Celecoxib, a COX-2 Selective Inhibitor, Induces Cell Cycle Arrest at the G2/M Phase in HeLa Cervical Cancer Cells

  • Celecoxib, a selective inhibitor of COX-2, showed cytotoxic effects in many cancer cell lines including cervical cancer cells. This study investigated the effect of celecoxib on cell cycle arrest in HeLa cervical cancer cells through p53 expression. In vitro anticancer activity was determined with the 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) method. A double staining method was applied to investigate the mechanism of cell death, cell cycling was analyzed by flow cytometryand immunocytochemistry was employed to stain p53 expression in cells. Celecoxib showed strong cytotoxic effects and induced apoptosis with an $IC_{50}$ value of $40{\mu}M$. It induced cell cycle arrest at G2/M phase by increasing level of p53 expression on HeLa cells.

Model based Facial Expression Recognition using New Feature Space (새로운 얼굴 특징공간을 이용한 모델 기반 얼굴 표정 인식)

  • This paper introduces a new model based method for facial expression recognition that uses facial grid angles as feature space. In order to be able to recognize the six main facial expression, proposed method uses a grid approach and therefore it establishes a new feature space based on the angles that each gird's edge and vertex form. The way taken in the paper is robust against several affine transformations such as translation, rotation, and scaling which in other approaches are considered very harmful in the overall accuracy of a facial expression recognition algorithm. Also, this paper demonstrates the process that the feature space is created using angles and how a selection process of feature subset within this space is applied with Wrapper approach. Selected features are classified by SVM, 3-NN classifier and classification results are validated with two-tier cross validation. Proposed method shows 94% classification result and feature selection algorithm improves results by up to 10% over the full set of feature.

  • Nowadays, many robots have evolved to imitate human social skills such that sociable interaction with humans is possible. Socially interactive robots require abilities different from that of conventional robots. For instance, human-robot interactions are accompanied by emotion similar to human-human interactions. Robot emotional expression is thus very important for humans. This is particularly true for facial expressions, which play an important role in communication amongst other non-verbal forms. In this paper, we introduce a method of creating lifelike facial expressions in robots using variation of affect values which consist of the robot's emotions based on emotional boundaries. The proposed method was examined by experiments of two facial robot simulators.

Self-expression Behavior of Smartphone Through In-depth Interview: Focused on Younger Generation Users

  • Today's young generation users are very active in expressing themselves through smart phones, form networks in this cyber space and express their identities freely. Therefore, this study aims to investigate how young people in their 20s are expressing themselves through smart phones and the differences from the attempt of self-expression in the existing cyber space. The research method was an in-depth interview, which was a qualitative research method, aimed at people in their 20s, so that they could freely talk about their thoughts and feelings. In-depth interviews are useful for users' psychological and in-depth analysis of behavior.

  • In this paper, we propose a method to manage production system easily for operators when either equipments or products are changed. And we explain the scheduling AI tool which realizes the proposal method. The tool's knowledge expression consists of models, rules, mathematical expression and fuzzy logic. The model expresses the relations between products and manufacture, and properties of products. The models are separated into three type, equipment model, operation model, and product model. These models are classified by applicable fields as the assembly process or continuous plant process, The model expression of each type is based on object oriented paradigm. We report systems utilizing our approach.

Vibrations of wind-turbines considering soil-structure interaction

  • Wind turbine structures are long slender columns with a rotor and blade assembly placed on the top. These slender structures vibrate due to dynamic environmental forces and its own dynamics. Analysis of the dynamic behavior of wind turbines is fundamental to the stability, performance, operation and safety of these systems. In this paper a simplied approach is outlined for free vibration analysis of these long, slender structures taking the soil-structure interaction into account. The analytical method is based on an Euler-Bernoulli beam-column with elastic end supports. The elastic end-supports are considered to model the flexible nature of the interaction of these systems with soil. A closed-form approximate expression has been derived for the first natural frequency of the system. This new expression is a function of geometric and elastic properties of wind turbine tower and properties of the foundation including soil. The proposed simple expression has been independently validated using an exact numerical method, laboratory based experimental measurement and field measurement of a real wind turbine structure. The results obtained in the paper shows that the proposed expression can be used for a quick assessment of the fundamental frequency of a wind turbine taking the soil-structure interaction into account.

