Effects of Nanocalcium Supplemented Milk on Bone Calcium Metabolism in Ovariectomized Rats

  • This study examined effects of calcium supplemented milk on bone loss in ovariectomized rats. Twenty four Sprague-Dawley female rats, 7 weeks-old, were divided into 4 groups, ovariectomized and fed diets containing: 1) control, no Ca supplemented milk, 2) ovx 1, Ca carbonate supplemented milk, 3) ovx 2, ionized Ca supplemented milk, and 4) ovx 3, nano Ca supplemented milk. All rats were fed 1 ml of milk containing 20 mg supplemented Ca. After 18 wk feeding, body weight gain and food efficiency ratio were significantly different between ovx 1 and ovx 3. Serum concentration of calcium and phosphorus were not different among groups. However, there was a significant difference in calcium content of dry femoral weight in ovx 3 compared with the control and ovx 2. In addition, femoral bone mineral density ($g/cm^2$) was significantly greater in ovx 3 than in other groups (p<0.05). The ovx 3 group showed the highest stiffness (N/mm), maximum energy (N) in femur and trabecular bone area (%). The present study indicated that nano Ca supplementation in milk may be an effective way to enhance bone calcium metabolism for ovariectomized rats.

  • Industrialization and development of technology satisfied various humanly needs and lay the basis of numerous benefit and profit. New technologies like these mostly required large amounts of fossil Fuel, Fossil energy depletion rate was increasing rapidly. However, technical development for Human race required absolute sacrifice of the environment. Especially, 'City' which had been focused as stage of human activities, allowed to continue to have fossil energy dependent activities, and it shows in many data that the city is responsible for the 75-80% of the green house gas by human. In order to solve the problem relating climate changes and energy, European countries already made progress studies on many of the low carbon green city and pilot case construction. Especially, Germany, Austria, Holland, Sweden who had been playing leader role on environmental awareness and ecological concept, are actively constructing low carbon green city project based on Ecological city planning scheme. These projects positively utilize planning scheme that limits carbon emission using Urban Metabolism concept. Therefore, throughout this study, I would like to present planning and direction of future domestic low carbon green city by analyzing theories relative theories and best practices in Europe.

칼슘 급원의 종류가 흰쥐의 체내 칼슘 및 골격대사에 미치는 영향 (The Effect of Various Types of Calcium Sources on Calcium and Bone Metabolism in Rats)

  • This study was performed to investigate the effect of various types of calcium sources on calcium and bone metabolism. Sprague-Dawley male rats weighing approximately 89.3g were divided into 4 groups and fed experimental diets containing 0.5% calcium for 5 weeks. Four different calcium salts were used for the study : calcium phosphate, calcium lactate, calcium gluconate, and calcium carbonate. Food intake showed no significant difference n accordance with the type of calcium salt, but bo요 weight gain and food efficiency were lower in calcium gluconate and calcium carbonate groups. There was significant differences in liver, thymus and epididymal fat pad weight with the of calcium salt ; the calcium gluconate group showed lower values compared to the other groups. Femur and scapular length were higher in calcium lactate and calcium carbonate groups. Wet weight, dry weight, and density of the femur tended to be lower in the calcium gluconate group than the other groups, but this difference was not statistically significant. The calcium content of the other groups. The calcium gluconate group showed higher urinary calcium and lower calcium absorption and balance. In conclusion, calcium and bone metabolism were different according to the type of calcium sources consumed.

중년 비만여성에게 적용한 비만관리프로그램이 체성분, 복부둘레 및 지질대사에 미치는 효과 (The Effects of an Obesity-Management Program on Body Composition, Abdominal Circumference, and lipid Metabolism for Middle-Aged Obese Women)

  • Purpose: This study was aimed to develop and evaluate an obesity-management program for middle-aged obese women. Methods: Two 12 week session of the obesity-management programs constituted of weekly education and exercise such as aquarobic, pilates & yoga, and power-walking during 60 min for 3 days a week from June to December, 2006. The subjects included 47 middle-aged obese women between 30 and 60 yr. The effectiveness of the program was evaluated according to the change of body weight, body component, abdominal circumference, and serum lipid concentration. Data were measured by Inbody, tape measure, and blood test. Data were analyzed by descriptive statistics, and a paired t-test with an SPSS/PC. Results: There were statistically significant decreases in body weight, body mass index, body fat mass, Hemoglobin, abdominal circumference, total cholesterol level, and low density lipoprotein after the program was completed. But there were not statistically significant difference in fat free mass, muscle mass and triglyceride. Conclusion: The results of this study showed that an obesity-management program have positive effects in body composition, abdominal circumference, and lipid metabolism for middle-aged obese women.

