한국산(韓國産) 녹두(綠豆)의 성숙과정중(成熟過程中) 유리(遊離) Amino Acid 함량(含量) 변화(變化)에 관(關)한 연구(硏究) (A Study on the Changes of Free Amino Acid Composition in Seeds of Korean Mung Bean During the Ripening Process)

  • 한국에서 생산된 녹두의 성숙 중에 아미노산의 변화를 구명하기 위하여 개화 후 10일, 15일 20 일, 25일과 30일의 5단계로 취하여 분석한 결과를 요약하면 다음과 같다. 1) 초기에 검출된 아미노산은 histidine, arginine, cystine, aspartic acid, threonine(Serine 포함), glutamic acid와 valine이었고 methionine sulfoxide와 methionine sulfone도 나타났다. 2) 15일에는 초기의 것에 glycine, isoleucine, tyrosine, phenylalanine과 methionine의 5종(種)이 더 검출되었으며, methionine sulfoxide와 methionine sulfone의 양이 감소하면서 methionine이 나타났다. 3) 중기에 leucine이 나타났고 성숙하면서 약간의 증가를 보였고 25일 이후에는 methionine sulfoxide와 methionine sulfone이 검출되지 않는 대신 methionine 앙이 증가하였다. 4) 개화 20일 이후의 아미노산은 완숙기까지 계속 존재하였고 증가된 아미노산은 leucine, valine, isoleucine과 methionine이었으며, 그 외 대부분은 감소 또는 그대로 남았다.

Partition of Amino Acids Requirement for Maintenance and Growth of Broilers II. Methionine

  • Purified diets containing five levels of methionine with 0.4% cystine were fed to growing chicks (8 days old male Arbor Acre strain) to evaluate methionine requirements for growth and maintenance. A model was developed to separate methionine requirement for maintenance from requirement for growth. From this model the daily methionine requirement for growth was 4.22 mg/g gain, and the daily methionine requirement for maintenance was 0.034 times metabolic body size ($W^{0.75}$). Based on nitrogen gain response, the methionine requirement for growth was 0.162 mg/mg N gain, and the daily maintenance requirement was 0.037 times metabolic body size. The plateau of plasma methionine concentration reached at 117.16 mg intake pre day. The total methionine requirement determined based on weight gain response was 138.29 mg/day or 0.33% of the diet and the one determined based on nitrogen gain response was 141.7 mg/day of 0.34% of the diet, respectively. As a percentage of protein, methionine was calculated to be 2.6%; the reported methionine content of carcass CP was 1.76%.

  • This study was performed to investigate the effects of different kinds of dietary protein [plant protein ; Isolated Soy Protein(ISP), animal protein ; casein] on protein and lipid metabolism in rats fed with coffee and/or methionine in diet during four weeks of growing period after weanling. Forty male growing rats fo Sprague-Dawley strain, weighing 92.5$\pm$1.8g, were distributed into 8 groups by randomized complete block design, and fed diets containing 15% of protein by weight either as ISP or casein and 10% ofcalories as corn oil, supplemented with coffee and /or methionine for 4 weeks. Coffee were added at a concentration of 1.4% of diet as instant coffee, and methionine were added to ISP or casein diet to be 0.6% of diet as DL-methionine. Results were followed ; Body weight gain, F.E. R and P.E.R tanded to be higher in methionine added groups than non-methionine groups. The nitrogen content of feces was significantly higher in coffee groups than non-coffee groups, and tended to be higher in ISP groups than casein groups. but was not significantly different with or without methionine. thus, apparent protein digestilbity was significantly lower in coffee groups than non-coffee groups and was significantly lower in ISP groups than cesein groups, but was not significantly different with or without methionine. Total cholesterol content of serum tended to be higher in coffee groups than non-coffee groups, and tended to be lower in methionine groups than non-methionine groups.

