Methylene blue-catalyzed photodecomposition of guanine (Methylene Blue에 의한 guanine의 광분해 현상)

  • 1) The photodecomposition rate of guanine being catalized by methylene blue ws 53.7% in contrast with 9.3% that of dark control for 180 min. 2) In guanine control, the decomposition rate was very low. For 180 min., the rate was 8.1% in illuminated sample and 3.9% in non-illuminated sample. 3) The decomposition rate of methylene blue was obviously interfered by the existence of guanine. In guanine and methylene blue mixture solution, the net decomposition rate, excluding that of dark control ws 39.2% and in dye only solution, it was 48.5%.

Improvement of Adsorbability of Methylene Blue on Bentonite Treated with Electrolyte Solution (Bentonite의 전각질수용액 처리에 의한 Methylene Blue 흡착성 개선)

  • Korean Yung-il bentonite was treated with potassium chloride, zinc chloride, calcium chloride, ferric chloride, or chromic chloride solutions respectively varying their concentration, treating temperature and treating time. The adsorbabilities of methylene blue on these pretreated bentonite were investigated. In the case of treatment with potassium chloride solution, the improvement of the adsorbahility of methylene blue on the products was observed, and in the best result the adsorbability was 1.6 times better than that on the original bentonite. With zinc chloride solution, the optimum adsorbability was a value of 1.7 times better than that on the original bentonite. With ferric chloride, chromic chloride or calcium chloride solution, slight improvement of the adsorbability was observed.

  • A methylene blue-PVA system has been tested as a krad range dosimeter. Mb dye films were fabricated by casting PVA in solution with the methylene blue. In the air the system was irradiated by X-ray and the decolorisation of the dye film was found to remain unchanged for sufficiently long time. The radiation response on optical density at 670 nm in the Mb-PVA system shows a quite good linearity and reproducibility in the krad range.

  • Many gastrointestinal muscles show electrical oscillation, so-called 'slow wave', originated from interstitial cells of Cajal (ICCs). Thus, a technique to freshly isolate the cells is indispensable to explore the electrophysiological properties of the ICCs. To apply an enzyme solution on the serosal surface for cell isolation, the intestine was inverted and 0.02% trypsin solution and 0.04% collagenase solution were applied to serosal cavity. After the enzyme treatment, mucosal layer was removed and longitudinal muscle layer was gently separated from the rest of tissue. The thin layer was stretched in the recording chamber and mounted on an inverted microscope. Using ${\beta}-escine$, perforated whole cell patch clamp technique was used. Under a microscope, the tissue showed smooth muscle cells and interstitial cells around the myenteric plexus. Under voltage clamp condition, three types of membrane potential were recorded. One group of interstitial cells, which were positive to methylene blue and CD34, showed spontaneous outward current. These cells had bipolar shape and were considered as fibroblast-like cells because of their peculiar shape and arrangement. Another group, positive to c-kit and methylene blue, showed spontaneous inward current. These cells had more rounded shape and processes and were considered as ICCs. The third, positive to c-kit and had granules containing methylene blue, showed quiet membrane potentials under the voltage-clamp mode. These cells appeared to be resident macrophages. Therefore, in the freshly isolated thin tissue preparation, methylene blue could easily identify three types of cells rather than morphological properties. Using this method, we were able to study electrical properties of fibroblast and residential macrophage as well as myenteric ICCs.

  • The frost depth is one of important factors in the design of structures such as roadways, buried pipeline, and foundations. A frost indicator with methylene blue solution has several advantages with respect to installation cost, maintenance, and simple measurement. However, as a geotechnical engineering aspect, the accuracy of the frost indicator has not been proved yet. This paper presents experimental and field investigations of the accuracy of the frost indicator and contour maps of maximum frost depth. The contour maps of maximum frost depth can be applied to design geo-infrastructure in South Korea.

  • The binding isotherms of Methylene Blue with Poly(vinyl pyrrolidone) (PVP) were determined in a Mcllvaine buffer of pH 8.0 at 3$0^{\circ}C$ by a dynamic dialysis methods. The isotherms showed a partition binding which was increase linearly with the increase of free dye concentration in solution. The Scatchard plots for Poly(vinyl pyrrolidone)-Methylene Blue gave a constant value. The results were interpreted by the McGhee and von Hippel theory considering non-cooperative binding. The intrinsic binding constant k, for Poly(vinyl pyrrolidone)-Methylene Blue was 6.02$\ell$/base mol.

