significantly affected by the changes of extracellular $Ca^{2+}$ concentrations. These results suggest that $I_h$ is inhibited by extracellular divalent cations ($Ca^{2+}$, $Mg^{2+}$ and $Ba^{2+}$) by interfering ion influxes in cultured rat DRG neurons.

음양곽(淫羊藿)이 토끼의 수축혈관에 미치는 이완효과 (Relaxation Effect of Epimedium Koreanum Extract on Rabbit Carotid Artery)

  • The purpose of this study is to investigate vasorelaxant effect of Epimedium koreanum(EK) extract on rabbit carotid artery. In this study, to determine vasorelaxant effect of EK extract on rabbit carotid artery, arterial rings with intact or damaged endothelium were used for experiment using organ bath, and were contracted by norepinephrine(NE). After being contracted, arterial rings were treated with EK extract in a dose-dependent manner To study its mechanism, the contracted arterial rings induced by NE were pretreated with indomethacin(IM), $N_{\omega}$-nitro-L-arginine(L-NNA), methylene blue(MB) or tetraethylammonium chloride(TEA) and 0.1 $mg/m{\ell}$ EK extract was added. To analyze the effect of the EK extract on influx of extracellular calcium chloride($Ca^{2+}$) in rabbit carotid artery, in $Ca^{2+}$-free krebs solution, krebs solution containing 1 mM $Ca^{2+}$ was infused into the contracted arterial ring by NE after pretreatment of EK extract. To measure the cytotoxicity of the EK extract, cell viability of human umbilical vein endothelial cell(HUVEC) was measured by MTT assay, and nitric oxide(NO) was measured by Griess reagent. The EK extract significantly was relaxed the arterial ring with intact endothelium contracted by NE, but the vasorelaxant effect of the EK extract was inhibited in the arterial rings with damaged endothelium. The vasorelaxant effect of the EK extract was not different between the IM-pretreatedand and non-treated arterial rings. The vasorelaxant effect of EK extract were significantly inhibited, when arterial rings were pretreated with L-NNA, TEA, MB. And in $Ca^{2+}$-free krebs solution, increasing of arterial contraction by $Ca^{2+}$ addition were also inhibited by the treatment of EK, but not significant. The treatment of EK extract was increased NO concentration in HUVEC. This study suggested that the vasorelaxant effect of EK extract would be related with EDHF and NO production and increasing of cyclic GMP.

Role of Calcium in Reperfusion Damage of Ischemic Myocardium; Influence on Oxygen Radical Production

  • The role of calcium in the production of oxygen radical which causes reperfusion damage of ischemic heart has been examined. The reperfusion damage was indrced in isolated Langendorff perfused rat hearts by aortic clamping for 60 min followed by reperfusion with oxygenated Krebs-Henseleit solution with or without 1.25 mM $CaCl_2.$ On reperfusion of the ischemic hearts with the calcium containing solution, the release of cytosolic enzymes (LDH and CPK) increased abruptly. These increased release of enzymes were significantly inhibited by additions of oxygen radical scavengers (SOD, 5,000 U; catalase, 12,500 U) into the reperfusion solution. In the hearts isolated from rats pretreated with allopurinol(20 mg/kg orally, 24 hr and 2 hr prior to the experiments), the levels of enzymes being released during reperfusion were significantly lower than that of the control. However, in the hearts perfused with the calcium-free but oxygenated solution, the increase in the release of cytosolic enzymes during reperfusion was neither inhibited by oxygen radical scavengers nor by allopurinol pretreatment. For providing the evidence of oxygen radical generation during the reperfusion of ischemic hearts in situ, the SOD-inhibitable reduction of exogenously administered ferricytochrome C was measured. In the hearts perfused with the calcium containing solution, the SOD-inhibitable ferricytochrome C reduction increased within the first minute of reperfusion, and was almost completely inhibited by allopurinol pretreatment. When the heart was perfused with the calcium free solution, however, the reduction of ferricytochrome C was not only less than that in the calcium containing condition, but also was not so completely inhibited by allopurinol pretreatment. By ischemia, xanthine oxidase (XOD) in the ventricular tissue was changed qualitatively, but not quantitatively. In the heart made ischemic with the calcium containing condition, the oxygen radical producing O-form of XOD increased, while the D- and D/O-form decreased. However, in the ischemic heart reperfused with the calcium free condition, the D/O-form of XOD was elevated without significant increase in O-form of the enzyme. It is suggested from these results that the calclum may play a contributing role in the genesis of reperfusion damage by promoting the conversion of xanthine oxidase from the D/O-form to the oxygen radical producing O-form in the ischemic myocardium.

