사육수의 고수온 스트레스가 벤자리(Parapristipoma trilineatum)에 미치는 생리학적 영향 (Physiological Responses of the Chicken Grunt Parapristipoma trilineatum to High Water Temperature Stress)

  • We investigated the effects of water temperature on physiological parameters in the chicken grunt Parapristipoma trilineatum. At high temperature, the aspartate aminotransferase (AST) and the alanine aminotransferase (ALT) levels were increased, suggesting that high temperature induced hepatic damage. In addition, total protein (TP) was high at high water temperatures, which were considered stressful in the breeding environment. At high water temperatures, triglycerides (TG) were low due to increased metabolic activity, which decreased the blood TG levels as TG were used as an energy source. There was no significant difference in the plasma osmolality or the blood ion concentrations with water temperature. In generally, lysozyme, a factor in innate immunity, increased with water temperature. However, lysozyme activity tended to decrease with increasing water temperature, but the difference was not significant. These results suggested that the decrease of biophylaxis at high temperature was affect the growth or survival of the population.

Comparison of the trometamol-balanced solution with two other crystalloid solutions for fluid resuscitation of a rat hemorrhagic model

  • Currently, the optimal resuscitation fluid remains debatable. Therefore, in the present study, we designed a trometamol-balanced solution (TBS) for use as a resuscitation fluid for hemorrhagic shock. Hemorrhagic shock was induced in 18 male Wistar-Kyoto rats, which were assigned to normal saline (NS), Ringer's solution (RS), and TBS groups. During the hemorrhagic state, their hemodynamic parameters were recorded using an Abbott i-STAT analyzer with the CG4+ cartridge (for pH, pressure of carbon dioxide, pressure of oxygen, total carbon dioxide, bicarbonate, base excess, oxygen saturation, and lactate), the CG6+ cartridge (for sodium, potassium, chloride, blood glucose, blood urea nitrogen, hematocrit, and hemoglobin), and enzyme-linked immunosorbent assay kits (calcium, magnesium, creatinine, aspartate aminotransferase, alanine aminotransferase, bilirubin, and albumin). Similar trends were found for the parameters of biochemistries, electrolytes, and blood gas, and they revealed no significant changes after blood withdrawal-induced hemorrhagic shock. However, the TBS group showed more effective ability to correct metabolic acidosis than the NS and RS groups. TBS was a feasible and safe resuscitation solution in this study and may be an alternative to NS and RS for resuscitation in hemorrhagic shock patients without liver damage.

Effect of Sharbat Afsantīn in Poly Cystic Ovarian Disease - An Open Observational Study

  • Objective: The objective of the study was to evaluate the effect of Sharbat Afsantin in Polycystic Ovarian Disease. Methods: An open observational study was carried out in the Department of Ilmul Qabalat wa Amraze Niswan. Diagnosed cases(n=30) of PCOD were included in the study. Patients with thyroid dysfunction, systemic diseases, on hormonal treatment in last three months, pregnancy and lactation were excluded. Research drug (Sharbat Afsantin) was administered orally in a dose of 25ml twice daily for 15 days/cycle for three consecutive cycles. Outcome measures were, changes in subjective parameters (duration of cycle, duration and amount of flow and weight reduction) and objective parameters {pictorial blood loss assessment chart (PBAC) score, basal metabolic index (BMI), modified Ferriman Gallwey (mFG) score, acanthosis nigricans scale and pelvic ultrasonography}. Data were analyzed using paired Student 't' test. Results: Changes in duration of cycle, duration and amount of flow were achieved in 83.3%, 50% and 40% patients respectively and weight reduction in 30% patients. Changes in PBAC score and BMI were achieved in 50% and 30% patients respectively and 30% patients showed normal findings on pelvic ultrasonography. Conclusion: Sharbat Afsantin can be used as an alternate remedy in PCOD patients, as it has significant effect to regularize menstruation by reduction in BMI and probably by improving insulin resistance in PCOD. No adverse effect of Sharbat Afsantin was noted during the trial.

Optical imaging of epileptic activity and epilepsy treatments in neocortex

  • Optical imaging offers excellent spatio-temporal sensitivity that is unparalleled by any other perfusion based imaging techniques. We used in vivo optical recording of intrinsic signals (ORIS) to map neurovascular hemodynamics of perfusion, oximetry and membrane potential during epileptic events in rat and mouse neocortex. Studies of hemodynamic changes with ORIS alone were also performed in human. Laboratory studies in rodent epilepsy models have demonstrated a persistent increase in deoxygenated hemoglobin (Hbr) and a decrease in tissue oxygenation during interictal spikes and ictal events. This "epileptic dip", like the "initial dip" recorded during normal sensory processing, implies that the enormous rise in cerebral blood flow (CBF) is inadequate to meet the increased metabolic demands associated with synchronized epileptic activity. These findings are critically important to the interpretation of the perfusion-based imaging studies, such as fMRI. In addition, we visualized the effect of direct cortical electrical stimulation, an alterative epilepsy treatment. The optical data following direct cortical electrical stimulation showed that hemodynamic signals are sensitive to different electrical stimulation parameters. Furthermore, our recent data demonstrated that the application of unilateral electrical stimulation is able to elicit bilateral hemodynamic responses in rat neocortex.