대두단백질(大豆蛋白質)에 DL-Methionine 의 보충(補充)이 흰쥐의 성장(成長), 체내(體內) 대사(代謝) 및 체조성(體組成)에 미치는 영향(影響) (Effect of Isolated Soyprotein Supplemented with DL- Methionine on the Growth, Metabolism and Body Composition in Albino Rats)

  • The effect of methionine supplementation to the isolated soyprotein(ISP) diet on the growth, body metabolism and composition of the Albino male rats was studied. Three levels(0.3,0.6 and 0.9%) of methionine were supplemented to the ISP diet with the constant levels of energy and protein of 3,600 kcal/kg and 20%, respectively. The body weight and weight gain of the growing rats were significantly increased by 0.3% methionine supplementation to the ISP diet compared to the ISP diet(P< 0.05).The effects of methionine supplementation to the ISP diet tended to be larger with increasing of the level of methionine supplementation, 0.6 and 0.9%, were statistically insignificant. Food and gross energy intake of growing rats fed the ISP diet or the ISP supplemented with methionine diet were lower than those fed the casein diet(P< 0.05). FER and PER of all the methionine supplemented diets were higher than those of the ISP or casein diet (P< 0.05) without significant differences among the supplementation levels of methionine to the diets. The weight gain of adult rats fed 0.9% methionine supplemented ISP diet were higher than those of the other treatments with significant difference. The effects of methionine supplementation to the ISP diet on the protein digestibility, BV, NPU, N-balance, N-retention, and body and liver compositions were not significant.

두유(豆乳)에 첨가된 Methionine이 유산균의 생육과 산생성에 미치는 영향 (Effects of Methionine Supplemented to Soy Milk on Growth and Acid Production by Lactic Acid Bacteria)

  • 본 연구는 농축대두단백으로 만든 두유에 L-methionine을 여러가지 농도로 첨가하여 6종의 유산균 (Lactobacillus 4종, Leuconostoc 1종, Streptococcus 1종)의 생육과 산생성에 미치는 영향을 조사하고 아울러 DL-methionine, D-methionine 또는 L-cysteine이 L. acidophilus에 미치는 영향을 조사한 것으로서 그 결과를 요약하면 다음과 같다. 두유에 첨가된 L-methionine은 6종의 유산균 가운데 4종 (L. bulgaricus, L. casei, Leuconostoc mesenteroides, S. lactis)의 생육과 산생성에 거의 영향을 미치지 않았으나 L. acidophilus 2종의 산생성을 억제시켰으며, 같은 L. acidophilus 중에서도 KFCC 12731은 AKU 1122보다 더 큰 영향을 받는 것으로 나타났다. 두유에 첨가된 DL-methionine은 L-methionine과 동일하게 L. acidophilus(KFCC 12731)의 산생성을 현저하게 억제시켰으나, D-methionine은 높은 농도에서도 생육과 산생성에 전혀 영향을 미치지 않았다. 한편 L-cysteine에 의하여 L. acidophilus (KFCC 12731)의 산생성이 다소 억제되었으나 그 정도는 L-methionine이 비하여 매우 완만한 것이었다.

Effects of Dietary Methionine Levels on Choline Requirements of Starter White Pekin Ducks

  • A $2{\times}5$ factorial experiment, using 2 dietary methionine levels (0.28% and 0.48%) and 5 dietary choline levels (0, 394, 823, 1,239, and 1,743 mg/kg), was conducted to study the effects of dietary methionine status on choline requirements of starter white Pekin ducks from 7 to 28 days of age. Four hundred eighty 7-d-old male White Pekin ducks were randomly allotted to ten dietary treatments, each containing 6 replicate pens with 8 birds per pen. At 28 d of age, weight gain, feed intake, and feed/gain were measured and the legs of all ducks from each pen were examined for incidence of perosis. Perosis and growth depression were observed in choline-deficient ducks and supplementation of choline reduced perosis and significantly increased weight gain and feed intake regardless of dietary methionine levels (p<0.05). In addition, significant positive effects of dietary methionine supplementation on weight gain, feed intake, and feed/gain were observed at any choline level (p<0.05). Supplementation of 1,743 mg/kg choline in diets alleviated the depression of weight gain and feed intake caused by methionine deficiency at 0.28% methionine level. The interaction between choline and methionine influenced weight gain and feed intake of ducks (p<0.05). At 0.28% methionine level, 1,743 mg/kg choline group caused 4.92% and 3.23% amount of improvement in weight gain and feed intake compared with 1,239 mg/kg choline group, respectively. According to the broken-line regression, the choline requirements of starter Pekin ducks for weight gain and feed intake were 1,472 and 1,424 mg/kg at 0.28% methionine level and 946 and 907 mg/kg at 0.48% methionine level, respectively. It suggested the choline recommendations of starter Pekin ducks on a semi-purified diet were 1448 mg/kg at 0.28% methionine level and 927 mg/kg at 0.48% methionine level, respectively. Compared with the adequate methionine level, menthionine deficiency markedly increased the choline requirements of ducks.

