GC를 이용한 극성용매의 분석방법 개발 연구 (Study on Development of Analytical Method for Polar Solvents by GC)

  • Objectives: The purpose of this study is to simplify and standardize analytical method of polar solvents(methanol, isopropanol, n-butanol, acetone, methylene chloride and MIBK) in the working environmental by GC. Because NIOSH methods are various and complicated. Methods: The method is using the same stationary phase(5% phenyl 95% dimethylpolysiloxane), absorbent(silica gel) and desorption solvent(DMSO) for above 6 solvents. For the 6 solvents desorption efficiency, calibration curve, and limit of detection were studied Results: As the results, 6 solvents{2 groups ; first group(methanol/isopropanol/butanol) and second group(acetone/methylene chloride/MIBK)} could be separated and quantified within 10 minutes. Desorption efficiency from silica gell of 6 solvents using dimethylsulfoxide(DMSO) was methanol 86.2%, isopropanol 103.2%, n-butanol 101.8%, acetone 98.2%, methylene chloride 103.9% and MIBK 106.2% in the range of 0.2, 0.5, 2.0 times of TWA, consequently, satisfied NIOSH estimation level(beyond 75%). Correlation coefficient(r)in the range of 0.2~2.0 times of TWA, was above 0.999 for 6 solvent. LOD(mg/DMSO ml) using calibration curve in the range of 0.2~2 times of TWA was methanol 0.11, isopropyl alcohol 0.20, n-butanol 0.03, acetone 0.50, methylene chloride 0.05, MIBK 0.04 respectably. Conclusions: This method can be used at the sampling and analytical method for polar solvents by GC. Also, will be able to be applied with NIOSH methods.

국내산 생약추출물의 항산화 효과 및 생리활성 (Antioxidative Activity and Physiological Activity of Some Korean Medicinal Plants)

  • 국내산 재배 생약류의 용매추출물의 항산화력 비교를 위해 28가지 생약류를 methylene chloride로 각각 추출한 후 linoleic acid에 1,000 ppm 첨가하여 항산화력을 비교하였다. 28종의 생약 methylene chloride 추출물들은 대조구에 비해 다소 차이는 있으나 모두 항산화력을 나타내었다. 황금추출물은 BHA와 비슷한 수준의 항산화력을 보였으며, 목단과 맥문동은 황금보다는 효과가 떨어졌으나 비교적 높은 항산화 효과를 나타내었다. Methylene chloride 추출물의 항산화 효과는 황금이 가장 뛰어난 것으로 나타났으므로 극성이 서로 다른 9가지용매로 추출하였을 때 전반적으로 극성이 높아질수록 수율이 높아졌다. 국내산 생약류의 생리활성을 알아보기 위하여 열수추출물의 아질산염 소거작용, 전자공여 작용 그리고 항보체 활성을 조사하였다.

뇌 신경교세포에서 가미보양환오탕(加味補陽還五湯) 분획물의 항염증 효과 비교 연구 (Anti-inflammatory Effects of Different Fractions Isolated from Modified Boyanghwano-tang Extract in LPS-stimulated Microglial Cells)

  • Objectives : In this study, the effects of different fractions isolated from modified Boyanghwanotang(mBHT) extract on LPS-induced inflammation in BV2 microglial cells were investigated. Methods : mBHT was extracted with water, and then fractionated with n-hexane, methylene chloride, ethylacetate and n-butanol. BV2 cells, a mouse microglia line were incubated with different concentrations of each fraction of mBHT for 30 min, and then stimulated with LPS for 24 h. Cell toxicity was determined by MTT assay. The concentration of nitric oxide (NO) was measured in culture medium by Griess reagent assay. The expression of inducible nitric oxide synthease (iNOS) protein was determined by Western blot. Results : Four fractions of mBHT were significantly inhibited LPS-induced NO productions in BV2 cells in a dose-dependent manner. The methylene chloride fraction of mBHT was most strongly inhibited the NO production compared with those of the others. The methylene chloride fraction of mBHT was also suppressed LPS-induced iNOS expression comparison of other fractions at same concentration ($50\;{\mu}g/ml$) in BV2 cells. Conclusions : The results showed that the methylene chloride fraction of mBHT may have an strong anti-inflammatory property through the inhibition of NO production and iNOS expression in activated microglia, and could a therapeutic potential for the treatment of various brain inflammatory diseases.

