Enzyme Activities Related to the Methanol Oxidation of Mycobacterium sp. strain JCl DSM 3803

  • Mycobacterium sp. strain JCl DSM 3803 grown in methanol showed no methanol dehydrogenase or oxidase activities found in mast methylotrophic bacteria and yeasts, respectively. Even though the methanol-grown cells exhibited a little methanol-dependent oxidation by cytochrome c-dependent methanol dehydrogenase and alcohol dehydrogenase, they were not the key enzymes responsible for the methanol oxidation of the cells, in that the cells contained no c-type cytochrome and the methanol oxidizing activity from the partially purified alcohol dehydrogenase was too low, respectively. In substrate switching experiments, we found that only a catalase-peroxidase among the three types of catalase found in glucose-grown cells was highly expressed, in the methanol-grown cells and that its activity was relatively high during the exponential growth phase in Mycobacterium sp. JCl. Therefore, we propose that catalase-peroxidase is an essential enzyme responsible for the methanol metabolism directly Of indirectly in Mycobacterium sp. JCl.

Electro-catalytic Performance of PtRu Catalysts Supported on Urea-treated MWNTs for Methanol Oxidation

  • In this work, nitrogen and oxygen functionalities was introduced to the graphite nanofibers (GNFs) and their effect on electrocatalytic performance of the GNF supports for direct methanol fuel cells (DMFCs) was invesigated. The nitrogen and oxygen groups were introduced through the urea treatments and acid treatment, respectively. And, PtRu catalysts deposited on modified GNFs were prepared by a chemical reduction method. The catalysts were characterized by means of elemental analysis, nitrogen adsorption, and X-ray photoelectron spetroscopy (XPS). The structure and morphological characteristics of the catalysts were characterized by X-ray diffraction (XRD) and transmission electron microscopy (TEM). As a result, the Pt-Ru nanoparticles were impregnated on GNFs with good formation in 3-5 nm. And, the cyclic voltammograms for methanol oxidation revealed that the methanol oxidation peak varied depending on changes of surface functional groups. It was thus considered that the PtRu deposition was related to the reduction of PtRu and surface characteristics of the carbon supports. The changes of surface functional groups were related to PtRu reduction, significantly affect the methanol oxidation activity of anode electrocatalysts in DMFCs.

  • 메탄은 풍부하고 재생 가능한 탄화수소이지만, 온실가스로서 지구 온난화를 발생시킨다. 따라서 메탄을 유용한 화학물질이나 에너지원으로의 변환이 필요하다. 메탄올은 메탄의 부분 산화 반응을 통해 합성할 수 있는 간단하고 풍부한화학물질이다. 메탄올은 화학 공급 원료나 수송 연료로 사용될 뿐만 아니라, 저온 연료 전지의 연료로도 적합하다. 그러나 메탄올의 전기화학 산화는 복잡하고 다단계의 반응이므로, 이 반응을 이해하고 최적화하기 위해서는 새로운 전기화학촉매와 반응 메커니즘의 연구가 필요하다. 본 총설에서는 메탄올 산화 반응 메커니즘 및 최근 연구 동향과 향후 연구 방향을 고찰하였다.

다공성 탄소나노섬유 지지체에 담지된 백금촉매의 메탄올 산화 특성 연구 (Methanol Electro-Oxidation Properties of Pt Electro-Catalysts Embedded by Porous Carbon Nanofiber Supports)

  • To improve the methanol electro-oxidation in direct methanol fuel cells(DMFCs), Pt electrocatalysts embedded on porous carbon nanofibers(CNFs) were synthesized by electrospinning followed by a reduction method. To fabricate the porous CNFs, we prepared three types of porous CNFs using three different amount of a styrene-co-acrylonitrile(SAN) polymer: 0.2 wt%, 0.5 wt%, and 1 wt%, respectively. A SAN polymer, which provides vacant spaces in porous CNFs, was decomposed and burn out during the carbonization. The structure and morphology of the samples were examined using field emission scanning electron microscopy and transmission electron microscopy and their surface area were measured using the Brunauer-Emmett-Teller(BET). The crystallinities and chemical compositions of the samples were examined using X-ray diffraction and X-ray photoelectron spectroscopy. The electrochemical properties on the methanol electro-oxidation were characterized using cyclic voltammetry and chronoamperometry. Pt electrocatalysts embedded on porous CNFs containing 0.5 wt% SAN polymer exhibited the improved methanol oxidation and electrocatalytic stability compared to Pt/conventional CNFs and commercial Pt/C(40 wt% Pt on Vulcan carbon, E-TEK).