Genetic Polymorphisms and Cancer Susceptibility of Breast Cancer in Korean Women

  • Breast cancer is the most prevalent cancer among women in Western countries, and its prevalence is also increasing in Asia. The major risk factor for breast cancer can be traced to reproductive events that influence the lifetime levels of hormones. However, a large percentage of breast cancer cases cannot, be explained by these risk factors. The identification of susceptibility factors that predispose individuals to breast cancer (for instance, if they are exposed to particular environmental agents) could possibly give further insight into the etiology of this malignancy and provide targets for the future development of therapeutics. The most interesting candidate genes include those that mediate a range of functions. These include carcinogen metabolism, DNA repair, steroid hormone metabolism, signal transduction, and cell cycle control. We conducted a hospital-based case-control study in South Korea to evaluate the potential modifying role of the genetic polymorphisms of selected low penetrance genes that are involved in carcinogen metabolisms (i.e., CYP1A1, CYP2E1, GSTM1/T1/P1, NAT1/2, etc.), estrogen synthesis and metabolism (i.e., CYP19, CYP17, CYP1B1, COMT, ER-$\alpha$, etc.), DNA repair (i.e., XRCC1/3, ERCC2/4, ATM, AGT, etc.), and signal transduction as well as others (i.e., TGF-$\beta$, IGF-1, TNF-$\beta$, IL-1B, IL-1RN, etc.). We also took into account the potential interaction between these and the known risk factors of breast cancer. The results of selected genes will be presented in this mini-review.

Identification of Differentially Expressed Proteins at Four Growing Stages in Chicken Liver

  • Because of high growth rate and large deposition of fat in the abdomen, the chicken has been used as a model organism for understanding lipid metabolism, fattening and growing. In this study, differentially expression of proteins in chicken liver, one of the important organs for lipid metabolism, has been investigated at four different growing stages. After separation of proteins using two-dimensional electrophoresis (2-DE), more than 700 protein spots were detected. Among them, 13 growing stage specific proteins in chicken liver were selected and further investigated by matrix-assisted laser adsorptions ionization-time of flight mass spectrometry (MALDI-TOF MS). Of these, 12 proteins were matched to existing proteins based on a database search. The identified fat-related proteins in this study were fatty acid synthase (FASN) and malic enzyme (ME1). These proteins were more highly expressed at week 32 than at other weeks. In order to confirm the differential expression, one of the proteins, FASN, was confirmed by western blotting. The identified proteins will give valuable information on biochemical roles in chicken liver, especially for lipid metabolism.

마우스 간의 황함유 아미노산 대사에 미치는 베타인의 용량의존성 영향 (Dose-dependent Effects of Betaine on Hepatic Metabolism of Sulfur Amino Acids in Mice)

  • Acute betaine treatment induces time-dependent changes in the hepatic glutathione (GSH), cysteine and S-adenosylmethionine (SAM) levels. Our previous study demonstrated that betaine administered $1{\sim}4$ hours prior to sacrifice decreased hepatic GSH levels, but these levels were increased when measured 24 hours following the treatment. The present study was aimed to determine dose-dependent effects of betaine on hepatic metabolism of sulfur amino acid in mice. Mice were sacrificed 2.5 or 24 hours after intraperitoneal treatment with betaine at different dose levels ranging from 50 to 1000 mg/kg. The concentrations of methionine and SAM were increased by a betaine dose of 100 mg/kg, and the concentrations of GSH and cysteine were decreased by a betaine dose of 200 mg/kg at 2.5 hours. These changes were augmented with increasing doses of betaine. At 24 hours following betaine treatment, increased GSH and decreased taurine levels were observed from dose levels of 400 mg/kg. Changes in hepatic activities of cystathionine beta-synthase, gammaglutamylcysteine ligase and cysteine dioxygenase were observed from dose levels of $200{\sim}400$ mg/kg of betaine administered 24 hours prior to sacrifice.

