The Biochemical Characteristics of Acetobacter sp. Isolated from Persimmen Vinegar (감식초에서 분리한 Acetobacter sp.의 생화학적 특성)

  • The following is experimental result of Biochemical characteristics of Acetobacter sp. Acetobacter sp. were gram negative, short rod, non-spore-forming and motile. It reacted positively catalase, methyl red, oxidation fementation, voges-proskauer and nitrate reduction tests and hydrogen sulfide test negative and ONPG negative. Acetobacter sp. showed normal growth curve in Carr broth and there was no significant difference between isolates and type strains such as Acetobacter aceti(KCTC 1010), Acetobacter diazotrophicus(KCTC 2859). Acetobacter liquefaciens(KCTC 2804) and Acetobacter pasteurianus(KCTC 1008).

The Synthesis of Naphthothiazolo Carbocyanine Dye (Naphthothiazolo Carbocyanine 증감색소의 합성)

  • In this study, sensitizing dye was prepared by the reaction of 2-methyl-3-sulfopropyl-4, 5-naphthothiazolium(inner salt) with triethyl orthoformate in the presence of triethylamine. The product was identified by using various analytical tools such as Elemental analyzer, IR spectrophotometer, UV-Vis spectrophotometer, $^1H-NMR$ spectrometer, TGA and DSC. Therefore, it was concluded that naphthothiazolo carbocyanine dye can be used as red-sensitizing dye for the spectral sensitization of photographic emulsion.

Effects of Shikonin Pigments from the Roots of Lithospermum eryhrorhizon on Rabbit Platelets (자근으로부터 혈소판에 작용하는 천연색소물질에 관한 연구)

  • Lithospermum erythrorhyzon has been used as a red fooddye and traditional Chinese medicine to treat wounds, skin diseases and burns. Platelet activation plays an important role in thrombosis and haemostasis. Here, we studied the inhibition of platelet activation and its active compound from the root of Lithospermum erythrorhyzon. Its ethyl acetate extract inhibited the aggregation of washed rabbit platelets induced by collagen or thrombin. Five naphthoquinone pigments , shikonin , acetylshikonin , is obutylshikonin, $\alpha$-methyl-n-butylshikonin and $\beta$,$\beta$-dimethylacrylshikonin were isolated by means of high pressure liquid chromatography. The structures were determined by comparison of their proton nuclear magnetic resonance spectra. The potency of their inhibition was in the following order : $\beta$,$\beta$-dimethylacrylshikonin$\geq$$\alpha$-methyl-n-butylshikonin>isobutylshikonin>acetylshikonin>shikonin. It is suggested that the size of the aliphatic hydroxy group of shikonin is important for the enhancement of potency.

The effect of methyl yellow concentrations on the optical properties of polyvinyl chloride-methyl yellow film as a radiation dosimeter

  • A dosimetry system's ability to monitor absorbed doses is crucial for the quality assurance and control of products that have been exposed to radiation. Here, we propose a polyvinyl chloride (PVC)-methyl yellow (MY) film as a potential dosimeter for irradiation facilities. The PVC-MY film was prepared with a simple solvent-casting method at different MY concentrations from 0.3 to 1.2 mM. To evaluate the dosimeter's performance, the film was exposed to gamma-irradiation with irradiation doses of 0-25 kGy. The optical characteristics of the films examined using a UV-Vis spectrophotometer showed that the color of the films changed from yellow to red with increasing irradiation doses, and the color change sensitivity of the films increased with increasing MY concentrations. The absorption spectra of the irradiated PVC-MY films showed three maximum absorption peaks at 417, 522, and 547 nm. As irradiation doses increased, the film absorption at 417 nm decreased, while the film absorption at 522 and 547 nm increased. The film's stability studies suggest storage of the irradiated film in a cold-dark conditions is recommended to prevent damage and maintain response stability. The findings in this study emphasize the potential of the PVC-MY film as a dosimeter for irradiation facilities.

Changes in Volatile Flavor Compounds in Red Snow Crab Chionoecetes japonicus Cooker Effluent during Concentration (붉은 대게 가공부산물 농축중의 휘발성 향기성분 변화)

  • To develop natural crab-like flavorants from red snow crab Chionoecetes japonicus cooker effluent (RSCCE), the flavor was analyzed during the concentration of RSCCE up to $40^{\circ}Brix$. Using solid phase microextraction (SPME)/gas chromatography (GC)/mass selective detection (MSD), 30 volatile flavor compounds were detected in four RSCCE samples (10, 20, 30, and $40^{\circ}Brix$). These comprised 12 aromatic compounds, 5 N-containing compounds, 2 5-containing compounds, 2 alcohols, 2 aldehydes, and 7 miscellaneous compounds. The amounts of all volatiles except alcohols and aldehydes increased significantly with the concentration (p<0.05). Of the volatiles detected, the most abundant was a dimethyl trisulfide with an odor like onion/cooked cabbage. Of the N-containing compounds (nutty, roasted peanut-like odor), 2-ethyl-5-methylpyrazine was the most abundant, followed by 2,5-dimethylpyrazine and 2-methyl-5-isopropylpyrazine in that order (p<0.05). The N- and S-containing compounds with characteristic odors detected in this experiment are thought to play a positive role in RSCCE during concentration.

