Biochemieal Effect on Methionine Metabolism to the Expperimental Rats on Rice Diet (백미사과(白米飼科)가 실험용백서(實驗用白鼠)의 Methionine대사(代謝)에 미치는 영향(影饗)에 대(對)한 생물화학적연구(生物化學的硏究))

  • Using the $Methionine-2-C^{14}$, the metabolism of methionine to the experimental rats on rice diet was studied comparing with that to the rats on stock diet in this paper. The National Institute of Health strain of weaning albino rats were housed into the individual cages deviding into 2 groups, the rice diet (RD) group and the stock diet (SD) group, and fed on rice diet and stock diet respectively for 10 weeks. On the day of experiment, the rats were parenterally administered. the $methionine-2-C^{14}$ solution after fasting over night. And then the rats were sacrificed by ether anesthesia by time being of one, three, six, and twelve hours each and the organs, pituitary gland, pancreas, spleen, liver, and kidney, were taken out for the determinations of radioactivities. And also the excretion of radioactivities through urine were determined by time being. The radioactivities were determined by Autoscaler SC-51 using the planchets. The results of radioactivities of urine excretion were shown at table 3 and the results of radioactivities distibutions in the organs by time being after parenteral administration of $methionine-2-C^{14}$ were shown table 4 in the original paper. According to the results, the following are summarized; 1. The growth experiment result of rats on. rice diet and stock diet were same as shown by the previous workers indicating significant growth inhibition at the rice diet group. 2. Due to the result of radioactivity excretion through urine after administration of $methionine-2-C^{14}$, it might he considered that methionine in the rice diet seems to be limited. However, it seems to be not 주 mostly limited. 3. And due to the results of radioactivity distribution in the organs by time being, the radioactivity in the liver tissue showed appearently higher readings at this methionine study compared with the results at the lysine study shown by HAW and his co-worker. This might be interpreted, though it is not clear, that liver might require methionine as a deficient amino acid at the tissue because methionine is limited at the rice diet.

Effects of Dietary Casein, Soy, and Methionine-Supplemented Soy on Serum Lipids Level in Rats

  • The objective of the current study was to determine the influences of dietary proteins and methionine on plasma lipid concentrations. Thirty growing male Sprague-Dawley rats were fed diets similar in all respects except that dietary protein was from either casein, soy protein isolate, or soy protein isolate supplemented with L-methionine (0.24 %). The animals were fed experimental diets ad libitum for nine weeks. Plasma total-cholesterol concentrations were unaffected by the protein source or methionine supplementation. Plasma triglyceride concentrations were lower in rats of methionine supplemented soy protein diets (76 mg/dL) than in the rats fed casein or soy diet (120 mg/dL, 109 mg/dL, respectively). These results indicate that soy protein reduces plasma triglycerides relative to casein in rats fed cholesterol free diets, and that methionine-supplemented soy diets decrease plasma triglyceride concentrations more than soy protein alone.

  • In the view of the fact that data are scare concerning the change of Methionine composition of poultry meat in function of storage temperature and cooking method. The present study was carried out to change in these compounds and the results were summarized as follows. 1. The result of measuring methionine compound of raw material stored under the condition of 5$^{\circ}C$, 1$0^{\circ}C$ for 6 weeks were increased as temperature increases. 2. The result of change in methionine compound of roasting were decreased than raw material It was greatly decreased in the storage for 42 days at 1$0^{\circ}C$ and slightly decreased in the storage for 14 days at 5$^{\circ}C$ 3. When the boiling in the water, methionine compound was decreased than roasting to compare with raw material 4. By the results of this study, one of the best cooking method is roasting, because of the change in methionine compound of roasting is slightly decrease.