Clofibrate의 유도체가 토끼의 혈소판 응집에 미치는 영향 (The Effects of Congeners of Clofibrate on Inhibition of Rabbit Platelet Aggregation)

  • Several clofibrate congeners (bezafibrate, gemfibrozil and fenofibrate) were investigated the relationship between effects on the aggregation induced by aggregating agents (thrombin, arachidonic acid, ADP and collagen) and arachidonic acid metabolism in rabbit homogenized platelet. In platelet aggregation study, all drugs produced no significant inhibition (data not shown) in arachidonic acid and thrombin. Also platelet aggregation by ADP was not changed in bezafibrate and Inhibited dose dependently in fenofibrate and gemfibrozil. Platelet aggregation by collagen was inhibited dose dependently and significantly (from p<0.5 to p<0.001) by gemfibrozil and fenofibrate at concentrations between 20 and 400 $\mu$M. In arachidonic acid metabolism study, synthesis of thromboxane $B_2$ was not changed in rabbit platelet membranes and that of prostaglandin $E_2$ and $F_{2{\alpha}}$ was slightly increased by all drugs. It was concluded that clofibrate congeners inhibited ADP and collagen induced rabbit platelet aggregation and inhibition of collagen induced aggregation was probably mediated through some mechanism (pathway) other than arachidonic acid metabolism, judging from arachidonic acid metabolites (thromboxane $B_2$, prostaglandin $E_2$and $F_{ 2{\alpha}}$) synthesis in rabbit homogenized Platelet.

Effects of Fermented Chub Mackerel Extract on Lipid Metabolism of Rats Fed a High-Cholesterol Diet

  • The present study was conducted to evaluate effect of fermented chub mackerel extract (FCME) on lipid metabolism of rats fed a diet supplemented with 1% cholesterol. Four week-old male rats were divided into three groups of 15 rats with 0, 0.1% or 0.2% FCME supplementation. In comparison with control, rats fed 0.2% FCME showed reduction of activities of acetyl-Coenzyme A carboxylase (p<0.05), 3-hydroxy-3-methyl glutaryl Coenzyme A reductase (p<0.01) and fatty acid synthetase (p<0.01). Rats fed 0.2% FCME also showed reductions in free cholesterol in the liver (p<0.05), and in concentrations of free cholesterol (p<0.05), LDL+VLDL-cholesterol (p<0.05), triglyceride (p<0.01) and phospholipid (p<0.01) in the plasma. Plasma HDL-cholesterol concentration was significantly (p<0.05) higher in treatment groups as compared with control group. Atherogenic index was also significantly lower in rats fed 0.1% or fed 0.2% supplement diet, whereas bile acid in feces was not significantly affected. The current study showed that 0.2% inclusion level of the fermented chub mackerel extracts might have hypolipidemic properties.

Benzo(a)pyrene에 의한 돌연변미월성과 지질과산화에 미치는 d-Limonene과 Cineole의 엑제 효과에 대한 비교연구 (Inhibition of Benzo(a)pyrene-Induced Mutagenicity and Lipid Peroxidation by d-Limonene and Cineole)

  • It has been reported that d-limonene inhibits chemical-induced rat mammary cancer by the mechanism of increases in detoxification enzymes such as glutathione S-transferases and that cineole fails to exhibit significant suppressive effect on chemical-induced carcinogenesis. The present study was designed to compare the effects of d-limonene and cineole on the benzo(a)pyrene (BP)-induced mutagenicity, BP metabolism and lipid peroxidation. Modified Ames assay was employed to evaluate the inhibitory effect of d-limonene and cineole on the BP-induced mutagenicity. The number of revertant-bearing wells was decreased by 44~77% in the presence of both BP and d-limonene compared with that of BP alone whereas cineole decreased the number of revertant-bearing wells by 28~45% at the concentrations between $2{\mu}m$m.TEX> and 2 mM. d-Limonene suppressed BP metabolism by 16, 54 and 67% at 1, 10 and 100 mM, respectively while cineole inhibited the metabolism by 16, 26 and 55% at the same concentrations. The $EC_{50}$ values for d-limonene and cineole in inhibiting lipid peroxidation were 2.0 mM and 16 mM respectively, as assayed by thiobarbituric acid method. The present study showed that d-limonene and cineole have common antimutagenic effects although d-limonone appeared to be more effective than cineole in suppressing mutation and lipid peroxidation. The results suggest that the antimutagenic effects of d-limonene and cineole may be associated with alternation in enzyme activities and with inhibition of lipid peroxidation.