대두유의 자동산화 및 가열산화에 미치는 정향추출물의 항산화 효과 (Effects of Clove Extracts on the Autoxidation and Thermal Oxidation of Soybean Oil)

  • This study was conducted to investigate the antioxidant activities of clove extracts in water, methanol and ether. The clove extracts, BHA and ${\alpha}$-tocopherol were added to each oil at a level of 200 ppm. The activities of the substrate oils and controls were tested under autoxidation and thermal oxidation conditions. The degree of the effects of the antioxidant activities under autoxidation condition were in the following order; ether extract 〉 methanol extract 〉BHA 〉 ${\alpha}$-tocopherol 〉 water extract = control group. The induction periods of the control, water, methanol and ether extracts, and BHA and ${\alpha}$-tocopherol were 9.5, 9.6, 10.7 11.8, 10.4 and 9.7 days, respectively. Under thermal oxidation condition, the methanol extract showed stronger antioxidant activity than those of the water and ether extracts. The antioxidant activity of the methanol extract was attributed to ${\alpha}$-tocopherol and BHA.

부하(溥荷)의 항산화효능(抗酸化效能)에 대한 연구 (I) (A Study on the Anti-Oxidation Effects of Menthae Herba (I))

  • Objectives : In order to develop effective and safe anti-oxidation, the effects of Menthae Herba (M. spicata L. and M piperita L.) were examined. Methods : M. spicata L. and M. piperita L. of Menthae Herba were used to make water extracts and methanol extracts, and then its anti-oxidative effects were compared in vitro. Results : 1. M. spicata L. was stronger than M. piperita L. in anti-oxidative effects of methanol extracts, and water extracts showed similar anti-oxidation effects. 2. Water extracts of M. spicata L. was stronger than M. piperita L. showed a stronger superoxide anit-oxidation than the same-density ascorbic acid, so that water extracts were proved to be stronger than methanol extracts. 3. Methanol extracts' DPPH anti-oxidation was similar in M. spicata L. and M piperita L.. As for water extracts, M. spicata L. showed higher anti-oxidation and M. piperita L. had no anti-oxidation. 4. As for anti-oxidation of LDL and linoleic acid of methanol extracts, both M. spicata L. and M. piperita L. were stronger than a-tocopherol. As for the water extracts, these two showed similar level of anti-oxidative effect on linoleic acid, which is weaker that mehtanol extracts. 5. M. spicata L. includes more phenol than M. piperita L. Conclusions : Menthae Herba (M. piperita L. M. spicata L.) had anti-oxidation effect, so that it can be medically developed for anti-oxidation.

메탄의 균일 및 접촉부분산화에 의한 메탄올 합성 (Homogeneous and Catalytic Methanol Synthesis by Partial Oxidation of Methane)

  • Methanol was synthesized by homogeneous and catalytic reactions of partial oxidation of methane. The effect of pressure, temperature and oxygen concentration on methanol synthesis was investigated. The catalyst used was Bi-Cs-Mg-Cu-Mo mixed oxide. The partial oxidation reaction was carried out in a fixed bed reactor at 20${\sim}$46 bar and $450{\sim}480^{\circ}C$ and oxygen concentration of 5.3${\sim}$7.7mol%. The results were compared with results of homogeneous reaction performed at the same conditions. Methane conversions of the homogeneous and catalytic reactions increased with temperature. Methanol selectivity of the homogeneous reaction decreased with increasing temperature. However, the methanol selectivity of catalytic reaction increased with temperature. For both homogeneous and catalytic reactions, the methane conversions were around 5%. This may be due to the low oxygen concentration. Methanol selectivity of the catalytic reaction was higher than that of homogeneous one.

  • The methanol partial oxidation using commercial $CuO/ZnO/Al_2O_3$ catalysts in a plug flow reactor was studied in the temperature range of $200{\sim}250^{\circ}C$ at atmospheric pressure, It was achieved the high activities by Cu-based catalysts and the selectivity of $CO_2$/$H_2$ was 100% when $O_2$ was fully convened. The reactivity changes and their hysteresis with increasing/decreasing temperatures were observed due to the chemical state differences between the oxidation and the reduction on the Cu surface, It was suggested as the two-step reaction: the complete oxidation and the following steam reforming for methanol, which was indicated by the distributions of final products vs. the residence time. In addition, the complete oxidation step was shown to be extremely fast and the total reaction rate can be controlled by the steam reforming reaction.

  • To prevent or reduce air pollutant from methanol fueled vehicles, methanol oxidation reaction was carried out using a heteropoly acid catalysts. Catalytic activities of catalysts have been experimented at atmospheric pressure in a fixed bed flow reactor. Catalysts were characterized by XRD, IR, thermal analysis, N $H_{3}$-TPD and GC pulse technique. Acidities of catalysts were highly affected by poly-atoms. Methanol conversion was much higher on catalyst with W than on catalyst with Mo as a poly-atoms. With the increase of copper content(X) in C $u_{x}$ $H_{{3-2x}}$PMo catalyst, acidity was decreased and oxidation ability was increased. Methanol conversion and product distribution were affected by the acidity and oxidation ability of catalyst. Especially, supported PdSiW(1wt%) catalyst has a very good methanol conversion and C $O_{2}$ selectivity as high as a commertial 3-way catalyst.t.
