Antioxidant Activity of Methylene Chloride Extracts Obtained from Glucose-Ammonia (1M+8M) Browning Mixtures (Glucose-Ammonia (1M+8M) 갈색화(褐色化) 반응액(反應液)에서 얻어진 Methylene Chloride 추출물(抽出物)의 항산화(抗酸化) 효과(?果))

  • An attempt was made to investigate the antioxidant activity of methylene chloride extracts of a glucose-ammonia browning reaction mixture, which contain pyrazines, important intermediate prod ucts of Maillard-type browning reactions. Methylene chloride extracts were obtained from a glucose-ammonia(1M+8M) browning reaction mixture, which had been heated at $100^{\circ}C$. for 4 hours. The molar ratio of the reactants and the reaction time were reported to be the optimum ratio and time for the maximum formation of pyrazines. The methylene chloride extracts and furfural (for comparison purpose) were added to edible rapeseed oil substrates, and the resulting substrates and the control were stored at $37.0{\pm}1.0^{\circ}C$. Peroxide values (POV), thiobarbituric acid values (TBA-values) and acid values (AV) of the substrates and the control were determined regularly during a 34-day storage period. The antioxidant activity of the methylene chloride extracts and furfural was estimated on the basis of POV, TBA-value and AV-development of the substrates and the control. It was found that the methylene chloride extracts of the glucose-ammonia (1M+8M) browning mixture possessed considerable antioxidant activity. Furfural also showed some activity.

Improvement of Adsorbability of Methylene Blue on Bentonite Treated with Electrolyte Solution (Bentonite의 전각질수용액 처리에 의한 Methylene Blue 흡착성 개선)

  • Korean Yung-il bentonite was treated with potassium chloride, zinc chloride, calcium chloride, ferric chloride, or chromic chloride solutions respectively varying their concentration, treating temperature and treating time. The adsorbabilities of methylene blue on these pretreated bentonite were investigated. In the case of treatment with potassium chloride solution, the improvement of the adsorbahility of methylene blue on the products was observed, and in the best result the adsorbability was 1.6 times better than that on the original bentonite. With zinc chloride solution, the optimum adsorbability was a value of 1.7 times better than that on the original bentonite. With ferric chloride, chromic chloride or calcium chloride solution, slight improvement of the adsorbability was observed.

Relationship between Exposure Concentrations to Methylene Chloride in Air and Carboxyhemoglobin Levels in Blood of Workers Engaged in Blowing, and Cleaning Operations (발포 및 세척 공정 근로자의 공기중 Methylene Chloride 노출 농도와 혈중 Carboxyhemoglobin 수준간의 상관관계에 관한 연구)

  • The objectives of this study were to evaluate exposures to airborne methylene chloride and postshift carboxyhemoglobin (COHb) in blood of workers engaged in processes using blowing or cleaning agents, and to investigate correlation between methylene chloride concentrations and the blood COHb levels of workers. The geometric mean (GM) of workers' exposures (8 hour-time weighted averages, TWA) to airborne methylene chloride during cleaning molds using rags wetted with the solvent in the manufacture of flexible polyurethane foam (GM = 61.4 ppm), during operating the dip tank for cleaning molds in the manufacture of lens (GM = 61.0 ppm), and during cleaning the blowing nozzles by spraying the solvent in the manufacture of shoes (GM = 117.2 ppm) were exceeded the American Conference of Governmental Industrial Hygienists (ACGIH) Threshold Limit Value ($TLV^{(R)}$)-Time Weighted Average (TWA) (50 ppm). The COHb levels were significantly different among groups (p<0.05). The average COHh levels in blood of non-smoking workers were 2.0% in. low-level (<50 ppm) exposure group, and 3.9 % in high-level (>50ppm) exposure group. The average COHb levels in smoking workers were 3.1% in low-level exposure group, and 4.8% in high-level exposure group. The blood COHb levels of no-exposed workers to methylene chloride were 1.8% in non-smoking group, and 2.8% in smoking group. It was found that the COHb level dependeds on the methylene chloride concentration and smoking habit, and was highly correlated with methylene chloride concentration in air. The correlation coefficient was 0.81 among non-smoking workers. The estimated COHb level (3.6%) and 95% upper confidence limit (4.0%) corresponding to TLV-TWA of methylene chloride exceeded the current ACGIH Biological Exposure Index (COHb 3.5%) for carbon monoxide. The estimated COHb level (5.4%) at 100 ppm exceeded the standard (5%) recommended by National Institute for Occupational Safety and Health (NIOSH) for preventing adverse cardiovascular effect. The estimated COHb value and 95% upper confidence limit at 25 ppm of the Occupational Safety and Health. Adminstration (OSHA) Permissible Exposure Limit-TWA (PEL-TWA) were 2.6% and 3.0%, respectively. It is suggested that COHb in blood be kept below 3.0% to comply with OSHA PEL-TWA.

Removal of Residual Solvents in Paclitaxel by Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 Paclitaxel의 잔류용매 제거)

  • Because of casehardenign effect of amorphous paclitaxel, residual solvents, methylene chloride and emthanol could not be reduced to the maximum value allowed, 600 ppm and 3,000 ppm, in accord with the guidelines issued by the International Conference on Harmonization (ICH, 1997), using rotary evaporation and successive drying in a vacuum oven. However, methylene chloride and methanol were reduced to 486 ppm and 403 ppm, respectively using supercritical $CO_2$ on purified paclitaxel. The optimum pressure and operating time were 80 bar and 30 min at fixed operating temperature ($40^{circ}C$). This approach serves as a novel application of supercritical fluid extraction to remove residual solvents from active pharmaceutical ingredients.