Inhibitory Effects of Herbal Extracts on Dopa Oxidase Activity of Tyrosinase

  • Tyrosinase catalyzes the rate-limiting steps in melanin biosynthesis which is involved in skin-coloring and local hyperpigmentation of human beings, and unfavorable darkening of food products, Inhibitory effects on 3,4-dihydroxy-phenylalanine (dopa) oxidase activity of tyrosinase by 594 kinds of herbal extracts prepared from herbal medicines and wild plants in Korea were estimated. Two herbal extracts prepared from radicis cortex of Morus alba and rhizoma of Curcuma longa were selected as those exhibiting potent inhibitory effects on the enzyme activity. These herbal extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol, and polar residue. The inhibitory effects on dopa oxidase activity of tyrosinase were shown in ethyl acetate fraction of Morus alba, and in methylene chloride fraction of Curcuma langa. The ethyl acetate fraction of Marus alba exhibited 50% of inhibition on dopa oxidase activity of tyrosinase at the concentration of 12 ${\mu}g/ml$, and methylene chloride fraction of Curcuma langa at 51 ${\mu}g/ml$.

낙상홍 추출물의 효능에 관한 연구 (A Study of Effects of Ilex serrata Thunb Extracts)

  • In this study, Ilex serrata Thunb was extracted with 70% methanol and concentrated under reduced pressure to measure its total polyphenol contents; furthermore, we examined the effect of electron donating ability of the various extracts. The extract was fractionated using different solvents such as methylene chloride, ethyl acetate, and methanol to investigate total polyphenol contents and electron donating abilities. The total polyphenol contents were $1,058.59{\pm}85.85{\mu}g\;GAE/mL$ in 70% methanol, $297.29{\pm}29.43{\mu}g\;GAE/mL$ in methylene chloride, $313.28{\pm}9.22{\mu}g\;GAE/mL$ in ethyl acetate, and $837.82{\pm}31.16{\mu}g\;GAE/mL$ in methanol. The total polyphenol contents showed significant differences (p<0.05) between the solvents. The electron donating abilities were $72.93{\pm}0.29%$ for 70% methanol extract, $14.02{\pm}2.01%$ for methylene chloride, $24.05{\pm}1.75%$ for ethyl acetate, and $84.08{\pm}1.95%$ for methanol. The electron donating abilities were significantly different (p<0.05) between the solvents. The two portions of Ilex serrata Thunb extracts obtained from methanol showed significantly relevant results in the total polyphenol contents and electron donating ability, which was higher than the original extract.

Relation of Dynamic Changes in Interfacial Tension to Protein Destabilization upon Emulsification

  • The objective of this study was to link conformational changes of proteins at a water/methylene chloride interface to their destabilization upon emulsification. When 4 aqueous protein solutions (bovine serum albumin, $\beta$-lactoglobulin, ovalbumin, or ribonuclease) were emulsified in methylene chloride, considerable proportions of all the proteins became water insoluble aggregates. There were also noticeable changes in the compositions of their water-soluble species. A series of water/methylene chloride interfacial reactions upon the proteins was considered a major cause of the phenomena observed. Based on this supposition, the interfacial tension was determined by a Kruss DVT-10 drop volume tensiometer under various experimental conditions. It substantiated that the interfacial tension was high enough to cause the adsorption of all the proteins. Under our experimental conditions, their presence in the aqueous phase resulted in reductions of the interfacial tension by the degrees of 8.5 - 17.1 mN $m^{-1}$. In addition, dynamic changes in the interfacial tension were monitored to compare relative rates at which the adsorbed proteins underwent conformational, structural rearrangements at the interface. Such information helped make a prediction about how easily proteins would denature and aggregate during emulsification. Our study indicated that emulsifying aqueous protein solutions in organic solvents should be handled with care, due to adverse interfacial effects.

더덕으로부터 면역세포 활성 성분의 분리 (Isolation of Active Components on Immunocytes from Codonopsis Lanceolatae)