  • The buccal mucosa was observed histochemically in male rats which were injected with methionine. The staining methods were proceed by means of periodicacid-Schiff reaction, toluidine blue stain, alloxan-Schiff reaction, Mallory's aniline blue stain, Lillie's modification of Bielschowsky method and hematoxylin-eosin stain. The results of the experiment were summarized as follows: 1) PAS reactions of basement membrane and lamina propria increased after 3 and 5 days of methionine administration. 2) Metachromasia of stratum spinosum increased after 3 days of methionine administration. 3) Alloxan-Schiff reactions of stratum granulosum and stratum spinosum increased after 7 days of methionine administration. 4) In the lamina propria, aniline blue staining of collagenous fibers increased after 7, 10 and 14 days of methionine administration.

  • This study was conducted to increase the content of taurine in milk from the dairy cows fed the rumen protected DL-methionine matrix and methionie hydroxy analog matrix for 42days. Dairy cows fed rumen protected DL-methionine matrix showed 23% increased milk taurine concentration compared to the value of the control groups. Dairy cows fed rumen protected methionine hydroxy analog matrix showed 53% increased milk taurine concentration compared to the value for the control groups (p<0.01).

  • The preventive effect of Sosiho-Tang extract administered concurrently with methionine on hepatotoxicity induced by carbon tetrachoride (20% $CCl_4$ in olive oil, 5 ml/kg) in rats was studied. S-GOT values were decreased significantly by the concurrent administration of Sosiho-Tang extract 100 mg/kg with methionine 100 mg/kg and also Sosiho-Tang 200 mg/kg with methionine 100 mg/kg. S-GPT values were decreased remarkably by the concurrent administration of Sosiho-Tang extract 200 mg/kg with methionine 100 mg/kg. In histological observations, the pattern of the contrast group indicated the marked necrosis of the parenchymatous cells around the centrilobular area and the fatty changes of the hepatic cells around the midzonal area, but the concurrent administration of Sosiho-Tang extract 200 mg/kg and methionine 100 mg/kg revealed better improvements than their respective single administration in all pathological aspects.

The Effects of Different Methionine and Lysine Levels in. 15% Iso-protein Diet on the Performance of Laying Hens (동일한 단백질 수준의 사료에서 Methionine과 Lysine수준이 산란계의 생산성에 미치는 영향)

  • The purpose of this study was to investigate the effects of dietary methionine and lysine levels on laying hen performance. The level of protein was fixed 15% during whole experiment period, but the levels of methionine and lysine were 0.30% and 0.58% (Low), 0.32% and 0.64% (Medium), 0.35% and 0 70% (High), respectively. Total 288 laying pullets of 22 weeks age were reared from January 28, 1989 to March 23, 1990 for 60weeks. The results obtained were summarized as follows : 1. The e99 Productions were highest in medium treatment in phase I (22~42weeks of age), phase II (42~62 weeks of age) and phase III (62~82weeks of age) and especially, there was significant difference among treatments during phase II (P<0.05). 2. Egg weight was significantly increased as the levels of methionine and lysine were increased up to methionine and lysine were 0.32% and 0.64%, respectively(P<0.01). 3. Daily egg mass was highest when the levels of methionine and lysine were 0.32% and 0.64%, respectively and there were significant differences among treatments during phase I and phase II (P<0.01) 4. Daily feed intake was increased as the levels of methionine and lysine were increased, and there was significant difference among treatments during phase III (P<0.05). 5. Feed efficiency was best in medium treatment in phase I and phase II (P<0.01) 6, Viability was highest in medium treatment, but there was no significant difference among treatments. 7. Nutrient utilizability of experimental diets was not significantly different among treatments. 8. Eviscerated yield was highest and abdominal fat accumulation was lowest in medium treatment, but there was no significant difference among treatments. 9. Egg shell quality and chemical composition of egg content were not different among treatments. 10. The feed cost per kg egg mass was lowest in medium treatment and there were significant differences among treatments in phase I, phase II and whole egg laying period(P<0.05)