A Study for Health Hazard Evaluation of Methylene Chloride Evaporated from the Tear Gas Mixture

  • This study explored the health hazard of those exposed to methylene chloride by assessing its atmospheric concentration when a tear gas mixture was aerially dispersed. The concentration of methylene chloride ranged from 311.1-980.3 ppm (geometric mean, 555.8 ppm), 30 seconds after the dispersion started. However, the concentration fell rapidly to below 10 ppm after dispersion was completed. The concentration during the dispersion did not surpass the National Institute for Occupational Safety and Health 'immediately dangerous to life or health' value of 2,300 ppm, but did exceed the American Conference of Governmental Industrial Hygienists excursion limit of 250 ppm. Since methylene chloride is highly volatile (vapor pressure, 349 mmHg at $20^{\circ}C$), the post-dispersion atmospheric concentration can rise instantaneously. Moreover, the o-chlorobenzylidenemalononitrile formulation of tear gas (CS gas) is an acute upper respiratory tract irritant. Therefore, tear gas mixtures should be handled with delicate care.

Field Application of an Alternative Cleaning Agent to Methylene Chloride (Methylene Chloride의 대체세정제 적용 사례 연구)

  • A study for replacing methylene chloride for an alternative cleaning agent which can be utilized for cleaning aluminium parts of hard disk drives was carried out at Suwon factory of Samsung Electro-mechanics Co. Ltd. As a result, quite a good cleaning agent was selected through data collection of alternative cleaning agents, their analysis of environment effects and safty factors and testing of their cleaning power. As compared with methylene chloride, the selected cleaning agent has better cleaning power. It is also more environmental-friendly, less toxic and more economical. And the operating cost reduction due to adoption of this alternative cleaning agent in Samsung Electro-mechanics Co. Ltd. was more than ₩l46,000,000 annually without any capital instrument in addition to improvement of environmental and safty conditions in the plant.

A Fatal Case of Methylene Chloride Poisoning (염화메틸렌 중독에 의한 사망 1례)

  • A 52-year-old man was found dead in the driver's seat of his tank lorry. The tank lorry was used to transport industrial wastewater to a disposal plant; the material was transferred into a storage tank with the help of compressed air. The wastewater contained methylene chloride and 2-chloropyridine. No respiratory protective equipment was used while working under these conditions. The autopsy report showed extensive edema and congestion of the brain, lung, and intraperitoneal organs. The concentrations of methylene chloride in lung and brain were reported at 398 and 67 mg/kg, respectively.

Anticancer Activity of Natural Products including Salvia miltiorrhiza (단삼 등 천연물의 항암작용)

  • The cellular growth inhibition of 20 natural products was screened using SRB (sulforhodamine B) assay against 4 human cancer cell lines(SNU-1, SNU-C$_{4}$, Hep3B, Kato III). Ethanol extracts of Salvia miltiorrhiza, Saussurea lappa and Chelidonium majus showed potent anticancer activity among them, and further, it was fractionated into methylene chloride, hexane and methanol. Methylene chloride and methanol fraction of Salvia radix showed significant inhibitory activity against 4 human cancer cell lines. The effect of Salvia miltiorrhiza on anticancer activity in vitro models was evaluated with methylene chloride fraction of Salvia miltiorrhiza. Life span of ICR mice implanted with sarcoma-180 was increased by 40-61% and BDF$^{1}$ mice implanted with L1210 was increased by 66-89% upon intraperitoneal administration with methylene chloride fraction of Salvia miltiorrhiza. Based on these result, we suggested that Salvia miltiorrhiza showed anticancer activity on the in vivo and in vitro models

  • The effect of methylene chloride/1,3-dioxolane co-solvent on the crystallization in the optical polycarbonate film was investigated. Increasing 1,3-dioxolane content in co-solvent resulted in the crystallization due to the lowering of solvent evaporation rate during film drying process. The crystallization in PC film could be minimized by either controlling of solvent composition and increasing solvent drying temperature. It was found that the surface roughness of solution casting PC film was affected by both crystallization and solvent evaporation rate. This morphological effect by alternative solvent is ascribed to a large decrease in light transmissivity on the optical PC film.

Antifungal Activity of Methylene Chloride Fraction of Pimpinella brachycarpa Against Aspergillus niger (참나물 Methylene Chloride 분획의 Aspergillus niger에 대한 항진균 활성)

  • In order to develop safe and economic novel antifungal agents, we prepared 73 methanol extracts from medicinal and edible herbs and examined their 365 solvent fractions using n-hexane, methylene chloride, ethylacetate, butanol and water residue based on the sequential organic solvent fraction method. When using the various fractions in the screening step for antifungal activity, we discovered ethylacetate fraction of Morus alba L., methylene chloride fraction of Pimpinella brachycarpa (MCPB), and n-hexane fraction of Salvia miltiorrhiza Bunge, which all have activities in methanol extracts, as potential sources of antifungal agents. Amongst these, the antifungal activity of P. brachycarpa has not to date been reported on. In addition, the mycelial growth inhibition and spore germination inhibition activities of MCPB against A. niger were confirmed by disc-diffusion assay in a 10 day culture. The MIC and MFC of MCPB were determined as 0.25 and 0.5 mg/ml, respectively. The MCPB has no hemolytic activity against human RBC at 0.5 mg/ml and glycoside-flavonoids are theorized to be active constituents. These results suggest that MCPB has a prominent antifungal activity and that the application of sequential organic solvent fractions, instead of simple natural product extracts, is useful in the screening process of novel bioactive substances.
