Functional Properties of Korean Atractylodes japonica Koidz (한국산 삽주의 기능성)

  • To study the availability of the Korean Atractylodes japonica Koidz. as ingredients for functional flood, functional properties of solvent extracts were investigated and the results were followed. Yield was 14.8% by ethanol extraction of fresh Korean A. Japonica and 17.7% by water fraction. Acetone extract and butanol fraction showed stronger activity of the hydrogen donating activities, each of 72.9% and 74.2%, respectively, in fresh Korean A. japonica and methanol extract and butanol fraction showed stronger activity of the nitrite scavenging effects, each of 95.0% and 79.2%. in fresh Korean A. japonica. Among the solvent extacts from fresh Korean a. japonica, extract by methanol showed strong antimicrobial activity in which clear zone showed 20 mm for Bacillus subtilis and 19 mm for Pseudomonas aeruginosa. Butanol fraction derived from methanol extract showed moderate antimicrobial activity : 18 mm clear zone for Bacillus subtilis and Vibrio parahaemolyticus. Minimum inhibitory concentrations of methanol extract and butanol fraction were about 2 mg/disc and 4 mg/disc against gram(+) bacteria and 6 mg/disc against gram(-) bacteria, respectively.

Recovery Yields of Protopectinase Depending on Treatments of Organic Solvents (유기용매의 처리에 따른 Bacillus subtillis IFO 12113 유래 Protopectinase의 회수)

  • To recover protopectinase (PPase) secreted from Bacillus subtilis IFO 12113, culture filtrate of the microorganism was treated with acetone, methanol, and ethanol, respectively. In the case of treatment with acetone at a ratio of 1: 1 (culture filtrate: acetone, v/v), PPase was purified 1.7-fold with 59.2% recovery The recovery of PPase was increased by increasing the acetone concentration. PPase was purified 4-fold with 100% recovery when the culture filtrate was precipitated with methanol at a ratio of 1 : 2 (culture filtrate: methanol, v/v). However, recovery of PPase was decreased by increasing the methanol concentration. PPase was purified 13.5-fold resulting in 68% recovery by the addition of ethanol with the final ratio 1 : 1(culture filtrate: ethanol, v/v) to the supernatant, which was obtained after precipitation of the culture filtrate with ethanol at a ratio of 1 : 0.5. These results show that methanol treatment is better than other organic solvent treatments for the simple recovery of PPase, whereas fractionated treatment of ethanol can recover PPase with higher purification fold.

Extraction of Triterpenoid Saponin (glycyrrhizin) from Liquorice by Co-solvent Modified Supercritical Carbon Dioxide (보조용매로 변형된 초임계 이산화탄소에 의한 감초의 triterpenoid saponin(glycyrrhizin)의 추출)

  • Effects of modifier and soaking on extraction of triterpenoid saponin (glycyrrhizin) from liquorice were examined using supercritical $CO_2(SC-CO_2)$ at 50 MPa, $60^{\circ}C$, and flow rate of 3 mL/min, and glycyrrhizin content was analyzed by HPLC. Additon of undiluted methanol, ethanol or isopropanol as modifier to $SC-CO_2$ had little influence on extraction yield of glycyrrhizin. Soaking process using water increased the extraction yield as the sample to solvent ratio was increased. Addition of 70% methanol, ethanol or isopropanol to $SC-CO_2$ significantly increased the extraction yields, with 70% methanol resulting in the highest yield. When water at 90% (w/w) of sample weight was used for soaking, the extraction yield and rate increased, 70% ethanol-modified $SC-CO_2$ was almost equal to that obtained using 70% methanol.

