  • Transporters are membrane proteins that mediate the transfer of substrate across the cellular membrane. In this overview, the characteristics and the toxicological relevance were discussed for various types of transporters. For drug transporters, the overview focused on ATP-binding cassette transporters and solute carrier family 21A/22A member transporters. Except for OCTN transporters and OATP transporters, drug transporters tend to have broad substrate specificity, suggesting drug-drug interaction at the level of transport processes (e.g., interaction between methotrexate and non-steroidal anti-inflammatory agents) is likely. For metal transporters, transporters for zinc, copper and multiple metals were discussed in this overview. These metal transporters have comparatively narrow substrate specificity, except for multiple metal transporters, suggesting that inter-substrate interaction at the level of transport is less likely. In contrast, the expressions of the transporters are often regulated by their substrates, suggesting cellular adaptation mechanism exists for these transporters. The drug-drug interactions in drug transporters and the cellular adaptation mechanisms for metal transporters are likely to lead to alterations in pharmacokinetics and cellular metal homeostasis, which may be linked to the development of toxicity. Therefore, the transporter-mediated alterations may have toxicological relevance.

  • It is important to uniformly supply the fuel gas into the reaction activity area in polymer electrolyte membrane fuel cell (PEMFC). Recent studies have shown that the cell performance can be significantly improved by employing metal foam gas distributor as compared with the conventional bipolar plate types. The metal foam gas distributor has been reported to be more efficient to fuel transport. In this study, three-dimensional computational fluid dynamics (CFD) simulations have been performed to examine the effects of metal foam flow field design on the fuel supply to the reaction site. Darcy's law is used for the flow in the porous media. By solving additional advection equation for fluid particle trajectory, the gas transport has been visualized and examined for various geometrical configuration of metal foam gas distributor.

(1). • Recently, many studies for the supported liquid membrane (SLM) using a carrier have been actively reported. Polymeric cation exchange membrane was synthsized by dissolving monensin carrier of antibiotic material in organic solvent. Then the SLM was applied to the Nernst-Planck and Fick equations and membrane potential and membrane permeability were measured respectively. The results showed the high selectivity towards alkali metal ions and the SLM showed linear relationship with low concentration. However, linear relationship did not appear at high concentration. This is explained by means of the new theory of the stagnant layer and the slop of logarithm value was linear for the up-hill transport phenomena of membrane transport.

  • In order to develop procedures for the rapid isolation of recombinant sugar transporter in functional form from away from the endogenous insect cell transporter, gene fusion techniques were exploited. Briefly, BamH1-digested human HepG2 type glucose transport protein cDNA was first cloned into a transfer vector pBlueBacHis, containing a tract of six histidine residues. Recombinant baculoviruses including the human cDNA were then generated by allelic exchange following transfection of insect cells with wild-type BaculoGold virus DNA and the recombinant transfer vector. Plaque assay was then performed to obtain and purify recombinant viruses expressing the human transport protein. All the cell samples that had been infected with viruses from the several blue plaques exhibited a positive reaction in the immnuassay, demonstrating expression of the glucose transport protein. In contrast, no color development in the immunoassay was observed for cells infected with the wild-type virus or no virus. Immunoblot analysis showed that a major immunoreactive band of apparent Mr 43,000~44,000 was evident in the lysate from cells infected with the recombinant baculovirus. Following expression of the recombinant fusion protein with the metal-binding domain and enterokinase cleavage site, the fusion protein was recovered by competition with imidizole using immobilized metal charged resin. The leader peptide was then removed from the fusion protein by cleavage with porcine enterokinase. Final separation of the recombinant protein of the interest was achieved by passage over $Ni^{2+}$-charged resin under binding conditions. The expressed transport protein bound cytochalasin B and demonstrated a functional similarity to its human counterpart.