폴리페놀 함량과 항산화력에 따른 로즈마리의 최적 추출조건 확립 (Optimal Extraction Conditions to Produce Rosemary Extracts with Higher Phenolic Content and Antioxidant Activity)

  • 본 연구에서는 로즈마리의 항산화 물질을 최대로 추출할 수 있는 최적의 추출 조건을 확립하였다. 열수 추출의 경우에는 $90^{\circ}C$, 30분 추출할 경우 항산화력($IC_{50}$ 값이 4.49 g DM/L)과 총 페놀함량(28.30 mg GAE/g DM)이 다른 온도와 시간보다 높은 수치를 나타내었다. 에탄올 추출의 경우에는 50%에탄올, $70^{\circ}C$, 10분의 경우가 항산화력($IC_{50}$ 값이 2.60 g DM/L), 총 페놀함량(40.28 mg GAE/g DM)이 다른 농도와 온도, 시간보다 높은 결과를 나타내었다 메탄올 추출 결과는 75% 메탄올, $60^{\circ}C$, 30분의 경우가 항산화력($IC_{50}$ 값이 2.31 g DM/L), 총 페놀함량(57.22 GAE/g DM)이 다른 농도와 온도, 시간보다 높은 수치를 나타내었다.

태음조위탕(太陰調胃湯)의 유탕억제효능(遺瘍抑制效能)에 관(關)한 연구(硏究) (A study on the ulcerous inhibitory effects of Taeumchowetang)

  • The experimental study used in this paper was done to examine the clinical effects of Taeumchowetang for the gastric ulcer and the function of gastrointestinal tract using rats and mice which were administered orally the water extraction from Taeumchowetang. Then, the counter-action of the water extraction on the isolated ileum and gastric fundus, the inhibitory effects of pylorus-ligated ulcer and indomethacin-induced ulcer, the associations with gastric juice secretion, total acidity, pepsin output, the transportability in the small and large intestine, were studied with administering acetylcholine chloride and barium chloride. In addition, it was investigated whether the central nervous system related to pain control and sleeping time was influenced by Taeumchowetang. The following results have been obtained; 1. As resulting from injection of acetylcholine chloride and barium chloride into the isolated ileum of rats and mice, Taeumchowetang led to have an inhibitory effect on the muscle contraction of the ileum. Then, acetylcholine chloride was measured as lower effect than barium chloride 2. For the inhibitory effect on contraction for the gastric fundus strip in rats by either acetylcholine chloride or barium chloride, the one showed low inhibitory effect, on the other hand the other showed density-dependent effect. 3. The water extraction was given on the pylorus ligated ulcer with using two different administration groups of 1,300mg/kg and 2,600mg/kg, each result was measured as 22.9% and 36.5% for an ulcerous inhibitory effect (p<0.05). 4. According to the two administration groups, the preventive effect was tabulated 18.1% and 29.3% for indomethacin-induced ulcer (p<0.05, P<0.01). 5. For associations with gastric juice secretion, total acidity, pepsin output in the administration group 2,600mg/kg, Taeumchowetang was recognized as having an inhibitory effects related to suppressive actions involving gastric juice secretion(p<0.05), and free acidity(p<0.01), but there was no significant association with total acidity and pepsin output. 6. To know the transportability in the intestine, BaSO4 solution was used. The transportability of the small intestine in the administration group 2,600mg/kg was 22.2% which was statistically significant compared with the large intestine's transportability(P<0.01). 7. In the administration group 1,300mg/kg and 2,600mg/kg, analgesic effect with against acetic acid was measured as being 16.8% and 24.4% which was shown as statistically siginificant(p<0.01). 8. No statistically significant association between Taeumchowetang and sleeping time was found in both 1,300mg/kg and 2,600mg/kg by administering phenobarbital-Na. According to the results of this study, Taeumchowetang has agreed with the effects of literature review. Further more in this study, Taeumchowetang also has preventive effects on pylorus-ligated ulcer. Hence, Teaumchowetang can be significant effect such as both anti ulcer agent and increasing gastric activity for the patients who suffer from gastric ulcer.