Metabolomics, a New Promising Technology for Toxicological Research

  • Metabolomics which deals with the biological metabolite profile produced in the body and its relation to disease state is a relatively recent research area for drug discovery and biological sciences including toxicology and pharmacology. Metabolomics, based on analytical method and multivariate analysis, has been considered a promising technology because of its advantage over other toxicogenomic and toxicoproteomic approaches. The application of metabolomics includes the development of biomarkers associated with the pathogenesis of various diseases, alternative toxicity tests, high-throughput screening (HTS), and risk assessment, allowing the simultaneous acquisition of multiple biochemical parameters in biological samples. The metabolic profile of urine, in particular, often shows changes in response to exposure to xenobiotics or disease-induced stress, because of the biological system's attempt to maintain homeostasis. In this review, we focus on the most recent advances and applications of metabolomics in toxicological research.

NMR Metabolomic Profiles for Quality Control of Korean Green Tea (Camellia sinensis) Classified by the Plucking Season

  • The plucking season of green tea leaves is one of the important parameters that decide their metabolic diversity, quality, and prices. The effects of plucking sghlwleasons on green tea metabolites were investigated through metabolite profiling by $^1H$ NMR spectroscopy. The orthogonal projection on latent structure-discriminant analysis (OPLS-DA) showed clear discriminations of green teas by three different grades depending on plucking seasons: Ujeon, Sejak, and Jungjak. These results suggested that the nine peak groups could be used for diagnostics for identification of high quality Ujeon grade of green tea.

Study of Antimicrobial Effects of Different Types of Glycyrrhiza Extracts by Microcalorimetry

  • Recently studies indicate that the microcalorimetry is a suitable measurement for metabolic activities of organisms by recording the rate of heat outputs. In this work, we investigated the growth thermogenic curves of Escherichia coli (E. coli) affected by three kinds of Glycyrrhiza extracts by microcalorimetry. The power-time and exponential phase power-time curves of the E. coli growth at various concentrations of extracts were generated. The kinetic parameters such as the growth rate constants (C), maximum power outputs (Pm), peak times (Tm), and inhibition ratios were calculated and the relationships between Pm or Tm and C were established. Also, the clear correlations among the antimicrobial effects, Pm and C were obtained. The results revealed the Glycyrrhiza extracts had inhibitory activities towards E. coli while the Glycyrrhiza polysaccharides showed the most potent effects.

Influence of Sodium Nitrate (NaNO$_3$) of Different Feeds on Growth and Bioenergetics of Bivoltine NB$_4$D$_2$ Race of the Silkworm, Bombyx mori L.

  • Dietary supplementation of sodium nitrate with different concentrations 50, 100, 200, 500, 700 and 1000 $\mu\textrm{g}$/ml of single, two, three and four feeds to fifth instar larvae of biovoltine NB$_4$D$_2$ race of the silkworm, B. mori resulted in significant increase in the food conversion, conversion rate and conversion efficiency $K_1$ and $K_2$. However, there were significant decrease in the food assimilation, assimilation rate and assimilation efficiency in the sodium nitrate treated groups as compared with that of the corresponding parameters of the carrier control. This indicates that the administration of sodium nitrate may stimulate metabolic activities, thereby influencing conversion of food into body weight in the bivoltine silkworm, B. mori.

Quantitative analysis of metabolites in Korean green tea using NMR

  • The plucking season of green tea leaves is one of the important parameters that decide their metabolic quality. Here, we performed the identification and quantity analysis of the metabolites of the green tea using NMR spectroscopy. We assigned the $^1H$ resonances for sixteen metabolites. This analysis found that four metabolites, gallic acid, quinic acid, theobromine and ECG, exhibited clear discrimination of green teas by the three different grades, Ujeon, Sejak and Jungjak. Our results suggest that these four metabolites could be used for diagnostics for quality control of green tea.

Comparative in vitro biotransformation of fipronil in domestic poultry using liver microsome

  • Domestic poultry are among the non-target species of exposure to fipronil, but limited information is available on the metabolic effects of fipronil exposure in avian. We investigated the comparative capacity of in vitro biotransformation of fipronil among chicken, duck, quail, goose, and rat. Interspecies differences in kinetic parameters were observed; the clearance rate calculations (Vmax/Km) indicated that chicken and duck are more efficient in the cytochrome P450-mediated metabolism of fipronil to sulfone than quail, goose and rat. The lower hepatic clearance of fipronil in quail, goose and rat, suggested that fipronil sulfone may serve as a biomarker to indicate fipronil exposure in these species.