  • The N hyperfine coupling constants ($a_N$) of di-t-butyl nitroxide (DTBN) radicals in water-solvent system were measured with EPR spectroscopy. Various kinds of the solvents with different polarity such as acetone, dimethylsulfoxide (DMSO), methanol, ethanol and 1-propanol were applied and studied. Equilibrium constants for the solvation equilibrium and the solvent parameters ($E_T$, molar transition energy) of various water-solvent system were obtained from the experimental results and are presented. The $a_N$ values were plotted as a function of mole fraction of the solvent. In case of water-DMSO, water-ethanol and water-1-propanol system, slight negative deviations from the straight line were observed. In water-acetone system, the absorption wavelength (${\lambda}$) due to ${\eta}{\rightarrow}{\pi}^{\ast}$ transition increased linearly with the increase of mole fraction of acetone. The relationship between $a_N$ of DTBN and ${\lambda}$ due to ${\eta}{\rightarrow}{\pi}^{\ast}$ transition in water-acetone and water-DMSO system was examined. It was found that the electronic structure of the nitroxide radicals is stablized from the fact that the N hyperfine coupling constants of DTBN radicals are greatly unaffected in the environment of water-solvent system.

Antioxidant Activities of Various Solvent Extracts from Canola Meal (추출용매별 canola meal의 항산화 활성)

  • This study was performed to investigate antioxidant activity of various solvent extracts (methanol, ethanol, acetone and water) from canola meal. As the concentration of all extracts increased, antioxidant activities increased gradually. Among solvent extracts from canola meal, methanol extract showed the highest antioxidant activities with $EC_{50}$ values of 0.9, 3.5, 1.3, and 8.5 mg/mL for DPPH radical scavenging ability, ABTS radical scavenging ability, reducing power, and chelating ability, respectively, and it had the highest total phenolics and total flavonoids contents. Furthermore, methanol extract was fractionated into three fractions (MF 1, MF 2, and MF 3) using Sephadex LH-20 column. The highest antioxidant activity was found on MF 2 for DPPH radical scavenging ability, on MF 3 for ABTS radical scavenging ability and reducing power, and on MF 1 for chelating ability at the same concentration of 1.5 mg/mL. The results indicated that each fraction containing various antioxidant materials seemed to have different influence at each antioxidant activity.

Optimization for Chia Seed Antioxidative Activity of Solvent Extraction Using the Response Surface Methodology (반응표면 분석법을 이용한 치아씨 항산화 활성 추출의 최적화 조건)

  • The purpose of this study was to determine the optimum conditions of solvent extraction type and solvent concentration (60, 70, 80%, v/v), extraction time (30, 80, 130 mins) and extraction temperature (10, 15, $20^{\circ}C$) in order to increase the antioxidant activity of the chia seed. The total polyphenol content and DPPH radical scavenging activity was measured by using response surface methodology (RSM) to establish the optimal conditions. Using ethanol and methanol extractions at extraction concentrations of 60%, and time and temperature of 130 mins and $20^{\circ}C$, the maximum total polyphenol content was 871.00 mg% ($R^2=0.9507$) and 557.70 mg% ($R^2=0.9784$) for ethanol and methanol extraction respectively. Using the same extraction conditions, the maximum level of DPPH radical scavenging activity was 72.14% ($R^2=0.9675$) and 52.79% ($R^2=0.9524$) for ethanol and methanol extraction respectively. The results indicate that ethanol extracts showed a higher antioxidant activity than methanol extracts. The ethanol extraction conditions of response surface analysis (RSA) were affected more by ethanol concentration than by extraction time or temperature. In contrast, the methanol extraction conditions of response surface analysis (RSA) were affected more by extraction time. Based on the RSM, the optimum ethanol extraction conditions were the following: extraction concentration, 63%: time, 100 mins: and temperature, $18^{\circ}C$. The optimum methanol extraction conditions were the following: extraction concentration, 65%; time, 120 mins; and temperature, $16^{\circ}C$.

Purification of Isoflavone from Soybean Hypocotyls using Various Resins

  • Isoflavone was extracted with various concentration of aqueous methanol using whole hypocotyls as the starting material. Whole hypocotyls were preferred as the raw material because the residue could be easily removed from the solvent after the extraction process. Extraction yield was almost constant at the methanol concentration of $20-80\%$. Most of the isoflavone was extracted within 1 hr, and the extraction yield remained almost constant thereafter. When the concentration of methanol was $80\%$, the content of total solid was reduced due to the reduced extraction of contaminating protein as the result of protein insolubilization. Among resins tested, Diaion HP-20, Amberlite XAD-16, and Amberlite IRC-50 showed the highest capacity to absorb the compound. Open column chromatography with Diaion HP-20 showed that $80\%$ aqueous ethanol was most efficient as the eluting solvent with final recovery of the phytochemical being more than $95\%$. Maximum adsorption of the phytochemical occurred at the acidic pH 2-4. When the spatial velocity was increased to 15 and more, the degree of adsorption was decreased, whereas below spatial velocity of 15, the adsorption capacity of isoflavone to the resin was almost constant. The purity of the isoflavone purified by column chromatography was $78\%$.