  • The purpose of this research was to isolate an active component on immunoytes from 70% MeOH extract of Codonopsis lanceolatae Radix(CLE). CLE was fractionated successively with hexane, methylene chloride, n-butyl alcohol and water, and then the water fraction was separated with molecularporous membrane tubing(m.w. 3,500). Each fraction(50mg/kg) was administered p.o. once a day for 7 days in BALB/c mice respectively. None of these fractions affected the apoptosis and mitochondrial transmembrane potential in thymocyte. Hexane and methylene chloride fractions decreased CD4$^{[-10]}$ CD8$^{+}$ single-positive cells, and the water fraction enhanced CD4$^{+}$ CD8$^{[-10]}$ single-positive cells in thymocyte. The proliferation of thymocytes was decreased by the fraction hexane, but was enhanced by the water fraction. Hexane, methylene chloride and butyl alcohol fractions suppressed the production of nitric oxide, which was not affected by the water fraction. Hexane and butyl alcohol fraction suppressed the phagocytic activity, but water fraction enhanced it. The components(m.w. 3,500 above) separated from the water fraction enhanced the proliferation of thymocyte, the population of CD4$^{+}$ CDB$^{[-10]}$ single-positive cells, and phagocytic activity in macrophage. These results suggest that the stimulative components of proliferation, TH population and phagorytc activity is in the water fraction, and the molecular weight is 3,500 above. 쌀과 콩 중 규제잔류 농약의 동시분석에 관한 연구 (A Study on the Simultaneous Analysis of Regulated Pesticides Residues from Rice and Soy Bean)

  • 국내에서 현재 규제되고 있는 농약에 대해 신속, 간단한 동시분석법이 연구되었다. 시료는 아세톤과 메탄올로 추출하고 물을 가한 다음, $CH_2Cl_2$로 분배시킨다. 추출물은 Bio-Beads S-X3 컬럼에서 cyclohexane-methylene chloride의 용출액 11ml를 받아 불순물을 분리, 제거하고 ECD/NPD가 병렬되게 BP-1(methyl silicone) 모세관 컬럼에 연결된 기체 크로마토그래피로 분석한다. 농약 첨가에 의한 평균 회수율은 쌀에서 83% 이상이었고, 콩에서 3종 농약을 제외하면 81% 이상이었다. 검출한계는 쌀에서 0.002 ppm(${\alpha}$-BHC 등)~0.3 ppm(carbaryl 등) 이었다.

Methyleue blue가 X線에 照射된 흰쥐의 肝臟 glycogen, 血糖 및 血液內 電解質에 미치는 影響 (Influence of Methylene Blue on Liver Glycogen, Blood Glucose, and Electrolytes in Serum of Rats Exposed to X-rays)

  • 成熱한 albino 雄性 흰쥐를 對照群과 實驗群으로 나누어 對照群에는 0.9% 生理食鹽水를, 實驗群에는 methylene blue (40mg/kg, 食鹽水에 溶解, pH 7.0)를 各各 皮下注射處理하였다. 30 分後에 兩群共히 每分當 33r 으로 總線量 300r 의 X-線을 一時 全身照射하였다. 照射後 24日間에 걸쳐 肝臟 glycogen 含量(Carroll et al. 方法), 血糖含量(Folin and Wu 方法) 및 K 와 Na 含量(flame photometer)과 Cl 含量(automatic chloride titrator)을 各各 測定하여 methylene blue의 放射線에 對한 防禦 果를 硏究調査하였다. 一般的으로 肝臟 glycogen 含量은 兩群共히 X-線照射直後 若干 減少하나 1 日區에서부터 急激한 增加를 나타내고 있으며 特히 實驗群인 methylene blue 處理群에서는 對照群인 生理食鹽水群에 比하여 增加의 遲滯性을 나타내었다. 血糖含量에 對해서는 兩群모두 X-線 照射直後 若干 減少하는 傾向을 보였으나 時日經過에 따라서 서서히 增加하며 特히 實驗根인 methylene blue 處理群은 對照群인 生理食鹽水에 比하여 增加에 遲滯性을 나타내었다. 또한 血液內 電解質含量에 있어서 K, Na 및 Cl 含量은 一般的으로 兩群모두 時日經過에 따라 큰 變化는 없었으나 特히 K와 Na의 含量은 初期狀態에서 若干의 變化가 있었고 Cl 에서는 別變化를 觀察할 수 없는것으로보아 methylene blue가 血液內 電解質含量에는 別影響이 없었다. 以上의 結果로 미루어 보아 炭水化物代謝에 있어서 methylene blue가 放射線에 對한 防禦 果가 있는 것으로 思料된다.

  • DMSO-oxalyl chloride at low temperature in methylene chloride reacted with isolated secondary hydroxyl groups in some monosaccharides to give alkoxysulfonium salts, convertible to carbonyls in high yields upon addition of triethylamine. And 1, 2:5, 6-di-O-isopropylidene-.alpha.- D-allofuranose which is the key intermediate in the synthesis of 3-O-acetyl-5-O-benzoyl- 2-deoxy-2- fluoro-D-arabinofuranosyl bromide, was also obtained by oxidizing 1, 2:5, 6-di-O-isopropylidene-.alpha.- D-glucofuranose with the oxidizing reagent, followed by reduction with sodium borohydride.