Inhibitory Effect of Kochujang Extracts on Chemically Induced Mutagenesis

  • Antimutagenic effects of 5 kinds of kochujang(Korean red pepper soybean paste) samples compared with doenjang(Korean soy paste) were studied using the Ames test with Salmonella typhimurium TA100 and the SOS chromotest, with E. Coli PQ37. Th eantimutatenic effects of methanol extracts from red pepper powder and meju(fermented soybean) powder, the major ingredients of the kochujang,were also evaluated for the mutagenicity of aflatoxin B1 (AFB1) in the Ames assay. The methanol extracts from the kochujang samples showed lower antimutagenicities than those of doenjang against AFB1 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the Ames assay. Traditional kochujang I and II exhibited strong antimutagenic activity against AFB1 and MNNG. The traditional kochujang samples against MNNG were aslo observed in the SOS chromotest system with the same fashions as shown in the Ames mutagenicity test. The methanol extracts from meju powder had the strongest inhibitory effects on mutabenicity induced by AFB1, however, those form red pepper powder showed lower inhibition rate than kochujang. These results suggest that traditional kochujang exhibit higher antimutagenic acitivity than the commercial variety, and that meju powder seems to be one of the major antimutagenic components in kochujang.

Effects of Irradiation and Fumigation on Physicochemical Properties of Red Pepper during Storage (감마선과 훈증처리가 건고추의 저장 중 이화학적 특성에 미치는 영향)

  • In order to study the quarantine and sanitization methods for dried red pepper, comparative effects of commercial fumigation (methyl bromide/MeBr, $phosphine gas/PH_{3}$ and gamma irradiation (5, 10 kGy) were investigated in terms of its physicochemical properties. There were no noticeable chances in pH and soluble solids among the untreated control, irradiated and fumigated samples soon after treatments, but some decrease was found in stored samples (especially soluble solid in fumigated samples) for 8 months under room temperature. Total sugar content was influenced by storage time rather than both treatments. Immediately after treatments, reducing sugar content was significantly reduced in the samples including pericarp when exposed to fumigants (p<0.05), while an apparent decrease was observed in the stored samples including seeds with negligible differences among treatment groups. The electron donating ability (EDA) of the extracts was high in the order of pericarp, whole pepper, powdered pepper and seeds, which was reduced during storage for 8 months particularly in the samples containing seeds. The EDA of irradiated samples during storage was equal to that of the control sample, whereas that of fumigated samples was relatively low (p<0.05).

White Organic Light-emitting Diodes using the Tandem Structure Incorporating with Organic p/n Junction

  • Efficient white organic light-emitting diodes are fabricated with the blue and red electroluminescent (EL) units electrically connected in a stacked tandem structure by using a transparent doped organic p/n junction. The blue and red EL units consist of the light-emitting layer of 1,4-bis(2,2-diphenyl vinyl)benzene (DPVBi) and 4-dicyanomethylene-2-methyl-6-[2-(2,3,6,7-tetrahydro-1H,5H-benzo[i,j] quinolizin-8-yl)vinyl]-4H-pyran) (DCM2) doped tris(8-hydroxyquinoline) aluminum $(Alq_3)$, respectively. The organic p-n junction consists of ${\alpha}-NPD$ doped with $FeCl_3$ (15 % by weight ratio) and $Alq_3$ doped with Li (10 %). The EL spectra exhibit two peaks at 448 and 606 nm, resulting in white light-emission with the Commission Internationale d'Eclairage (CIE) chromaticity coordinates of (0.36, 0.24). The tandem device shows the quantum efficiency of about 2.2 % at a luminance of 100 $cd/m^2$, higher than individual blue and red EL devices.

Extraction of pesticide residues in medical herbs by microwave (Microwave를 이용한 한약재 중의 잔류농약 추출)

  • This study was conducted to elucidate extraction efficiency by microwave in comparison with Soxhlet for extraction of pesticide residues in dried medical herbs; red-ginseng, white-ginseng, Bupleuri Radix, Angelica gigas Nakai, Rehmannia glutinosa. The acetone extraction by microwave of tolclofos-methyl and quintozene in medical herbs was efficient. The extraction efficiency by microwave with power 45 to 150 watts, extraction time 1 to 5 minutes and solvent volume 30 ml was compared with that of Soxhlet with extraction time 7 hours and solvent volume 150 ml. The extraction efficiency by microwave with extraction time 3 to 5 minutes was similar with extraction time of 7 hours by Soxhlet. When medical herbs spiked with tolclofos-methyl and quintozene was analyzed to how the extraction efficiency of microwave by kind of medical herbs, the extraction efficiency by microwave with extraction time of 3 to 5 minutes was the same as Soxhlet extraction. The optimal condition for extraction of tolclofos-methyl and quintozene in medical herbs by microwave was 45 to 90 watts of power supply, 3 to 5 minutes of extraction time and acetone 30 ml of solvent volume.