  • Poly(ethylene terephthalate)(PET) filaments were treated in binary mixtures of organic solvents such as benzyl alcohol/perchloroethylene(BA/PER), benzyl alcohol/trichloroethylene(BA/TRI), benzyl alcohol/ethylene chloride(BA/EC), methanol/perchloroethylene (ME/PER), and methanol/trichloroethylene(ME/TRI). From the equilibrium swelling of PET, total and partial isotherms of PET for sorption of solvent mixtures and isotherms for distribution of the components between the phases were taken. Also the shrinkage and the crystallinity of PET treated with binary mixtures were investigated. The results obtained were summerized as follows: 1. All isotherms for distribution of the components between the phase deviated from the diagonal in system of PET-binary mixtures. Especially in the binary mixtures of ME/PER or ME/TRI, selectivo sorption of chlorinated hydrocarbon PER and TRI by PET occured. 2. The shrinkage of PET treated in binary mixrures was increased to compare with single solvent-treated, but the composition of binary mixtures corresponding to maximum values of shrinkage was not always agreed with the composition of binary mixtures corresponding to maximum values of shrinkage was not always agreed with the composition of binary mixtures exhibiting of the maximum swelling of PET. 3. The crystallinity of PET treated in binary mixtures generally increased than that of single solvent-treated.

The Grunwald-Winstein Relationship in the Solvolysis of β-Substituted Chloroformate Ester Derivatives: The Solvolysis of 2-Phenylethyl and 2,2-Diphenylethyl Chloroformates

  • Solvolysis rate constants of 2-phenylethyl-(2-$PhCH_2CH_2OCOCl$, 1) and 2,2-diphenylethyl chloroformate (2,2-$Ph_2CHCH_2OCOCl$, 2), together with the previously studied solvolyses of ${\alpha}$- and ${\beta}$-substituted chloroformate ester derivatives, are reported in pure and binary solvents at $40.0^{\circ}C$. The linear free energy relationship (LFER) and sensitivities (l and m) to changes in solvent nucleophilicity ($N_T$) and solvent ionizing power ($Y_{Cl}$) of the solvolytic reactions are analyzed using the Grunwald-Winstein equation. The kinetic solvent isotope effects (KSIEs) in methanol and activation parameter values in various solvents are investigated for 1 and 2. These results support well the bimolecular pathway with same aspects. Furthermore, the small negative values of the entropies of activation of solvolysis of 1 and 2 in the highly ionizing aqueous fluoroalcohols are consistent with the ionization character of the rate-determining step, and the KSIE values of 1.78 and 2.10 in methanol-d indicate that one molecule of solvent acts as a nucleophile and the other acts as a general-base catalyst. It is found that the ${\beta}$-substituents in alkyl chloroformate are not the important factor to decide the solvolysis reaction pathway.

Solvent effect on enzymatic steroid transformation

  • As part of our endeavor to increase the productivity of steroid by enzymatic transformation of corticosteroids, attempts have been made to increase the solubility of steroids by using some organic solvents. When the solubility of steroids is the rate limiting factor in the steroid transformation, it was found that the use of solvents significantly improved the yield. Hydrocorisone as a substrate and 3-ketosteroid .DELTA.$^{1}$ dehydrogense as an immobilized whole cell enzyme were employed as the model system for this study. It was found that the yield of product, prodnisolone, goes through a maximum with an increase in the solvent concentration. At a high solvent concentration, the solvent showed a toxic effect and it causes a decrease in the product yield by the second order inhibition mechanism. Among the solvents evaluated, methanol and ethanol were found to be the best. These alcohols are not only good solvents but also showed minimal toxic effect. Based on the experimental results, it was concluded that the productivity of steroid can be increased by usign well selcted solvents systems for the enzymatic transformation of steroids.