Absidia coerulea와 Thraustochitrium Sp. 에 의한 납과 코발트의 생물흡착 (Biosorption of Lead and Cobalt by Absidia coerulea and Thraustochitrium sp.)

  • 중금속 폐수 및 방사성 폐액 속에 포함된 납과 코발트를 제거하기 위해 생물흡착제로 Absidia coerulea와 Thraustochitrium sp.를 사용하여 흡착실험을 진행하였다. 납과 코발트 제거의 최적 pH 영역은 blank의 경우 각각 8.4~11.2, 10.5~11.5, Absidia coerulea는 6.5~11.4, 8.6~12.0, 그리고 Thraustochitrium sp.는 4.2~10.5, 8.9~11.6이었다. 초기 pH 5.0에서 1 g/L의 생물흡착제를 투입했을 때 Absidia coerulea와 Thraustochitrium sp.는 104와 125 mg/g dry biomass의 납을 제거할 수 있었으며, 초기 pH 6.0에서 코발트는 단지 2와 20 mg Co/g dry biomass만을 제거할 수 있었다. 초기 500 mg Pb/L, pH 5.0에서 최적 생물흡착제 투입량은 Absidira coerulea와 Thraustochitrium sp.가 각각 0.2 g/L이었으며, 초기 200 mg Co/L, pH 6.0에서는 3.0 g/L이었다 . Absidia coerulea와 Thraustochitrium sp.는 납에 대해서 높은 흡착능을 보였으나 코발트에 대해서는 비교적 낮은 흡착능을 보였다.

수직형 폴리머 마이크로링 공진기 기반의 글루코스 바이오 센서 (Refractometric Glucose Biosensor Incorporating a Vertically Coupled Microring Resonator in Polymeric Waveguides)

  • 본 논문에서는 굴절률 감지 방식의 수직 결합형 폴리머 마이크로링 공진기 기반 글루코스 바이오 센서를 제안하고 구현하였다. 이 센서에서 상부 클래딩 역할을 하는 글루코스 분석 대상물의 농도 변화는 링 공진기의 공진파장 이동을 측정함으로써 얻어진다. 특히, 센서의 감도를 향상시키기 위해 다음과 같은 방법을 사용하였다. 첫째로, 분석 대상물에 근접한 유효굴절률을 갖는 폴리머 도파로 구조를 공진기 센서에 도입하였다. 둘째로, 분석 물질의 접촉면적을 확대하기 위해 측면 클래딩층이 충분히 식각된 pedestal 링 도파로 구조를 사용하였다. 제안된 공진기 센서는 빔전파 방법을 사용하여 설계하고 분석하였다. 사용된 코어 및 클래딩용 폴리머의 굴절률은 각각 1.430과 1.375였으며, 링 반경은 $400\;{\mu}m$였다. 이 때 얻어진 도파로의 유효굴절률은 ${\sim}1.390$였으며 글루코스 수용액의 굴절률 1.333와 매우 근접하였다. 제작된 소자의 기본적인 전달특성은 FSR(free spectral range) 0.66 nm, 대역폭 ${\sim}0.15\;nm$, Q 값 10,000 이었다. 1550 nm 광파장 대역에서 얻어진 측정 결과를 살펴보면, 센서 감도는 ${\sim}0.28\;pm$/(mg/dL)였으며, 이에 따른 굴절률 변화 감도는 ${\sim}200\;nm/RIU$였다.