Effects of Kaempferol on Lippolysaccharide-induced Inflammation in Mouse Brain (Kaempferol이 LPS로 유도된 생쥐 중추신경계 염증에 미치는 영향)

  • Objectives : Brain inflammation early activates the microglia and activated microglia secrete a variety of pro-inflammatory cytokines. Kaempferol, which is a flavonoid in Cuscutae Semen, shows a wide range of physiological activities, including neurons protection and anti-inflammatory actions through inhibition of pro-inflammatory mediators. The present study examined the modulatory effect of kaempferol on cytokines [tumor necrosis factor- alpha ($TNF-{\alpha}$), interleukin-1beta ($IL-1{\beta}$) and interleukin-6 (IL-6)] and cyclooxygenase-2 (COX-2) mRNA expression and microglia activation in the brain tissue of the mouse. Methods : Kaempferol was administered orally three doses of 10, 20 and 30 mg/kg respectively, once 1 hour before the lippolysaccharide(LPS) (3 mg/kg, i.p.) injection. Brain tissue was removed at 4 hours after LPS injection. Cytokines and COX-2 mRNA expression in the brain tissue was measured by the quantitative real-time polymerase chain reaction (PCR) method. Iba1 expression was calculated by western blotting method. Microglia was observed with immunohistochemistry. Immunohistochemistry stained microglia was analyzed by using ImageJ software. Results : Kaempferol 20 and 30 mg/kg was significantly attenuated the expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 mRNA. Kaempfrol 10, 20 and 30 mg/kg significantly attenuated COX-2 mRNA expression in the brain tissue. Kaempferol 30 mg/kg significantly suppressed the increase of Iba1 protein expression by LPS. Kaempferol 30 mg/kg significantly decreased the number of microglia in the cerebral cortex and the number and cell size of microglia in the hypothalamic region and the area percentage of ionized calcium binding adaptor molecule 1(Iba1)-expressed microglia in the hippocampus. Conclusions : This results indicate that kaempferol plays an anti-inflammatory role in the brain.

Concentration Range Analysis for Fluorescence Expression of Indocyanine Green (Indocyanine green 형광조영제의 형광발현을 위한 농도 범위 분석)

  • In the characteristic of the brain malignant, the blood vessels and tumors have the same color and shape, and the boundary distinction is not clear, Therefore, it is difficult to observe the naked eye. Because of the high invasiveness, the risk of recurrence is high. Therefore, complete resection of the tumor is essential. The method for distinguishing the boundary between blood vessels and tumors is a fluorescence contrast method using indocyanine green (ICG), a fluorescence contrast agent. In ICG, the concentration range analysis is very important because the fluorescence expression state varies depending on the concentration. However, since the analysis result of the fluorescence expression condition is insufficient according to the current concentration, this paper proposes by analyzing the initial protocol of the concentration range. 780 nm infrared light was irradiated to the ICG sample to observe the fluorescence expression through a near infrared (NIR) camera. The wavelength is measured by using a spectrum instrument (ocean view) to observe the fluorescence expression wavelength of 811nm. As a result of analyzing the mol concentration according to each sample, the fluorescence expression range was found to be 16.15-0.16M and the optimum fluorescence concentration on the brightest part was found to be 3.23-0.81M.

Automatic 3D Facial Movement Detection from Mirror-reflected Multi-Image for Facial Expression Modeling (거울 투영 이미지를 이용한 3D 얼굴 표정 변화 자동 검출 및 모델링)

  • This thesis presents a method for 3D modeling of facial expression from frontal and mirror-reflected multi-image. Since the proposed system uses only one camera, two mirrors, and simple mirror's property, it is robust, accurate and inexpensive. In addition, we can avoid the problem of synchronization between data among different cameras. Mirrors located near one's cheeks can reflect the side views of markers on one's face. To optimize our system, we must select feature points of face intimately associated with human's emotions. Therefore we refer to the FDP (Facial Definition Parameters) and FAP (Facial Animation Parameters) defined by MPEG-4 SNHC (Synlhetic/Natural Hybrid Coding). We put colorful dot markers on selected feature points of face to detect movement of facial deformation when subject makes variety expressions. Before computing the 3D coordinates of extracted facial feature points, we properly grouped these points according to relative part. This makes our matching process automatically. We experiment on about twenty koreans the subject of our experiment in their late twenties and early thirties. Finally, we verify the performance of the proposed method tv simulating an animation of 3D facial expression.

A Research of the Architectural Embodiment Design-Method of Won-Buddhism Doctrine (원불교 교리의 현대적 건축 구현 방법 연구)

  • Each human era has its own unique architectural embodiment of religious philosophy at the time. Though, many modern religions have developed their own architectural expression based upon each own religious doctrine, one of the main native Korean religions, Won-Buddhism, has not developed its own proper architectural expression yet. So, the goal of this research is to find the probable architectural design methods for Won-Buddhism doctrines. 1) The history of Won-Buddhism architecture is comparatively analyzed to find the architectural trend of Won-Buddhism. 2) Modern religious architectural cases are analysed and compared between plastic expression and spatial expression types. 3) Won-Buddhism doctrines are analysed to find key characteristics to be used as architectural design concepts and method. Based upon analyses, the main characteristics of Won-Buddhism doctrine are synchronicity, circularity, and liminality, which are found to be similar to the trans-boundary characteristics of contemporary architecture. In the future, more study on these characteristics and how they are materialized, the probable design methodology for Won-Buddhism Doctrines can be proposed.