  • The electrochemical reduction of methylene blue (MB) in 1.0${\times}$10-2 M KNO3 aqueous solution was investigated by direct current (DC), differential pulse (DP) polarography, cyclic voltammetry (CV) and controlled potential coulometry (CPC). The electrode reduction of melthylene blue was processed CE reaction mechanism by two electrons transfer at the first reversible wave (- 0.18 volts vs. Ag/AgCl). MB was strongly adsorbed on the stationary mercury electrode and the reduction product of conptrolled potential electrolysis was rapidly auto-oxidized in air to the original methylene blue. Upon the basis of interpretation of cyclic voltammogram with pH change, possible CE electrode reaction mechanism was suggested.

  • This study was undertaken to examine the intensity of involvement of inducible nitric oxide synthase (iNOS) and cyclic GMP signal transduction pathway as one of the mechanisms of vaso-relaxative action of bacterial lipopolysaccharide (LPS) on the canine femoral artery strips. Canine femoral arteries were isolated and spiral strips of 10 mm long and 2 mm wide were made in the Tyrode solution of $0-4^{\circ}C$. The strips were prepared for isometric myography in Biancani's isolated muscle chamber containing 1 ml of Tyrode solution, which was maintained with pH 7.4 by aeration with 95% $O_2$/5% $CO_2$ at $37^{\circ}C$ and nitric oxide (NO) production was measured simulltaneously with isolated nitric oxide meter. LPS induced NO production, suppressed the phenylephrine (PE) induced contraction and enhanced the acetylcholine (ACh) induced relaxation. $N^G$-nitro-L-arginine methyl ester (L-NAME), an NOS inhibitor, methylene blue, a guanylyl cyclase inhibitor, potentiated PE induced contraction and suppressed ACh induced relaxation on the LPS treated strips. The inhibitory potency of methylene blue for LPS induced vascular hyporesponsiveness was stronger than that of L-NAME. These results suggest that in canine femoral artery, both iNOS and cyclic GMP signal trnasduction pathway are related with LPS induced vascular hyporeponsiveness, but in minor with iNOS and in major with cyclic GMP signal trnasduction pathway.

On Improvement of the Adsorption by Chemical Treatment on Yeong-Il Bentonite (영일산 Bentonite의 화학적 처리에 의한 흡착력 개선)

  • The adsorption of Methylene Blue on Yeong-Il bentonite which was treated by aqueous NaOH, $Na_2SO_4 or NaHSO_4 solution respectively, varying concentrations, temperature and time, was studied. In case of treatment with NaHSO_4 solution, slight improvement of the adsorption of Methylene Blue on bentonite was observed. With Na_2SO_4$ solution, the best results obtained when bentonite was treated with 1 N of the solution for 2hr at $100^{\circ}C$, and the adsorption capacity of the product was 3 times better than that of original bentonite. At the higher concentration and the higher temperature than above, faujasite was formed. With NaOH solution, the best condition was in 1N solution for 1hr at $100^{\circ}C$ and the adsorption capacity of the product was 3.3 times better than that of original bentonite. At the higher concentration of the treating agent and at the higher temperature than above, hydroxysodalite was formed.

Removal Characteristics of Crystal Violet and Methylene Blue from Aqueous Solution using Wood-based Activated Carbon (목질계 활성탄에 의한 수중의 Methylene blue와 Crystal violet의 제거 특성)

  • The adsorption ability of wood-based activated carbon to adsorb methylene blue (MB) and crystal violet (CV) from aqueous solution has been investigated. Adsorption studies were carried out on the batch experiment at different initial MB and CV concentrations (MB=150 mg/L~400 mg/L, CV=50 mg/L~350 mg/L), contact time, and temperature. The results showed that the MB and CV adsorption process followed the pseudo-second-order kinetic and intraparticle diffusion was the rate-limiting step. Adsorption equilibrium data of the adsorption process fitted very well to both Langmuir and Freundlich model. The maximum adsorption capacity ($q_m$) by Langmuir constant was 416.7 mg/g for MB and 462.4 mg/g for CV. The thermodynamic parameters such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$ and ${\Delta}G^{\circ}$ were evaluated. The MB and CV adsorption process was found to be endothermic for the two dyes.