The Optimum Methionine to Methionine Plus Cystine Ratio for Growing Pigs Determined Using Plasma Urea Nitrogen and Nitrogen Balance

  • The objective of this study was to determine the optimum ratio of methionine to methionine plus cystine for growing pigs. A nitrogen balance trial was conducted using a total of 21 barrows (Large WhiteLandrace) over two replicates. The initial body weight was $20.36{\pm}1.22kg$ (mean${\pm}$SD) in the first replicate and $23.54{\pm}1.02kg$ (mean${\pm}$SD) in the second. For each replicate, the 21 pigs were randomly assigned to one of seven dietary treatments with three observations per treatment. The diets included a methionine and cystine-deficient basal diet with all other essential nutrients meeting nutrient requirements and six diets formulated with graded levels of DL-methionine (0.00, 0.03, 0.06, 0.10, 0.13, 0.16%) and $L-Cystine{\cdot}HCl{\cdot}H_2O$ (0.19, 0.15, 0.11, 0.07, 0.04, 0.00%). This resulted in ratios of methionine to methionine plus cystine of 41.3, 29.6, 35.3, 41.2, 46.0, 51.6 and 57.5%. Each experimental period lasted 12 days consisting of a seven-day adaptation period followed by a five-day total collection of urine and feces. During the collection period, pigs were fed 900 g/day for the first replicate and 1,200 g/day for the second replicate. The feed was provided in three equal portions at 0800, 1500, and 2200 h daily. Pigs had ad libitum access to water after feeding. There was a linear (p<0.01) and quadratic (p<0.01) effect on daily gain and feed conversion as the ratio of methionine to methionine plus cystine increased. Pigs receiving the diets providing a methionine to methionine plus cystine ratio of 51.6% had the best daily gain and feed conversion. Plasma urea nitrogen was also lowest for this treatment. Nitrogen retention increased (p<0.01) as the relative proportion of methionine increased up to 51.6% and then a downward trend occurred at 57.5%. The quadratic regression model, as well as one- and two- slope regression line models, were used to determine the optimum ratio of methionine to methionine plus cystine. Eliminating the 35.3% methionine to methionine plus cystine treatment resulted in $R^2$ values in excess of 0.92. The optimal ratio of methionine to methionine plus cystine was estimated to be 54.15% for nitrogen retention and 56.72% for plasma urea nitrogen.

유지의 산화에 대한 Methionine과 Lysine의 항산화 효과 (Antioxidative Effect of Methionine and Lysine on Oil Rancidity)