  • The classical concept for iron uptake into mammalian cells has been the endocytosis of transferrin( $T_{f}$ )-bound F $e^{3+}$ via the $T_{f}$ - $T_{f}$ receptor cycle. In this case, we could not explain the uptake of F $e^{2+}$ ion and the export of iron from endosome. Studies on iron transport revealed that other transport system exists in epithelial cells of the intestine. One of non- $T_{f}$ -receptor-mediated transport systems is Nramp2/DMT1/DCT1 which transports M $n^{++}$, $Mg^{++}$, Z $n^{++}$, $Co^{++}$, N $i^{++}$ or C $u^{++}$ ion as well as F $e^{+2}$ ion. DMT1 was cloned from intestines of iron-deficient rats and shown to be a hydrogen ion-coupled iron transporter and a protein regulated by absorbed dietary iron. DMT1 is founded in other cells such as cortical and hippocampal glial cells as well as endothelial cells in duodenum. Two F $e^{3+}$ ion bound to transferrin( $T_{f}$ ) are taken up via the $T_{f}$ - $T_{f}$ receptor cycle in the intestinal epithelial cell. F $e^{3+}$ in endosome was converted to F $e^{2+}$ ion, and then exported to cytosol via DMT1. F $e^{2+}$ ion is taken up into cytosol via DMT1. Several other transporters such as FET, FRE, CCC2, AFT1, SMF, FTR, ZER, ZIP, ZnT and CTR have been reported recently and dysfunction of the transporters are related with diseases containing Wilson's disease, Menkes disease and hemochromatosis. Evidences from several studies strongly suggest that DMT1 is the major transporter of iron in the intestine and functions critically in transport of other metal ions.

  • An IP-2 type container is used for the transport of low-level radioactive materials. An IP-2 type metal container was modeled and analyzed for the dropping conditions in normal transport conditions required by NSSC Notice 2014-50, "Regulations on the packaging and transport of radioactive materials." Structural analyses were performed by finite element method, and the acceptability was reviewed by comparing the results with technical standards and maximum allowable stresses of each material. Structural members of the container were modeled as 3D solid elements, and the possible dropping directions were considered in these analyses. Results show that maximum stresses, maximum deformed gaps in the cover, and maximum estimated thickness reduction in structural members were well below the allowable limits; thus, the structural integrity of the container was confirmed.

  • Organic thin film electroluminescent(EL) cells were fabricated. Their output characteristics and luminance versus voltage characteristics were measured with different work function metal electrodes. The EL structure was Indium-Tin-Oxide(ITO)/hole transport layer/emission layer(electron transport layer)/metal electrode. PMMA+TPD(0.5 wt%), MC homopolymer+TPD(0.005 wt%) and (MC/MMA) copolymer+TPD(0.005 wt%) were used as hole transport layer. Ca, Mg, Mg:Ag(10:l) and Al were used as metal electrode. I-V output showed exponential feature, and the threshold voltage of 5 volts and the luminance of over 700 $Cd/m^{2}$ at 10 volts were observed.

  • The transport phenomena of alkali metal chlorides through poly(2 hydroxyethyl methacrylate) hydrogel membrane have been studied using electrodialysis. The hydrogel membranes were prepared by the polymerization of 2-hydroxvethyl methacrylate in the presence of 45%(V/V) H2O and ethyleneglycodimethacrylate. The initiator used in the polymerization was azobismethylisobutyrate (AMIB) prepared from azobisiobtyronitrile (AIBN) using Mortimer method. The permeability of alkali metal chlorides such LiCl, NaCl and KCI at 50 voltage was obtained. The permeability of NaCl was also observed at 30, 40, 50, and 60 voltages respectively. The concentration of solutes permeated through the membrane was measurer by flame photometry. The experimental results have been discussed with the comparison of apparent solute molecular size, the self-diffusion coefficient of solutes, the transport number of cations in aqueous solution. These indic aloes that poly(2 hydroxyethyl methacrylate) hydrogel membrane shows a specific selectivity for sodium ion.

  • Due to their better photosensitivity in X-ray, the amorphous selenium based photoreceptor is widely used on the X-ray conversion materials. It was possible to control the charge carrier transport of amorphous selenium by suitably alloying a-Se with other elements(e.g. As, Cl). The charge transport properties of amorphous Selenium is decided on hole which is induced from metal to selenium in metal-selenium junction and which is transferred in a-Se bulk. This phenomenon is resulted of changing electric field owing to increasing of space charge by deep trap of a-Se bulk. In this paper, We dopped the chlorine to compensate deep hole trap and deposited blocking layer using dielectric material to prevent from increasing space charge for injection charge between metal electrode and a-Se layer. We compared space charge and the decreasing of trap density through measuring dark and photo current. 缀Ѐ㘰〻ሀ䝥湥牡氠瑥捨湯汯杹