  • This research is to examine the detoxication effect of methionine on cadmium intoxication For this purpose, this paper provides an analysis of the data on the groups of mice (ICR), one group of mice treated with 40 ppm of cadmium only. and other groups of mice combined-treated with cadmium and 0.1%, 0.25%, 0.5% and 1% methionine. After breeding for 40 days, the data on the growth of mice, changes in activation of alkaline phosphatase in blood, and the cadmium content in the liver and kidney, are analysed. The results were as follows: 1) The growth-rate of mice, in the cadmium only injected group, was declined by 9% in comparison with the control group after 40 days. But the two groups of cadmium with 0.5% and 1% methionine showed the rise of 9% ana 14% respectively above the growth-rate of the control group. The results from the groups of cadmium with 0.1% and 0.25%, methionine were similar to that from the cadmium only injected group. 3) Changes in activation of alkaline phosphatase in blood decreased to 86.45% in the cadmium only injected group behind the 100% activation of the control group. The groups of cadmium with 0.1% and 0.25% methionine showed no difference with the former group. But the groups of cadmium with 0.5% and 1% methionine recovered to the 93.14% and 96.08% of activation respectively. 3) The mean content of cadmium in the liver was $0.028{\pm}0.001{\mu}g/g$ in the control group. The cadmium only injected group showed the mean cadmium content of $2.80{\pm}0.62{\mu}g/g$ in the liver, which was similar to $2.82{\pm}1.03{\mu}g/g$ in the group of cadmium with 0.1% methionine, and $2.56{\pm}0.77{\mu}g/g$ in the group of cadmium with 0.25% methionine. But the groups of cadmium with 0.5% and 1% methionine showed the reduction of cadmium contents in the liver to $1.84{\pm}0.56{\mu}g/g$ and $1.74{\pm}0.35{\mu}g/g$ respectively. In the kidney, the groups of cadmium with 0.1%, 0.25% and 0.5% methionine shelved the similar cadmium content to the group treated with cadmium only. But the group of cadmium with 1% methionine showed a small increase to $4.13{\pm}1.00{\mu}g/g$ in comparison with the group treated with cadmium only. This analysis proves that the mobility and diffusion of cadmium in tile tissues advance faster ill the group treated with cadmium and methionine than in the group treated with cadmium only.

  • Cephalosporium acremonium MAR-80, a strain of methionine analogue-resistant mutant, showed good activity of sulfate utilization as only sulfur source. The effect of methionine on the sulfate uptake system was investigated by using $Na_{2}^{35}SO_{4}$ as a tracer in the resting cell system. From this result, it was revealed that sulfate permease of this strain was less repressed and/or less inhibited by methionine than parent type. This deregulation was due to low actibity of methionine uptake, which was operated by somewhat simple diffusion. From these studies, it could by anticipated that the improved productivity of cephalosporin C and lower dependence of cephalosporin C production on methionine were related to increased uptake rate of sulfate.

Disruption of the metC Gene Affects Methionine Biosynthesis in Pectobacterium carotovorum subsp. carotovorum Pcc21 and Reduces Soft-Rot Disease

  • Plant pathogenic Pectobacterium species cause severe soft rot/blackleg diseases in many economically important crops worldwide. Pectobacterium utilizes plant cell wall degrading enzymes (PCWDEs) as the main virulence determinants for its pathogenicity. In this study, we screened a random mutant, M29 is a transposon insertion mutation in the metC gene encoding cystathionine β-lyase that catalyzes cystathionine to homocysteine at the penultimate step in methionine biosynthesis. M29 became a methionine auxotroph and resulted in growth defects in methionine-limited conditions. Impaired growth was restored with exogenous methionine or homocysteine rather than cystathionine. The mutant exhibited reduced soft rot symptoms in Chinese cabbages and potato tubers, maintaining activities of PCWDEs and swimming motility. The mutant was unable to proliferate in both Chinese cabbages and potato tubers. The reduced virulence was partially restored by a complemented strain or 100 µM of methionine, whereas it was fully restored by the extremely high concentration (1 mM). Our transcriptomic analysis showed that genes involved in methionine biosynthesis or transporter were downregulated in the mutant. Our results demonstrate that MetC is important for methionine biosynthesis and transporter and influences its virulence through Pcc21 multiplication in plant hosts.