  • 대두유와 라아드의 항온저장 및 가열시 methionine과 lysine의 첨가농도에 따른 항산화 효과 및 기존 항산화제들의 항산화력에 대한 상승효과를 비교 연구하였다. 유지의 항온(60$\pm$2$^{\circ}C$) 저장시 methionine과 lysine을 각 농도별로 첨가한 경우 모든 시료에서 항산화 효과를 보였으며 그 중에서 methionine을 1% 첨가하였을 때 가장 효과가 좋았고 그 정도는 TBHQ를 첨가한 경우와 유사하였으며 tocopherol 보다 월등히 높은 항산화 효과를 보였다. 그리고 이들의 항산화 효과는 농도증가에 따라 비례적으로 증가하였으나 농도간에 큰 차이는 없었다. 즉 이들의 항산화 효과는 TBHQ>methionine 1%>methione 0.1%> methionine 0.02%> lysine 1%>ly-sine 0.1%>Lysine 0.02%>tocopherol 순이었다. 또한 아미노산, TBHQ, tocopherol 모두 식물성유 보다 동물성유지에서 더 큰 항산화력을 나타내었다. 유지 가열(180$\pm$2$^{\circ}C$)시 methionine과 lysine을 각 농도별로 첨가한 모든 시료에서 항산화 효과가 나타났고 TBHQ는 자동사놔시와는 달리 항산화 효과가 크게 떨어졌으며 이들의 항산화 효과의 크기 순서는 methione 1%>methionine 0.1%>methionine 0.02%> lysine 1%>lysine 0.1%>lysine 0.02%>tocopherol, TBHQ 순이었다. Methionine 및 lysine을 tocopherol, ascorbic acid, cit-ric acid와 혼합하여 대두유에 첨가하였을 때 tocoperol 과의 혼합물이 가장 높은 상승효과를 나타내었으며 asco-rbic acid 및 citric acid와의 혼합물도 약하지만 상승효과를 나타내었다. 따라서 methionine과 lysine은 유지에 대하여 항온저장 또는 가열시 모두 항산화 효과가 있으며 특히 가열 시와 동물성 유지에 대하여 그 효과가 우수하고 식품 항산화제에 대한 상승효과도 크다는 것을 알 수 있었다.

  • To study the effects of dietry methionine level on lipid peroxidation of rats, rats were fed vitamin E, selenium and methionine - deficient diet or the same diet supplemented with various levels(0.3%, 0.6%, 0.9%) of methionine for 6 weeks. The biochemincal and mophological changes in the rat liver were investigated. Lipid peroxide levels in plasma and hepatic mitochondrial fraction of MF rats were more increased than those of control rats. However, supplementation with 0.6% methionine modified this increment. Catalase activity was decreased in hepatic mitochondrial fraction from rats fed MF diet. Methionine supplementation did not induce this enzyme. The ultrastructural evidence for lipid peroxidation was found in plasma membranes facing sinusoids. The most striking changes in including disruption and loss of microvilli and development of numerous lipid droplets occurred in rats fed MF diet. These changes were not effectively prevented by the same diet supplemented with 0.3% or 0.9% methionine, but supplementation with 0.6% methionine modulated more or less the changes.

취장 및 간장에 미치는 식이성(食餌性) Ethionine 과 Methionine 의 독성에 관한 연구 (Studies on the Toxicity of Dietary Ethionine and Methionine to Pancreas and Liver)

  • It has been demonstrated repeatedly that the administration of ethionine, a methionine analogue, will produce destruction of pancreatic tissue and liver cells. Destruction of acinar cells of pancreas by the administration of excess methionine similar to that seen after feeding diets supplemented with ethionine was also reported, but the liver was not involved by this amino acid. In an attempt to reproduce the results of these investigators the tissue damages were found to be slight and seen only irregulary in rats receiving ethionine on a complete diet and the result of tissue damages by excess methionine were also controversial. The present studies describe the toxicity of dietary supplemented ethionine and methionine to liver and pancreas in rats fed a low protein diet. Hundred five albino rats weighing around 120 gm were divided into three groups as follows; 1) Control group: A low protein diet containing 8% casein was fed throughout the experimental period. 2) Methionine group: A low protein diet (7% casein) supplemented with 1% methionine was used. 3) Ethionine group: A similar diet as methionine group except the supplementation of 1% ethionine instead of methionine was used. Five animals per wee from each group were killed for 6 weeks. The liver and pancreas were fixed in 10% formalin and histologic sections were prepared and stained with hematoxylin eosin. Serum amylase was expressed as much of glucose liberated from a starch substrate. The glucose was determined by the method of Nelson (1944). Serum glutamic pyruvic transaminase (SGPT) and glutamic oxaloacetic transaminase (SGOT) were determined by the technique described in Sigma Bulletin.
